RESUMO

The cardioprotective mechanism of KATP channel openers and especially their influence on mitochondrial respiration has not been clarified yet. In this article we investigated the effect of DiazoFm and DiazoFp, the new fluor-containing analogues of diazoxide and the potential mitochondrial KATP channel openers, on the oxidative phosphorylation in the isolated mitochondria. It was shown that the influence of KATP channel openers on ADP-stimulated oxygen consumption (State 3) depended on the substrates we used (succinate or 2-oxoglutarate sodium). We have shown that the depression of State 3 was less when we used DiazoFm (30 MM) and DiazoFp (30 MM) in comparison with Diazoxide in experiments where succinate was used. The fluor-containing KATP, channels openers did not significantly change the activity of succinate dehydrogenase in comparison with diazoxide (it decreased succinate dehydrogenase activity by 27%). Thus, the fluor-containing analogues of diazoxide did not significant influence on the complex II of the respiratory chain. In the other experiments when we used 2-oxoglutarate sodium as an oxidative substrate, DiazoFp increased ADP-stimulated oxygen consumption by 33%. All the studied KATP openers have an uncoupling effect, regardless the substrates we used. This effect was more significant when we used succinate as a substrate. We have shown that the uncoupling effect of oxidative phosphorylation is a consequence of K channels activation. This statement was proved by 5-hydroxydecanoate (200 MM) with depressed influence of Diazoxide and its fluoring-containing analogues. Conclusion. The fluor-containig KATP channels openers had not direct influence on the respiratory chain in mitochondria, but activation mitochondrial KATP channels by them lead to uncoupling phosporylation and respiration.

RESUMO

The influence of omega-3 polyunsaturated fatty acids (omega-3 PUFA) on the activity of glutathione reductase, glutathione transferase and glutathione peroxidase in the liver cytosole and red blood cells of normal rats and animals with experimental chronic bronchitis. omega-3 PUFA ("Tekom" medication) activate glutathione reductase of liver cytosole and glutathionperoxidase in the red blood cells in rats. In the rats with chronic inflammatory process in bronchia omega-3 PUFA corrects the glutathione-dependent systems of detoxication. Effects were more expressed in the liver cytosole in comparison with the red blood cells. The using of omega-3 PUFA as a means for treatment and prophylaxis was more effective than for treatment only.

RESUMO

Blood serum was studied for its antitryptic activity in 161 patients with chronic bronchitis, 87 individuals with malignant tumors of the respiratory organs, 70 blood donors, with regard to phenotypic variant of alpha 1-inhibitor of proteinases. Difference was revealed in antitryptic activity, associated with the particular features of the pathologic process and phenotypic variant of the inhibitor. There exists in the carriers of homozygote latent insufficiency of the inhibitory defence of the organism against the proteolytic effects.

RESUMO

Quantity and activity of components in the monoxygenase system of the liver microsomes have been studied for their dependence on the phenotype of acetylation. Results of investigations are presented. The majority of these enzymes exist as numerous molecular forms: isozymes. It is shown that activity of aminopyrine-N-demethylase and NADPH-cytochrome P-450 reductase depends on the phenotype of acetyltransferase.

RESUMO

The study established that shifts in oxidant-antioxidant system induced by chronic obstructive bronchitis are correctable by He-Ne laser irradiation. This conclusion was based on biochemical indices of the blood measured in experimental animals: lipid peroxidation, general antitryptic activity of the serum, activity of the enzymes superoxide dismutase, gamma-glutamyl transferase, pseudocholine esterase, alpha-oxybutyrate dehydrogenase, glucose 6-phosphate dehydrogenase.

RESUMO

The authors investigated the clinical and biochemical efficacy of millimeter-wave resonance therapy in patients with destructive pulmonary tuberculosis associated with different concomitant diseases. It was established that inclusion of millimeter-wave resonance therapy in the complex treatment of this patient category favoured the efficacy of tuberculosis treatment. The treatment produced also a favourable effect on concomitant diseases.

RESUMO

The activity of the key enzyme of the pentoso-phosphate cycle--glucoso-6-phosphatedehydrogenase was examined in patients with chronic inflammatory pulmonary diseases. Results indicate that with exacerbation of the process in the bronchopulmonary system there occurs an increase of the activity of the mentioned enzyme. Examination of the activity of this enzyme may be of value for optimization of treatment measures in patients with inflammatory pulmonary diseases.

RESUMO

Tubaside causes disturbances in oxidative phosphorylation of the liver mitochondria. Sodium succinate administered during tubaside intoxication favours normalization of the oxygen uptake rate in state 3 and the ADP phosphorylation rate. Sodium succinate weakens the toxic effect of tubaside and favours normalization of p-hydroxylation in the liver. The animals, which were given tubaside in combination with sodium succinate are characterized by a higher survival rate.

RESUMO

A single administration of ethionamide (LD50) to guinea pigs causes an inhibition of p-hydroxylation and N-demethylation in the liver. Sodium succinate favours the activation and, when it is used for correction of the ethionamide toxic effect, it normalizes the above-mentioned processes.

RESUMO

The data of studies on proton metabolism in mitochondria of the guinea pig lungs and liver with application of benemycin and sodium succinate show that benemycin does not affect the metabolism of protons in the lungs and liver mitochondria. Sodium succinate activates the release of protons from the lungs and liver mitochondria in state 4 according to Chance. When using sodium succinate for reducing the effect of benemycin on theorganism it is found that sodium succinate inhibits the utilization of protons of the lungs and liver mitochondria in state 3 according to Chance and activates this release in state 4, thus favouring an increase in their concentration in the extra-mitochondrial space. Sodium succinate increases survival rate of animals which were administered benemycin to.
