RESUMO

Forest soils harbor hyper-diverse microbial communities which fundamentally regulate carbon and nutrient cycling across the globe. Directly testing hypotheses on how microbiome diversity is linked to forest carbon storage has been difficult, due to a lack of paired data on microbiome diversity and in situ observations of forest carbon accumulation and storage. Here, we investigated the relationship between soil microbiomes and forest carbon across 238 forest inventory plots spanning 15 European countries. We show that the composition and diversity of fungal, but not bacterial, species is tightly coupled to both forest biotic conditions and a seven-fold variation in tree growth rates and biomass carbon stocks when controlling for the effects of dominant tree type, climate, and other environmental factors. This linkage is particularly strong for symbiotic endophytic and ectomycorrhizal fungi known to directly facilitate tree growth. Since tree growth rates in this system are closely and positively correlated with belowground soil carbon stocks, we conclude that fungal composition is a strong predictor of overall forest carbon storage across the European continent.

OBJECTIVE: Major depressive disorder (MDD) is one of the most common psychiatric issues in hemodialysis population. However, the research on proper diagnostic tools and its treatment is still insufficient. The study was performed to investigate the safety and effectiveness of sertraline and agomelatine in a group of hemodialysis patients. PATIENTS AND METHODS: 78 adult patients from one dialysis centre in Poland were included into the study. The Beck Depression Inventory II (BDI-II) was used to screen for depressive symptoms and was followed by the clinical interview with the psychiatrist. Nine patients diagnosed with major depressive disorder received antidepressant treatment with sertraline or agomelatine, according to the best clinical practice. The additional treatment with vortioxetine was used if the initial one was not effective. The time of observation was 24 weeks. The psychiatric follow up as well as the laboratory data were obtained during the course of observation. RESULTS: All patients receiving sertraline achieved remission of depressive symptoms. In patients receiving agomelatine no remission was observed despite dose augmentation. The side effects of antidepressants were mild and did not result in treatment discontinuation. No abnormalities in liver enzymes levels were observed. In five cases the significant decrease of haemoglobin level was noticed, with no cases of bleeding reported. CONCLUSION: In patients receiving sertraline the antidepressant effect was satisfactory. No remission of depressive symptoms was observed in patients taking agomelatine. The side effects of antidepressants were mild and transient. Further research on depression treatment in hemodialysis patients is needed, including newer medications.

The present study investigates the effects of the simultaneous use of two additives, an organosilane warm mix asphalt (WMA) agent and a grade-bumping polyolefin compound, on the conventional and high-temperature performance properties of a paving grade 50/70 bitumen and a polymer-modified 45/80-55 bitumen. The WMA agent and polyolefin additive were introduced to the binders at rates of up to 0.3% and 2%, respectively. The base asphalt binders and their blends with the additives were tested before and after aging in a rolling thin film oven test at a temperature of 143 °C. The effects of the investigated additives were found to be dependent on the type of base binder and its aging state. It was generally observed that the WMA additive decreased the performance of the asphalt binders and limited the effects of the other additive, which increased the high-temperature stiffness and non-recoverable compliance of the blends. This interaction amounted to as much as an approx. 20% decrease in high-temperature stiffness and non-recoverable compliance of the binders. The additives caused a small increase in the elasticity of the binders and improved their creep performance when measured in multiple stress creep recovery tests.

The present paper investigates the viscoelastic stress-strain responses of laboratory and plant produced warm mix asphalt mixtures containing basalt fiber dispersed reinforcement. The investigated processes and mixture components were evaluated for their efficacy in producing highly performing asphalt mixtures with decreased mixing and compaction temperatures. Surface course asphalt concrete (AC-S 11 mm) and high modulus asphalt concrete (HMAC 22 mm) conventionally and using a warm mix asphalt technique with foamed bitumen and a bio-derived fluxing additive. The warm mixtures included lowered production temperature (by 10 °C) and lowered compaction temperatures (by 15 °C and 30 °C). The complex stiffness moduli of the mixtures were assessed under cyclic loading tests at combinations of four temperatures and five loading frequencies. It was found that the warm produced mixtures were characterized by lower dynamic moduli than the reference mixtures in the whole spectrum of loading conditions, however, the mixtures compacted at the 30 °C lower temperature performed better than the mixtures compacted at 15 °C lower temperature, specifically when highest testing temperatures are considered. The differences in the performance of plant and laboratory produced mixtures were ascertained to be nonsignificant. It was concluded that the differences in stiffness of hot mix and warm mixtures can be attributed to the inherent properties of foamed bitumen mixtures and that these differences should shrink in time.

This paper presents the results of an analysis of the changes in the stiffness of asphalt binders modified with a bio-flux additive and subjected to the processes of foaming and short-term ageing. The purpose of the analyses was to determine changes in the rheological properties of asphalt binder as a result of technological processes characteristic of hot and warm mix asphalt technology. Three asphalt binders with similar penetration but varying degrees of polymer modification were tested: 50/70, 45/80-55 polymer-modified bitumen, and 45/80-80 highly modified bitumen. Tests were carried out on four groups of binders: original binders, foamed binders after 14 days of storage, non-foamed binders after Rolling Thin Film Oven Test (RTFOT) ageing, and foamed binders after 14 days of storage subjected to RTFOT ageing. The master curves of the complex shear modulus G* were analysed, and three indexes of binder stiffening were determined, characterising the investigated effects. The tests showed that some of the stiffening indices significantly depended on the degree of polymer modification and the content of the bio-flux additive. Moreover, it was found that the foaming process in the case of paving-grade bitumen and polymer-modified bitumen did not contribute to the additional stiffening of the binders.

The study investigated the effects of laboratory ageing on the fluxed and water-foamed asphalt binders in scope of Fourier transform infrared spectroscopic measurements of ageing indicators and changes in their chemical composition. The investigated binders included two paving grades, two polymer modified asphalt binders, and a highly modified asphalt binder. The bio-flux additive was produced from rapeseed methyl esters in an oxidation reaction in the presence of a metal catalyst and organic peroxide. The use of the bio-origin additive, in particular oil derivatives, was aimed at softening and better foaming of asphalt binders. This modification is possible due to the good mixability of vegetable oils with an asphalt binder, which gives a homogeneous product with reduced stiffness. The study involved the rolling thin film oven, short-term, and the pressure ageing vessel, long term, and ageing to induce oxidation on the evaluated asphalt binders. The addition of the bio-flux additive has significantly decreased the measured content of ketone compounds related to oxidation in both non-aged and aged asphalt binders, although this effect after ageing were far smaller in magnitude. Additionally, both ageing processes decreased significantly the absorbances in the ester spectral bands specific to the bio-flux additive. All mentioned effects were similar in magnitude in all tested asphalt binders.

The animal gut microbiota consist of many different microorganisms, mainly bacteria, but archaea, fungi, protozoans, and viruses may also be present. This complex and dynamic community of microorganisms may change during parasitic infection. In the present study, we investigated the effect of the presence of microsporidians on the composition of the mosquito gut microbiota and linked some microbiome taxa and functionalities to infections caused by these parasites. We characterised bacterial communities of 188 mosquito females, of which 108 were positive for microsporidian DNA. To assess how bacterial communities change during microsporidian infection, microbiome structures were identified using 16S rRNA microbial profiling. In total, we identified 46 families and four higher taxa, of which Comamonadaceae, Enterobacteriaceae, Flavobacteriaceae and Pseudomonadaceae were the most abundant mosquito-associated bacterial families. Our data suggest that the mosquito gut microbial composition varies among host species. In addition, we found a correlation between the microbiome composition and the presence of microsporidians. The prediction of metagenome functional content from the 16S rRNA gene sequencing suggests that microsporidian infection is characterised by some bacterial species capable of specific metabolic functions, especially the biosynthesis of ansamycins and vancomycin antibiotics and the pentose phosphate pathway. Moreover, we detected a positive correlation between the presence of microsporidian DNA and bacteria belonging to Spiroplasmataceae and Leuconostocaceae, each represented by a single species, Spiroplasma sp. PL03 and Weissella cf. viridescens, respectively. Additionally, W. cf. viridescens was observed only in microsporidian-infected mosquitoes. More extensive research, including intensive and varied host sampling, as well as determination of metabolic activities based on quantitative methods, should be carried out to confirm our results.

The present paper investigates the effects of simultaneous mechanical foaming using water and fluxing with a bio-derived agent on the properties of three distinct asphalt binders: 50/70 paving-grade bitumen, 45/80-55 polymer-modified bitumen, and 45/80-80 highly modified asphalt binder. The testing involved classical tests for assessing binder consistency (penetration at 25 °C, ring and ball softening point, Fraass breaking point, and dynamic viscosity) as well as performance tests (high and low Superpave critical temperatures and multiple stress creep recovery). The tests included assessment directly after asphalt binder foaming and were repeated after a 14-day period. It was shown that bitumen foaming had only short-term effects on the asphalt binders, which did not persist in the repeated tests after 14 days. The fluxing agent that was utilized caused significant changes in the consistency of all asphalt binders. The changes in the performance characteristics of the 50/70 and 45/80-55 binders were severe and amounted to a significant decrease in high-temperature performance of these binders. On the other hand, an improvement in all performance characteristics in the case of the 45/80-80 asphalt binder was observed as a result of the applied processes, particularly when measured 14 days after foaming. This study shows that the simultaneous use of foaming and the fluxing additive decreased the dynamic viscosity of the 45/80-80 binder, while improving its properties relating the pavement performance.

Contribution of UCP1 single nucleotide polymorphisms (SNPs) to susceptibility for cardiometabolic pathologies (CMP) and their involvement in specific risk factors for these conditions varies across populations. We tested whether UCP1 SNPs A-3826G, A-1766G, Ala64Thr and A-112C are associated with common CMP and their risk factors across Armenia, Greece, Poland, Russia and United Kingdom. This case-control study included genotyping of these SNPs, from 2,283 Caucasians. Results were extended via systematic review and meta-analysis. In Armenia, GA genotype and A allele of Ala64Thr displayed ~2-fold higher risk for CMP compared to GG genotype and G allele, respectively (p<0.05). In Greece, A allele of Ala64Thr decreased risk of CMP by 39%. Healthy individuals with A-3826G GG genotype and carriers of mutant allele of A-112C and Ala64Thr had higher body mass index compared to those carrying other alleles. In healthy Polish, higher waist-to-hip ratio (WHR) was observed in heterozygotes A-3826G compared to AA homozygotes. Heterozygosity of A-112C and Ala64Thr SNPs was related to lower WHR in CMP individuals compared to wild type homozygotes (p<0.05). Meta-analysis showed no statistically significant odds-ratios across our SNPs (p>0.05). Concluding, the studied SNPs could be associated with the most common CMP and their risk factors in some populations.

BACKGROUND: Microsporidia is a large group of eukaryotic obligate intracellular spore-forming parasites, of which 17 species can cause microsporidiosis in humans. Most human-infecting microsporidians belong to the genera Enterocytozoon and Encephalitozoon. To date, only five microsporidian species, including Encephalitozoon-like, have been found in hard ticks (Ixodidae) using microscopic methods, but no sequence data are available for them. Furthermore, no widespread screening for microsporidian-infected ticks based on DNA analysis has been carried out to date. Thus, in this study, we applied a recently developed DNA metabarcoding method for efficient microsporidian DNA identification to assess the role of ticks as potential vectors of microsporidian species causing diseases in humans. METHODS: In total, 1070 (493 juvenile and 577 adult) unfed host-seeking Ixodes ricinus ticks collected at urban parks in the city of Poznan, Poland, and 94 engorged tick females fed on dogs and cats were screened for microsporidian DNA. Microsporidians were detected by PCR amplification and sequencing of the hypervariable V5 region of 18S rRNA gene (18S profiling) using the microsporidian-specific primer set. Tick species were identified morphologically and confirmed by amplification and sequencing of the shortened fragment of cytochrome c oxidase subunit I gene (mini-COI). RESULTS: All collected ticks were unambiguously assigned to I. ricinus. Potentially zoonotic Encephalitozoon intestinalis was identified in three fed ticks (3.2%) collected from three different dogs. In eight unfed host-seeking ticks (0.8%), including three males (1.1%), two females (0.7%) and three nymphs (0.7%), the new microsporidian sequence representing a species belonging to the genus Endoreticulatus was identified. CONCLUSIONS: The lack of zoonotic microsporidians in host-seeking ticks suggests that I. ricinus is not involved in transmission of human-infecting microsporidians. Moreover, a very low occurrence of the other microsporidian species in both fed and host-seeking ticks implies that mechanisms exist to defend ticks against infection with these parasites.

This study aimed to simulate oak and beech forest growth under various scenarios of climate change and to evaluate how the forest response depends on site properties and particularly on stand characteristics using the individual process-based model HETEROFOR. First, this model was evaluated on a wide range of site conditions. We used data from 36 long-term forest monitoring plots to initialize, calibrate, and evaluate HETEROFOR. This evaluation showed that HETEROFOR predicts individual tree radial growth and height increment reasonably well under different growing conditions when evaluated on independent sites. In our simulations under constant CO2 concentration ([CO2]cst) for the 2071-2100 period, climate change induced a moderate net primary production (NPP) gain in continental and mountainous zones and no change in the oceanic zone. The NPP changes were negatively affected by air temperature during the vegetation period and by the annual rainfall decrease. To a lower extent, they were influenced by soil extractable water reserve and stand characteristics. These NPP changes were positively affected by longer vegetation periods and negatively by drought for beech and larger autotrophic respiration costs for oak. For both species, the NPP gain was much larger with rising CO2 concentration ([CO2]var) mainly due to the CO2 fertilisation effect. Even if the species composition and structure had a limited influence on the forest response to climate change, they explained a large part of the NPP variability (44% and 34% for [CO2]cst and [CO2]var, respectively) compared to the climate change scenario (5% and 29%) and the inter-annual climate variability (20% and 16%). This gives the forester the possibility to act on the productivity of broadleaved forests and prepare them for possible adverse effects of climate change by reinforcing their resilience.

The paper investigates the phenomena of adhesion in binder-aggregate systems produced to reflect warm-mix asphalt with water-foamed bitumen. The investigated materials included limestone and quartzite aggregates and a total of four asphalt binders: 50/70 and 45/80-55 bituminous binders obtained from two sources. The adhesive bonding between the asphalt binders and aggregates was evaluated in boiling water stripping tests, which results were quantified using digital image analysis. The bituminous binders were additionally tested for their dynamic viscosities, and their chemical composition was probed using FTIR spectroscopy. The tests were carried out using traditional liquid and foamed bituminous binders on samples prepared at temperatures characteristic of hot-mix asphalt and warm-mix asphalt production (20 °C decrease). The use of foamed binders yielded higher values of residual asphalt binder coverage of the aggregates. Limestone aggregates provided superior adhesion, with the lowest result amounting to approximately 88%, while with quartzite aggregates, the results ranged from approximately 40% to 87%. The refinery from which the asphalt binders were sourced had a significant influence on the results; however, the additional rheological and chemical analyses were insufficient to explain those differences. It was concluded that the process of asphalt binder foaming, per se, may have a beneficial impact on the resistance of the asphalt binder-aggregate system to the action of water.

OBJECTIVE: The aim: The study aimed to assess the content of selected toxic compounds in mineral and spring waters available on the Polish market regarding potential health risks to consumers. PATIENTS AND METHODS: Materials and methods: Selected mineral and spring waters available on the Polish market were the study's objects. The content of such chemical compounds as arsenic, cadmium, lead, copper, and mercury in selected mineral and spring waters was analyzed. The content of metals in the samples was determined by inductively coupled plasma mass spectrometry (ICP-MS). Additionally, a literature review was performed to determine nitrates contamination of bottled waters available on the Polish market. Based on the collected data, an assessment of exposure and health risk to consumers was performed. RESULTS: Results: Arsenic had the highest mean concentration in the analyzed water samples. Consumption of such contaminated waters may be a significant health risk factor for consumers. Literature data indicate a relatively low content of nitrates in bottled waters available on the Polish market. Consumption of such mineral waters is not a significant source of exposure and does not translate into a significant health risk for consumers. CONCLUSION: Conclusions: To ensure consumers' health safety, there is a need to monitor the content of potentially harmful compounds in mineral and spring waters available on the Polish market.

The thermodynamic properties of phenacetin in solid state and in saturated conditions in neat and binary solvents were characterized based on differential scanning calorimetry and spectroscopic solubility measurements. The temperature-related heat capacity values measured for both the solid and melt states were provided and used for precise determination of the values for ideal solubility, fusion thermodynamic functions, and activity coefficients in the studied solutions. Factors affecting the accuracy of these values were discussed in terms of various models of specific heat capacity difference for phenacetin in crystal and super-cooled liquid states. It was concluded that different properties have varying sensitivity in relation to the accuracy of heat capacity values. The values of temperature-related excess solubility in aqueous binary mixtures were interpreted using the Jouyban-Acree solubility equation for aqueous binary mixtures of methanol, DMSO, DMF, 1,4-dioxane, and acetonitrile. All binary solvent systems studied exhibited strong positive non-ideal deviations from an algebraic rule of mixing. Additionally, an interesting co-solvency phenomenon was observed with phenacetin solubility in aqueous mixtures with acetonitrile or 1,4-dioxane. The remaining three solvents acted as strong co-solvents.

In this study, the temperature-dependent solubility of nicotinamide (niacin) was measured in six neat solvents and five aqueous-organic binary mixtures (methanol, 1,4-dioxane, acetonitrile, DMSO and DMF). It was discovered that the selected set of organic solvents offer all sorts of solvent effects, including co-solvent, synergistic, and anti-solvent features, enabling flexible tuning of niacin solubility. In addition, differential scanning calorimetry was used to characterize the fusion thermodynamics of nicotinamide. In particular, the heat capacity change upon melting was measured. The experimental data were interpreted by means of COSMO-RS-DARE (conductor-like screening model for realistic solvation-dimerization, aggregation, and reaction extension) for concentration dependent reactions. The solute-solute and solute-solvent intermolecular interactions were found to be significant in all of the studied systems, which was proven by the computed mutual affinity of the components at the saturated conditions. The values of the Gibbs free energies of pair formation were derived at an advanced level of theory (MP2), including corrections for electron correlation and zero point vibrational energy (ZPE). In all of the studied systems the self-association of nicotinamide was found to be a predominant intermolecular complex, irrespective of the temperature and composition of the binary system. The application of the COSMO-RS-DARE approach led to a perfect match between the computed and measured solubility data, by optimizing the parameter of intermolecular interactions.

The process of water-based foaming of bitumen produces binders that can be incorporated in cold recycled asphalt mixes and pavement upper structural layers made of half-warm mix asphalt prepared at 100-130 °C. During the foaming process, cold water and air act on hot bitumen (160-170 °C), which results in the explosive vaporization of water leading to changes in the binder structure. The impact of foaming on the properties of bitumen 70/100 was evaluated by investigating the binder characteristics before and after foaming. Determination of two foaming parameters, maximum expansion and half-life, was followed by measurements of penetration at 25 °C, softening point, Fraass breaking point, and dynamic viscosity at 60, 90, and 135 °C. Rheological and low-temperature tests were also performed before and after foaming bitumen 70/100. The Bending Beam Rheometer method was applied to determine the low temperature stiffness modulus. A DHR-2 rheometer was used to determine the dynamic modulus and phase angle of the tested binder. The Black and master curves before and after foaming were plotted in the 2S2P1D model and the model parameters were analysed. Analysis of the test results confirmed the effects of the foaming process on the basic, low-temperature, and rheological characteristics of the bitumen.

This study explores the effects of foaming on three selected bituminous binders: 50/70 paving grade bitumen, 45/80-55 polymer modified bitumen and 45/80-80 HiMA binder. The first part of the investigations included the evaluation of the foaming performance in terms of foaming temperature and foaming water content with the utilization of desirability functions and based on the equality of maximum expansion ratio and bitumen foam half-life. The second part of the study investigated the effects of foaming on the chemical structure of the binders using Fourier-transform infrared spectroscopy. The results of the spectroscopic measurements permitted calculation of structural indices specific to functional groups associated with bitumen oxidation, as well as those indicative of elastomeric modification. The results have shown that the different types of bitumen exhibited different foaming characteristics, which was most evident in bitumen foam half-lives, with the HiMA binder performing the best. The spectrometric measurements did not show any significant effects of foaming on the chemical structure of the evaluated binders related to oxidative stress, neither were any major changes in the PmB-specific regions found.

OBJECTIVES: The purpose of this meta-analysis was to reveal a potential association of the four functional polymorphisms in human Beta-defensin 1 (DEFB1) gene: rs1047031(c*5G > A) at 3'UTR and rs11362 (-20 G > A), rs1800972(-44 C > G), and rs1799946 (-52 G > A) at 5'UTR with the risk of common oral cavity pathologies that included periodontitis, caries, lichen planus, and recurrent aphthous stomatitis. METHODS: The relevant studies were obtained by the two researchers from PubMed, Scopus, and Web of Science up to April 29, 2020. The manual search of the reference lists was also performed. Studies on DEFB1 gene polymorphisms and oral cavity disorders, using the case-control genetic association analysis approach, and published as full texts in English were included. To assess the association strength, odds ratios (ORs) with their 95% confidence intervals (CIs) were extracted. RESULTS: Thirteen publications met the inclusion criteria and were incorporated in this meta-analysis. Statistically significant values of the association tests were found only for the rs1047031 polymorphism. Allele distribution in the rs1047031 polymorphism was significantly associated with susceptibility to oral cavity pathologies (adjusted P value = 0.003). The rare variant allele carriers had a significantly higher risk for oral disasters under recessive (CC vs CT + TT), and CC vs CT models. No significant correlations between rs11362, rs1800972, and rs1799946 and the risk of oral pathologies were revealed. CONCLUSIONS: Significant association between rs1047031 polymorphism and risk of oral pathologies has been found, and therefore, we suggest to include this polymorphism in future research concerning the genetic background of the oral cavity diseases.

DNA metabarcoding offers new perspectives, especially with regard to the high-throughput identification and diagnostics of pathogens. Microsporidia are an example of widely distributed, opportunistic and pathogenic microorganisms in which molecular identification is important for both environmental research and clinical diagnostics. We have developed a method for parallel detection of both microsporidian infection and the host species. We designed new primer sets: one specific for the classical Microsporidia (targeting the hypervariable V5 region of small subunit [ssu] rDNA), and a second one targeting a shortened fragment of the COI gene (standard metazoan DNA-barcode); both markers are well suited for next generation sequencing. Analysis of the ssu rDNA data set representing 607 microsporidian species (120 genera) indicated that the V5 region enables identification of >98% species in the data set (596/607). To test the method, we used microsporidians that infect mosquitoes in natural populations. Using mini-COI data, all field-collected mosquitoes were unambiguously assigned to seven species; among them almost 60% of specimens were positive for at least 11 different microsporidian species, including a new microsporidian ssu rDNA sequence (Microsporidium sp. PL01). Phylogenetic analysis showed that this species belongs to one of the two main clades in the Terresporidia. We found a high rate of microsporidian co-infections (9.4%). The numbers of sequence reads for the operational taxonomic units suggest that the occurrence of Nosema spp. in co-infections could benefit them; however, this observation should be retested using a more intensive host sampling. Our results show that DNA barcoding is a rapid and cost-effective method for deciphering sample diversity in greater resolution, including the hidden biodiversity that may be overlooked using classical methodology.
