RESUMO

Negative emotional stimuli are particularly salient events that receive privileged access to neurocognitive resources. At the neural level, the processing of negative stimuli relies on a set of sensory, limbic, and prefrontal areas. However, controversies exist on how demographic and task-related characteristics modulate this brain pattern. Here, we used activation likelihood estimation (ALE) meta-analysis and replicator dynamics to investigate the processing of negative visual stimuli in healthy adults. Our findings endorse the central role of the amygdala. This result might reflect how this structure modulates perceptual and attentional mechanisms in response to emotional stimuli. Additionally, we characterize how the neural processing of negative visual stimuli is influenced by the demographic factors of age and sex as well as by task-related characteristics like stimulus type, emotion category, and task instruction, with the amygdala showing comparable engagement across different sexes, stimulus types, and task instructions. Our findings practically inform experimentation in the affective neurosciences but also suggest brain circuits for neurobiological investigations of affective symptomatology.

SUMMARY Environmental samples are a cost-saving and easy-to-use approach to diagnose paratuberculosis at the herd level. Detailed knowledge concerning its uncertainties in herds with a low prevalence of Mycobacterium avium spp. paratuberculosis (MAP) is required to design sampling strategies in control programmes. This study aimed to calculate a threshold level of the apparent within-herd prevalence (WHPapp) as determined by individual mycobacterial cultivation (faecal culture; FC) of all cows thus allowing the detection of a herd as MAP-positive at a certain probability level (P d). Out of 200 environmental samples taken twice from five predefined locations in a barn, 25 were positive by FC and 60 were positive by a quantitative real-time PCR method (qPCR). A logistic regression model was used to calculate the WHPapp threshold of detection. For 50% P d, a WHPapp threshold of 2.9% was calculated for the combination of three samples (milking area, main cow alleyways, holding pen) tested simultaneously both by FC and qPCR. The threshold increased to 6.2% for 90% P d. Repeated environmental sampling did not reduce the WHPapp threshold. Depending on the particular needs for prevalence estimation or in control programmes (single or repeated sampling) the provided WHPapp thresholds at different P d will enable decisions to be made about various sampling strategies.

A production process, using upshock fermentation and osmotic downshock, for the effective production/excretion of mannosylglycerate (MG) by the trehalose-deficient mutant of the strain Thermus thermophilus RQ-1 has been developed. In the first phase of fed-batch fermentation, the knockout mutant was grown at 70 degrees C on a NaCl-free medium. After the culture reached the end of the exponential growth phase, upshift in temperature and NaCl concentration was applied. The temperature was increased to 77 degrees C, and NaCl was added up to 3.0% and kept constant during the second phase of fermentation. Although this shift in cultivation parameters caused a dramatic drop of cell density, a significant improvement in accumulation of MG up to 0.64 micromol/mg protein compared to batch fermentations (0.31 micromol/mg protein) was achieved. A total yield of 4.6 g MG/l of fermentation broth was obtained in the dialysis bioreactor with a productivity of 0.29 g MG l(-1) h(-1). The solute was released from the harvested biomass by osmotic downshock using demineralized water at 70 degrees C. More than 90% of the intracellularly accumulated solute was recovered from the water fraction. The process was very efficient, as hyperosmotic shock, release of the solute, and reiterative fed-batch fermentation could be repeated at least four times.

Host snail demography and trematode parasitism were followed for one host generation in a shallow brackish lagoon of the western Baltic Sea. In addition, a laboratory experiment was simultaneously conducted to quantify the effects of parasitic infection on host fecundity. Hydrobia ventrosa of the cohort of 1996 had a maximum life-span of up to 2 years and reproduced between May and November of their second calendar year in 1997. Snails died after reproduction. The first trematode infections appeared in May 1997 when the snails started to mature. Total trematode prevalence peaked in summer and declined during winter to the lowest level in early spring 1998. Eight taxa of larval trematodes were found. Egg production of females with trematode infections was significantly reduced. Among females with pre-patent infections, about 20% were still able to produce eggs. Among females with patent infections merely 9% could lay eggs, compared to an average of about 51% in uninfected females. Taking into account a summer prevalence of about 25%, parasitic infections caused an overall reduction in egg production of the snail host population of about 15%. The reduction in host fecundity as a result of larval trematode infection did not measurably affect the population dynamics of H. ventrosa, because other environmental factors, especially winter severity and available food supply, were concluded to be much more relevant.
