RESUMO

A new methyl-dinitro-phenylhydrazinyl-pyridine derivative [2-methyl-3,5-dinitro-6-(2-phenylhydrazinyl)pyridine] was synthesised and characterised by means of structural and spectroscopic measurements. The X-ray diffraction studies revealed that the compound crystallises in the centrosymmetric monoclinic space group P21/n, with two symmetry-independent molecules in the asymmetric unit with Z = 8. Hydrazo bridge C-NH-NH-C links two fragments composed of phenyl ring and pyridine unit substituted with methyl and nitro groups. Such a structure was confirmed by 1H and 13C NMR studies as well as IR, Raman, UV-Vis, and emission spectra. The results were analysed using the quantum-chemical DFT calculations. The paper reports the vibrational characteristics and discusses dynamical properties of this moiety. The full set of the normal modes typical of the hydrazo bridge was identified and assigned to respective IR and Raman bands. The results of structural and spectroscopic studies were used to find the dependence between the conformation of the θ-NH-NH-ϕ system and its optic properties. The experimental UV-Vis and emission spectra were discussed in terms of the calculated singlet and triplet states that allowed assigning the unique spectral pattern originating from the electrons of the hydrazo-bridge system.

Silicones are commonly used as adhesives when high-quality materials are required due to harsh environmental conditions such as high temperature, humidity, etc. To ensure high resistance to environmental conditions, including high temperatures, modifications of silicone adhesives are made using fillers. The characteristics of a modified silicone-based pressure-sensitive adhesive with filler are the focus of this work. Functionalized palygorskite was prepared in this investigation by grafting 3-mercaptopropyltrimethoxysilane (MPTMS) onto palygorskite (palygorskite-MPTMS). The palygorskite was functionalized using MPTMS under dried conditions. FTIR/ATR spectroscopy, thermogravimetric analysis, and elemental analysis were all used to characterize the obtained palygorskite-MPTMS. MPTMS loading onto palygorskite was also proposed. The results demonstrated that palygorskite's initial calcination favors the grafting of functional groups on its surface. New self-adhesive tapes based on palygorskite-modified silicone resins have been obtained. This functionalized filler allows for the improvement of the compatibility of palygorskite with specific resins for application in heat-resistant silicone pressure-sensitive adhesives. The new self-adhesive materials showed increased thermal resistance while maintaining good self-adhesive properties.

The plants of the genus Rubus (R.) are applied as antiseptic agents in the treatment of skin diseases. Despite the great interest in plants of this genus, there are few reports on the antioxidant and biological activities of preparations obtained from the leaves of these plants. Therefore, we decided to evaluate the antioxidant activity of preparations from leaves of wild plant species of the genus Rubus using the frequently applied DPPH, ABTS, and FRAP methods, as well as to determine the total polyphenol content using the Folin−Ciocalteau method and perform qualitative evaluation by gas chromatography−mass spectrometry (GC-MS). The bactericidal and fungicidal activities of the obtained preparations were evaluated by applying laboratory tests: using the disc and the well methods based on the standards EN 13697:2019, EN 13697:2015, and EN 1500:2013. Microbiological tests of the plant preparations against bacteria, fungi, and yeasts isolated from the environment and against reference strains were performed. Moreover, antimicrobial testing of antibiotics against the tested strains was performed for comparison. The n-octanol/water partition coefficient of the obtained preparations was determined by the shake-flask method to determine their lipophilicity. According to the results, a high content of polyphenols and other antioxidant and biologically active compounds can be thought of as the parameter responsible for the effective activity of plant preparations obtained from wild plant species of the genus Rubus. The methods for determining bactericidal and fungicidal activity clearly demonstrates that preparations with reduced ethanol content exhibit bactericidal and fungicidal activity on surfaces. Testing of hand disinfection by means of rubbing with the preparations confirmed their antimicrobial activity against Escherichia coli K12 NCTC 10538. The obtained results show that the tested preparations exhibit on average two times lower activity against the reference bacterial strains than comparable antibiotics. The preparations obtained from the leaves of R. idaeus L. and R. fruticosus L. could complement classical antibiotics. While environmental bacteria showed a similar response to the preparations and antibiotics, their sensitivity was about one-third less than that of the reference strains. Our studies have shown that the obtained preparations are highly hydrophilic (logP < 0). Thus, these preparations can only be used in lipid bilayers in the aqueous core of liposomes, not in the lipid envelope.

For this study, the thermal degradation of palm, coconut, rice bran, and rapeseed (canola) oils was studied. Products formed during deep-frying were identified using chemical methods and these results were verified with those derived from FT-IR (Fourier-transform infrared spectroscopy) studies. Mathematically processed spectral data were analyzed in terms of the breaking of double bonds, the decomposition of the carotenoids, and the reduction of the C=O carbonyl group. Clearly visible changes in the position and intensity of some bands were used for explaining the structural changes in the studied oils. These changes prove that during the heating of the oils, decomposition of the plant fat into fatty acids appears, together with the reduction of the number of certain bonds (e.g., C=C, =C-H, and C=O) and cracking of the acylglycerol chains. The iodine values of the heated oils, determined from the FT-IR spectra measurements, show a significant decrease in their degree of unsaturation level. These effects, visible in the FT-IR spectra, confirm the chemical and structural changes derived from the chemical and physicochemical studies of the plant oils. The influence of heating time on the band intensity of proteins was also studied.

Epilobium angustifolium L. is an ethnomedicinal plant known as a medicinal plant in many regions of the world, among others, in various skin diseases. Despite the great interest in this plant, there are still few reports of biological activity of ready-made dermatological or cosmetical preparations containing the E. angustifolium extracts. The antioxidant, anti-ageing, anti-inflammatory, antibacterial properties and toxicity, wound healing, and skin permeation of topical hydrogels containing E. angustifolium extracts (HEas) was assessed. First, the plant extracts were prepared using three solvents: 70% (v/v) ethanol, 70% (v/v) isopropanol and water, next by preparing hydrogels witch by dry extracts (HEa-EtOH), (HEa-iPrOH) and (HEa-WA), respectively. Finally, the content of selected phenolic acids in the HEas was evaluated by high-performance liquid chromatography (HPLC). All the HEas were characterized by high antioxidant activity. The most increased antibacterial activity was observed for a strain of Streptococcus pneumoniae ATCC 49619, Escherichia coli, Enterococcus faecalis ATCC 29212, Enterococcus faecium, Sarcina lutea ATCC 9341 and Bacillus pseudomycoides, while the strains of Streptococcus epidermidis, Bacillus subtilis, and Staphylococcus aureus were the least sensitive. All the HEas showed a reduction in the activity of lipoxygenase enzymes, proteases, and inhibition of protein denaturation. The HEa-EtOH and HEa-iPrOH also enhanced the wound healing activity of HDF cells. Additionally, in vitro penetration studies were performed using the Franz diffusion cells. These studies showed that the active ingredients contained in E. angustifolium penetrate through human skin and accumulate in it. Furthermore, the hydrogels containing E. angustifolium extracts showed a broad spectrum of activity. Therefore, they can be considered as an interesting alternative for dermatologic and cosmetic preparations.

Ferulic acid (FA) has been widely used in the pharmaceutical and cosmetics industry due to its, inter alia, antioxidant, antiaging and anti-inflammatory effects This compound added to cosmetic preparations can protect skin because of its photoprotective activity. However, the usefulness of FA as a therapeutic agent is limited due to its low solubility and bioavailability. The paper presents the synthesis, identification, and physicochemical properties of new FA derivatives with propyl esters of three amino acids, glycine (GPr[FA]), L-leucine (LPr[FA]), and L-proline (PPr[FA]). The NMR and FTIR spectroscopy, DSC, and TG analysis were used as analytical methods. Moreover, water solubility of the new conjugates was compared with the parent acid. Both ferulic acid and its conjugates were introduced into hydrogel and emulsion, and the resulting formulations were evaluated for stability. Additionally, in vitro penetration of all studied compounds from both formulations and for comparative purposes using Franz diffusion cells was evaluated from the solution in 70% (v/v) ethanol. Finally, cytotoxicity against murine fibroblasts L929 was tested. All of the analyzed compounds permeated pig skin and accumulated in it. LPr[FA] and PPr[FA] were characterized by much better permeability compared to the parent ferulic acid. Additionally, it was shown that all the analyzed derivatives are characterized by high antioxidant activity and lack of cytotoxicity. Therefore, they can be considered as an interesting alternative to be applied in dermatologic and cosmetic preparations.

Coordination polymers with multiple non-centrosymmetric phases have sparked substantial research efforts in the materials community. We report the synthesis and properties of a hitherto unknown cadmium dicyanamide coordination polymer comprising benzyltrimethylammonium cations (BeTriMe+). The room-temperature (RT) crystal structure of [BeTriMe][Cd(N(CN)2)3] (BeTriMeCd) is composed of Cd centers linked together by triple dca-bridges to form one-dimensional chains with BeTriMe+ cations located in void spaces between the chains. The structure is polar, the space group is Cmc21, and the spontaneous polarization in the c-direction is induced by an arrangement of BeTriMe+ dipoles. BeTriMeCd undergoes a second-order phase transition (PT) at T1 = 268 K to a monoclinic polar phase P21. Much more drastic structural changes occur at the first-order PT observed in DSC at T2 = 391 K. Raman data prove that the PT at T2 leads to extensive rearrangement of the Cd-dca coordination sphere and pronounced disorder of both dca and BeTriMe+. On cooling, the HT polymorph transforms at T3 = 266 K to another phase of unknown symmetry. Temperature-resolved second harmonic generation (TR-SHG) studies at 800 nm confirm the structural non-centrosymmetry of all the phases detected. Optical studies indicate that BeTriMeCd exhibits at low temperatures an intense emission under 266 nm excitation. Strong temperature dependence of both one-photon excited luminescence and SHG response allowed for the demonstration of two disparate modes of optical thermometry for a single material. One is the classic ratiometric luminescence thermometry employing linear excitation in the ultraviolet region while the other is single-band SHG thermometry, a thus far unprecedented subtype of nonlinear optical thermometry. Thus, BeTriMeCd is a rare example of a dicyanamide framework exhibiting polar order, SHG activity, photoluminescence properties and linear and nonlinear optical temperature sensing capability.

[DMHy]Mn(HCOO)3 (DMHy+ = dimethylhydrazinium cation) is an example of an organic-inorganic hybrid adopting perovskite-like architecture with the largest organic cation used so far in the synthesis of formate-based hybrids. This compound undergoes an unusual isosymmetric phase transition at 240 K on heating. The mechanism of this phase transition has a complex nature and is mainly driven by the ordering of DMHy+ cations and accompanied by a significant distortion of the metal-formate framework in the low temperature (LT) phase. In this work, the Density Functional Theory (DFT) calculations and factor group analysis are combined with experimental temperature-dependent IR and Raman studies to unequivocally assign the observed vibrational modes and shed light on the details of the occurring structural changes. The spectroscopic data show that this first-order phase transition has a highly dynamic nature, which is a result of balanced interplay combining re-arrangement of the hydrogen bonds and ordering of DMHy+ cations. The tight confinement of organic cations forces simultaneous steric deformation of formate ions and the MnO6 octahedra.

RESUMO

Recently discovered hybrid perovskites based on hypophosphite ligands are a promising class of compounds exhibiting unusual structural properties and providing opportunities for construction of novel functional materials. Here, we report for the first time the detailed studies of phonon properties of manganese hypophosphite templated with methylhydrazinium cations ([CH3NH2NH2][Mn(H2PO2)3]). Its room temperature vibrational spectra were recorded for both polycrystalline sample and a single crystal. The proposed assignment based on Density Functional Theory (DFT) calculations of the observed vibrational modes is also presented. It is worth noting this is first report on polarized Raman measurements in this class of hybrid perovskites.

Subcompartments of the plasma membrane are believed to be critical for lymphocyte responses, but few genetic tools are available to test their function. Here we describe a previously unknown X-linked B cell-deficiency syndrome in mice caused by mutations in Atp11c, which encodes a member of the P4 ATPase family thought to serve as 'flippases' that concentrate aminophospholipids in the cytoplasmic leaflet of cell membranes. Defective ATP11C resulted in a lower rate of phosphatidylserine translocation in pro-B cells and much lower pre-B cell and B cell numbers despite expression of pre-rearranged immunoglobulin transgenes or enforced expression of the prosurvival protein Bcl-2 to prevent apoptosis and abolished pre-B cell population expansion in response to a transgene encoding interleukin 7. The only other abnormalities we noted were anemia, hyperbilirubinemia and hepatocellular carcinoma. Our results identify an intimate connection between phospholipid transport and B lymphocyte function.

To identify genes and mechanisms involved in humoral immunity, we did a mouse genetic screen for mutations that do not affect the first wave of antibody to immunization but disrupt response maturation and persistence. The first two mutants identified had loss-of-function mutations in the gene encoding a previously obscure member of a family of Rho-Rac GTP-exchange factors, DOCK8. DOCK8-mutant B cells were unable to form marginal zone B cells or to persist in germinal centers and undergo affinity maturation. Dock8 mutations disrupted accumulation of the integrin ligand ICAM-1 in the B cell immunological synapse but did not alter other aspects of B cell antigen receptor signaling. Humoral immunodeficiency due to Dock8 mutation provides evidence that organization of the immunological synapse is critical for signaling the survival of B cell subsets required for long-lasting immunity.

T cell antigen receptor (TCR) signaling in CD4(+)CD8(+) double-positive thymocytes determines cell survival and lineage commitment, but the genetic and molecular basis of this process is poorly defined. To address this issue, we used ethylnitrosourea mutagenesis to identify a previously unknown T lineage-specific gene, Themis, which is critical for the completion of positive selection. Themis contains a tandem repeat of a unique globular domain (called 'CABIT' here) that includes a cysteine motif that defines a family of five uncharacterized vertebrate proteins with orthologs in most animal species. Themis-deficient thymocytes showed no substantial impairment in early TCR signaling but did show altered expression of genes involved in the cell cycle and survival before and during positive selection. Our data suggest a unique function for Themis in sustaining positive selection.

Null mutations that cripple T cell receptor (TCR) signaling explain rare primary immunodeficiencies, but it is not understood why more common polymorphisms that lead to subtle TCR signaling defects are paradoxically associated with autoimmunity. Here we analyzed how a series of Zap70 variants with step-wise decreases in TCR signaling impacted upon opposing TCR functions of immunity and tolerance. One Zap70 variant, murdock, moderately decreased TCR signaling and thymic selection without compromising immunological tolerance, whereas a more severe Zap70 defect, mrtless, abolished thymic-positive selection and led to immunodeficiency. Signaling capacities between these two thresholds disproportionately compromised negative selection and Foxp3(+) regulatory T cell formation, creating a cellular imbalance between immunogenic and tolerogenic functions that resulted in the excessive production of autoantibodies and immunoglobulin E (IgE). The pleiotropic functions of ZAP-70 and their differential response to graded variation provide a paradigm for understanding the complex outcomes of human genetic variation.

RESUMO

Polycrystalline infrared and polarized FT-Raman spectra have been measured for 2-adamantylamino-5-nitropyridine, a novel organic material for laser Raman converters. The assignment of IR and Raman bands is given on the basis of DFT calculations. The spectroscopic studies have not indicated the presence of any significant intermolecular interactions in the crystal structure of this compound. The lines observed in the stimulated Raman spectrum of this crystal are assigned to the respective molecular vibrations.

In a genome-wide ENU mouse mutagenesis screen a recessive mouse mutation, unmodulated, was isolated with profound defects in humoral immune responses, selective deficits in B cell activation by antigen receptors and T cell costimulation by CD28, and gradual development of atopic dermatitis with hyper-IgE. Mutant B cells are specifically defective in forming connections between antigen receptors and two key signaling pathways for immunogenic responses, NF-kappaB and JNK, but signal normally to calcium, NFAT, and ERK. The mutation alters a conserved leucine in the coiled-coil domain of CARMA-1/CARD11, a member of the MAGUK protein family implicated in organizing multimolecular signaling complexes. These results define Carma-1 as a key regulator of the plasticity in antigen receptor signaling that underpins opposing mechanisms of immunity and tolerance.
