RESUMO

Twenty-three silver fox x hamster somatic cell hybrid clones were used to assign 15 fox genes: GPI to chromosome 1; PGD to chromosome 2; MDH2 to chromosome 3; ESD to chromosome 6; LDHB to chromosome 8; NP to chromosome 10; LDHA to chromosome 11; APRT, ENO1, and PGM1 to chromosome 12; IDH1 and MDH1 to chromosome 16; and GLA, G6PD, and HPRT to the X chromosome. High-resolution G-banding of human, cat, mink, and fox chromosomes containing homologous regions (according to genetic maps) revealed regions of putative homology. The results lend support to the suggestion that the most considerable karyotypic reorganization of the ancestral genome in the order Carnivora occurred during Canidae formation. The details of karyotypic evolution in mammals are discussed.

RESUMO

A fast electrophoretic variant of hypoxanthine phosphoribosyltransferase (HPRT) has been detected in Mus musculus bactrianus, a mouse subspecies from Middle Asia (USSR). The electrophoretic HPRT pattern yielded by hybrids between the somatic cell of LMTK- (deficient in thymidine kinase) and the splenocytes of a male of M. m. bactrianus was five-banded. The pattern obtained from the germ cells of the ovaries from 14.5-day-old embryos from laboratory CBA mice X M. m. bactrianus crosses was also composed of five bands. The hybrids between the somatic cells of human fibroblasts X LMTK- cells gave a three-banded electrophoretic HPRT pattern because the asymmetrical heteropolymeric isozymes are probably unstable. Taken together, all the evidence is in favor of a tetrameric structure of mammalian HPRT.

The electrophoretic mobilities of 52 enzymes and proteins were used as measures of the genetic similarity between the sibling species Microtus arvalis and M. subarvalis. The two vole species differed in the electrophoretic mobilities of seven (glucose-6-phosphate dehydrogenase, adenylate kinase, diaphorase, lactate dehydrogenase-A, alpha-galactosidase, 6-phosphogluconate dehydrogenase, and hemoglobin) of these markers. This allowed us to accept the seven markers assayed as species-specific markers. Based on the frequency distribution of the genes at the polymorphic loci of M. arvalis and M. subarvalis, the degree of their genetic similarity was estimated as 0.312 and the genetic distance as 1.164 by Nei's formula. The estimates for genetic similarity were close to those obtained for species recognized as distinct.
