Dynamic nuclear architecture and chromatin organizations are the key features of the mid-prophase I in mammalian meiosis. The chromatin undergoes major changes, including meiosis-specific spatiotemporal arrangements and remodeling, the establishment of chromatin loop-axis structure, pairing, and crossing over between homologous chromosomes, any deficiencies in these events may induce genome instability, subsequently leading to failure to produce gametes and infertility. Despite the significance of chromatin structure, little is known about the location of chromatin marks and the necessity of their balance during meiosis prophase I. Here, we show a thorough cytological study of the surface-spread meiotic chromosomes of mouse spermatocytes for H3K9,14,18,23,27,36, H4K12,16 acetylation, and H3K4,9,27,36 methylation. Active acetylation and methylation marks on H3 and H4, such as H3K9ac, H3K14ac, H3K18ac, H3K36ac, H3K56ac, H4K12ac, H4K16ac, and H3K36me3 exhibited pan-nuclear localization away from heterochromatin. In comparison, repressive marks like H3K9me3 and H3K27me3 are localized to heterochromatin. Further, taking advantage of the delivery of small-molecule chemical inhibitors methotrexate (heterochromatin enhancer), heterochromatin inhibitor, anacardic acid (histone acetyltransferase inhibitor), trichostatin A (histone deacetylase inhibitor), IOX1 (JmjC demethylases inhibitor), and AZ505 (methyltransferase inhibitor) in seminiferous tubules through the rete testis route, revealed that alteration in histone modifications enhanced the centromere mislocalization, chromosome breakage, altered meiotic recombination and reduced sperm count. Specifically, IOX1 and AZ505 treatment shows severe meiotic phenotypes, including altering chromosome axis length and chromatin loop size via transcriptional regulation of meiosis-specific genes. Our findings highlight the importance of balanced chromatin modifications in meiotic prophase I chromosome organization and instability.

It is of interest to compare two myofunctional appliances (frankal appliance and twin bloc) and two fixed orthodontic appliances (PowerScope and Forsus) in management of class II div 1 malocclusion. A total of 56 Class II division 1 malocclusion patients indicated for treatment with myofunctional appliances and fixed functional appliances were randomized. They were equally divided among frankal appliance (n=14), twin block appliance (n=14), PowerScope (American Orthodontics) (n=14), Forsus (3M Unitek Corp) groups (n=14). Skeletal and dentoalveolar effects of all appliances were compared. SNB increased remarkably by 4.2° in the Twin block group and it was high among all treatment groups. There was a significant decrease in vertical dimensions (SN-GoGn) in the Twin block (p = 0.002). Early treatment of Class II due to mandibular retrusion with Twin block functional appliance is recommended due to its favorable skeletal effect.

Rapid development of the industrial and domestic sectors has led to the rise of several energy and environmental issues. In accordance with sustainable development and waste minimization issues, biohydrogen production along with biomethane production via two-stage fermentation process using microorganisms from renewable sources has received considerable attention. In the present study, biohythane production with simultaneous wastewater treatment was studied in a two-stage (Biohydrogen and Biomethane) fermentation process under anaerobic conditions. Optimization of high organic content (COD) distillery spent wash effluent (DSPW) with dilution using sewage wastewater was carried out. Addition of leachate as a nutrient source was also studied for effective biohythane production. The experimental results showed that the maximum biohythane production at optimized concentration (substrate concentration of 60 g/L with 30% of leachate as a nutrient source) was 67 mmol/L bio-H2 and with bio-CH4 production of 42 mmol/L. A rapid and sensitive RP-HPLC method with UV detection (260nm) for routine analysis of voriconazole in a pharmaceutical formulation (Vfend((R))) was developed. Chromatography was performed with mobile phase containing a mixture of acetonitrile and water (50:50, v/v) with flow rate was of 1.0mlmin(-1). Quantitation was accomplished with internal standard method. The procedure was validated for linearity (correlation coefficient=0.9999), accuracy, robustness and intermediate precision. Experimental design was used for validation of robustness and intermediate precision. To test robustness, three factors were considered. Percentage of acetonitrile in mobile phase, flow rate and p(H); an increase in the flow rate results in a decrease of the drug found concentration, while the percentage of organic modifier and p(H) have no important effect on the response. For intermediate precision measure the variables considered were: analyst, equipment and number of days. The R.S.D. value (0.45%, n=24) indicated a good precision of the analytical method. The proposed method was simple, highly sensitive, precise and accurate and retention time less than 4min indicating that the method is useful for routine quality control.
