RESUMO

Beach sand and seawater taken from the coastlines of the North-East Pacific Ocean and Hawaii State were investigated to determine the causes of global chemical contamination from polystyrene (PS). All samples were found to contain styrene monomer (SM), styrene dimers (SD), and styrene trimers (ST) with a concentration distribution of styrene analogues in the order of ST > SD > SM. The contamination by styrene analogues along the West Coast proved more severe than in Alaska and other regions. The Western Coastlines of the USA seem be affected by both land- and ocean-based pollution sources, which might result from it being a heavily populated area as the data suggest a possible proportional relationship between PS pollution and population. Our results suggest the presence of new global chemical contaminants derived from PS in the ocean, and along coasts.

The pollution caused by plastic debris is an environmental problem with increasing concern in the oceans. Among the plastic polymers, polystyrene (PS) is one of the most problematic plastics due to the direct public health risk associated with their dispersion, as well as the numerous adverse environmental impacts which arise both directly from the plastics and from their degradation products. Little is known about their potential distribution characteristics throughout the oceans. For the first time, we report here on the regional distribution of styrene monomer (SM), styrene dimers (SD; 2,4-diphenyl-1-butene, SD1; 1,3-diphenyl propane, SD2), and styrene trimer (2,4,6-triphenyl-1-hexene: ST1), as products of PS decomposition determined from samples of sand and seawater from the shorelines of the North-West Pacific ocean. In order to quantitatively determine SM, SD (=SD1+SD2), and ST1, a new analytical method was developed. The detection limit was 3.3 µg L(-1), based on a signal-to-noise ratio of three, which was well-suited to quantify levels of SM, SD, and ST1 in samples. Surprisingly, the concentrations of SM, SD, and ST1 in sand samples from the shorelines were consistently greater than those in seawater samples from the same location. The results of this study suggest that SM, SD, and ST1 can be widely dispersed throughout the North-West Pacific oceans.

Using our original in vitro assay system with goldfish scales, we examined the direct effect of prostaglandin E2 (PGE2) on osteoclasts and osteoblasts in teleosts. In this assay system, we measured the activity of alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) as respective indicators of each activity in osteoblasts and osteoclasts. ALP activity in scales significantly increased following treatment at high concentration of PGE2(10⁻7 and 10⁻6 M) over 6 hrs of incubation. At 18 hrs of incubation, ALP activity also significantly increased in the PGE2 (10⁻9 to 10⁻6 M)-treated scale. In the case of osteoclasts, TRAP activity tended to increase at 6 hrs of incubation, and then significantly increased at 18 hrs of incubation by PGE2 (10(-7) to 10⁻6 M) treatment. At 18 hrs of incubation, the mRNA expression of osteoclastic markers (TRAP and cathepsin K) and receptor activator of the NF-κB ligand (RANKL), an activating factor of osteoclasts expressed in osteoblasts, increased in PGE2 treated-scales. Thus, PGE2 acts on osteoblasts, and then increases the osteoclastic activity in the scales of goldfish as it does in the bone of mammals. In an in vivo experiment, plasma calcium levels and scale TRAP and ALP activities in the PGE2-injencted goldfish increased significantly. We conclude that, in teleosts, PGE2 activates both osteoblasts and osteoclasts and participates in calcium metabolism.

The distribution and abundance of marine litter on 26 beaches along the Sea of Japan were investigated from September to November 2000. The mean concentration of stranded litter in Japan and Russia was 2144 and 1344 g/100 m(2), respectively. The mean number of pieces of stranded litter in Japan and Russia was 341 and 20.7/100 m(2), respectively. The most abundant type of stranded litter was plastic, which accounted for 40-80 % of the total items in terms of weight and number. The mean concentration of buried litter in Japan and Russia was 9.03 and 2.70 g/m(2), respectively. The total weight ratio of buried litter to stranded litter in the samples was 0.65, suggesting the significance of buried litter when evaluating the status of litter on beaches. Resin pellets were found on 12 Japanese beaches, but on none of the Russian beaches.

RESUMO

The increasing number of existing and new chemicals demands ecotoxicological data as well as toxicological data for pre- and postmarketing risk assessments. Although human health has been the major concern in Japanese environmental management, ecosystem health is becoming the big issue as the need for preserving the diversity of ecosystems has been recognized. This recognition is changing the regulatory framework in Japan, resulting in new actions toward establishment of water-quality standards for aquatic organisms and ecotoxicological assessment of existing chemicals. At the same time, the need to assess complex liquids that contain several kinds of chemicals is increasing. The ecotoxicological study of Japanese effluents shows that the present chemical-specific standards are not enough to protect aquatic ecosystems. These two factors encourage the application of ecotoxicological tests as well as the toxicological data.
