AIM: To experimentally define the optimum modes ofoperation ofthe nonspecific detector IBAC integrated with C100 sampler and BioCapture sampler in model experiments during the work in various environmental conditions and at dispersion of the biological aerosol. MATERIALS AND METHODS: The nonspecific detector IBAC with the C100 sampler and the BioCapture sampler were used for air-monitoring test on existence of pathogenic biological agents (PBA). For preparation of the bioaerosols the strain of Salmonella typhimurium 9640 and a bull seralbumin were used. RESULTS: It is experimentally established that programming of IBAC on turning on the bioalarm system when maintaining concentration of bioaerosol in air above threshold level during 10 sec. is optimum. Its sensitivity makes 1x10³ m.c./ml of aerosol at this mode. The possibility of operation of air-monitoring instrumentation in various environmental conditions inside and outside is shown. CONCLUSION: As a result of the experi- ment the effectiveness of usage of the nonspecific detector IBAC with the C100 sampler and BioCapture sampler for air-monitoring on existence of PBA is proved. IBAC with C100 can be used as an inventory ofthe station ofair-monitoring during the public events, and BioCapture is suitable for equipment of the groups of epidemiological investigation.

AIM: Determination of the degree of phylogenetic relationship of Yersinia pestis strains iso- lated from the territories of natural foci of plague from the Caucasus using VNTR-typing by 25 loci (MLVA25). MATERIALS AND METHODS: 26 strains of Y pestis from Russian natural foci of the Caucasus were used in the study. 25 loci of tandem repeats in Y pestis genome by Le Fleche scheme were used for execution of multi-locus VNTR-analysis. Deciphering of nucleotide sequences was carried out in automatic sequencer ABI 3130 Genetic Analyser. Analysis of confinement of clusters to certain territories, objects and time of isolation of strains was carried out. using Arc GIS 10.1 program. RESULTS: Groups of MLVA25-types of various levels of discrimination were formed: clusters, groups and subgroups. Clusters were formed by strains ofvarious taxonomic membership: main and subspecies of Y pestis. Subgroups reflect membership of strains in certain foci, and MLVA25-types - the degree of genetic relationship. CONCLUSION: Genetic <> of plague causative agents obtained using MLVA25-types circulating in various natural-focal territories allow to solve problems ofboth theoretical and practical character: from interpretation of microevolution processes to the search of the source of infection and ways of its spread during possible epidemic complications.

Peri- and postmenopausal women commonly suffer from climacteric symptoms. We evaluated the effectiveness and safety of dietary supplement Amberen to relieve vasomotor and psychosomatic symptoms during the course of a 3-month, randomized, double-blind, placebo-controlled study. General clinical assessment, evaluation using the Greene climacteric test and Spielberger-Hanin test, determination of plasma levels of gonadotropins, estradiol, leptin and apolipoproteins were used to evaluate 42-60-year-old women with vasomotor and psychosomatic menopausal symptoms. One hundred and twenty-five women were enrolled in the study and randomized between two groups. Based on the Greene test results, there was a statistically significant improvement (р < 0.05) in 13 out of 21 menopausal symptoms in women who took Amberen. During the course and by the end of the study, patients showed statistically significant changes in the levels of estradiol, gonadotropins and leptin, and decreases in body weight and waist circumference. Spielberger-Hanin test showed that Amberen stabilizes patients' psychological state with a statistically significant decrease in anxiety, increased stress resistance and improved adaptability. Comparative analysis of the vital signs measurements, blood tests and urinalysis did not show any negative effects of Amberen on the patients. Our findings indicate that Amberen can be considered a method of choice to relief mild/moderate climacteric symptoms.

Comparative analysis of the MLVA25- and MLVA7-typing ability to evaluate focal belonging of Y. pestis strains by the example of bv. medievalis isolates from the Central-Caucasian highland natural plague focus was carried out. The MLVA25-types of-82 isolates from this area were determined and included into the database containing information on 949 Y. pestis strains from other natural foci of Russia and other countries. Categorical-UPGMA dendrograms were created on the bases of the data concerning all 25 VNTR loci or only seven of them, which were recommended by the experts of the Russian Research Anti-Plague Institute "Microbe" for differentiation of the Y. pestis strains according to their affiliation to specific foci. The obtained data indicated greater possibility of diagnostic mistakes in the case of the MLVA7-typing and supported expediency of division of the Central-Caucasian highland natural plague focus into two sub-foci.

The reactions of the regional lymph nodes, caused by implantation of the autologous multipotent stromal cells of bone marrow origin (AMSCBMO) to accelerate the healing of mandibular bone defect were studied by fluorescent microscopy in inbred male Wag rats aged 6 months (n=62). After the introduction of polyhydroxyalkanoate transplant containing adsorbed AMSCBMO with a transfected Green Fluorescent Protein (GFP) gene into a damaged bone area, the lymphoid nodules in submandibular lymph nodes demonstrated the appearance of numerous large macrophages containing multiple oval fluorescent inclusions in the cytoplasm. The number of these macrophages increased within 2 weeks after surgery and then began to decline. Apparently, AMSCBMO introduced in this way, were partially absorbed by macrophages. After destruction of the structures formed from AMSCBMO, the debris was also phagocytized by macrophages. In either case, these macrophages appeared in the germinal centers of lymphoid nodules in lymph nodes, where the induction of immune responses against DNA and GFP protein was probable.

The phylogenetic position of the genera Rossithidium and Psammothidium was studied using the plastid gene rbcL. The genus Rossithidium is not an independent taxon and should be transferred to Psammothidium. Taxa from Psammothidium genus formed a sister, but independent clade to taxa with Achnanthidium. Our data has shown that the order Achnathales was not the natural taxon and should be reduced, but the three families included in it were natural. Genera like Psammothidium (including Rossithidium) and Lemnicola should be included into the family Achnanthidiaceae. Therefore, raphe does not play an important role in diatom taxonomy and its loss occurred many times during the evolution of diatoms.

AIM: Analysis ofresults of epidemiologic monitoring especially dangerous, natural-foci and other infectious diseases, asill as epizootologic activity of natural foci of infection on,the terri- tory, of city-resort Sochi. MATERIALS AND METHODS: Laboratory studies of 820. samples by PCR; im- mune- and bacteriologic methods were carried out, among those 344 - clinical material, 12 - water from open bodies and 321 - field material. Molecular-genetic identification of-143 strains of Vibrio-cholerae, isolated from open water bodies of city-resort Sochi, was-carried out,. RESULTS: Circulation of causative agents of Q fever, tularemia and hemorrhagic fever with renal syndrome of Dobrava-Adler genotypes was established, -as well as rickettsia of tick spotted fever group Predomination of rotaviruses (70.9%) was detected during study of samples of clinical material in etiologic structure of sporadically emerging acute intestine infections. Relatively high temperature values of river water during summer months (from 18, to 30 C), exit of sulfide mineral waters into the riverbed, that create-alkaline medium, seasonal reduction of river debit and speed of water current facilitated sustenance of contamination of water of Agura river by V.cholerae. CONCLUSION: Activity of natural foci of the infectious disease and contamination of Agura iver by V.cholerae Ο1, (atoxigenic) gives evidence on the necessity of constant control of epidemiologic situation by dangerous and natural-foci infectious diseases on the territory of city-resort Sochi, as well as study regional features, of their causative agents, including using genetic methods.

RESUMO

This paper considers the experience of genotyping and sequencing technologies in laboratories of specialized anti-epidemic team (SAET) during the XXII Olympic Winter Games and XI Paralympic Winter Games of 2014 in Sochi. The work carried out during the pre-Olympic period on performance of readiness by SAET for these studies is analyzed. The results of genotyping strains of pathogens during the Olympic Games are presented. A conclusion about the effectiveness of the use of molecular genetic techniques in terms of SAET is made.

Morphological changes in tissues adjacent to the implant were studied 1, 2, 6, and 12 months after implantation of biodegradable suture materials (catgut thread, DemeTECH polyfilament thread, and Surgilactin monofilament thread) into subcutaneous fat of rats. Tissue reaction to implantation of different suture materials developed as usual in response to a wound process and to a foreign body. By the end of month 1. the stage of traumatic perifocal inflammation was replaced in all groups by the proliferative phase with formation of new vessels and connective tissue. A trend to reduction of the productive inflammation activity from month 1 to month 12 was noted. Complete degeneration of the material after 12 months with complete recovery of the histoarchitecture of the adjacent tissues (restitution) was detected only after implantation of DemeTECH polyfilament thread.

The method of light microscopy was used to study the reactions of the surrounding tissues to the implantation of non-degradable surgical materials (prolene 3/0 thread made from polypropelene, the composite grid made from polypropelene or polyester with polylactic acid) in the subcutaneous adipose tissue of rats 1, 2, 6 and 12 months after grafting. No less than 6 rats were examined in every time point. It was shown that the tissue reactions to implantation of different non-degradable surgical materials follows the general pattern, characteristic of wound process and foreign body reaction. By the first month of observation in both groups of animals the stage of traumatic perifocal inflammation was replaced by the proliferative phase with the formation of new blood vessel and connective tissue. The tendency to reduction of the activity of productive inflammation was observed from the 1st to the 12th month together with the increased fibrous scar transformation of the granulation tissue.

Polylactide degradation processes after its implantation into the subcutaneous fat of rats were studied by microscopy. After implantation polylactide was encapsulated in connective tissue. If a fragment of polylactide had sharp edges, injuring the tissue, a thick capsule formed with pronounced inflammatory infiltration and sclerosis of adjacent tissues. If the implant had no sharp edges, the capsule was thin and the level of inflammation the minimum. Foreign body giant cells were found in the capsule and near it in all cases. The activity of the inflammatory process gradually reduced over 6 months. The intensity of inflammation sharply increased by the 12th month after implantation, when activity of phagocyte enzyme and deformation by the capsule led to fragmentation or liquefaction of polylactide.

Contemporary experience of organization of atmospheric air monitoring for the presence of pathogenic biological agents (PBA) is generalized in the article. Proposals for the use of posts of atmospheric air control during mass events are presented. Issues of equipping the instrument base for air sampling and PBA detection are examined.

Operation experience of specialized anti-epidemic brigades (SAEB) for the support of sanitary-epidemiologic welfare during various emergencies is presented. The order of functioning and SAEB objectives during mass events is examined. Analysis of exercise of Stavropol Institute of Plague Control SAEB was carried out, proposals for augmentation of SAEB equipment table by instruments and reagents for indication and genotyping of pathogen strains are examined.

Recent literature on the morphological results of the application of polymers produced from lactic acid (polylactides--PLA), contains a plethora of experimental and clinical data on the efficiency and safety of their use for the medico-biological purposes. However, the question on the actual rate of biological disintegration of PLA materials remains unanswered. There are conflicting views on the inflammatory reaction ranging from a complete negation of an inflammatory response to PLA implantation up to the reports describing the expressed aseptic inflammatory reactions caused by presence of this material in tissues. Some researchers report the total absence of the foreign-body reactions to this class of implants, while some others indicate the obligatory formation of the foreign-body giant cells. Further research is necessary to precisely predict degradation processes of and to detect all the potential risks associated with the use of PLA-based materials.

The reactions of rat regional lymph nodes, caused by implantation of the autologous multipotent mesenchymal stromal cells of a bone marrow origin (AMMSCBM) for acceleration of bone defect regeneration in bottom jaw experiment were studied by methods of fluorescent light microscopy. After introduction in an injury site of a bottom jaw bone of polyhydroxyalkanoate with adsorbed AMMSCBM with a transfected GFP gene the numerous large macrophages with a set of oval fluorescent inclusions in cytoplasm appear in lymph nodules of submandibular lymph nodes. The number of such macrophages increases within 2 weeks after operation, and further starts decreasing. Probably, entered via such way the AMMSCBM partially are phagocytized by macrophages. At destruction of the structures created from AMMSCBM, debris also are phagocytized by macrophages. In that and other case these macrophages appear in the germinative centers of lymph nodules in lymph nodes where initiation of immunity reactions against DNA and same GFP isn't excluded.

Light microscopy was used to study connective tissue capsules formed around different silicone breast implants. It has been found that due to the activity of microfibroblasts to minimize the volume of foreign bodies, the capsule gradually shrinks; its inner surface deforms and develops an undulating appearance with a multitude of outgrowths or protrusions inside. With the passage of time, silicone migrates to the capsule tissue and outside where it is absorbed by phagocytes. The inerter to a living organism the implant material is, the less it will stimulate a macrophage response. The task of designing new implants is to search for maximally bioinert materials that are strong enough to be compressed by the capsule and be fragmented and, moreover, elastically similar to normal breast tissue.

Tissue reactions, accompanying the degradation processes of polylactide (PLA) implants after their placement into the subcutaneous tissue, were studied by the methods of light microscopy in 6 month-old Wag rats (n=24) weighing 180-200 g. It was found that PLA implants became encapsulated by connective tissue. If the implant or its fragment had sharp margins that damaged the tissues, a thick capsule was formed with a distinct inflammatory infiltration and the sclerosis of the surrounding tissues. If the implant had no sharp margins, the capsule was thin, and the activity of inflammation was minimal. In all cases, the giant cells of foreign body type were present in the capsule and next to it. Gradually, during 6 months, the activity of inflammatory process subsided. Inflammation sharply increased 12 months after the implantation, when, as a result of effect of enzymes released by phagocytes and deformation by a capsule, PLA implants were either fragmented or is liquified. PLA-based materials are not totally biodegradable, and may persist in the body for a long time.
