CONTEXT: In this paper, the adsorption characteristics of five sulfonamide antibiotic molecules on carbon nanotubes were investigated using density functional theory (DFT) calculations. The adsorption configurations of different adsorption sites were optimized, and the most stable adsorption configuration of each sulfonamide molecule was determined by adsorption energy comparison, and the relative adsorption stability of five sulfonamide molecules on carbon nanotubes was determined by comparing their adsorption energies, i.e., sulfamethazine > sulfadiazine > sulfamerazine > sulfamethoxazole > sulfanilamide. The electron densities of the adsorption configurations were then calculated to confirm that the adsorption of five sulfonamide drug molecules on carbon nanotubes should be physical adsorption. Moreover, the adsorption energy of five sulfonamide molecules on carbon nanotubes in the aqueous environment was larger than that in the vacuum even though the adsorption process remain to be physical adsorption. The adsorption characteristics of the five sulfonamide molecules in various acid-base environments were finally investigated. In contrast, the adsorption energies of the five drug molecules in acid-base environments were significantly reduced, indicating that carbon nanotubes may need to have a suitable pH range to achieve the optimal adsorption effect when they are used for the treatment of sulfonamide antibiotics. METHODS: In this paper, we use density functional theory (DFT) with PBE functional to study the adsorption properties of five sulfonamides on carbon nanotubes. The structural optimization and the calculation of electronic structural properties are carried out by CP2K package (version 7.1), adopting the DZVP-MOLOPT-SR-GTH basis set and Goedeck-Teter-Hutter (GTH) pseudo potential. Grimme's D3 correction is used to during all the calculations to correctly capture the influence of the van der Waals interactions.

Background: This study determined the predictive value of CRMP4 promoter methylation in prostate tissues collected by core needle biopsies for a postoperative upgrade of Gleason Score (GS) to ≥8 in patients with low-risk PCa. Method: A retrospective analysis of the clinical data was conducted from 631 patients diagnosed with low-risk PCa by core needle biopsy at multiple centers and then underwent Radical Prostatectomy (RP) from 2014-2019. Specimens were collected by core needle biopsy to detect CRMP4 promoter methylation. The pathologic factors correlated with the postoperative GS upgrade to ≥8 were analyzed by logistic regression. The cut-off value for CRMP4 promoter methylation in the prostate tissues collected by core needle biopsy was estimated from the ROC curve in patients with a postoperative GS upgrade to ≥8. Result: Multivariate logistic regression showed that prostate volume, number of positive cores, and CRMP4 promoter methylation were predictive factors for a GS upgrade to ≥8 (OR: 0.94, 95% CI: 0.91-0.98, P=0.003; OR: 3.16, 95% CI: 1.81-5.53, P<0.001; and OR: 1.43, 95% CI: 1.32-1.55, P<0.001, respectively). The positive predictive rate was 85.2%, the negative predictive rate was 99.3%, and the overall predictive rate was 97.9%. When the CRMP4 promoter methylation rate was >18.00%, the low-risk PCa patients were more likely to escalate to high-risk patients. The predictive sensitivity and specificity were 86.9% and 98.8%, respectively. The area under the ROC curve (AUC) was 0.929 (95% CI: 0.883-0.976; P<0.001). The biochemical recurrence (BCR)-free survival, progression-free survival (PFS), and cancer-specific survival (CSS) were worse in patients with CRMP4 methylation >18.0% and postoperative GS upgrade to ≥8 than in patients without an upgrade (P ≤ 0.002). Conclusion: A CRMP4 promoter methylation rate >18.00% in prostate cancer tissues indicated that patients were more likely to escalate from low-to-high risk after undergoing an RP. We recommend determining CRMP4 promoter methylation before RP for low-risk PCa patients.

BACKGROUND: The treatment of ketamine-induced bladder contractures remains poorly studied. We therefore evaluated the efficacy of cystectasia with a sodium hyaluronate balanced solution in this kind of bladder contracture. METHODS: Eighteen patients presenting with ketamine-induced bladder contracture between July 2010 and February 2018 were selected and analysed. Ketamine was discontinued in all patients, who were then treated with weekly cystectasia (0.09% sodium hyaluronate balanced solution) 3 times. The volume of the first perfusion was twice the preoperatively measured bladder capacity, and the volume of the subsequent two perfusions was increased by 100 mL each time. The Pelvic Pain and Urgency/Frequency (PUF) symptom score, O'Leary-Sant Interstitial Cystitis (IC) Symptom Index (ICSI), IC Problem Index (ICPI), Quality of Life (QOL) score, and bladder capacity were recorded before surgery and 3 and 12 months after the 3rd expansion. RESULTS: No significant complications were observed during the 3 expansions. Fourteen patients completed the full follow-up schedule. Preoperatively and at the 3- and 12-month follow-up evaluations performed after the 3rd expansion, the PUF symptom scores were 20.4±3.6, 11.5±3.1, and 13.2±3.3, respectively; the mean ICSI was 13.6±2.8, 7.7±2.3, and 8.2±2.5, respectively; the mean ICPI was 10.6±2.6, 7.3±2.1, and 7.7±2.5, respectively; and the mean QOL scores were 6.0±0, 2.1±0.5, and 2.7±0.8, respectively; and the mean bladder catheter volume was 83±27, 234±56, and 228±52 mL, respectively. There were significant differences between all preoperative and postoperative values. CONCLUSIONS: Cystectasia with a sodium hyaluronate balanced solution is an effective treatment modality for ketamine-induced bladder contracture.

The treatment of sewage with high-nitrogen/-phosphorus and low-carbon remains a challenge. A novel iron-carbon galvanic cells strengthened anaerobic/anoxic/oxic process (Fe/C-A2O) was developed for high-nitrogen/-phosphorus and low-carbon sewage treatment. The cost-effective iron-scraps (ISs) was recycled as Fe(0)-source under the mediation of Fe/C galvanic cell reaction to develop effective Fe(0)-oxidizing autotrophic-denitrification and -dephosphorization. Utilizing practical high-nitrogen/-phosphorus and low-carbon sewage as target wastewater, the performance, impact factors, contribution of Fe/C galvanic cell reactions, microbial characteristics, strengthening mechanisms, and application potential of Fe/C-A2O process were investigated. The Fe/C-A2O process achieved high TN and TP removal efficiencies of 92.0 ± 1.3% and 97.2 ± 0.9% with removal loads of 0.176 ± 0.002 kg TN/(m3·d) and 0.017 ± 0.002 kg TP/(m3·d), respectively. Optimal HRT of 12 h, DO of 4.0-4.5 mg/L, and reflux-ratio of 4:1 were obtained, and no sludge-reflux was required. Autotrophic-denitrification and -dephosphorization supported by the Fe/C galvanic cell reactions contributed 63.1% and 75.3% of TN and TP removal, respectively. Microbial characterization revealed the dominance of autotrophic denitrifiers (e.g., Thiobacillus), AOB (e.g., Nitrosomonas), NOB (e.g., Nitrospira), and heterotrophic denitrifiers (e.g., Zoogloea). The mechanism analysis demonstrated that Fe/C galvanic cells strengthened nitrogen removal by raising Fe2+/H2-supported autotrophic denitrification; and strengthened dephosphorization by introducing Fe3+-based PO43--precipitation and enhancing the denitrifying phosphate-accumulation by denitrifying phosphate-accumulating organisms (DPAOs). Based on the efficiency and cost evaluation, the ISs-based Fe/C-A2O process showed significant application potential as an upgrade strategy for traditional A2O process in advanced high-nitrogen/phosphorus and low-carbon sewage treatment.

The Shen-Fu region is an important urban area in northeast China. We report on a study of the distribution of polybrominated diphenyl ethers (PBDEs) in representative topsoil from this region. In the summer of 2016, 72 soil samples from three cities (Shenyang, Fushun, and Shen-Fu New City) were collected, which covered four land use types:urban, rural residential, cultivated, and woodland. We report on the concentrations, compositions, and distributions of 14 PBDEs in soil and explore their sources, and additionally undertake human exposure analysis and health risk assessments. The results showed that the concentration of ∑14PBDEs in the topsoil ranged from 0.279-50.719 ng·g-1(dry weight), with a mean of (10.466±9.246) ng·g-1. The concentrations of PBDEs was ranked for the cities as:Fushun > Shenyang > Shen-Fu New City > background, and for different land use types as:urban land > rural residential land > cultivated land > forest. Deca-PBDE had the highest proportion of all congeners, accounting for 81.25%-89.23% of all PBDEs. Source analysis indicated that commercial Deca-PBDE was the main source, contributing 66.06% of the total Deca-PBDE according to principal component analysis/multiple linear regression (PCA-MLR). Among five different age groups assessed for exposure, children in Fushun had the highest exposure dose:(20.98±25.01) ng·(kg·d)-1. In terms of different land types, the highest exposure dose was for children living in urban areas:(18.54±20.27) ng·(kg·d)-1. The non-oncogenic health risks in the Shen-Fu region are of a relatively low level.

Wastewater from high-speed railway trains represents a mobile reservoir of microorganisms with antibiotic resistance. It harbors abundant and diverse antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs). This study investigated the removal of ARB and ARGs in a pilot-scale reactor, which consisted of an anaerobic/anoxic/oxic process, anaerobic/anoxic/aerobic process, and ozone-based disinfection to treat 1 m3/day wastewater from an electric multiple unit high-speed train. Further, the high prevalence of two mobile genetic elements (intI1 and Tn916/615) and five ARGs (tetA, tetG, qnrA, qnrS, blaNDM-1, and ermF) was investigated using quantitative PCR. Significant positive correlations between ARGs (tetA, blaNDM-1, and qnrA) and intI1 were identified (R2 of 0.94, 0.85, and 0.70, respectively, P < 0.01). Biological treatment could significantly reduce Tn916/1545 (2.57 logs reduction) and Enterococci (2.56 logs reduction of colony forming unit (CFU)/mL), but the qnrS abundance increased (1.19 logs increase). Ozonation disinfection could further significantly decrease ARGs and Enterococci in wastewater, with a reduction of 1.67-2.49 logs and 3.16 logs CFU/mL, respectively. Moreover, food-related bacteria families which may contain opportunistic or parasitic pathogens (e.g., Moraxellaceae, Carnobacteriaceae, and Ruminococcaceae) were detected frequently. Enterococci filtered in this study shows multi-antibiotic resistance. Our study highlights the significance to mitigate antibiotic resistance from wastewater generated from high-speed railway trains, as a mobile source.

In a previous study by the present authors, it was identified that the expression of engrailed-2 (EN2) gene was downregulated in clear cell renal cell carcinoma (cc-RCC). The aim of the present study was to determine whether aberrant methylation was the mechanism underlying the silencing of EN2 gene in cc-RCC. A total of forty paired cc-RCC tissues, four cc-RCC cell lines and one normal human proximal tubule epithelial cell line were evaluated for EN2 gene methylation status using methylation-specific polymerase chain reaction (PCR). Following treatment with 5-Aza-dc, reverse transcription-quantitative PCR and western blot analysis were performed to examine the expression of EN2. Furthermore, cell proliferation, apoptosis and invasion assays were conducted to analyze the inhibitory effects of EN2 re-expression in 786-O cells. The results of the present study demonstrated that hyper-methylation of EN2 was identified in 12/40 cc-RCC tissues and all cc-RCC cell lines. The methylation status of the EN2 gene was revealed to be associated with histological grade and tumor size in cc-RCC. Following 5-Aza-dc treatment, demethylation of the EN2 gene was identified in 786-O cells, in conjunction with EN2 re-expression. Furthermore, re-activation of the EN2 gene markedly inhibited the proliferative and invasive capacities of cc-RCC. The results of the present study demonstrated that the EN2 gene promoter was hyper-methylated in cc-RCC, which may underlie the silencing of the EN2 gene in cc-RCC.

Our preliminary study indicated that Engrailed-2 (EN2) is downregulated but also ectopically expressed in clear-cell renal cell carcinoma (CCRCC), and the absence of EN2 expression was associated with poor histological grade. However, the specific roles of EN2 in CCRCC have yet to be elucidated. In the present study, we examined the effects of inhibiting EN2 expression by human renal tubular epithelial cells (HK-2) and overexpressing EN2 by human clear-cell renal cells (786-O). Results showed that EN2 inhibition accelerated HK-2 cell proliferation, shortened the cell cycle, reduced apoptosis, and acted more invasively. By contrast, EN2 overexpression in 786-O cells decelerated the proliferative ability of 786-O, increased the percentage of cell apoptosis, and weakened the invasive ability. Overall, the results demonstrated that EN2 might play an anti-oncogenic role in oncogenesis and development of CCRCC, thereby maintaining the normal growth of human renal tubular epithelial cells.

The aim of this study was to assess the diagnostic value of RASSF1A methylation in renal cell carcinoma. Systematically search were performed using the Pubmed, ProQest and Web of Science for all articles on the association between RASSF1A methylation and renal cell carcinoma before 15 April 2015. After the filtration, 13 studies involving 677 cases and 497 controls met our criteria. Our meta-analysis suggested that hypermethylation of RASSF1A gene was associated with the increased risk of RCC(OR:4.14, 95%CI:1.06-16.1). Stratified analyses showed a similar risk in qualitative detection method(OR:28.4, 95%CI:10.2-79.6), body fluid sample(OR:12.8, 95%CI:5.35-30.8), and American(OR:10.5, 95%CI:1.97-55.9). Our result identified that RASSF1A methylation had a strong potential in prediction the risk of Renal cell carcinoma.

Engrailed-2 (EN2), a member of the homeobox family of genes, encodes a homeodomain-containing transcription factor that is thought to be a potential oncogene in a number of cancers. Because the role of EN2 in clear cell renal cell carcinoma (CCRCC) has not been determined, we investigated its expression in CCRCC tissues and cell lines. Using immunohistochemical (IHC) staining, we found that EN2 protein was expressed in normal renal cells and tubules, but was frequently down-regulated in tissues from patients with CCRCC and in CCRCC cell lines. In addition, we found that EN2, which functions in the nucleus, was completely localized to the cytoplasm of CCRCC cells as detected by IHC and immunofluorescence staining. Furthermore, expression of EN2 protein was negatively correlated with increasing histological grade of CCRCC tumors (P = 0.003). The exact role of EN2 expression in renal carcinoma carcinogenesis requires further investigation.

Ring-opening isomerization from ring-shaped isomers to chain-shaped isomers of N(8)H(8) has been studied by a density function B3LYP method at 6-311+ +G** level. 20 ring-shaped isomers have been found to be able to transform into chain-shaped isomers, with 20 possible transition states got by ring-opening structure optimization. Furthermore, the ring-openings have been found in the longer N-N single bond by analyzing the length change of N-N bond of ring-shaped isomers in ring-opening processes. In addition, with the activation energies in ring-opening processes, the differences of the activation energies in isomerization between the isomers have been found according to the classification of rings. The activation energies in ring-opening isomerization of six-membered ring-shaped isomers are higher than that of the four-membered ring-shaped isomers. It indicates that six-membered ring-shaped isomers difficult in ring-opening in the isomerization are the steadiest ring-shaped isomers of N(8)H(8) while four-membered ring-shaped isomers easy in ring-opening are the most unstable.

The PBS material that in the form of insoluble biodegradable polymers pellets was investigated as the solid carbon source and the biofilm carrier for nitrate removal from wastewater. The denitrification of nitrate removal and kinetic process were carried out in a packed-bed reactor in order to remove nitrate in recirculation aquaculture system. The experimental results indicated that the optimal influent loading rate was in the range of 0.107-1.098 kg/(m3 x d), when the water temperature was (29 +/- 1) degrees C and the influent nitrate concentration was in the range of 25-334 mg/L. The maximum nitrate volumetric removal rate of 0.577 kg/(m3 x d) was achieved at the influent loading rate of 1.098 kg/(m3 x d). When the influent loading rate exceeded 1.098 kg/(m3 x d), the nitrate volumetric removal rate was declined. The kinetic experimental results show that the denitrification rate of PBS as the solid carbon source and the biofilm carrier corresponds to first-order kinetics. Based on the kinetics characteristics, constants n and K used in Eckenfelder model were deduced, which can be successfully applied for the prediction of effluent nitrate concentration. The two groups' predictive values and actual values were analyzed by using SPSS 16.0 software for Paired-Samples t test analysis. The Paired-Samples t test analysis indicates that the corresponding p > 0.05 values are 0.553 and 0.632, which proved that no significant differences exist between the predictive values and actual values of the model.

We have constructed a novel refractive index (RI) sensor based on a fiber optic Fabry-Perot interferometer (FPI) by splicing a section of hollow core fiber between a single-mode fiber and a photonic crystal fiber (PCF). Owing to the air holes in the cladding of the PCF, various substances, such as liquids and gases with different RI, can enter or leave the in-fiber air cavity, which makes the device usable as a refractometer. In this paper, the fiber optic FPI sensor has been used to monitor the RI changes of air with different pressures, and the experimental results show that such a sensor has an RI sensitivity of 805.1 microm/RIU, and hysteresis is not observed. Moreover, the easy fabrication method gives the in-fiber refractometer many potential applications in the sensing field.

Because of the decrease in pasture and woodland proportion, soil loss is severe along the Yellow River basin. The Yellow River delta has been one of the deltas that form land most quickly. Research on the pollution of new land is important for its use and appraising the environment and food pollution along Yellow River basin. The contents of heavy metals (Mn, Cu, Zn, As, Cd, Pb, Hg and Cr) in new soils (2006) and old soils (1996) from Yellow River delta, were analyzed by the method of ICP-MS/ICP-AES, in order to determine the level of pollution along Yellow River basin. The results showed that the heavy metals pollution along Yellow River basin is increasingly severe, especially the concentrations of Mn, Cu, Zn, Cr and Cd in soil forming in 2006 are significantly higher than those in 1996, which should be noticed by our government.

The mechanism of the carbon-nitrogen coupling reaction of 2-iodo-selenophene with benzamide catalyzed by CuI has been investigated with density functional theory at the GGA/PW91/DND and GGA/PBE/DNP levels. The geometric configurations of the reactants, intermediates, transition states, and products were optimized and verified by means of vibration frequency calculations. A four-step mechanism was proposed for the reaction. The first step was the rate-control step. Two possible pathways in the fourth step were investigated, and the main pathway was identified by comparing their activation and dissociation energies. For comparison, the same calculations were performed to the reaction without the CuI activator. The activation barrier with CuI is 76 kJ mol(-1) smaller than that without CuI. It turns out that CuI can promote the reaction by lowering the activation energy. Our calculations reveal the crucial role of CuI in the reaction and agree well with experimental findings.
