This experiment was conducted to evaluate reproductive and productive responses of Bos indicus and B. taurus beef cows according to body condition score (BCS) at initiation of the annual breeding season. Non-pregnant, suckling multiparous cows were assigned to a fixed-time artificial insemination treatment regimen followed by natural service. Cows were classified as having a BCS < 5.0 (B. taurus, n = 158; B. indicus, n = 248) or BCS ≥ 5.0 (B. taurus, n = 173; B. indicus, n = 122). Calving rate was greater (P < 0.03) in cows with a BCS ≥ 5.0 (93.7 % and 85.1 % in B. taurus, 83.6 % and 73.3 % in B. indicus), which also calved earlier (P < 0.01) compared with cows with a BCS < 5.0. Cows with a BCS ≥ 5.0 weaned more (P ≤ 0.05) calves compared with those with a BCS < 5.0, which were older (P ≤ 0.04) and weighed more (P ≤ 0.09) at weaning (258 and 252 kg in B. taurus, 213 and 203 kg in B. indicus). Kilograms of calf weaned/cow in the breeding herd were greater (P ≤ 0.03) in cows with a BCS ≥ 5.0 compared with those with a BCS < 5.0 (221 and 200 kg in B. taurus, 159 and 129 d in B. indicus). Collectively, cows with a BCS ≥ 5.0 at initiation of the breeding season had greater reproductive and overall productivity during a cow-calf cycle, and these outcomes were observed in B. taurus and B. indicus cowherds.

There is a need to increase efficiency of beef production. Decreasing losses of CH4 and improving byproduct utilization are popular strategies. Two feed additives were tested to find potential solutions. Three randomized complete block design experiments were performed using batch culture systems to evaluate the effects of bismuth subsalicylate (BSS) and calcium-ammonium nitrate (CAN) on in vitro ruminal fermentation of bahiagrass hay and supplemental molasses. The first experiment contained four treatments: (1) basal substrate; (2) basal substrate with 0.75% urea (DM basis); (3) basal substrate with 1.2% CAN and 0.38% urea (DM basis); and (4) basal substrate with 2.4% CAN (DM basis). Treatments 2, 3, and 4 were isonitrogenous. The second experiment had a 4 × 3 factorial arrangement of treatments with 4 concentrations of BSS (0.00, 0.33, 0.66, and 1.00%; DM basis) and 3 concentrations of CAN (0.0, 1.2, and 2.4%; DM basis). The third experiment had the following treatments: (1) basal substrate; (2) basal substrate with 0.05% BSS (DM basis); (3) basal substrate with 0.10% BSS (DM basis); and (4) basal substrate with 0.33% BSS (DM basis). For all experiments, basal substrate consisted of Pensacola bahiagrass hay (Paspalum notatum Flüggé; 80% substrate DM) and molasses (20% substrate DM). All data were analyzed using the MIXED procedure of SAS. In Exp. 1, in vitro organic matter (OM) digestibility (IVOMD) was linearly reduced (P < 0.001) with the inclusion of CAN, and CH4, in mmol/g OM fermented, was decreased linearly (P < 0.001). The volatile fatty acid (VFA) profile was not impacted by the inclusion of nonprotein nitrogen (NPN) or CAN (P > 0.05). In Exp. 2, except for CH4 production (P < 0.05), there were no BSS × CAN interactions. Linear reductions in total gas production (P < 0.001), IVOMD (P < 0.001), and total concentration of VFA (P = 0.007) were observed with the inclusion of BSS up to 1%. The inclusion of BSS decreased H2S production in a quadratic manner (P = 0.024). In Exp. 3, IVOMD was not impacted by the inclusion of BSS (P > 0.05); however, production of H2S was linearly decreased (P = 0.004) with the inclusion of BSS up to 0.33%. In conclusion, in vitro fermentation was negatively impacted by the inclusions of BSS, up to 1%, and CAN, up to 2.4%; however, BSS decreased production of H2S when included up to 0.33% without impeding fermentation, while CAN decreased CH4 production.

Recent research from our group demonstrated that Bos indicus-influenced suckled beef cows had greater resilience to withstand nutrient restriction and establish pregnancy compared with B. taurus cows exposed to the same conditions. To further understand these findings, differences in metabolic profile between these same B. indicus-influenced and B. taurus females were explored. Suckled beef cows (n = 134) were enrolled in a completely randomized design with a 2 × 2 factorial arrangement of treatments. On day -21, Angus (AN; Bos taurus) and Brangus (BN; B. indicus-influenced) cows were randomly assigned to 1) a diet that met daily energy maintenance requirements (MAINT), or 2) a diet that restricted intake to 70% of the daily energy maintenance requirements (RESTR). Cows were exposed to an estrus synchronization protocol and received an embryo 7 d after ovulation was pharmacologically induced on day 0. Blood samples were collected on days -21 and 19 to determine circulating concentrations of non-esterified fatty acids (NEFA), ß-hydroxybutyrate (BHB), insulin, glucose, and IGF-1. Pregnancy status after embryo transfer was determined on day 28. As a consequence of the proposed diets, cows in the RESTR diet had less body condition score (BCS) on day 19 (P = 0.008) across breed types. Moreover, BCS change from day -21 to 19 was included as independent covariate into subsequent analyses, allowing for the comparison of breed types under an equivalent level of body reserve mobilization. A breed × diet interaction was observed for plasma insulin (P = 0.03) and IGF-1 (P = 0.04) on day 19, where AN-RESTR cows had less plasma concentrations on day 19 compared with AN-MAINT cows. Diets did not impact (P > 0.10) plasma insulin and IGF-1 concentrations in BN cows. No diet or breed effects were observed in circulating concentrations of NEFA, BHB, and glucose (P > 0.10). Across breed types and nutritional treatment, there was positive linear effect (P ≤ 0.04) of plasma concentrations of insulin and IGF-1 on the probability of pregnancy to fixed-time embryo transfer. In summary, the negative impacts of nutrient restriction on the somatotropic axis, independently of body tissue mobilization, were heightened in Bos taurus females compared with B. indicus-influenced cohorts, which corroborate with the differences observed in fertility between these subspecies.

This study was conducted to determine effects of pre-synchronization of ovulation timing among heifers and delayed fixed-time artificial insemination (TAI) with sex-sorted semen on proportion of heifers pregnant after TAI (PR/AI). Heifers were assigned to one of eight treatments: 1 and 2), 7-d CO-Synch + CIDR treatment regimen with administration of gonadotropin-releasing hormone and a CIDR insert on Day 0, prostaglandin F2α (PGF) at CIDR removal on Day 7, and TAI occurring 54 h later with conventionally processed (CTRL54-CNV) or sex-sorted semen (CTRL54-SEX); 3 and 4), same as CTRL54 but TAI delayed to 72 h with conventionally processed (CTRL72-CNV) or sex-sorted semen (CTRL72-SEX); 5 and 6), same as CTRL54 but additional administration of PGF on Day -7 and TAI with conventionally processed (PRE54-CNV) or sex-sorted semen (PRE54-SEX); 7 and 8), same as PRE54 treatments but TAI delayed to 72 h with conventionally processed (PRE72-CNV) or sex-sorted semen (PRE72-SEX). Proportion of heifers pregnant after TAI was greater (P ≤  0.02) with conventionally processed semen compared with sex-sorted semen, yet PR/AI did not differ (P =  0.14) between heifers in PRE72-CNV and PRE72-SEX groups. There were greater PR/AI in the PRE72-SEX (P =  0.03) than CTRL54-SEX group (46.1 % and 36.9 %) and there was no difference (P =  0.31) in PR/AI between CTRL54-CNV and PRE72-SEX groups (50.4 % and 46.1 %). In conclusion, pre-synchronization of ovulation timing among heifers combined with delayed TAI resulted in increased PR/AI with sex-sorted semen compared with the 7-d CO-Synch+CIDR treatment regimen.

To determine the effects of two presynchronization strategies in conjunction with delayed fixed-time artificial insemination (TAI) on pregnancy rates to TAI (PR/AI), 1700 Angus beef heifers at three locations in South Dakota were enrolled in a completely randomized design with a 2 by 2 factorial arrangement of treatments. Within location, all heifers were randomly assigned to one of four treatments: 1) PG54 (n = 434), heifers were administered prostaglandin F2α (PGF; 25 mg im) 7 d prior [Day -14] to the initiation of the 7-d CO-Synch + controlled internal drug releasing (CIDR) protocol wherein they received gonadotropin-releasing hormone (GnRH; 100 µg im) and a CIDR insert on Day -7, PGF at CIDR removal on Day 0, and a second injection of GnRH concurrently with TAI 54 ± 2 h later; 2) PG72 (n = 426), heifers were exposed to the same treatment as PG54, however, TAI was performed 72 ± 2 h after CIDR removal; 3) PG-CIDR54 (n = 422), same as PG54 but heifers received a CIDR insert on Day -14 rather than Day -7, in addition to PGF administration; 4) PG-CIDR72 (n = 418), same as PG-CIDR54, however, TAI was performed 72 ± 2 h after CIDR removal. Estrus detection patches were applied to all heifers on Day 0 and were evaluated for activation at TAI. Pregnancy was diagnosed via transrectal ultrasonography between 30 and 47 d after TAI. The percentage of heifers exhibiting estrus between Day 0 and TAI was greater (P < 0.01) in the PG72, PG-CIDR54, and PG-CIDR72 treatments compared to the PG54 treatment (78.11, 86.59, and 91.09 vs. 31.05%, respectively). Furthermore, estrus response was greater (P < 0.01) in PG-CIDR72 heifers when compared to PG72. Pregnancy rates to TAI differed among treatments and were greater (P < 0.05) in the PG72 and PG-CIDR54 treatments when compared to PG-CIDR72 (48.8 and 50.4 vs. 38.4%, respectively), and were greater (P = 0.03) in PG-CIDR54 vs. PG54 (43.1%). Moreover, a tendency (P = 0.10) was determined on PR/AI between PG54 and PG72. In conclusion, presynchronization strategies and prolonged exposure to exogenous progesterone have the potential to alter estrus expression and improve fertility in replacement beef heifers.

To evaluate the effects of a polyunsaturated fatty acids (PUFA) supplement on reproductive parameters of suckled beef cows, two experiments were conducted. In Experiment (Exp.) 1, 60 primiparous cows were randomly assigned to one of two treatments: CTRL - 1.36 kg/day of corn gluten feed (CGF) and MEGR - 1.36 kg/day of CGF and 0.23 kg/day of calcium salts of soybean oil. Supplementation occurred from 30 days before fixed-time artificial insemination (TAI) until 7 days post-TAI. The expression of interferon-stimulated genes (ISG) was measured on days 18 and 21. Pregnancy rates were diagnosed on days 30 and 100. Treatment altered plasma fatty acid profile (P<0.05), however, did not change cow BW (P=0.52) or body condition score (BCS) (P=0.52). Treatment did not alter (P=0.12) pregnancy rates to TAI or final pregnancy rates (P=0.56). Treatments did not impact messenger RNA (mRNA) expression of the ISG OAS1 or MX2 on days 18 (P=0.67; P=0.96, respectively) or 21 (P=0.72; P=0.17, respectively). Length of gestation was greater (P=0.02) for MEGR, however, treatments did not alter calf birth weight (P=0.20). In Exp. two, 66 multiparous cows were assigned to one of two treatments: MEG - 0.65 kg/day of CGF+0.23 kg/day of calcium salts of palm oil and MEGR - 0.65 kg/day of CGF+0.23 kg/day of Ca salts of soybean oil. Cows were supplemented from 30 days prepartum to 30 days postpartum. On day 35 after TAI, pregnancy status, embryo crown-to-rump length (CRL), and plasma concentrations of pregnancy-specific protein-B (PSPB) were evaluated. Treatment altered plasma fatty acid profile (P<0.05). In addition, cows from the MEG treatment had greater BW (P<0.01) and BCS (P<0.01) than those in the MEGR treatment, as well as heavier calves at weaning (P=0.03). Treatment did not affect resumption of estrous cycle (P=0.29). There were no differences in pregnancy rates to TAI (P=0.87) or final pregnancy rates (P=0.29). No differences between treatments were detected on CRL (P=0.24) and plasma concentrations of PSPB (P=0.46). Birth weight (P=0.12) and calving distribution (P=0.52) were not altered. We concluded that PUFA supplementation altered plasma fatty acid profile, however, did not impact the remaining reproductive parameters evaluated.

A 3 yr study evaluated the effects of three preweaning injections of bovine ST, administered 14 d apart, on growth and reproductive performance of Bos indicus-influenced beef heifers. On d 0 of each year, suckling Angus × Brangus heifers (n = 15 heifers/treatment/yr) were stratified by BW (147 ± 20 kg) and age (134 ± 11 d) and randomly assigned to receive an s.c. injection of saline (SAL; 5 mL; 0.9% NaCl) or 250 mg of sometribove zinc (BST; Posilac, Elanco, Greenfield, IN) on d 0, 14, and 28. Heifers and respective dams were managed as a single group on bahiagrass (Paspalum notatum) pastures from d 0 until weaning (d 127). From d 127 to 346, heifers were grouped by treatment, allocated to bahiagrass pastures (1 pasture/treatment/yr) and fed a molasses-based supplement (2.9 kg/heifer daily; DM basis) until d 346. Blood samples were collected on d 0, 14, 28, 42, and then every 9-10 d from d 179 to 346. In yr 3, liver biopsy samples were collected on d 0, 42, and 263. Heifers were exposed to mature Angus bulls from d 263 to 346. Growth performance and physiological parameters were analyzed using the MIXED procedure, whereas reproductive variables were analyzed using the GLIMMIX procedure of SAS. Effects of treatment × year and treatment × year × time were not detected for any variable measured in this study (P ≥ 0.14), except for calving percentage (P = 0.03). Heifers assigned to BST injections had greater overall plasma concentrations of IGF-1 and ADG from d 0 to 42 (P ≤ 0.05), less ADG from d 42 to 127 (P = 0.04), but had similar BW at weaning and postweaning ADG (P ≥ 0.25) compared to SAL heifers. Heifers assigned to BST tended to achieve puberty 26 d earlier (P = 0.10), had greater percentage of pubertal heifers on d 244, 263, 284, and 296 (P ≤ 0.04), tended to have greater overall pregnancy percentage (P = 0.10), and had greater (P ≤ 0.05) calving percentages in yr 1 and 2 (but not yr 3; P = 0.68) compared to SAL heifers. Liver mRNA expression of GHR-1B and IGF-1 on d 0 and 42 did not differ between treatments (P ≥ 0.15), but was greater for BST vs. SAL heifers on d 263 (P ≤ 0.02). Hence, administering three injections containing 250 mg of sometribove zinc at 14 d intervals before weaning (between 135 and 163 d of age) induced long-term impacts on liver gene expression and may be a feasible management practice to enhance puberty and pregnancy attainment in B. indicus-influenced replacement beef heifers.

To determine the effects of estrus synchronization (ES) and fixed-time artificial insemination (TAI) on calving distribution in Bos indicus influenced heifers, 751 Bos taurus × Bos indicus beef heifers were enrolled in a completely randomized design at 2 locations from January to May of 2016. Within location, all heifers were randomly assigned to one of two treatments: 1) SYNCH (n = 371); heifers were exposed to the 5-day CO-Synch + CIDR protocol where they were treated with 100 µg of GnRH, 25 mg of PGF2α, and a controlled internal drug releasing insert (CIDR) on d 0; heifers received 50 mg of PGF2α at CIDR removal on d 5, and were treated with 100 µg of GnRH and TAI 66 ± 2 h later on d 8; or 2) CONTROL (n = 380); heifers were exposed to natural service without ES or TAI. On d 9, all heifers were exposed to bulls for the remainder of the breeding season at each location. Blood samples were collected on d -9 and on d 0 to determine pretreatment estrous cyclicity (progesterone ≥ 1.0 ng/mL). Pregnancy was diagnosed via transrectal ultrasonography 54 d after TAI by determining the presence of a viable fetus. Fetal age was estimated based on fetal size and structural features at the time of pregnancy diagnosis. Pregnancy rates on d 54 differed (P < 0.001) between locations, but did not differ (P = 0.777) between CONTROL and SYNCH treatments. Pregnancy rates on d 54 were greater (P < 0.001) in cycling compared with non-cycling heifers (63.9 vs 42.4%). A greater (P < 0.05) proportion of SYNCH heifers became pregnant in the first 19 d of the breeding season compared with CONTROL heifers (52.2 vs 46.4%). Overall breeding season pregnancy rates did not differ (P = 0.982) between treatments. In summary, ES and TAI increased the percentage of heifers that conceived in the first 19 d of the breeding season, and therefore, potentially altered the calving distribution by ensuring that more heifers calve early during the subsequent calving season.

A 3-yr study evaluated the effectiveness of puberty induction using exogenous reproductive hormones in beef heifers undergoing different post-weaning growth rates. On d 0 of each year, grazing Angus × Brahman heifers (60 heifers/yr) were stratified by initial BW and age (BW = 253 ± 30 kg; age = 310 ± 18 d), and randomly assigned, in a 3 × 2 factorial design, to achieve a low (LOW; 0.45 kg/d), medium (MED; 0.73 kg/d), or high (HIGH; 1.00 kg/d) growth rate from d 0 to 168 (5 heifers/pasture; 4 pastures/growth rate/yr). Dietary supplements were offered 3 times weekly (Mondays, Wednesdays, and Fridays) at 0800 h to provide 1.8, 2.9, and 4.4 kg of supplement DM per heifer daily from d 0 to 168. On d 0 of each year, 2 pastures/growth rate treatment were randomly assigned to receive (SYNC) or not (NOSYNC) a puberty induction protocol, which consisted of intravaginal controlled internal drug release (CIDR) insertion on d 65, CIDR removal on d 72, followed by 100-µg i.m. injection of GnRH on d 74, and 25-mg i.m. injection of PGF on d 84. Immediately after the puberty induction protocol, all heifers were grouped by growth rate treatment (1 group/growth rate/yr) into a single pasture and exposed to yearling Angus × Brahman bulls from d 84 to 168 (1 bull/group/yr). Interactions among growth rate and puberty induction protocol were not detected for any variable ( ≥ 0.26). Overall heifer ADG achieved only 51 to 56% of targeted ADG, and percentage of mature BW on d 84 did not differ between MED and HIGH heifers ( = 0.30), but both were heavier than LOW heifers ( < 0.01). Percentage of pubertal heifers on d 84 was greater for MED and HIGH vs. LOW heifers ( ≤ 0.05), whereas puberty induction protocol increased puberty attainment on d 84 ( < 0.01). Overall pregnancy and calving percentages after first breeding season were not affected by growth rate or puberty induction protocol ( ≥ 0.25). However, heifers calved 11 d earlier when enrolled in the puberty induction protocol ( = 0.02). In summary, puberty induction and HIGH and MED growth rates resulted in earlier attainment of puberty and pregnancy in -influenced beef heifers. However, neither growth rate or puberty induction impacted overall heifer pregnancy and calving rates on their first breeding season.

The objective of this study was to determine if supplementation frequency of rumen-protected fat (RPF) influences circulating serum concentrations of fatty acids (FA), NEFA, and urea nitrogen in beef heifers and lactating cows. In Exp. 1, 12 early gestation beef heifers were supplemented 0.5 kg of corn gluten feed (CGF) daily during a 2-wk adaptation period. During the last 3 d of adaptation, blood samples were collected immediately before supplementation and then 8 and 16 h postsupplementation daily. Each heifer was then randomly assigned to 1 of 3 supplementation frequency treatments of RPF (3, 5, or 7 d/wk) for 3 wk in a 3 × 3 Latin square design with 3 periods (4 heifers per treatment per period), with each treatment receiving the same amount of RPF and CGF per wk (1.0 and 2.7 kg as fed, respectively). Blood samples were collected during the final 3 d of each supplementation period as described in the adaptation period. In Exp. 2, 18 Angus crossbred cows in early lactation were supplemented with 4.54 kg (as fed) of CGF weekly either at 3, 5, or 7 d/wk during a 2-wk adaptation period. Blood samples were collected during the last 3 d of adaptation as in Exp. 1. For the subsequent 3 wk, RPF (530, 318, and 227 g/d when supplemented) was added to the CGF supplement so that each supplementation frequency received 1.59 kg as fed/wk of RPF. Blood samples were collected during the last 3 d of supplementation as in Exp. 1. Serum FA profiles on a random subsample of 9 heifers in Exp. 1 and all animals in Exp. 2 were determined via gas chromatograph (GC), and values were analyzed using the MIXED procedure of SAS. In Exp. 1, there were no differences ( ≥ 0.53) in serum FA profile across supplementation frequencies. There was a decrease in serum 18:2, 18:1 -9 and total FA during the sampling time (time effect, < 0.02). In Exp. 2, there was treatment × time effect ( ≤ 0.001) for both 18:2 and total FA measured. The 7 d/wk frequency had a greater concentration of C18:2 and total FA at 8 h of the sampling period when compared to the other 2 frequencies. These results demonstrate that supplementation of RPF at 3, 5 or 7 d/wk resulted in no changes in serum FA profiles in growing heifers. In early lactating beef cows, there were minor time point changes in serum FA due to supplementation frequency; however, the vast majority of time points were not different, indicating no substantial changes in serum FA profiles.

The objectives of this experiment were to determine the effects of follicular wave (first or second) on diameter of the dominant follicle, concentrations of progesterone and estradiol and the hepatic enzymes that inactivate them, thickness of the endometrium, and pregnancy rates to AI. Beef heifers ( = 101) and cows ( = 106) were randomly assigned to 1 of 2 treatments: insemination to the first follicular wave (FFW) or insemination to the second follicular wave (SFW). Estrous cycles of females were synchronized to ensure appropriate timing for the treatments. The MIXED procedure of SAS was used for analysis. A similar proportion of females in each treatment responded to presynchronization; however, females in the FFW group ovulated in response to the first injection of GnRH of the CO-Synch protocol more frequently. Only females ( = 94) that properly responded to ovulation synchronization were included in further analyses. Cows in the FFW group tended ( 0.06) to have larger ovulatory follicles 36 h post-PGF of the CO-Synch protocol compared to cows in the SFW group (14.22 ± 0.42 and 11.83 ± 0.49, respectively), whereas heifers were similar between treatment groups. Three d prior to AI, circulating concentrations of progesterone were lesser ( 0.01) in females in the FFW (3.63 ± 0.80 ng/mL) than in the SFW (7.12 ± 0.83 ng/mL), whereas concentrations of estradiol tended ( 0.08) to be greater in those in the FFW (82.72 ± 6.48 pg/mL) than in the SFW (65.55 ± 6.74 pg/mL). Concentrations of cytochrome P450 1A in the liver were lesser ( 0.01) in females in the FFW than those in the SFW (0.68 ± 0.08 vs. 0.96 ± 0.06, respectively). Endometrial thicknesses were similar between treatments but were thicker ( < 0.0001) in cows (9.73 ± 0.24 mm) than heifers (7.22 ± 0.26 mm). When considering all females or only those that were properly presynchronized, pregnancy rates were similar between treatments. However, when evaluating females that ovulated to the assigned follicular wave, there was a treatment by parity interaction ( = 0.04) with heifers in the FFW having a lesser pregnancy rate (25.9%) than heifers in the SFW (72.0%) while cows in both treatment groups were intermediate (45.4% in FFW and 50.0% in SFW). The differences in concentrations of steroids between treatment groups may affect fertility of heifers; however, additional research is necessary.

Two experiments were performed to evaluate the effects of 1) increasing supplementation doses of a 50:50 (as-fed) liquid supplement of molasses and crude glycerol (M:G) on ruminal fermentation parameters and blood urea nitrogen (BUN) in beef steers consuming Tifton 85 bermudagrass ( spp.) hay and 2) different proportions of molasses and crude glycerol in a liquid supplement on in vitro fermentation and gas production kinetics. For Exp. 1, 8 ruminally cannulated, Angus-crossbred steers were used in a duplicated 4 × 4 Latin square design, had ad libitum access to Tifton 85 bermudagrass hay, and were randomly assigned to 1 of 4 treatments: 1) CTRL, no supplementation; 2) SUP1, 0.45 kg/d (as fed) of 50:50 M:G; 3) SUP3, 1.36 kg/d (as fed) of 50:50 M:G; and 4) SUP5, 2.27 kg/d (as fed) of a 50:50 M:G. For Exp. 2 in vitro batch cultures were conducted to test the same treatments from Exp. 1 and effects of different proportions of a M:G mixture (100:0, 75:25, 50:50, 25:75, and 0:100) when added to a hay substrate simulating the proportions of hay and liquid supplement used in SUP5. In Exp. 1, increasing doses of liquid supplement linearly decreased ( < 0.001) concentrations of NH-N, BUN, and acetate molar proportions, whereas propionate ( = 0.002) and butyrate ( < 0.001) molar proportions increased linearly. Treatment × time interactions were observed for ruminal pH ( < 0.001), where the greatest decrease was observed at 3 h postfeeding for animals consuming SUP5 (from 6.82 at 0 h to 6.32 at 3 h). In Exp. 2, decreases in acetate molar proportions ( < 0.001) and increases ( < 0.001) in propionate and butyrate molar proportions were also observed for either increasing doses of a 50:50 mixture or increasing proportions of glycerol in the mixture. Total VFA and in vitro organic matter digestibility were increased linearly ( < 0.001) as doses of a 50:50 mixture increased. Increasing doses of 50:50 M:G to growing beef heifers consuming bermudagrass hay caused a shift in VFA profile toward increases in propionate and decreases in acetate molar proportions. This was also confirmed in vitro, as the proportions of crude glycerol increased in a molasses:crude glycerol mix. Thus, molasses and crude glycerol combined seem to be useful to enhance performance in growing cattle consuming forage-based diets.

This study was designed to evaluate the effect of concentrations of P4 during a fixed-time artificial insemination (TAI) protocol and 48 hours temporary calf removal (CR; i.e., calves were temporally weaned for 48 hours) or treatment with 300 IU of eCG on ovulation time in suckled beef cows. One hundred and fourteen Nellore cows were estrous synchronized by receiving an intravaginal P4 releasing device containing 1.9 g of P4 (CIDR) plus 2.0-mg estradiol benzoate on Day 0, 12.5-mg dinoprost tromethamine on Day 7, and CIDR removal plus 0.5-mg estradiol cypionate on Day 9. Concentrations of P4 (P4 conc.) were manipulated by insertion of a new CIDR containing 1.9 g of P4 (CIDR1) or a CIDR previously used for 9 days (CIDR2), 18 days (CIDR3), or 27 days (CIDR4). On Day 9, within CIDR uses, cows received either CR or eCG. On Day 11 (48 hours after CIDR removal), all cows received a single TAI. Estrus was observed twice, a day between Day 9 and TAI. Ovulation by 60 hours or 72 hours after CIDR removal was defined as the disappearance of the largest follicle (LF) detected on Day 11 and presence of CL on Day 18, determined by transrectal ultrasonography. Cows ovulating after 72 hours were deemed to have a dominant follicle at 60 hours and 72 hours but also had a CL on Day 18. Serum P4 conc. were evaluated on Days 9, 11, and 18. Cows from CIDR4 + CR treatment ovulated at 60 hours (53.3%) tended to be greater (P = 0.07) than CIDR4 + eCG (21.4%). For the remaining treatments, this effect was not detected (CIDR1 + eCG: 6.3% vs. CIDR1 + CR: 0.0%; CIDR2 + eCG: 6.7% vs. CIDR2 + CR: 12.5%; CIDR3 + eCG: 0.0% vs. CIDR3 + CR: 25%). As a consequence, the percentage of cows from CIDR4 + CR treatment ovulating by 72 hours (26.7%) was lesser (P < 0.05) than that of cows in the CIDR4 + eCG treatment (78.6%), but for the remaining CIDR uses ovulation was not affected. The analysis of ovulating time revealed that earlier ovulation was associated (P < 0.01) to: (a) lesser P4 conc. on Day 9 (2.69b, 3.36a, and 3.82a ng/mL, standard error of the mean [SEM]: 0.12); (b) greater LF on Day 11 (12.46a, 12.09a, and 11.06b mm, SEM: 0.34); (c) greater estrus rate (94.1%a, 80.0%a, and 28.6%b), for ovulation at 60 hours, 72 hours, or >72 hours, respectively. Thereafter, previously used CIDR resulted (P < 0.01) in lesser P4 conc. on Day 9 (4.84a, 3.24b, 3.00b, and 2.50c ng/mL, SEM: 0.12), greater LF (10.50b, 12.07a, 11.98a, and 12.33a mm, SEM: 0.18) but only CIDR4 increased (P < 0.05) estrus rate (53.1%b, 65.6%b, 57.9%b, and 90.3%a), for CIDR1, CIDR2, CIDR3, and CIDR4, respectively. In conclusion, cows with low concentrations of P4 during a TAI protocol exhibited increased follicle diameter and an increased rate of estrus. This resulted in a greater percentage of cows ovulating by 60 hours when they had experienced calf removal.

The effects of administration of recombinant bovine ST (bST) on plasma hormone concentrations of cows, conceptus development, and postnatal calf performance were examined. Lactating beef cows ( = 190) were exposed to a fixed-time AI (TAI) protocol from d -10 to 0 (TAI on d 0). Cows were blocked by breed and stratified by days postpartum and then randomly assigned to receive, subcutaneously 1) 2 injections of saline (1 mL of 0.9% saline), 1 on d 0 at TAI and a second injection on d 14 (CTRL; = 53); 2) an injection of 325 mg of bST on d 0 and a saline injection on d 14 (bST0; = 48); 3) a saline injection on d 0 and an injection of 325 mg of bST on d 14 (bST14; = 49); or 4) 2 injections of 325 mg of bST, 1 on d 0 and a second injection on d 14 (bST0+14; = 40). Pregnancy status, crown-to-rump length (CRL) on Day 35, and crown-to-nose length (CNL) on Day 65 were determined via transrectal ultrasonography. Blood samples were collected on d 0, 7, 14, 21, 35, and 65, relative to TAI, to determine plasma concentrations of progesterone (P4), IGF-1, and pregnancy-specific protein B (PSPB) and also on d 18 and 21 for isolation of peripheral blood leukocytes for RNA extraction and measurement of interferon-stimulated genes transcript abundance. Individual calf BW was determined at birth and every 30 d until weaning. A subset of 24 calves was randomly selected for liver biopsies at birth to determine mRNA expression of target genes. Administration of bST to cows increased ( < 0.0001) concentrations of plasma IGF-1 for 14 d after injection compared with CTRL but did not affect fetal CRL and CNL ( = 0.23). Cows receiving bST only on d 0 had a greater ( = 0.05) transcript abundance in myxovirus resistance 2 on d 21 compared with 2bST cows (2.0- and 0.8-fold for bST0 and 2bST, respectively), whereas cows receiving bST14 and CTRL were intermediate (1.2- and 0.9-fold, respectively). Calf BW did not differ ( ≥ 0.100) among treatments on d 0, 30, 60, 90, 120, and 150 relative to birth. Injection of bST only on d 0 tended ( = 0.062) to increase calf liver mRNA expression of at birth compared with the calves born to cows in other treatments. Therefore, during a TAI protocol, the administration of 1 or 2 injections of 325 mg of bST to lactating beef cows enhanced their plasma concentrations of IGF-1 but failed to improve fetal size and plasma concentrations of maternal PSPB and P4 and had no effect on postnatal calf growth performance.

The role of progesterone in maintaining pregnancy is well known in the bovine. Subtle differences exist between dairy and beef cows because of differing concentrations of progesterone during recrudescence of postpartum estrous cycles, rate of follicular growth and maturation, proportions of 2- and 3-follicular wave cycles, and other effects on pregnancy outcomes per artificial insemination (P/AI). Because proportions of anovulatory cows before the onset of the artificial insemination (AI) period are greater and more variable in beef (usually ranging from 30 to 70%) than dairy (25%) cows, AI programs were developed to accommodate anovulatory and cycling beef cows enrolled therein. Incorporating a progestin as part of an AI program in beef cows improved P/AI by reducing the proportion of cows having premature luteal regression and short post-AI luteal phases. In both genotypes, prolonged dominant follicle growth in a reduced progesterone milieu resulted in increased (1) LH pulses, (2) preovulatory follicle diameter, and (3) concentrations of estradiol and a subsequently larger corpora lutea (CL). In contrast, the progesterone milieu during growth of the ovulatory follicle in an ovulation control program does not seem to affect subsequent P/AI in beef cows, whereas in dairy cows follicle development in an elevated compared with a low progesterone environment increases P/AI. Progesterone status in beef cows at the onset of ovulation synchronization is not related to P/AI in multiparous cows, whereas P/AI was suppressed in primiparous cows that began a timed AI program in a low-progesterone environment. In timed AI programs, elevated concentrations of progesterone just before PGF2α and reduced concentrations at AI are critical to maximizing subsequent P/AI in dairy cows, but seemingly much less important in beef cows. By inducing ancillary CL and increasing concentrations of progesterone, human chorionic gonadotropin may increase P/AI when administered to beef cows 7d after AI or at embryo transfer, and its success seems to depend on induction of ancillary CL, whereas in dairy cows increased fertility was detected in cows with multiple CL, human chorionic gonadotropin-enhanced progesterone from original CL, or both. Pregnancy losses after AI are less frequent in beef cows and are not associated with pre-AI progesterone or cycling status, whereas losses in dairy cows are inversely related to progesterone and adversely affected in anovular dairy cows. Genotype and nutritional management likely influence several physiological differences including circulating concentrations of progesterone and responses to supplemental progesterone.

Two experiments were conducted to determine the effect of administering PGF at the initiation of the 7-d CO-Synch+controlled internal drug release (CIDR) fixed-timed AI (TAI) protocol on pregnancy rates of suckled beef cows and replacement heifers. Within location, cows were stratified by days postpartum (DPP), BCS, and parity (Exp. 1; = 1,551) and heifers were stratified by BCS (Exp. 2; = 999) and randomly assigned to 1 of 2 treatments: 1) CO-Synch+CIDR (100-µg injection of GnRH at CIDR insertion [d -10] with a 25-mg injection of PGF at CIDR removal [d -3] followed by injection of GnRH and TAI on d 0) or 2) PG-CO-Synch+CIDR (a 25-mg injection of PGF on d -10 of the CO-Synch+CIDR protocol). Follicle diameter and corpus luteum (CL) development were assessed on d -10 and -3, and pregnancy status was determined on d 30 to 35. Blood was collected on d -20, -10, -3, and 0 relative to TAI to determine concentrations of progesterone (P4). In Exp. 1, TAI pregnancy rates did not differ ( = 0.667) between treatments and were affected by BCS ( = 0.003) and DPP ( = 0.006). Concentrations of P4 were greater ( < 0.0001) on d -3 for CO-Synch+CIDR than for PG-CO-Synch+CIDR (4.1 ± 0.2 and 3.4 ± 0.2 ng/mL, respectively). Follicle diameter on d -3 differed ( = 0.05) between PG-CO-Synch+CIDR (13.4 ± 0.3 mm) and CO-Synch+CIDR (12.5 ± 0.3 mm) treatments. Cows with P4 > 2.5 ng/mL on d -10 had greater ( = 0.024) pregnancy rate to TAI (56.5%) compared with cows with 2.5 ng/mL < P4 > 1 (43.0%), whereas cows with P4 < 1 ng/mL were intermediate (51.6%). Cows with a CL on d -10 had greater ( = 0.012) pregnancy rates to TAI than cows without a CL (66.3 vs. 39.4%, respectively). In Exp. 2, TAI pregnancy rates did not differ ( = 0.316) between treatments. Concentrations of P4 differed ( < 0.0001) on d -3 with greater concentrations of P4 for CO-Synch+CIDR than for PG-CO-Synch+CIDR (3.75 ± 0.20 ng/mL and 3.60 ± 0.21 ng/mL, respectively). Follicle diameter was similar ( = 0.749) between treatments on d -10 and -3. Regardless of treatment, cyclic status tended ( = 0.062) to improve pregnancy rates to TAI (55 vs. 45%, for cycling and noncycling heifers, respectively). We concluded that addition of PGF to the 7-d CO-Synch+CIDR protocol decreased concentrations of P4 in cows and heifers and increased follicle diameter at CIDR removal in cows but failed to increase TAI pregnancy rates.

Chitosan was evaluated as a feed additive to mitigate in vivo CH4 emissions in beef cattle. Twenty-four crossbred heifers (BW = 318 ± 35 kg) were used in a randomized block design replicated in 2 periods. The design included a 2 × 3 factorial arrangement of treatments, which included diet (high concentrate [HC] or low concentrate [LC]) and 0.0, 0.5, or 1.0% of chitosan inclusion (DM basis). Diets were offered ad libitum and individual intake was recorded. An in vitro experiment to analyze chitosan's effect on fermentation parameters and gas production kinetics was performed. A diet effect (P < 0.01) was observed for CH4 emissions expressed as grams/day, grams/kilogram of BW0.75, and grams/kilogram of DMI. Heifers consuming the LC diet produced 130 g of CH4/d vs. 45 g of CH4/d in those consuming the HC diet. Incubation fluid pH increased linearly (P < 0.05) when chitosan was included in HC substrates. In vitro CH4 production was not affected (P > 0.10) by chitosan in HC substrate; however, when incubated with the LC substrate, CH4 production increased quadratically (P < 0.01) as chitosan inclusion increased. A digestibility marker × diet interaction occurred (P < 0.05) for DM, OM, CP, NDF, and ADF digestibility. Diet × chitosan interactions (P < 0.05) occurred for DM, OM, NDF, and ADF digestibility when Cr2O3 was used. When TiO2 was used, diet × chitosan interactions (P < 0.05) were observed for NDF and ADF. However, using indigestible NDF as an internal marker, DM and OM digestibility were improved (P < 0.05) by 21 and 19%, respectively, when chitosan was included in LC diets. In conclusion, feeding up to 1% of chitosan (DM basis) to heifers consuming a LC diet increased apparent total tract digestibility of nutrients. Enteric CH4 emissions were not affected by chitosan feeding, regardless of type of diet, and heifers consuming a 36% concentrate diet produced 2.6 times more methane per day than those consuming an 85% concentrate diet.

Our objective was to assess the effects of progesterone before initiating an estrus- or ovulation-synchronization program in addition to the influence of parity, BCS, and days postpartum on resulting pregnancy rates per AI. Experimental data were combined from 73 herd-year studies consisting of more than 8,500 suckled beef cows exposed to variants of the CO-Synch program. Blood was harvested from samples collected at 10 and 0 d before the onset of CO-Synch, and progesterone concentrations of the samples were determined. The progesterone environment preceding synchronization was assessed in 3 ways on the basis of progesterone concentrations measured in the 2 defined blood samples. All binomial logistic regression models used procedure GLIMMIX in SAS and included the fixed effects of program duration, inclusion of progesterone via an intravaginal insert, parity, days postpartum at AI, BCS, and appropriate interactions. In addition, model 1 included 3 categories of progesterone concentrations (low [<1 ng/mL], medium [1.00 to 3.99 ng/mL], and high [≥4.00 ng/mL] concentrations) at 10 and 0 d before synchronization and their interaction. Model 2 included 4 categories defining the stage of the estrous cycle (late diestrus, early diestrus, and proestrus-estrus-metestrus) or anestrus, at which cows started the synchronization program. Model 3 defined cows as cycling or noncycling at the onset of the program. Significant effects of progesterone supplementation, which hormone was used to initiate the timed AI program, parity, BCS, days postpartum, and progesterone status assessed in 3 ways were consistent in nearly all models. Progesterone status at the onset of synchronization was not important to pregnancy outcomes in multiparous cows, whereas pregnancy rate per AI was suppressed in primiparous cows that began in a low-progesterone environment (proestrus, estrus, metestrus, or anestrus). A significant 3-way interaction of parity, BCS, and days postpartum in 2 models reinforced the importance of these factors to AI pregnancy outcomes. Ancillary analyses identified the significant effects of cycling status and BCS as well as days postpartum on luteolytic response to PGF(2α). Pregnancy loss of 2.7% to 4.2% was detected to occur between a positive pregnancy diagnosis at 35 d post-AI and later stages of pregnancy. We concluded that progesterone status at the onset of the synchronization program is critical to pregnancy outcomes in primiparous but not multiparous cows.

Two experiments were performed to evaluate the effects of different levels of supplementation with a 50:50 (as-fed) mixture of molasses:crude glycerol on animal performance, total tract digestibility of nutrients, and ruminal in situ degradability of nutrients in beef heifers and steers consuming Tifton 85 Bermuda grass (Cynodon spp.) hay. For Exp. 1, 24 Angus crossbred heifers (380 ± 31 kg BW) were used in a generalized randomized block design. For Exp. 2, 8 ruminally cannulated Angus crossbred steers (323 ± 42 kg BW) were used in a 4 × 4 duplicated Latin square design. For both experiments, animals were housed in individual pens at the University of Florida Feed Efficiency Facility, had ad libitum access to Tifton 85 Bermuda grass hay, and were randomly assigned to 1 of 4 treatments: 1) CTRL, no supplementation; 2) SUP1, 0.45 kg/d (as fed) of 50:50 mixture; 3) SUP3, 1.36 kg/d (as fed) of 50:50 mixture; and 4) SUP5, 2.27 kg/d (as fed) of a 50:50 mixture. Individual feed intake was recorded. Total DMI increased linearly (P = 0.005) as the level of supplementation increased. Hay intake ranged from 1.36 (CTRL) to 1.23% (SUP5) of BW, and was not affected (P ≥ 0.10) by liquid supplementation. Final BW was not affected by liquid supplementation ( ≥ 0.10). There was a linear increase (P = 0.027) in ADG as the liquid supplementation amounts increased. Liquid supplementation did not affect G:F (P ≥ 0.10). Apparent total tract digestibility of DM, OM, NDF, and ADF increased linearly (P < 0.001), while CP total tract digestibility decreased linearly (P = 0.002) as the level of supplementation increased. Ruminal pH was decreased linearly (P = 0.012) as the level of supplementation increased. No effect (P ≥ 0.10) of liquid supplementation was detected on lag time for NDF and ADF content of bermudagrass hay; however, rate of degradation (Kd) of NDF tended (P = 0.076) to be affected cubically by liquid supplementation. In addition, liquid supplementation linearly decreased (P < 0.05) ED of OM, CP, NDF, and ADF. In conclusion, supplementing up to 2.27 kg/d of a 50:50 mixture of molasses:crude glycerol may stimulate microbial growth and fermentative activity, thereby increasing nutrient digestibility. Increased fiber digestion, along with energy supplementation, led to increased ADG in heifers consuming Bermuda grass hay.
