During routine cadaveric dissection, accessory hypothenar muscles were incidentally discovered in two cadavers, both males, aged 86 and 92. Both muscles originated from the palmaris longus tendon in the distal portion of the forearm and were identified as accessory abductor digiti minimi (AADM) muscles, based on their association with abductor digiti minimi. While AADM is a common variant in the antebrachium, it is less typical for them to originate from the palmaris longus tendon. The presence of such an AADM could complicate surgical procedures requiring resection of the palmaris longus tendon. Moreover, the surrounding neurovasculature - namely the ulnar nerve as it passes through the ulnar canal between the pisiform and hook of the hamate - could be compressed by contractions of an AADM with such a proximal origin. This can manifest as ulnar neuropathies resulting in pain, weakness, or protracted flexion of the fourth and fifth digits (ulnar claw). Our description of these muscles adds to previous accounts of variation of the palmaris longus and abductor digiti minimi muscles while considering potential clinical implications.

The blacklegged tick, Ixodes scapularis Say, is the primary vector of several tick-borne pathogens, including those causing Lyme disease and babesiosis, in the eastern United States and active collection methods for this species include dragging or wild animal sampling. Nest boxes targeting mice may be an alternative strategy for the surveillance and collection of immature I. scapularis feeding on these hosts and would be much safer for animals compared to small mammal trapping. We constructed double-walled insulated nest boxes (DWINs) with collection tubes mounted below the nesting chamber and deployed eleven in southern Wisconsin from June until September of 2020. The DWINs were occupied by Peromyscus spp. and birds (wren species, Troglodytidae family). We collected 192 ticks from collection tubes, all of which were identified as either I. scapularis (95%) or Dermacentor variabilis Say (Acari: Ixodidae) (5%). Only 12% (21/182) and 20% (2/10) of I. scapularis and D. variabilis were blood-fed, respectively. The high proportion of unfed ticks found in collection tubes may be due to grooming by hosts inside the nest boxes. Alternatively, immature ticks may have climbed trees and entered the DWIN seeking a host. Results suggest that nest boxes could be a tool for finding ticks in areas of low density or at the leading edge of invasion, when small mammal trapping or drag sampling is not feasible.

The therapeutic landscape of chronic myeloid leukemia (CML) has profoundly changed over the past 7 years. Most patients with chronic phase (CP) now have a normal life expectancy. Another goal is achieving a stable deep molecular response (DMR) and discontinuing medication for treatment-free remission (TFR). The European LeukemiaNet convened an expert panel to critically evaluate and update the evidence to achieve these goals since its previous recommendations. First-line treatment is a tyrosine kinase inhibitor (TKI; imatinib brand or generic, dasatinib, nilotinib, and bosutinib are available first-line). Generic imatinib is the cost-effective initial treatment in CP. Various contraindications and side-effects of all TKIs should be considered. Patient risk status at diagnosis should be assessed with the new EUTOS long-term survival (ELTS)-score. Monitoring of response should be done by quantitative polymerase chain reaction whenever possible. A change of treatment is recommended when intolerance cannot be ameliorated or when molecular milestones are not reached. Greater than 10% BCR-ABL1 at 3 months indicates treatment failure when confirmed. Allogeneic transplantation continues to be a therapeutic option particularly for advanced phase CML. TKI treatment should be withheld during pregnancy. Treatment discontinuation may be considered in patients with durable DMR with the goal of achieving TFR.

Horizontal silage bunkers produce leachate that contains contaminants that can be detrimental to the environment if released untreated. Vegetated filter strips are used to treat silage bunker runoff to prevent contamination of surface waters via infiltration, however increased infiltration poses risks to groundwater, particularly for nitrate (NO3-). Vegetated filter strip plots with a sandy loam soil, half of which are amended with biochar, were investigated to assess the treatment of silage bunker runoff over 20 application events. The subsurface effluent biological oxygen demand (BOD5), chemical oxygen demand (COD), and total phosphorus (TP) were reduced on average by 40%, 46%, and 75%, respectively, and there was no statistical difference between treatments. The total nitrogen (TN) was reduced by 49 and 64% for control and biochar plots, respectively, which was significantly different between treatments. Biochar significantly reduced nitrate nitrogen (NO3--N) leaching by 40% compared to the control, however, the NO3--N concentration in leachate was still high ranging from 0.19 to 191.04 mg NO3--N L-1 and 0.18-108.89 mg NO3--N L-1 for control and biochar plots, respectively. A mass balance suggests the primary mechanism for a decrease in TN and NO3--N leaching from biochar amended plots was greater retention of NO3--N and organic N (ORG-N) within the soil/biochar matrix. The development of oxygenated functional groups and/or formation of organomineral layer on the biochar surface likely enhanced N retention.

Frameworks for limiting ecosystem exposure to excess nutrients and acidity require accurate and complete deposition budgets of reactive nitrogen (Nr). While much progress has been made in developing total Nr deposition budgets for the U.S., current budgets remain limited by key data and knowledge gaps. Analysis of National Atmospheric Deposition Program Total Deposition (NADP/TDep) data illustrates several aspects of current Nr deposition that motivate additional research. Averaged across the continental U.S., dry deposition contributes slightly more (55%) to total deposition than wet deposition and is the dominant process (>90%) over broad areas of the Southwest and other arid regions of the West. Lack of dry deposition measurements imposes a reliance on models, resulting in a much higher degree of uncertainty relative to wet deposition which is routinely measured. As nitrogen oxide (NOx) emissions continue to decline, reduced forms of inorganic nitrogen (NHx = NH3 + NH4+) now contribute >50% of total Nr deposition over large areas of the U.S. Expanded monitoring and additional process-level research are needed to better understand NHx deposition, its contribution to total Nr deposition budgets, and the processes by which reduced N deposits to ecosystems. Urban and suburban areas are hotspots where routine monitoring of oxidized and reduced Nr deposition is needed. Finally, deposition budgets have incomplete information about the speciation of atmospheric nitrogen; monitoring networks do not capture important forms of Nr such as organic nitrogen. Building on these themes, we detail the state of the science of Nr deposition budgets in the U.S. and highlight research priorities to improve deposition budgets in terms of monitoring and flux measurements, leaf- to regional-scale modeling, source apportionment, and characterization of deposition trends and patterns.

Biochar amendments can reduce nitrate (NO3) leaching in agricultural soil. It has been hypothesized that functional groups on the biochar surface from oxidation can increase NO3 sorption. This study evaluates the effect of chemical oxidation of biochar on NO3 sorption characteristics. Eight biochars, made from wood and corn cobs, underwent sodium hypochlorite (NaClO) and hydrogen peroxide (H2O2) oxidation and then assessed for NO3 sorption capacity using batch isotherm methods. The unoxidized and oxidized biochar produced at low temperatures (400 °C) had no significant NO3 sorption. Oxidized biochars produced at higher temperatures (600 °C and 700 °C) had calculated maximum NO3 sorption capacities (Smax) ranging from 0.50 to 3.97 mg NO3-N g-1. Biochar oxidations with 50 mmol NaClO g-1 (N50) in combination with an acid wash (AW) had the largest estimated sorption capacities of 3.68, 3.97, and 1.46 mg NO3-N g-1 for CTN50,AW, BW3N50,AW, and CC3N50,AW, respectively. Sorption capacity of wood-based biochars was higher than corn cob biochars due to increased oxidation as measured by total acid group content (TAGC). Wood biochar Smax values were correlated with ΔTAGC (R2 = 0.86), with a slope of 1.2 µmol NO3-N µmol TAGC-1 suggesting that cationic bridging of NO3 to oxidized sites is the primary mechanism for NO3 sorption.

We conducted a systematic review and network meta-analysis to determine the comparative efficacy of antibiotics used to control bovine respiratory disease (BRD) in beef cattle on feedlots. The information sources for the review were: MEDLINE®, MEDLINE In-Process and MEDLINE® Daily, AGRICOLA, Epub Ahead of Print, Cambridge Agricultural and Biological Index, Science Citation Index, Conference Proceedings Citation Index - Science, the Proceedings of the American Association of Bovine Practitioners, World Buiatrics Conference, and the United States Food and Drug Administration Freedom of Information New Animal Drug Applications summaries. The eligible population was weaned beef cattle raised in intensive systems. The interventions of interest were injectable antibiotics used at the time the cattle arrived at the feedlot. The outcome of interest was the diagnosis of BRD within 45 days of arrival at the feedlot. The network meta-analysis included data from 46 studies and 167 study arms identified in the review. The results suggest that macrolides are the most effective antibiotics for the reduction of BRD incidence. Injectable oxytetracycline effectively controlled BRD compared with no antibiotics; however, it was less effective than macrolide treatment. Because oxytetracycline is already commonly used to prevent, control, and treat BRD in groups of feedlot cattle, the use of injectable oxytetracycline for BRD control might have advantages from an antibiotic stewardship perspective.

Vaccination against putative causal organisms is a frequently used and preferred approach to controlling bovine respiratory disease complex (BRD) because it reduces the need for antibiotic use. Because approximately 90% of feedlots use and 90% of beef cattle receive vaccines in the USA, information about their comparative efficacy would be useful for selecting a vaccine. We conducted a systematic review and network meta-analysis of studies assessing the comparative efficacy of vaccines to control BRD when administered to beef cattle at or near their arrival at the feedlot. We searched MEDLINE, MEDLINE In-Process, MEDLINE Daily Epub Ahead of Print, AGRICOLA, Cambridge Agricultural and Biological Index, Science Citation Index, and Conference Proceedings Citation Index - Science and hand-searched the conference proceedings of the American Association of Bovine Practitioners and World Buiatrics Congress. We found 53 studies that reported BRD morbidity within 45 days of feedlot arrival. The largest connected network of studies, which involved 17 vaccine protocols from 14 studies, was included in the meta-analysis. Consistent with previous reviews, we found little compelling evidence that vaccines used at or near arrival at the feedlot reduce the incidence of BRD diagnosis.

Big data are frequently used in many facets of business and agronomy to enhance knowledge needed to improve operational decisions. Livestock operations collect data of sufficient quantity to perform predictive analytics. Predictive analytics can be defined as a methodology and suite of data evaluation techniques to generate a prediction for specific target outcomes. The objective of this manuscript is to describe the process of using big data and the predictive analytic framework to create tools to drive decisions in livestock production, health, and welfare. The predictive analytic process involves selecting a target variable, managing the data, partitioning the data, then creating algorithms, refining algorithms, and finally comparing accuracy of the created classifiers. The partitioning of the datasets allows model building and refining to occur prior to testing the predictive accuracy of the model with naive data to evaluate overall accuracy. Many different classification algorithms are available for predictive use and testing multiple algorithms can lead to optimal results. Application of a systematic process for predictive analytics using data that is currently collected or that could be collected on livestock operations will facilitate precision animal management through enhanced livestock operational decisions.

The primary aim of the present study was to determine the impact of acute changes in shear rate patterns, in particular retrograde shear rate, on microvascular function in 15 healthy, young men and women as determined via the post-occlusive near-infrared spectroscopy (NIRS) microvascular reactivity response. Microvascular reactivity, via NIRS-derived measurements of post-occlusion tissue saturation index (TSI%) and total microvascular hemoglobin+myoglobin concentration ([Hb]total), were assessed in each participant before and immediately after exposure to a 30min retrograde shear treatment. Retrograde shear was achieved via a blood pressure cuff placed below the knee inflated to 75mmHg. One leg was exposed to the retrograde shear (Treatment leg) and the contralateral leg served as a non-treatment control. In the Treatment leg, significant increases in retrograde shear rate occurred during the retrograde intervention. Following the intervention, the area under the TSI% post-occlusion response curve, which represents the total microvascular reactivity response, and the absolute peak TSI% response were significantly increased compared to pre-intervention in the Treatment leg, but not the Control leg. The absolute peak [Hb]total response was significantly increased post-intervention in both legs. These results are in contrast to our hypothesis that 75mmHg cuff inflation, designed to increase retrograde shear rate in the femoral artery would negatively affect post-occlusive microvascular reactivity. These data suggest that the current method of increasing retrograde shear rate in the intact human does not adversely impact NIRS derived measurements of microvascular reactivity.

Some cattle production experts believe that cow-calf producers should breed replacement heifers (nulliparous cows) before cows (primiparous and multiparous cows), sometimes referred to as providing a heifer lead time (tHL). Our objective was to model the effects different durations of tHL may have on measures of herd productivity, including the percent of the herd cycling before the end of the first 21 d of the breeding season (%C21), the percent of the herd pregnant at pregnancy diagnosis (%PPD), the distribution of pregnancy by 21-d breeding intervals, the kilograms of calf weaned per cow exposed (KPC), and the replacement percentage (%RH), using a deterministic, dynamic systems model of cow-calf production over a 10-yr horizon. We also wished to examine differences in the effect of tHL related to the primiparous duration of postpartum anestrus (dPPA). The study model examined 6 different dPPA for primiparous cows (60, 70, 80, 90, 100, or 110 d). The multiparous cow duration of postpartum anestrus was set to 60 d. The breeding season length for nulliparous cows was 63 d, as was the breeding season length for primiparous and multiparous cows. Nulliparous cows were modeled with a tHL of 0, 7, 14, 21, 28, 35, or 42 d. Results are reported for the final breeding season of the 10-yr horizon. Increasing tHL resulted in a greater %C21 for the herd and for primiparous cows. Length of tHL had minimal impact on the %PPD unless the dPPA was 80 d or greater. For a dPPA of 110 d, a 0 d tHL resulted in the herd having 88.1 %PPD. When tHL was 21 d, the %PPD increased to 93.0%. The KPC was 161.2 kg when the dPPA was 110 d and tHL was 0 d and improved to 183.2 kg when tHL was increased to 42 d. The %RH did not vary much unless the dPPA was 90 d or greater, but increasing tHL resulted in decreased %RH. Based on the model results, increasing tHL improves the production outcomes included in the analysis, but herds with dPPA of 90 d or greater had the greatest degree of improvement. For these herds, approximately two-thirds of the improvement in outcomes by increasing tHL from 0 d to 42 d was realized when tHL was 21 d. Costs are likely incurred when implementing tHL in a breeding management program, and an ideal tHL likely depends on the dPPA of the herd, the expected improvement in productivity, and the costs associated with increasing tHL. Determining the dPPA of a herd could help veterinarians and producers develop optimal herd management strategies.

Dairy products are major components of the human diet but are also important contributors to global environmental impacts. This study evaluated greenhouse gas (GHG) emissions, net energy intensity (NEI), and land use of confined dairy systems with increasing levels of pasture in the diet. A Wisconsin farm was modeled to represent practices adopted by dairy operations in a humid continental climate typical in the Great Lakes region and other climates that have large differences in seasonal temperatures. Five grazing scenarios (all of which contained some portion of confinement) were modeled based on different concentrations of dry matter intake from pasture and feed supplementation from corn grain, corn silage, and soybean meal. Scenarios that incorporate grazing consisted of 5 mo of pasture feeding from May to September and 7 mo of confined feeding from October to April. Environmental impacts were compared within the 5 scenarios that incorporate grazing and across 2 entirely confined scenarios with and without on-farm electricity production through anaerobic digestion (AD). To conduct a fair comparison, all scenarios were evaluated based on the same total amount of milk produced per day where resource inputs were adjusted according to the characteristics of each scenario. A cradle-to-farm gate life cycle assessment evaluated the environmental burdens that were partitioned by allocation between milk and meat and by system expansion when biogas-based electricity was produced. Overall, results for all scenarios were comparable. Enteric methane was the greatest contributor to GHG emissions, and the production of crops was the most energy-intense process. For the confined scenario without AD, GHG emissions were 0.87 kg of CO2 equivalents, NEI was 1.59 MJ, and land use was 1.59 m2/kg of fat- and protein-corrected milk (FPCM). Anaerobic digestion significantly reduced emissions to 0.28 kg of CO2 equivalents/kg of FPCM and reduced NEI to -1.26 MJ/kg of FPCM, indicating a net energy producing system and highlighting the potential of AD to improve the sustainability of confined systems. For scenarios that combined confinement and grazing, GHG emissions ranged from 0.84 to 0.92 kg of CO2 equivalents, NEI ranged from 1.42 to 1.59 MJ, and land use ranged from 1.19 to 1.26 m2/kg of FPCM. All environmental impacts were minimized in scenarios that supplemented enough feed to increase milk yield but maintained dry matter intake from pasture at a level high enough to reduce material and energy use.

Following the Deepwater Horizon (DWH) event in 2010, there were several lines of evidence indicating the presence of marine oil snow sedimentation and flocculent accumulation (MOSSFA). A significant amount of marine oil snow formed in the water column of the northern Gulf of Mexico (nGoM), settled rapidly, and ultimately accumulated in the sediments of the nGoM. This study utilized a commonly used radioisotope tracer (excess 210Pb, 210Pbxs) from 32 sediment cores collected from 2010 to 2013 to characterize the spatial extent of MOSSFA on the seafloor. Relative to pre-DWH conditions, an increase in 210Pbxs flux occurred in two distinct regions: (1) in the western portion of the study area on an east-northeast to west-southwest axis, stretching 230 km southwest and 140 km northeast of the DWH wellhead, and (2) in the eastern portion of the study area on a 70 km northeast to southwest axis near the DeSoto Canyon. The total sedimentary spatial extent of MOSSFA, as calculated by increased 210Pbxs flux after 2010, ranged from 12 805 to 35 425 km2. 210Pbxs flux provides a valuable tool for documenting the spatial extent of MOSSFA following DWH and will continue to aid in the determination of advective transport and ultimate depocenters of MOSSFA material.

The objective of this project was to identify risk factors associated with the probability of failing to resolve clinical signs of bovine respiratory disease (BRD) after initial antimicrobial treatment. A retrospective data analysis was performed with 194,062 animals treated for BRD with an antimicrobial from 10 U.S. feedlots from 2005 to 2009. A binary variable classified cattle as a treatment failure if they were retreated for BRD or died following an initial BRD treatment. A generalized logistic mixed model was created to evaluate associations of quarter of the year at feedlot arrival, arrival weight, sex, risk classification (high vs. low), days on feed at first BRD treatment, and rectal temperature at first BRD treatment and all 2-way interactions with the probability of first treatment failure for BRD. A total of 64,683 out of 194,062 (33.3%) of cattle initially treated for BRD were classified as treatment failures due to death or retreatment. Multiple 2-way interactions were present. High-risk calves had greater probability of first treatment failure compared with low-risk calves, but this effect was modified by quarter of arrival, days on feed at first treatment, and rectal temperature category. The effect of arrival weight category was modified by days on feed at first treatment of BRD and quarter of arrival for probability of first treatment failure. Multiple arrival and treatment characteristics were associated with the probability of first BRD treatment failure. Knowledge of these factors can lead future prospective studies to improve the prediction of treatment failure and provide baseline results for the industry.

The duration of postpartum anestrus (dPPA) is important to consider for reproductive performance and efficiency in cow-calf operations. We developed a deterministic, dynamic systems model of cow-calf production over a 10-yr horizon to model the effects that dPPA had on measures of herd productivity, including the percentage of cows cycling before the end of the first 21 d of the breeding season (%C21), the percentage of cows pregnant at pregnancy diagnosis (%PPD), the distribution of pregnancy by 21-d breeding intervals, the kilograms of calf weaned (KW), the kilograms of calf weaned per cow exposed (KPC), and the replacement percentage. A 1,000-animal herd was modeled, with the beginning and ending dates for a 63-d natural service breeding season being the same for eligible replacement heifers (nulliparous cows) and cows (primiparous and multiparous cows). Herds were simulated to have a multiparous cow dPPA of 50, 60, 70, or 80 d, with the dPPA for primiparous cows being set to 50, 60, 70, 80, 90, 100, or 110 d. Only combinations where the primiparous dPPA was greater than or equal to the multiparous dPPA were included, resulting in 22 model herds being simulated in the analysis. All other model parameters were held constant between simulations. In model season 10, the %C21 was 96.2% when the multiparous cow and primiparous cow dPPA was 50 d and was 48.3% when the multiparous cow and primiparous cow dPPA was 80 d. The %PPD in model season 10 for these same herds was 95.1% and 86.0%, respectively. The percentage of the herd becoming pregnant in the first 21 d of the breeding season also differed between these herds (61.8% and 31.3%, respectively). The 10-yr total KW was more than 275,000 kg greater for the herd with a 50-d multiparous cow and primiparous cow dPPA when compared with the herd with the 80-d multiparous and primiparous cow dPPA and had a model season 10 KPC of 180.8 kg compared with 151.4 kg for the longer dPPA. The model results show that both the multiparous cow and primiparous cow dPPA affect herd productivity outcomes and that a dPPA less than 60 d results in improved production outcomes relative to longer dPPA. Veterinarians and producers should consider determining the dPPA to aid in making management decisions to improve reproductive performance of cow-calf herds.

The objective of this project was to evaluate the effects of antimicrobials approved for parenteral metaphylactic use in feeder and stocker calves on morbidity and mortality for bovine respiratory disease with the use of a mixed treatment comparison meta-analysis. An initial literature review was conducted in April 2016 through Pubmed, Agricola, and CAB (Commonwealth Agricultural Bureau) for randomized controlled trials for metaphylaxis antimicrobial administered parentally to incoming feedlot or stocker calves within 48 h of arrival. The final list of publications included 29 studies, with a total of 37 trials. There were 8 different metaphylactic antimicrobials. Final event outcomes were categorized into bovine respiratory disease (BRD) morbidity cumulative incidence d 1 to ≤ 60 of the feeding period, BRD morbidity cumulative incidence d 1 to closeout of the feeding period, BRD mortality cumulative incidence d 1 to closeout of the feeding period, and BRD retreatment cumulative incidence morbidity d 1 to closeout of the feeding period. Network meta-analysis combined direct and indirect evidence for all the event outcomes to determine mean odds ratio (OR) with 95% credibility intervals (CrIs) for all metaphylactic antimicrobial comparisons. The "upper tier" treatment arms for morbidity d 1 to ≤ 60 included tulathromycin, gamithromycin, and tilmicosin. For BRD mortality cumulative incidence d 1 to closeout and BRD retreatment morbidity d 1 to closeout, classifying the treatment arms into tiers was not possible due to overlapping 95% CrIs. The results of this project accurately identified differences between metaphylactic antimicrobials, and metaphylactic antimicrobial options appear to offer different outcomes on BRD morbidity and mortality odds in feedlot cattle.

Experts have suggested that microscale biogas systems offer a source of renewable energy that improves indoor air quality, but such impacts have not been directly measured. This study documented cooking behaviors and measured 2.5-µm particulate matter (PM2.5 ), carbon monoxide (CO), and sulfur dioxide (SO2 ) concentrations within 14 institutional kitchens in Kampala, Uganda, that prepare meals using biogas (n=5), a mixture of biogas and fuelwood (n=3), and fuelwood (n=6). Small institutions (10-30 people) with biogas kitchens had 99% lower concentrations of PM2.5 (21 µg/m3 ) than fuelwood kitchens (3100 µg/m3 ). Larger institutions (>100 people) had biogas systems that produced insufficient gas and relied on fuelwood to meet over 90% of their energy needs. PM2.5 concentrations in these biogas-firewood kitchens were equivalent to concentrations in fuelwood kitchens. Although concentrations of hydrogen sulfide (H2 S) in biogas were as high as 2000 ppm, 75% of systems had undetectable H2 S levels (<100 ppm) in the biogas. Kitchens using biogas with high H2 S had correspondingly higher SO2 concentrations in the kitchen air. However, even the highest SO2 concentration in biogas kitchens (150 µg/m3 ) was lower than SO2 concentration in fuelwood kitchens (390 µg/m3 ). The results suggest that biogas systems can offer air quality improvements if sized properly for energy demands.

Myosin Va (MyoVa) is an actin-based molecular motor abundantly found at the centrosome. However, the role of MyoVa at this organelle has been elusive due to the lack of evidence on interacting partners or functional data. Herein, we combined yeast two-hybrid screen, biochemical studies and cellular assays to demonstrate that MyoVa interacts with RPGRIP1L, a cilia-centrosomal protein that controls ciliary signaling and positioning. MyoVa binds to the C2 domains of RPGRIP1L via residues located near or in the Rab11a-binding site, a conserved site in the globular tail domain (GTD) from class V myosins. According to proximity ligation assays, MyoVa and RPGRIP1L can interact near the cilium base in ciliated RPE cells. Furthermore, we showed that RPE cells expressing dominant-negative constructs of MyoVa are mostly unciliated, providing the first experimental evidence about a possible link between this molecular motor and cilia-related processes.

Asparaginase, which depletes asparagine and glutamine, activates amino-acid stress response. Oxidative stress mediated by excessive reactive oxygen species (ROS) causes enhanced mitochondrial permeabilization and subsequent cell apoptosis and is considered as a plausible mechanism for drug-induced hepatotoxicity, a common toxicity of asparaginase in adults with acute lymphoblastic leukemia (ALL). Studies investigating the pharmacogenetics of asparaginase in ALL are limited and focused on asparaginase-induced allergic reaction common in pediatric patients. Here, we sought to determine a potential association between the variant rs4880 in SOD2 gene, a key mitochondrial enzyme that protects cells against ROS, and hepatotoxicity during asparaginase-based therapy in 224 patients enrolled on CALGB-10102, a treatment trial for adults with ALL. We report that the CC genotype of rs4880 is associated with increased hepatotoxicity following asparaginase-based treatment. Thus, rs4880 likely contributes to asparaginase-induced hepatotoxicity, and functional studies investigating this single-nucleotide polymorphism (SNP) are needed to develop therapeutic approaches that mitigate this toxicity.
