RESUMO

Spontaneous liver steatosis can be experimentally induced in domestic Greylag geese by combining a short photoperiod with a sequence of feed restriction followed by ad libitum corn feeding. This could offer an alternative to the conventional "foie gras" production system based on overfeeding. The present work aimed at comparing the compositional characteristics, sensory profile, and acceptability by a consumer panel of fatty livers obtained by overfeeding and spontaneous fattening. In all, 210 male geese were used: 125 geese were raised over a 31-wk period to produce fatty liver without overfeeding ("alternative livers") and 85 were raised using conventional methods with overfeeding ("conventional livers"). Mean liver weight was over 1 kg (1,102 g) in the conventional group and 445 g in the alternative group. The characteristics of the livers were studied in 2 subpopulations: 44 conventional livers representative of the experimental population (mean liver weight 1,064 g) and 42 alternative livers weighing more than 400 g (mean 702 g). Compared with the alternative livers, livers from the conventional group showed significantly (P < 0.05) higher dry matter and lipid contents, lighter color (L*), and lower yellowness (b*). The neutral lipids of alternative livers contained significantly less triglycerides and free fatty acids and significantly more cholesterol and cholesterol esters than those from conventional livers. Detailed analysis of the fatty acid composition of triglycerides showed that the proportion of mono- and polyunsaturated fatty acids was significantly higher in the alternative livers. However, covariance analysis suggested that these differences in lipid composition were mainly due to differences in lipid content between both types of livers. The evaluation of cooked livers by a trained expert panel revealed significant differences in the sensory profile between the conventional and alternative livers. The acceptability by a consumer panel was significantly lower in alternative compared to conventional livers. This difference was not related to weight and/or lipid content since livers of similar weight range (800 to 1,000 g) were compared and showed clear-cut differences for hedonic scores.

The human androgen receptor (AR) gene contains a highly polymorphic CAG repeat in exon 1 that is inversely correlated with AR transcriptional activity in vitro. Several studies have shown that fewer CAG repeats are associated with an increased risk as well as more aggressive forms of prostate cancer. More recently, AR allele length was also inversely correlated with the histological grade of breast cancer, but no association was found between the AR-CAG polymorphism and the risk of either breast or ovary cancer. On the contrary, it was proposed that a longer CAG repeat sequence might be associated with an increased risk of breast cancer in BRCA1 mutation carriers, thus suggesting a different role of the AR-CAG polymorphism in sporadic and inherited breast cancers. With the intent of better understanding the role of the AR-CAG polymorphism as a cancer risk modifier, we defined the AR genotype of 151 patients (101 with breast and 50 with ovary cancer) belonging to high-risk breast/ovary cancer families. No difference in CAG repeat length was found between either breast and ovary cancer patients or age at diagnosis of both tumors. These results were also confirmed in a sub-group of 47 breast cancer cases, that either carried a BRCA gene mutation (11 cases) or were identified by very stringent operational criteria as hereditary breast cancers. Even though a substantially larger sample size would be required to reach conclusive evidence, our findings suggest that the AR-CAG polymorphism does not act as a modifier of tumor onset or tumor phenotype in breast/ovarian cancer families.

Using capillary zone electrophoresis, the urine creatinine (uCr) assay was validated in extemporaneous diluted urine, both in healthy subjects and athletes, with the uCr concentration as a reference value to compare excretion rates of other metabolites in the same samples. The electrokinetic sample injection was carried out at 10 kV per 10 s; UV absorbance detection was at 254 nm. Using standard samples, the creatinine migration mean time in 100 mmol/L acetate buffer, pH 4.4, was 3.3+/-0.2 min; the repeatability for absolute migration mean time was 0.6% and peak height repeatability was 2.9%. The correlation coefficient of the standard curve was r = 0.999 and the detection limit was 23.1 micromol/L. Intra- and interassay coefficients of variation (CV) were 3.0 and 3.6%, respectively; recovery was 99+/-3% and linearity was r= 0.98. Normal urine samples were diluted 1:80 in run buffer. The present CE urine creatinine assay showed a good correlation with HPLC and with Jaffe methods (r = 0.98 and r = 0.97, respectively; p < 0.0001). The uCr in the morning urine samples of 34 healthy males (M), 38 healthy females (F), and 83 male athletes (A) was 10.4+/-6.1 mmol/L, 10.8+/-8.1 mmol/L and 13.2+/-6.5 mmol/L, respectively. The uCr difference (p < 0.02) between M and A and a correlation (p < 0.05) with age in A were observed.
