RESUMO

Several methods have been proposed currently for evaluating the crack width of a mortar specimen. Among these, the water permeability test is widely used to estimate crack width because water permeability is directly related to the average crack width of a specimen through which water passes. However, the viscosity of water makes precise crack width measurement challenging. The possible inflow (outflow) of foreign (healing) substances could affect the test results. To circumvent this limitation, this study proposes a gas diffusion test using oxygen rather than water as the medium. The proposed method includes a process that could compensate for gas diffusion from specimen parts other than the crack, allowing for a more precise estimation of crack width. The crack width can indeed be estimated with an error of 4% or less.

RESUMO

Increased antimicrobial resistance presents a major threat to public health, and it is a global health problem due to the rapid globalization and transmission of infectious diseases. However, fast and precise diagnosis tool is lacking, and inappropriate antibiotic prescription leads to the unforeseen production of drug-resistant bacteria. Here, we report a Rapid and Accurate Nanoelectrokinetic Diagnostic System (RANDx) for detecting drug-resistant bacteria, which cause a common infectious disease called Urinary Tract Infection (UTI), within 7 min. We develop nanoelectrokinetic paper-based analytic device (NEK-PAD) as a sample prep module of RANDx and obtain >100-fold post-wetting preconcentration by balancing between ion concentration polarization (ICP) and radial imbibition for a constant flow rate. Simultaneously with preconcentration, our cathodic nanochannel design enables NEK-PAD to extract drug-resistant enzymes without denaturation and accelerate enzyme-linked reactions under electrical spontaneous heating at approximately 37 °C. Finally, using a cell phone camera, we detect label-free drug-resistant bacteria as low as 104 cfu/mL, which is higher than clinically required threshold (>105 cfu/mL) by enhancing 1000 times of the limit of detection (LOD) of colorimetric nitrocefin assay. We believe that the RANDx will be an innovative precision medicine tool for UTI and other infectious diseases in limited remote settings.

Research activities that have focused on the development and understanding of self-healing concrete have proposed various technologies intended to enhance self-healing capacity. The self-healing performance cannot be identified sufficiently with either a single test or a specific parameter because there are a number of factors that influence the performance of self-healing. Thus, it has become necessary to provide standardized test methods that make it possible to verify and compare the performance of self-healing materials. In this paper, self-healing mortars based on inorganic admixtures, which are developed for sealing 0.3 mm cracks with a healing index of 90%, are produced and used to validate the water permeability test and to propose protocols for the evaluation of self-healing performance. The healing performances of three self-healing mortars and a plain mortar as a reference are evaluated with a comparative study. The equivalent crack width, which can be estimated from the water flow rate, is suggested as a rational evaluation index. Finally, a self-healing performance chart is proposed to comprehensively show the healing performance of cement-based materials.

The separation of nanoparticles has drawn critical attention in various microfluidic applications including chemical analysis, diagnostics and environmental monitoring. Thus, a number of nanoparticle separation methods have been extensively proposed. However, most of the conventional methods require complicated structured devices, expensive manufacturing processes, and external power sources. While a spontaneous diffusiophoretic separation device based on an ion exchange mechanism could overcome such drawbacks, the recovery of separated particles and the inevitable development of an acidic environment due to the release of H+ from the cation exchange membrane limit its practical applicability. Therefore, in this work, we present a simple but robust nanoparticle separation method based on spontaneously induced diffusiophoresis, which is operated in a continuous manner to overcome the limitations of conventional methods. First, we confirmed that the particle exclusion distance followed the previously developed scaling law of diffusiophoresis. Consequently, we demonstrated the separation of nanoparticles of 40 nm, 200 nm and 2 µm diameter by utilizing the fact that the exclusion distances of various particles were proportional to their diffusiophoretic mobility. Furthermore, the use of Tris buffer increased the diffusiophoretic migration of nanoparticles due to the enhanced concentration gradient, and enabled the produced solution to be compatible with pH-sensitive bio-samples. Therefore, we expect this continuous and spontaneous diffusiophoretic separation platform to be useful in practical applications for analyzing various nano-meter scale bio-particles.

RESUMO

Maintenance of structures using self-healing concrete technologies has recently been actively studied. However, unlike the technological development of self-healing concrete, research focused on evaluating the self-healing performance is insufficient. Although water permeability experiments are widely used, the reliability of the test results may be reduced due to the viscosity of water and the possibility of elution of material inside the specimen. In this study, we propose a gas diffusion test for estimating the crack width and eventually for application to evaluation of the self-healing performance. The results verified that the proposed method can be effectively applied to the estimation of crack width.

Manipulating mechanism of particle's motion has been extensively studied for the sample preparation in microfluidic applications including diagnostics, food industries, biological analyses and environmental monitoring. However, most of conventional methods need additional external forces such as electric field or pressure and complicated channel designs, which demand highly complex fabrication processes and operation strategies. In addition, these methods have inherent limitations of dilution or mixing during separation or preconcentration step, respectively, so that a number of studies have reported an efficient selective preconcentration process, i.e. conducting the separation and preconcentration simultaneously. In this work, a power-free spontaneous selective preconcentration method was suggested based on leveraging convective flow over diffusiophoresis near the water-absorbing nanoporous ion exchange medium, which was verified both by simulation and experiment. Especially, the velocity of the convective flow by an imbibition deviated from the original tendency of t-1/2 due to non-uniformly patterned nanoporous medium that has multiple cross-sectional areas. As a result, the direction of particle's motion was controlled at one's discretion, which led to the spontaneous selective preconcentration of particles having different diffusiophoretic constant. Also, design rule for maximizing the efficiency was recommended. Thus, this selective preconcentration method would play as a key mechanism for power-free lab on a chip applications.

While the water impermeable constraint has been conventionally adopted for analyzing the transport phenomena at the interface of electrolyte/nanoporous medium, non-negligible water-permeance through the medium results in significant effect on ion and particle transportation. In this work, a rigorous theoretical and experimental analysis of the water-permeance effect were conducted based on a fully-coupled analytical/numerical method and micro/nanofluidic experiments. The regime diagram with three distinctive types of concentration boundary layers (ion depletion, ion accumulation, and intermediate) near the ion exchange nanoporous medium was proposed depending on the medium's permselectivity and the water-permeance represented by an absorbing parameter. Moreover, the critical absorbing parameters which divide the regimes were analytically obtained so that the bidirectional motion of particles were demonstrated only by altering the water-permeance without external stimuli. Conclusively, the presenting analysis of non-negligible water-permeance would be a substantial fundamental of transport phenomena at the interface of the ion exchange medium and electrolyte, especially useful for the tunable particle/ion manipulations in intermediate Peclet number environment.

This study examined the mechanism of action of a local anesthetic, lidocaine·HCl. Energy transfer between the surface fluorescent probe, 1-anilinonaphthalene-8-sulfonic acid, and the hydrophobic fluorescent probe, 1,3-di(1-pyrenyl) propane, was used to determine the effect of lidocaine·HCl on the thickness (D) of the synaptosomal plasma membrane vesicles (SPMV) isolated from the bovine cerebral cortex, and liposomes of the total lipids (SPMVTL) and phospholipids (SPMVPL) extracted from the SPMV. The thickness (D) of the intact SPMV, SPMVTL and SPMVPL were 1.044±0.008, 0.914±0.005 and 0.890±0.003 (arbitrary units, n=5) at 37℃ (pH 7.4), respectively. Lidocaine·HCl decreased the thickness of the neuronal and model membrane lipid bilayers in a dose-dependent manner with a significant decrease in the thickness, even at 0.1 mM. The decreasing effect of lidocaine·HCl on the membrane thickness might be responsible for some, but not all of its anesthetic action.
