RESUMO

Metal fume fever (MFF) is a work-related disease caused by the inhalation of metal particles, including zinc oxide. Chronic asthma may develop as a long-term consequence of exposure, particularly for welders and metal workers who are most at risk. In this study, we investigated the effects of ZnO fume inhalation on multiple inflammation-related cytokine- and cytokine receptor genes in mice from lung and lymph node samples, to explore the role of these in the pathogenesis of MFF. In our experiments, the animals were treated with a sub-toxic amount of ZnO fume for 4 h a day for 3 consecutive days. Sampling occurred 3 and 12 h post-treatment. We are the first to demonstrate that ZnO inhalation causes extremely increased levels of IL-17f gene expression at both sampling time points, in addition to increased gene expression rates of several other interleukins and cytokines, such as IL-4, IL-13, CXCL5, CSF-3, and IFN-γ. Our animal experiment provides new insights into the immunological processes of early metal fume fever development. IL-17f plays a crucial role in connecting immunological and oxidative stress events. The increased levels of IL-4 and IL-13 cytokines may explain the development of long-term allergic asthma after exposure to ZnO nanoparticles, which is well-known among welders, smelters, and metal workers.

Concentrations of 4 potentially toxic elements (As, Cd, Hg, Pb) were investigated in the feather, liver, kidney, and bone of great cormorants (Phalacrocorax carbo). The tissue samples were taken at the Central Tisza - Jászság Nature Conservation Area in Hungary. They were analysed by inductively coupled plasma optical emission spectroscopy (ICP-OES). The goal of the investigation was to analyse the metal burden of the above-mentioned elements in the various tissues of these wild birds and to provide important information for monitoring the environmental pollution.Amongst the examined potentially toxic elements no statistical gender difference was observed, so the data were not separated based on them during the statistical analysis. The concentration of mercury was the highest in the feather, followed by the liver, kidney, and bone. The lead was detected in the feather with the highest level followed by the kidney, liver, and bone. The cadmium was determined in all investigated tissues with the next descending order: kidney > bone > liver > feather. Highest arsenic concentration was measured in the feather, followed by liver, kidney, and bone with the same concentration.The detected concentrations of the investigated potentially toxic elements in different tissues of great cormorants (feathers, liver, kidney, bone) means that the living area of this birds is not highly contaminated to induce health problems or toxic signs, or even other undesirable effect in the animals.

The popularity of seafoods is high due to their superb dietary properties and healthy composition. However, it is crucial to understand whether they adequately contribute to our essential nutritional needs. Small amounts of essential metals are indispensable in the human body to proper physiological functioning; their deficiency can manifest in various sets of symptoms that can only be eliminated with their intake during treatment or nutrition. However, the excessive consumption of metals can induce undesirable effects, or even toxicosis. Shellfish, oyster, and squid samples were collected directly from a fish market. After sample preparation, the concentration of essential metals (cobalt, chromium, copper, manganese, molybdenum, nickel, and zinc) was detected by Inductively Coupled Plasma Optical Emission Spectrometry. The results were analyzed statistically using ANOVA and two-sample t-tests. The average concentration of the investigated essential elements and the calculated burden based on the consumption were below the Recommended Dietary Allowances and Tolerable Upper Intake Levels. Based on these results, the trace element contents of the investigated seafoods do not cover the necessary recommended daily intake of them, but their consumption poses no health hazard due to their low levels.

The consumption of marine fishes has a positive effect on a consumer's health; however, it poses a potential risk due to their level of heavy metals in their body. Heavy metals can be naturally found in the environment, but their concentration can be increased with anthropogenic activities. Samples of tuna (Thunnus albacares) were collected at a fishery market. The potentially toxic elements (arsenic, cadmium, lead, and mercury) were determined with a validated method in the flesh of fish using inductively plasma optical emission spectrometry after microwave digestion. Generally, the average concentration of them was below the official limit values regulated by the European Union, except for lead. Based on the concentrations of arsenic (inorganic derivates: 0.05 ± 0.02 mg/kg) and cadmium (0.03 ± 0.01 mg/kg) in the tuna fish samples, and their calculated EDI values (As: 0.03-0.09 µg/kg/day; Cd: 0.05-0.07 µg/kg/day), the investigated food could be declared safe for human consumption. Generally, mercury content was below the official regulated limit, and the calculated EDI value was below the dietary reference value (0.3 µg/kg/day) in most of the samples (90%), exceeding it only in two samples (0.69 and 0.82 µg/kg/day); thus, they may not be harmful to the consumer. The concentration of lead above the official maximum limit (0.30 mg/kg) in 40% of tuna samples (0.30-1.59 mg/kg), as well as the exceeding of the dietary reference value for lead (adult: 0.16 µg/kg/day; children: 0.26 µg/kg/day) based on the calculated EDI values (0.28-1.49 µg/kg/day), draw attention to the importance of environmental pollution and the protection of consumers' health.

Metal fume fever is a well-known occupational disease that arises from prolonged exposure to subtoxic levels of zinc oxide-containing fumes or dust. This review article aims to identify and examine the possible immunotoxicological effects of inhaled zinc oxide nanoparticles. The current most widely accepted pathomechanism for the development of the disease involves the formation of reactive oxygen species following the entry of zinc oxide particles into the alveolus resulting the release of pro-inflammatory cytokines by activation of the Nuclear Factor Kappa B transcriptional signal, thus evoking the symptoms. The role of metallothionein in inducing tolerance is believed to be a key factor in mitigating the development of metal fume fever. The other, poorly proven hypothetical route is that zinc-oxide particles bind to an undefined protein in the body as haptens to form an antigen and act as an allergen. After activation of the immune system, primary antibodies and immune complexes are developed and type 1. hypersensitivity reaction occurs, that can cause asthmatic dyspnoea, urticaria and angioedema. The development of tolerance is explained by the formation of secondary antibodies against primary antibodies. Oxidative stress and immunological processes cannot be completely separated from each other, as they can induce each other.

The study was performed on 10 female and 10 male wild boars (Sus scrofa) after shooting during the regular hunting season to investigate the concentration of metals in the muscle and fat tissue. The concentrations of essential and non-essential elements were determined (arsenic (As), cadmium (Cd), chromium (Cr), copper (Cu), lead (Pb), manganese (Mn), mercury (Hg), and zinc (Zn)) using inductively coupled plasma optical emission spectrometry. The concentrations of As, Hg, and Cd were below the limit of detection (As, Hg: < 0.5 mg/kg, Cd: < 0.05 mg/kg) in every tissue sample of both sexes. The lead was detected as 0.36 ± 0.16 mg/kg and 0.22 ± 0.06 mg/kg in the muscle of females and males, respectively, showing a significant difference between the sexes (p = 0.0184). The measured concentration of Cr was 0.14 ± 0.08 mg/kg and 0.13 ± 0.06 mg/kg, and that of copper was 1.22 ± 0.14 mg/kg and 1.06 ± 0.16 mg/kg in the muscle of females and males, respectively. The same tendency was observed in the case of copper content of fat tissues (female: 0.13 ± 0.10 mg/kg; male: 0.13 ± 0.04 mg/kg; p = 0.2707). Manganese concentration of muscle was 0.45 ± 0.30 mg/kg (female) and 1.36 ± 0.96 mg/kg (male), and that of fat tissue was 0.32 ± 0.22 mg/kg (female) and 0.74 ± 0.75 mg/kg (male). The Zn was detected as 56.75 ± 7.86 mg/kg and 1.83 ± 0.76 mg/kg in the muscle and fat of females and 52.12 ± 11.51 mg/kg and 1.94 ± 0.57 mg/kg in males, respectively. Based on data, the consumption of fat and muscle tissues of the wild boars tested can be food toxicologically objectionable, mainly due to the lead content, and thus pose a risk to frequent consumers of this type of game meat.

During plant cultivation, the pesticides can get into the tissue of vegetables due to crop protection processes, and thus into the food chain. Therefore, they constitute a potential risk to the consumer's health. Depletion of pesticides [spirotetramat (Movento), azoxystrobin and difenoconazole (Amistar Top)] was monitored by testing tomatoes treated individually or simultaneously and tomato juices prepared from the treated tomatoes. The investigations aimed to reveal any kinetic interaction between the compounds tested and changes in their elimination, and thus to assess their compliance with the official Maximum Residue Limits (MRLs). The co-presence of pesticides prolonged the elimination of the individual compounds which reached significantly higher residue levels (P < 0.0001) in tomato, especially difenoconazole (45%) and azoxystrobin (50%) on day 8 after treatment that can cause food safety issues to the human consumers. However, the concentrations of pesticides applied alone or simultaneously were found to be below the corresponding MRL values after the withdrawal period in all investigated tomato and tomato juice samples. Accordingly, the investigated pesticides can be safely used simultaneously, their concentrations are in compliance with the legal regulations and thus their concomitant presence does not pose any risk to the consumers' health.

Cd, Hg, and Pb concentrations were investigated in Sardina pilchardus fish. Samples originated from the Atlantic Northeast fishing area 27 and were analysed by inductively coupled plasma optical emission spectroscopy. The aim of the study was to estimate the health impact of consumption, based on the concentrations and calculated exposure for each element. Based on the average total arsenic concentration of 3.26 ± 0.39 mg/kg in the samples, the estimated inorganic arsenic content and calculated PTWI suggested that the samples do not pose a health risk for consumers. Cadmium content of the samples was also not considered hazardous, as both the measured concentrations and PTMI values were below the specified limits. Lead concentration exceeded the maximum limit in 73% of the samples, while calculated average weekly intake was below the recommended level. The measured mercury concentrations exceeded the maximum limit in only 5% of the samples.

The most important welding processes used are the gas metal arc (GMA) welding, the tungsten inert gas (TIG) welding, and the manual metal arc (MMA) welding processes. The goal of our investigation was to monitor the distribution of iron (Fe), manganese (Mn), calcium (Ca), and magnesium (Mg) in the lung, spleen, liver, and kidney of mice after inhalation exposure of different welding methods using different steel base materials. The treatment groups were the following: MMA-mild steel, MMA-molybdenum-manganese (MoMn) alloy, TIG-mild steel, and TIG-stainless steel. The samples were taken 24 and 96 h after the treatments. Most importantly, it was found that the Mn concentration in the lung' samples of the MMA-mild steel and the MMA-MoMn groups was increased extremely at both sampling times and in the spleen' samples also. In the TIG groups, the rise of the Mn concentration was only considerable in the lungs and spleens at 24 h, and emerged concentration was found in the liver in 96 h samples. Histopathology demonstrated emerged siderin content in the spleens of the treated animals and in siderin filled macrophages in the lungs mostly in all treated groups. Traces of high-level glycogen retention was found in the MMA groups at both sampling times. Similar glycogen retention in TIG-Ms and TIG stainless group's liver samples and emerged number of vacuoles, especially in the hepatocytes of the TIG-stainless steel 96 h group were also found. The mentioned results raise the consequence that there is a considerable difference in the kinetics of the Mn distribution between the MMA- and the TIG-fume-treated groups. Hence, the result suggests that manganese has a particle-size-dependent toxico-kinetics property. The anomaly of the glycogen metabolism indicates the systemic effect of the welding fumes. Also, the numerous vacuoles mentioned above show a possible liver-specific adverse effect of some components of the TIG-stainless steel welding fumes.

Concentrations of 12 metals (As, Ba, Cd, Co, Cr, Cu, Hg, Mn, Mo, Ni, Pb and Zn) were examined in the pectoral and thigh muscle of great cormorants (Phalacrocorax carbo). The samples were collected from Central Tisza-Jászság Nature Conservation Area in Hungary. The tissue samples were analysed by inductively coupled plasma optical emission spectroscopy (ICP-OES). The aim of the study was to examine the impacts of heavy metal pollution on the water birds, determine the concentrations of the abovementioned metals in the different muscle tissues of these wetland birds, and provide the basic materials for monitoring the environmental pollution. Among the investigated elements/metals, the detected concentrations of As, Ba, Cd, Co, Cr, Mo and Ni were below the detection limit. Higher concentration of Cu, Hg, Mn and Pb was measured in the pectoral muscle compared to the thigh muscle, but only in the case of Cu and Mn were found significant differences between the tissues. In the case of the Zn concentration, the higher value was detected in the thigh muscle. There were no statistical differences between males and females in either metal concentrations.

The aim of this study was to investigate the transfer of cephalexin, penicillin-G, and ampicillin & cloxacillin from cow's milk to cheese and whey. For this purpose, raw milk was artificially contaminated to different antibiotic levels and then heat-treated to prepare fresh cheese from it. Antibiotic levels of the milk, whey and cheese were measured with LC-MS/MS. The extent of heat degradation was not sufficient to remove the antibiotic residues from milk. Antibiotic concentrations in whey and fresh cheese were in good accordance with the concentration of the same compound in milk suggesting that contamination of the milk will result in contamination of the product. The investigated antibiotics were transferred less into the cheese curd (1.6-12.5% of the original amount), than into the whey (33.2-74.1%). For penicillin-G even 100% (complete removal) was experienced.

Single and simultaneous toxic effects of glyphosate (Amega Up, 360 g L-1, 4%) and copper sulphate (0.01%) were studied in avian embryos treated either with injection directly into the air chamber or by immersion application for 30 min on day 0 of incubation. Alterations of the chicken embryos were evaluated during necropsy performed on day 19 of incubation, together with mortality, body weight and the type of developmental abnormalities. Based on the results, the injection application appeared to be more toxic than the immersion method, as it induced increased mortality and reduced the average body weight, and resulted in a higher incidence of congenital anomalies. Supposedly, a toxicodynamic interaction occurs between copper sulphate and glyphosate, which may reduce the vitality of embryos and thus decrease the number of offspring in wild birds.

Plastic has been an incredibly useful and indispensable material in all aspects of human life. Without it many advances in medicine, technology or industry would not have been possible. However, its easy accessibility and low cost have led to global misuse. Basically, the production of the plastics from different chemical agents is very easy but unfortunately difficult to reuse or recycle, and it is thrown away as litter, incinerated or disposed of in landfill. Plastic once in the environment begins to degrade to very small sizes. Thus, many animals mistake them for food, so plastic enters a marine, terrestrial or freshwater food web. These microplastics although chemically inert have been shown to act as tiny "bio-sponges" for harmful chemicals found in the environment changing the nature of a plastic particle from chemically harmless to potentially toxic. It was believed that microparticles would simply pass through the gastrointestinal tract of animals and humans with no biological effect. However, studies have shown that they are sometimes taken up and distributed throughout the circulatory and lymphatic system and may be stored in the fatty tissues of different organisms. The result of the uptake of them showed potential carcinogenic effects, liver dysfunction and endocrine disruption. This review focuses on micro- and nanoplastics and their way entering marine and freshwater food webs, with particular attention to microplastic trophic transfer, their toxic side effects and influence to the human consumer in health and safety in the future.

Eye irritation potency of pesticides (fungicides, herbicides, insecticides) was comparatively tested by HET-CAM and ICE method. Based on the results of the tests the statistical analysis of agreement between classification using individual methods was done by Goodman-Kruskal's rank correlation and determination (calculation) of Cohen's kappa coefficient. Statistical analysis of agreement between classification revealed significant correlation between results of in vivo and in HET-CAM assays (76%). There was no significant correlation between result of in vivo and in ICE methods (64%). Weakest correlation was found between the data from in vitro HET-CAM and ICE tests. The percentage of agreement between two in vitro data was 48%. They may be recommended as a part of a battery of tests to reduce experimentation on mammals and to limit or eliminate pain and injury inflicted on experimental animals. The HET-CAM test is a useful tool for studying in vivo the potential conjunctival irritation, while the ICE test can be used to study corneal irritant effects in detail.

It is undeniable that with the popularity of sushi and sashimi over the last decade the consumption of raw fish has extremely increased. Raw fish is very appreciated worldwide and has become a major component of human diet because of its fine taste and nutritional properties. Possible hazards concerning fish safety and quality are classified as biological and chemical hazards. They are contaminants that often accumulate in edible tissue of fish and transmit to humans via the food chain affecting the consumer's health. Although their concentration in fish and fishery products are found at non-alarming level of a daily basis period, they induce hazardous outcome on human health due to long and continuous consumption of raw fish. Regular sushi and sashimi eaters have to be aware of the contaminants found in the other components of their dish that often add up to acceptable residue limits found in fish. Hence, there is the urge for effective analytical methods to be developed as well as stricter regulations to be put in force between countries to monitor the safety and quality of fish for the interest of public health.

The aim of our study was to investigate the concentrations of toxic metals in the feathers of predatory birds in the Hortobágyi Madárpark (Bird Hospital Foundation). Samples were collected from different predatory birds originated from the eastern and north-eastern region of Hungary. Inductively coupled plasma optical emission spectrometry was used to determine the concentration of toxic metals. The mean values varied between bird species groups, their concentrations were between 0.29 ± 0.24 and 0.40 ± 0.30 mg/kg for arsenic (As), 0.09 ± 0.03 and 0.20 ± 0.18 mg/kg for cadmium (Cd), 1.15 ± 1.40 to 2.30 ± 1.52 mg/kg for lead (Pb) and 0.58 ± 0.31 to 2.19 ± 1.25 mg/kg for mercury (Hg), respectively. The measured values are not over the considered threshold values for these toxic metals and in accordance with similar concentrations of them recorded in similar species within Europe. No significant differences were found in their concentration between genders or age in the species. According to the detected concentrations of these metals, their levels accumulated in the feather of the investigated birds do not indicate the possibility of poisoning.

Heavy-metal content (arsenic, cadmium, lead and mercury) was investigated and evaluated in shellfish, oysters and squids. The samples were collected weekly for 20 weeks from a fishery product market in Hungary. The concentration of heavy metals was measured by Inductively Coupled Plasma Optical Emission Spectrometer (ICP-OES) analysis after sample preparation. The results were analysed statistically by one-way ANOVA method. The average concentration of arsenic (3.01 ± 1.46 mg kg-1) in shellfish was significantly higher (p < 0.001) compared to oysters (2.88 ± 1.12 mg kg-1) and squids (1.28 ± 0.52 mg kg-1). The level of mercury was below the limit of detection (0.5 mg kg-1) in each sample and there was no statistical significance in the concentrations of cadmium (p = 0.351) and lead (p = 0.412) in the species investigated. The provisional tolerable intake values were also calculated. Based on the obtained results of the heavy-metal content of the investigated samples, the seafood is considered to be safe for human consumption. However, prolonged ingestion of oysters and squids at these levels may contribute to the consumers' cadmium burden.

Concentrations of different heavy metals (As, Cd, Cr, Cu, Hg, Pb, Zn) were examined in the contour feathers of long-eared owl (Asio otus), little owl (Athene noctua), tawny owl (Strix aluco), barn owl (Tyto alba), Eurasian sparrowhawk (Accipiter nisus), rook (Corvus frugilegus), hooded crow (Corvus cornix), carrion crow (Corvus corone), common buzzard (Buteo buteo) and barn swallow (Hirundo rustica). The samples were collected from the Hortobágyi Madárpark (Bird Hospital Foundation) in Hungary. The bird species were classified into six groups based on their nourishment. Feathers were analysed by inductively coupled plasma optical emission spectroscopy (ICP-OES). The aim of our study was to determine the concentration of the above-mentioned heavy metals in the six different groups and to compare them by the groups, to find a possible connection between the concentrations and the age of birds and to get some information about the heavy metal burden of the environment. The highest As concentration was measured in little owl (0.65 ± 0.56 mg/kg). The highest Cd, Cr and Pb concentration was found in the feathers of barn swallow (0.13 ± 0.06 mg/kg; 1.69 ± 0.44 mg/kg; 5.36 ± 1.46 mg/kg), while the highest Cu and Hg concentration (65.45 ± 17.66 mg/kg; 2.72 ± 1.08 mg/kg) in sparrowhawk feathers and the highest Zn concentration in owls (157.21 ± 57.3 mg/kg). Statistically significant difference has been determined between the juvenile and adult crows in the case of Cd (p = 0.011). The higher concentration was measured in adults (0.14 ± 0.04 mg/kg) than that in juveniles (0.08 ± 0.02 mg/kg). Based on our results, the examined area is not contaminated by these heavy metals on that level, which can cause any adverse effect or poisoning in birds, so this region is safe to wildlife.

The muscle, liver, kidney and fat samples of 20 roe deer of both sexes originating from a hunting area in central Hungary were investigated for the presence of heavy metals such as As, Cd, Hg and Pb, and their contents were evaluated for possible health risk to consumers. Both As and Hg were found at a level below the limit of detection (< 0.5 mg/kg wet weight) in all samples. The median of the measured Cd concentrations was significantly higher in both the kidney and the liver (p = 0.0011) of bucks than of does. In bucks, Cd levels exceeded the respective maximum limits laid down in the European legislation in four kidney and three muscle samples, whereas in does, the measured concentrations were below the respective limits in all samples. The detected amounts of Pb exceeded the maximum limits in the kidney of one buck and eight does, in the liver of two bucks and six does, in the muscle of six bucks and nine does, whereas in all fat tissues of both bucks and does. The concentration of Pb (p = 0.02) was significantly greater in the kidney of does compared to roebucks. Based on data obtained from the present study, the consumption of organs and tissues of the investigated roe deer could be objectionable from food-toxicological point of view and may pose risk to the high consumers of wild game due to their cadmium and lead contents.
