Tricholoma matsutake, a highly valued delicacy in Japan and East Asia, is an ectomycorrhizal fungus typically found in a complex soil community of mycorrhizae, soil microbes, and host-tree roots referred to as the shiro in Japan. A curious characteristic of the shiro is an assortment of small rock fragments that have been implicated as a direct source of minerals and trace elements for the fungus. In this study, we measured the mineral content of 14 samples of shiro soil containing live matsutake mycelium and the extent to which the fungus can absorb minerals directly from the rock fragments. X-ray powder diffraction identified major phases of quartz, microcline, orthoclase, and albite in all shiro samples. PCR-denaturing gradient gel electrophoresis (DGGE) fingerprinting and direct sequencing confirmed the presence of T. matsutake on 32 of 33 rock fragments. Piloderma sp. co-occurred on 40% of fragments and was positively correlated with locations known to produce good mushroom crops. The ability of T. matsutake to absorb trace elements directly from rock fragments was examined in vitro on nutrient-agar plates supplemented with rock fragments from the shiro. In comparison to the mineral content of tissues grown on control media, the concentration of Al, Cu, Fe, Mn, P, and Zn increased from 1.1 to 106.4 times for both T. matsutake and Piloderma sp. Mineral content of dried sporocarps sampled from the study site partially reflected the results of the in vitro study. We discuss the implications of our results with respect to the natural development and artificial culture of this important fungus.

Mine tailings impoundments are a source of leachates known as acid mine drainage (AMD) which can pose a contamination risk for surrounding surface and groundwater. Methodologies which can help management of this environmental issue are needed. We carried out a laboratory study of the spectral induced polarization (SIP) response of tailings from the Haveri Au-Cu mine, SW Finland. The primary objectives were, (1) to determine possible correlations between SIP parameters and textural properties associated with oxidative-weathering mechanisms, mineralogical composition and metallic content, and (2) to evaluate the effects of the pore water chemistry on SIP parameters associated with redox-inactive and redox-active electrolytes varying in molar concentration, conductivity and pH. The Haveri tailings exhibit well defined relaxation spectra between 100 and 10,000Hz. The relaxation magnitudes are governed by the in-situ oxidative-weathering conditions on sulphide mineral surfaces contained in the tailings, and decrease with the oxidation degree. The oxidation-driven textural variation in the tailings results in changes to the frequency peak of the phase angle, the imaginary conductivity and chargeability, when plotted versus the pore water conductivity. In contrast, the real and the formation electrical conductivity components show a single linear dependence on the pore water conductivity. The increase of the pore water conductivity (dominated by the increase of ions concentration in solution) along with a transition to acidic conditions shifts the polarization peak towards higher frequencies. These findings show the unique sensitivity of the SIP method to potentially discriminate AMD discharges from reactive oxidation zones in tailings, suggesting a significant advantage for monitoring threatened aquifers.

The occurrence and rates of terminal electron acceptor processes, and recharge processes in the unsaturated zone of a boreal site contaminated with petroleum hydrocarbons in the range C(10) to C(40) were examined. Soil microcosms were used to determine the rates of denitrification, iron (Fe) reduction, sulfate (SO(4)) reduction, and methanogenesis in two vertical soil profiles contaminated with oil, and in a noncontaminated reference sample. Furthermore, the abundances of the 16S rRNA genes belonging to Geobacteracaea in the samples were determined by real-time quantitative polymerase chain reaction (PCR). Analyses of ground water chemistry and soil gas composition were also performed together with continuous in situ monitoring of soil water and ground water chemistry. Several lines of evidence were obtained to demonstrate that both Fe reduction and methanogenesis played significant roles in the vertical profiles: Fe reduction rates up to 3.7 nmol h(-1) g(-1) were recorded and they correlated with the abundances of the Geobacteracaea 16S rRNA genes (range: 2.3 x 10(5) to 4.9 x 10(7) copies g(-1)). In the ground water, ferrous iron (Fe(2+)) concentration up to 55 mg L(-1) was measured. Methane production rates up to 2.5 nmol h(-1) g(-1) were obtained together with methane content up to 15% (vol/vol) in the soil gas. The continuous monitoring of soil water and ground water chemistry, microcosm experiments, and soil gas monitoring together demonstrated that the high microbial activity in the unsaturated zone resulted in rapid removal of oxygen from the infiltrating recharge thus leaving the anaerobic microbial processes dominant below 1.5 m depth both in the unsaturated and the saturated zones of the subsurface.
