RESUMO

An increase in the total choline-containing compound content is a common characteristic of cancer cells, and aberrant choline metabolism in cancer is closely associated with malignant progression. However, the potential role of choline-induced global transcriptional changes in cancer cells remains unclear. In this study, we reveal that an elevated choline content facilitates hepatocellular carcinoma (HCC) cell proliferation by reprogramming Krüppel-like factor 5 (KLF5)-dominated core transcriptional regulatory circuitry (CRC). Mechanistically, choline administration leads to elevated S-adenosylmethionine (SAM) levels, inducing the formation of H3K4me1 within the super-enhancer (SE) region of KLF5 and activating its transcription. KLF5, as a key transcription factor (TF) of CRC established by choline, further transactivates downstream genes to facilitate HCC cell cycle progression. Additionally, KLF5 can increase the expression of choline kinase-α (CHKA) and CTP:phosphocholine cytidylyltransferase (CCT) resulting in a positive feedback loop to promote HCC cell proliferation. Notably, the histone deacetylase inhibitor (HDACi) vorinostat (SAHA) significantly suppressed KLF5 expression and liver tumor growth in mice, leading to a prolonged lifespan. In conclusion, these findings highlight the epigenetic regulatory mechanism of the SE-driven key regulatory factor KLF5 conducted by choline metabolism in HCC and suggest a potential therapeutic strategy for HCC patients with high choline content.

Ongoing warming will influence plant photosynthesis via thermal effects and by enhancing water deficit. As the primary limiting factor for the growth and development of plants in arid deserts, water may alter the potential warming effects on plant photosynthesis and lead to increased uncertainty in plant dynamics. Here, we used open-top chambers (OTCs) to evaluate the impacts of in situ warming (+0.5 and +1.5 °C) on the photosynthesis and growth of two representative desert plants, Artemisia ordosica and Grubovia dasyphylla, from wet to dry spells. The plant traits associated with photosynthetic diffusive and biochemical processes were also measured to explore the underlying mechanisms involved. We found that warming significantly increased the net photosynthetic rate (Anet) during wet spells under 1.5 °C warming in both plants, while only increased that of A. ordosica under 0.5 °C warming. During dry spells, Anet decreased both in A. ordosica and G. dasyphylla, with the rates of declining being 48 % and 41 %, respectively, higher than control under warming. Consequently, warming significantly amplified photosynthetic responses to drought events, which offset the positive warming effects during wet spells and led to unchanged plant biomass in both species. Besides, alterations in plant traits tended to be associated with positive warming effects during wet spells, and the negative effects of drought were mainly due to stomatal limitation. Our results emphasised that the potential benefits of warming during wet spells may be reversed during drought events. Thus, the adverse effects of ongoing warming on desert productivity may increase during dry spells in growing seasons and during dry years.

The growth and survival of woody plant species is mainly driven by evolutionary and environmental factors. However, little is known about the hydraulic mechanisms that respond to growth limitation and enable desert shrub survival in arid habitats. To shed light on these hydraulic mechanisms, 9-, 31-, and 56-year-old Caragana korshinskii plants that had been grown under different soil water conditions at the southeast edge of the Tengger Desert, Ningxia, China were used in this study. The growth of C. korshinskii was mainly limited by soil water rather than shrub age in non-watered habitats, which indicated the importance of maintaining shrub survival prior to growth under drought. Meanwhile, higher vessel density, narrower vessels and lower xylem hydraulic conductivity indicated that shrubs enhanced hydraulic safety and reduced their hydraulic efficiency in arid conditions. Importantly, xylem hydraulic conductivity mediated by variation in xylem hydraulic architecture regulated photosynthetic carbon assimilation and growth of C. korshinskii. Our study highlights that the synergistic variation in xylem hydraulic safety and hydraulic efficiency is the hydraulic mechanism limiting growth and maintaining survival of C. korshinskii under drought, providing insights into the strategies for growth and survival of desert shrubs in arid habitats.

Hybrid speciation plays an important role in species diversification. The establishment of reproductive isolation is crucial for hybrid speciation, and the identification of diverse types of hybrids, particularly homoploid hybrid species, contributes to a comprehensive understanding of this process. Reaumuria songarica is a constructive shrub widespread in arid Central Asia. Previous studies have inferred that the R. songarica populations in the Gurbantunggut Desert (GuD) originated from homoploid hybridizations between its eastern and western lineages and may have evolved into an incipient species. To further elucidate the genetic composition of different hybrid populations and to determine the species boundary of this hybrid lineage, we investigated the overall phylogeographic structure of R. songarica based on variation patterns of five cpDNA and one nrITS sequences across 32 populations. Phylogenetic analyses demonstrated that within the GuD lineage, the Wuerhe population evolved directly from ancestral lineages, whereas the others originated from hybridizations between the eastern and western lineages. PCoA and genetic barrier analysis supported the subdivision of the GuD lineage into the southern (GuD-S) and northern (GuD-N) groups. Populations in the GuD-S group had a consistent genetic composition and the same ancestral female parent, indicating that they belonged to a homoploid hybrid lineage. However, the GuD-N group experienced genetic admixture of the eastern and western lineages on nrITS and cpDNA, with some populations inferred to be allopolyploid based on ploidy data. Based on cpDNA haplotypes, BEAST analyses showed that the GuD-S and GuD-N groups originated after 0.5 Ma. Our results suggest that multiple expansions and contractions of GuD, driven by Quaternary climatic oscillations and the Kunlun-Yellow River tectonic movement, are important causes of the complex origins of R. songarica populations in northern Xinjiang. This study highlights the complex origins of the Junggar Basin flora and the underappreciated role of hybridization in increasing its species diversity.

DNA- and RNA-binding proteins (DRBPs) are versatile proteins capable of binding to both DNA and RNA molecules. In this study, we identified fibrillarin (FBL) as a key DRBP that is upregulated in liver cancer tissues vs. normal tissues and is correlated with patient prognosis. FBL promotes the proliferation of liver cancer cells both in vitro and in vivo. Mechanistically, FBL interacts with the transcription factor KHSRP, thereby regulating the expression of genes involved in glucose metabolism and leading to the reprogramming of glucose metabolism. Specifically, FBL and KHSRP work together to transcriptionally activate the glycolytic enzyme PFKFB4 by co-occupying enhancer and promoter elements, thereby further promoting liver cancer growth. Collectively, these findings provide compelling evidence highlighting the role of FBL as a transcriptional regulator in liver cancer cells, working in conjunction with KHSRP. The FBL/KHSRP-PFKFB4 regulatory axis holds potential as both a prognostic indicator and a therapeutic target for liver cancer. SIGNIFICANCE: A novel role of FBL in the transcriptional activation of PFKFB4, leading to glucose metabolism reprogramming in liver cancer.

Although the effects of human-enhanced atmospheric nitrogen (N) deposition are well documented, the response of dryland soils to N deposition remains unclear owing to the divergence in hydrological outputs and soil heterogeneity. We selected a typical desert steppe in western China to simulate the effects of long-term N deposition by applying 0 (CK), 3.5, 7, and 14 g N m-2 yr-1 for 12 consecutive years. We found that, compared with the CK plots, the total N content of the upper (0-10 cm) and lower (10-20 cm) soil layers in fertilized plots increased by 8.3-14.6 % and 2.4-8.2 %, respectively. Correspondingly, the available, NH4+-, and NO3--N contents in the upper soil significantly increased by 25.5-68.3 %. However, in the lower soil, available and NO3--N contents were significantly lower than those in the CK plots, and their variation trend was opposite to that of NH4+-N, implying N turnover and leaching. As a result, the upper and lower soil pH in fertilized plots significantly decreased by 0.36-0.53 and 0.31-0.37 units; however, their CaCO3 content significantly increased by 9.8-22.8 % and 7.2-30.3 %, respectively. The total phosphorus (P) content in the upper and lower soil layers in fertilized plots significantly increased and decreased by 3.6-51.3 % and 16.7-62.5 %, respectively, however, both significantly decreased along the N fertilization gradient. Furthermore, the upper and lower soil organic carbon (SOC) content in the fertilized plots significantly increased by 57.7-78.1 % and 19.2-27.4 %, respectively. Pearson's correlation analysis revealed that available soil P was significantly negatively correlated with plant shoot Mn content (a proxy for rhizosphere carboxylates), whereas dissolved OC, SOC, and CaCO3 were significantly positively correlated, suggesting that Ca cycling is involved in P cycling and SOC sequestration. Our study suggests that long-term N input exacerbates P limitation in desert steppes, however, enhances SOC sequestration.

Recently developed optogenetic technology, which allows high-fidelity control of neuronal activity, has been applied to investigate the neural circuits underlying sensory processing and behavior. The nasal cavity is innervated by the olfactory nerve and trigeminal nerve, which are closely related to common symptoms of rhinitis, such as impairment of smell, itching, and sneezing. The olfactory system has an amazing ability to distinguish thousands of odorant molecules at trace levels. However, there are many issues in olfactory sensing mechanisms that need to be addressed. Optogenetics offers a novel technical approach to solve this dilemma. Therefore, we review the recent advances in olfactory optogenetics to clarify the mechanisms of chemical sensing, which may help identify the mechanism of dysfunction and suggest possible treatments for impaired smell. Additionally, in rhinitis patients, alterations in the other nerve (trigeminal nerve) that innervates the nasal cavity can lead to hyperresponsiveness to various nociceptive stimuli and central sensitization, causing frequent and persistent itching and sneezing. In the last several years, the application of optogenetics in regulating nociceptive receptors, which are distributed in sensory nerve endings, and amino acid receptors, which are distributed in vital brain regions, to alleviate overreaction to nociceptive stimuli, has gained significant attention. Therefore, we focus on the progress in optogenetics and its application in neuromodulation of nociceptive stimuli and discuss the potential clinical translation for treating rhinitis in the future.

OBJECTIVE: We review the prevalence of allergic diseases in children across prenatal exposures to heavy metals. METHODS: This systematic review and meta-analysis is registered in the PROSPERO database (CRD42023478471). A comprehensive search of PubMed, Web of Science, Medline and Cochrane library was conducted from the database inception until 31 October 2023. The Newcastle-Ottawa Quality Assessment Scale (NOS) was used to assess the quality of included studies. We used a random-effects model to summarize the effects from the studies. RESULTS: A total of 16 studies were included, 120,065 mother-child pairs enrolled. The NOS scores indicated that the quality of the literature included in the study was of a high standard. CONCLUSION: The final results indicate that prenatal exposure to Pb increased the incidence of wheeze and Eczema in infants, and exposure to Ni and CD increased the incidence of AD in infants.

Backgrounds: Observational studies suggest that air pollutants, including particulate matter and nitrogen compounds, could elevate asthma and allergic rhinitis health risks. Nevertheless, the exact nature of the causal relationship between air pollution and asthma and allergic rhinitis remains unknown. This study utilizes the Mendelian randomization (MR) technique to explore the potential causal links between air pollution components (PM2.5, PM2.5-10, PM10, NO2, and nitrogen dioxide) and the incidence of allergic rhinitis and asthma. Methods: A MR study utilized summary statistics from GWAS that are publicly accessible. The inverse variance weighting (IVW) approach served as the foundational analysis technique. To ensure robustness, supplementary methodologies such as the weighted median, MR-Egger regression, simple mode, and weighted model were also applied. Heterogeneity was evaluated using Cochran's Q test, and the presence of pleiotropy was determined through MR-Egger regression. The MR-PRESSO test was employed for outlier detection, and the analysis's sensitivity was scrutinized via a leave-one-out strategy. Results: The IVW technique showed a strong correlation between PM10 and asthma (OR = 0.625, 95% CI = 0.396-0.988, p = 0.044). No significant associations were found between asthma and other air pollutants such as PM2.5, PM2.5-10, NO2, or nitrogen dioxide. Similarly, allergic rhinitis showed no causal relationships with any studied air pollution metrics. Pleiotropy was absent in the findings. Sensitivity analyses, employing the leave-one-out method, confirmed the stability of these results, unaffected by individual single nucleotide polymorphisms (SNPs). Conclusion: This Mendelian randomization study establishes a causal link between PM10 exposure and asthma, suggesting that interventions to reduce air pollution may decelerate the adverse progression of asthma.

With the gradual depletion of natural gold ore, waste printed circuit boards (WPCBs) have become one of the most attractive alternatives to gold ore. Here, a series of quaternary phosphonium adsorbents with a large size were successfully synthesized by adjusting the number of functional groups and carbon chain length of functional monomers, which can be used for selective recovery of gold(III) from WPCBs leaching solution. The quaternary phosphonium adsorbent (PS-TEP) prepared by the nucleophilic substitution reaction between triethyl phosphine with the smallest volume and chloromethylated polystyrene (PS-Cl) exhibited the best gold loading capacity (617.90 mg g-1). The adsorption mechanism of gold(III) on PS-TEP surface mainly involves anion exchange between AuCl4- and Cl- in the adsorbent. The charge level of the H atom closest to -CH2-P+ group directly determines the strength of the interaction between the adsorbent and the gold ion. Multiwfn and VMD programs visually confirm the weak interaction between PS-TEP+ and AuCl4-. After 5 adsorption-stripping cycles, the adsorption rate of gold(III) in solution remained at about 99 %. In addition, PS-TEP exhibited good gold(III) selectivity in both simulated and actual WPCBs gold leaching solutions. These results indicate that the large-particle PS-TEP with high capacity is suitable for selective gold recovery from WPCBs leaching solution.

The causal link between long terminal repeat (LTR) retrotransposon-derived lncRNAs and hepatocellular carcinoma (HCC) remains elusive and whether these cancer-exclusive lncRNAs contribute to the effectiveness of current HCC therapies is yet to explore. Here, we investigated the activation of LTR retrotransposon-derived lncRNAs in a broad range of liver diseases. We found that LTR retrotransposon-derived lncRNAs are mainly activated in HCC and is correlated with the proliferation status of HCC. Furthermore, we discovered that an LTR retrotransposon-derived lncRNA, LINC01446, exhibits specific expression in HCC. HCC patients with higher LINC01446 expression had shorter overall survival times. In vitro and in vivo assays showed that LINC01446 promoted HCC growth and angiogenesis. Mechanistically, LINC01446 bound to serine/arginine protein kinase 2 (SRPK2) and activated its downstream target, serine/arginine splicing factor 1 (SRSF1). Furthermore, activation of the SRPK2-SRSF1 axis increased the splicing and expression of VEGF isoform A165 (VEGFA165). Notably, inhibiting LINC01446 expression dramatically impaired tumor growth in vivo and resulted in better therapeutic outcomes when combined with antiangiogenic agents. In addition, we found that the transcription factor MESI2 bound to the cryptic MLT2B3 LTR promoter and drove LINC01446 transcription in HCC cells. Taken together, our findings demonstrate that LTR retrotransposon-derived LINC01446 promotes the progression of HCC by activating the SRPK2/SRSF1/VEGFA165 axis and highlight targeting LINC01446 as a potential therapeutic strategy for HCC patients.

ETHNOPHARMACOLOGICAL RELEVANCE: Previous studies have revealed that a high-fat diet (HFD) promotes the progression of colorectal cancer (CRC) in close association with disturbances in the intestinal flora and metabolic disorders. Xianglian pill (XLP) is a well-established traditional prescription with unique advantages in controlling intestinal flora imbalance and inflammation. However, its therapeutic effects on HFD-related CRC remain largely unknown. AIM OF THE STUDY: The primary objective of this research was to investigate the anticancer mechanism of XLP in countering HFD-related CRC. MATERIALS AND METHODS: The protective effect of XLP was evaluated using azoxymethane (AOM) and dextran sulfate sodium (DSS)-induced CRC model of mice exposed to a HFD. The degree of colorectal carcinogenesis, including body weight, colon length, and histopathology, was measured in mice treated with XLP and untreated mice. The effect of XLP on gut microbiota and its metabolites was detected using 16S rDNA and liquid chromatography/mass spectrometry analysis. Furthermore, a "pseudo-sterile" mouse model was constructed using antibiotics (Abx) to verify whether the gut microbiota and metabolites play a role in the pathogenesis of CRC. RESULTS: XLP inhibited colorectal tumorigenesis in a dose-dependent fashion. Our findings also highlighted that XLP protected the integrity of the intestinal barrier by reducing the expression of pro-inflammatory cytokines, such as IL-6 and TNF-α, as well as the infiltration of pro-inflammatory macrophages. Mechanistically, XLP inhibited the TLR4/MyD88 pathway. Notably, the XLP treatment increased the proportion of probiotics (particularly Akkermansia) and significantly reduced fecal deoxycholic acid (DCA), a microbiota-derived metabolite of bile acids (BA) closely related to Muribaculaceae. Furthermore, after Abx treatment, XLP showed no clear antitumor effects on CRC. Simultaneously, DCA-supplemented feedings promoted colorectal tumorigenesis and provoked obvious colonic inflammation, M1 macrophage infiltration, and colonic injury. In vitro, the results of RAW-264.7 macrophages and normal intestinal epithelial cells treated with DCA corroborated our in vivo findings, demonstrating consistent patterns in inflammatory responses and intestinal barrier protein expression. CONCLUSION: Our findings suggest that XLP inhibits colorectal cancer associated with HFD via inactivating TLR4/MyD88 by remodeling gut microbiota composition and BA metabolism.

This article presents a comprehensive dataset from the annual reports of China's public-listed companies, the China Stock Market and Accounting Research Database, and the Wind database, focusing on digital transformation and strategic risk taking. This dataset covers 14 years from 2008 to 2021 with 17,089 firm-year observations. Digital transformation is calculated using text mining techniques and keyword frequency analyses based on the firms' annual reports. Then, strategic risk taking is a composite strategic index that combines long-term debt, R&D expenditure, and capital expenditure. The dynamic capability is measured by a comprehensive index that includes three dimensions: absorptive capacity, adaptive capability, and innovation capability. This dataset can serve as a reference base for future studies on the effect of digital transformation on corporate strategic behavior. It can also be integrated into building core competencies to assist managers in identifying the role of dynamic capability.

Thiosulfate gold leaching is one of the most promising green cyanide-free gold extraction processes; however, the difficulty of recovering Au(I) from the leaching system hinders its further development. This study prepared aminoguanidine-functionalized microspheres (AGMs) via a one-step reaction involving nucleophilic substitution between aminoguanidine hydrochloride and chloromethylated polystyrene microspheres and used AGMs to adsorb Au(I) from thiosulfate solutions. Scanning electron microscopy, Brunauer-Emmett-Teller analysis, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy were used to analyze the structure and properties of AGMs. Experiments were designed to investigate the effects of pH, temperature, initial Au(I), and thiosulfate concentrations on the gold adsorption performance of AGMs. Results demonstrated that AGMs can efficiently adsorb Au(I) from thiosulfate solutions in a wide pH range. The adsorption process conforms to the pseudo-second-order kinetic model and Langmuir isotherm model, with a maximum capacity of 22.03 kg/t. Acidic thiourea is an effective desorbent, and after four adsorption-desorption cycles, the adsorption rate of Au(I) by AGMs is 78.63%, which shows AGMs have good cyclic application potential. Based on the results of characterization, experiments, and density functional theory calculations, the mechanism for the adsorption of [Au(S2O3)2]3- on AGMs involves anion exchange. Importantly, AGMs exhibited satisfactory adsorption property for Au(I) in practical Cu2+-NH3(en)-S2O32- systems. This study provided experimental reference for the recovery of Au(I) from thiosulfate solution.

BACKGROUND: Early-onset schizophrenia (EOS) is a type of schizophrenia (SCZ) with an age of onset of < 18 years. An abnormal inflammatory immune system may be involved in the occurrence and development of SCZ. We aimed to identify the immune characteristic genes and cells involved in EOS and to further explore the pathogenesis of EOS from the perspective of immunology. METHODS: We obtained microarray data from a whole-genome mRNA expression in peripheral blood mononuclear cells (PBMCs); 19 patients with EOS (age range: 14.79 ± 1.90) and 18 healthy controls (HC) (age range: 15.67 ± 2.40) were involved. We screened for differentially expressed genes (DEGs) using the Limma software package and modular genes using weighted gene co-expression network analysis (WGCNA). In addition, to identify immune characteristic genes and cells, we performed enrichment analysis, immune infiltration analysis, and receiver operating characteristic (ROC) curve analysis; we also used a random forest (RF), a support vector machine (SVM), and the LASSO-Cox algorithm. RESULTS: We selected the following immune characteristic genes: CCL8, PSMD1, AVPR1B and SEMG1. We employed a RF, a SVM, and the LASSO-Cox algorithm. We identified the following immune characteristic cells: activated mast cells, CD4+ memory resting T cells, resting mast cells, neutrophils and CD4+ memory activated T cells. In addition, the AUC values of the immune characteristic genes and cells were all > 0.7. CONCLUSION: Our results indicate that immune system function is altered in SCZ. In addition, CCL8, PSMD1, AVPR1B and SEMG1 may regulate peripheral immune cells in EOS. Further, immune characteristic genes and cells are expected to be diagnostic markers and therapeutic targets of SCZ.

To cultivate excellent lily germplasms, an interspecific hybrid (LC×SQ-01) was successfully obtained by using a cut-style pollination method in which the rare wild lily Lilium callosum was used as the female parent and the cut flower L. longiflorum 'Snow Queen' was used as the male parent. The morphological features of LC×SQ-01 included height, leaf length, and width, which were observed to be between those of the parents in the tissue-cultured seedlings. The height and leaf length of LC×SQ-01 were more similar to those of the male parent, and the width was between the widths of the parents for field-generated plants. The epidermal cell length and the guard cell and stoma sizes were between those of both parents in tissue-cultured and field-generated plants. In addition, the shapes of the epidermal cells and anticlinal wall in LC×SQ-01 were more analogous to those in the male parent, while the stoma morphology was different from that of both parents. Fourteen pairs of polymorphic SSR primers were identified in both parents, and the validity of LC×SQ-01 was demonstrated by PCR amplification using five pairs of SSR primers. Flow cytometry and root tip squashing assays revealed that LC×SQ-01 was a diploid plant, similar to its parents. Furthermore, the LC×SQ-01 hybrid was more resistant to B. cinerea than its parents, and it also showed much greater peroxidase (POD) and catalase (CAT) activity than the parents. These results lay a foundation for breeding a new high-resistance and ornamental lily variety.

As a sustainable, nontoxic and environmentally friendly cyanide-free gold leaching agent, thiosulfate has been applied to some extent in the field of hydrometallurgy. However, the difficult recovery of gold ions in gold leaching solutions limits further application of thiosulfate gold leaching technology. This study demonstrated the feasibility of gold recovery by sodium dimethyldithiocarbamate (SDD) precipitation and recycling of ammonia and a lixiviant in solution. SDD achieved the purpose of recovering gold by forming granular precipitates with gold ions in solution. It can almost completely recover gold ions in 2.5-17.34 mg/L of gold leaching solution within 1 min at 25 °C, in which a gold recovery capacity of 7.99 kg/t is achieved. The leaching rate of gold ore did not change significantly after recycling the residual ammonia and thiosulfate in the leaching solution after gold recovery by SDD, and its leaching rate basically remained at 81%. The mechanism of SDD recovering Au was determined to involve the ligand exchange of SDD- and Au[(S2O3)2]3-. Moreover, the interaction mechanism between SDD and Au(I) was further validated by density functional theory calculations. Considering its low cost, simple technology, and environmental friendliness, the SDD precipitation process has the potential for large-scale application in gold recovery from thiosulfate gold leaching solutions.

INTRODUCTION: Epithelial barrier disruption is the initial cause of various diseases. We previously reported that acupoint catgut embedding (AE) improves tight junction proteins (TJs) in rats with allergic rhinitis. However, whether AE improves the epithelial barrier in local allergic rhinitis (LAR) remains unknown. METHODS: A total of 36 Sprague Dawley (SD) male rats aged 5-7 weeks were divided into 6 groups with 6 rats each: control group, LAR model group, false acupoint embedding + LAR group, acupoint embedding + LAR group, capsaicin + LAR group, and tunicamycin + acupoint embedding + LAR group. Behavioral observation, ELISA to detect inflammatory factors in nasal lavage fluid and serum IgE, nasal mucosal permeability test, hematoxylin-eosin staining, PCR to detect Substance P (SP), Western blot, and immunofluorescence to detect endoplasmic reticulum stress (ERS) index and TJs were used to investigate the mechanism of AE in LAR. RESULTS: AE improved the symptoms and pathological features of nasal mucosa of LAR rats, reduced the inflammatory factors (IL4, IL5, IL13) of nasal lavage fluid, and showed no significant change in serum IgE levels in all groups. In addition, AE decreased the expression of SP in nasal mucosa of LAR rats, inhibited ERS, increased the expression of tight junction protein, reduced the permeability of nasal mucosa, and improved the function of nasal mucosal barrier. CONCLUSION: This study confirms that AE can improve the nasal mucosal barrier function of LAR by reducing the expression of SP, inhibiting ERS and increasing the expression of TJs, thus enhancing the nasal mucosal barrier function.

Inflammatory depression is a treatment-resistant subtype of depression. A causal role of the gut microbiota as a source of low-grade inflammation remains unclear. Here, as part of an observational trial, we first analyze the gut microbiota composition in the stool, inflammatory factors and short-chain fatty acids (SCFAs) in plasma, and inflammatory and permeability markers in the intestinal mucosa of patients with inflammatory depression (ChiCTR1900025175). Gut microbiota of patients with inflammatory depression exhibits higher Bacteroides and lower Clostridium, with an increase in SCFA-producing species with abnormal butanoate metabolism. We then perform fecal microbiota transplantation (FMT) and probiotic supplementation in animal experiments to determine the causal role of the gut microbiota in inflammatory depression. After FMT, the gut microbiota of the inflammatory depression group shows increased peripheral and central inflammatory factors and intestinal mucosal permeability in recipient mice with depressive and anxiety-like behaviors. Clostridium butyricum administration normalizes the gut microbiota, decreases inflammatory factors, and displays antidepressant-like effects in a mouse model of inflammatory depression. These findings suggest that inflammatory processes derived from the gut microbiota can be involved in neuroinflammation of inflammatory depression.

OBJECTIVES: Insomnia has been the subject of much systematic research because it is a risk factor for a variety of diseases. There is some evidence that gamma sensory stimulation therapy has also been demonstrated to improve sleep quality for people with Alzheimer's disease. However, it is unclear whether this method is effective for treating insomnia. The principal objective of this project was to investigate the efficacy and safety of gamma sensory flicker in improving the sleep quality of insomnia patients. METHODS: Thirty-seven participants with insomnia were recruited for this prospective observational study. For a duration of 8 weeks, participants were exposed to flicker stimulation through a light and sound device. RESULTS: During the main phase of the study, adherence rates averaged 92.21%. Additionally, no severe adverse events were reported for flicker treatment. Analysis of sleep diaries indicated that 40 Hz flickers can enhance sleep quality by reducing sleep onset latencies, and arousals, and increasing total sleep duration. CONCLUSIONS: Gamma sensory flicker improves sleep quality in people suffering from insomnia.
