RESUMO

Chromium (Cr) is a heavy metal with a high toxicity and pathogenicity. Microbial reduction is an effective strategy to remove Cr(VI) at contaminated sites but suffers from the low populations and activities of Cr-reducing microorganisms in soils. This study proposed an in situ sonoporation-mediated gene transfer approach, which improved soil Cr(VI) reduction performance by delivering exogenous Cr-transporter chrA genes and Cr-reducing yieF genes into soil microorganisms with the aid of ultrasound. Besides the increasing populations of Cr-resistant bacteria and elevated copy numbers of chrA and yieF genes after sonoporation-mediated gene transfer, three new Cr-reducing strains were isolated, among which Comamonas aquatica was confirmed to obtain Cr-resistant capability. In addition, sonoporation-mediated gene transfer was the main driving force significantly shaping soil microbial communities owing to the predominance of Cr-resistant microbes. This study pioneered and evidenced that in situ soil sonoporation-mediated gene transfer could effectively deliver functional genes into soil indigenous microbes to facilitate microbial functions for enhanced bioremediation, e.g., Cr-reduction in this study, showing its feasibility as a chemically green and sustainable remediation strategy for heavy metal contaminated sites.

Chromium (Cr) can enter groundwater through rainfall infiltration and significantly affects human health. However, the mechanisms by which soil colloids affect chromium transport are not well investigated. In this study, column experiments were conducted to simulate the chromium (Cr) transport mechanism in two typical soils (humic acid + cinnamon soil and montmorillonite + silt) in the vadose zone of a contaminated site and the effects of acid rain infiltration conditions. The results showed that Mt colloids have less influence on Cr. The fixation of Cr by colloid mainly occurs in the cinnamon soil layer containing HA colloid. The adsorption efficiency of Cr was increased by 12.8% with the addition of HA. In the HA-Cr system, the introduction of SO42- inhibited the adsorption of Cr, reducing the adsorption efficiency from 31.4% to 24.4%. The addition of Mt reduced the adsorption efficiency of Cr by 15%. In the Mt-Cr system, the introduction of SO42- had a promoting effect on Cr adsorption, with the adsorption efficiency increasing from 4.4% to 5.1%. Cr release was inhibited by 63.88% when HA colloid was present, but the inhibition owing to changes in acidity was only 14.47%. Mt colloid promotes Cr transport and increases the leaching rate by 2.64% compared to the absence of Mt. However, the effect of acidity change was not significant. Intermittent acid rain will pose a higher risk of pollutant release. Among the influencing factors, the type of colloid had the most significant influence on the efficiency of Cr leaching. This study guides the quantitative assessment of groundwater pollution risk caused by Cr in the vadose zone.

The presence of residual Cr(VI) in soils causes groundwater contamination in aquifers, affecting the health of exposed populations. Initially, permeable reactive barriers(PRB) effectively removed Cr(VI) from groundwater. However, as PRB clogging increased and Cr(VI) was released from upstream soils, the contamination plume continued to spread downstream. By 2020, the level of contamination in the downstream was nearly identical to that in the upstream. The study results show that during normal operation, the PRB can successfully remove Cr(VI) from contaminated groundwater and reduce the carcinogenic and non-carcinogenic risks to humans from the downstream side of groundwater. However, the remediated groundwater still poses an unacceptable risk to human health. The sensitivity analysis revealed that the concentration of the pollutant was the most sensitive parameter and interacted significantly with other factors. Ultimately, it was determined that the residual Cr(VI) in the soil of the study region continues to contaminate the groundwater and constitutes a serious health danger to residents in the vicinity. As remediated groundwater still poses a severe threat to human health, PRB may not be as effective as people believe.

The global outbreak of coronavirus infectious disease-2019 (COVID-19) draws attentions in the transport and spread of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) in aerosols, wastewater, surface water and solid wastes. As pathogens eventually enter the subsurface system, e.g., soils in the vadose zone and groundwater in the aquifers, they might survive for a prolonged period of time owing to the uniqueness of subsurface environment. In addition, pathogens can transport in groundwater and contaminate surrounding drinking water sources, possessing long-term and concealed risks to human society. This work critically reviews the influential factors of pathogen migration, unravelling the impacts of pathogenic characteristics, vadose zone physiochemical properties and hydrological variables on the migration of typical pathogens in subsurface system. An assessment algorithm and two rating/weighting schemes are proposed to evaluate the migration abilities and risks of pathogens in subsurface environment. As there is still no evidence about the presence and distribution of SARS-CoV-2 in the vadose zones and aquifers, this study also discusses the migration potential and behavior of SARS-CoV-2 viruses in subsurface environment, offering prospective clues and suggestions for its potential risks in drinking water and effective prevention and control from hydrogeological points of view.

The microbial community and functional flora in rare earth mining areas are correlated, but the characteristics and metabolic pathways of pollutant in such mining areas are still poorly known. The heavy metals, rare earth elements, and microorganisms present after mining of rare earth mine sites were analyzed. After mining, all sampling sites exhibited low pH and low total organic carbon levels, accompanied by high iron and aluminum concentrations. The development of vegetation is closely related to the development of microorganisms. In the complex environment of rare earth mining areas, Proteobacteria exhibit an absolute competitive advantage. During mine environmental recovery, the relative abundances of Acidobacteria and Chloroflexi will increase markedly, and with further restoration the relative abundance of Firmicutes will gradually decrease. Many genera of bacteria related to the N cycle and heavy metal metabolism were detected in the study area, indicating the important metabolic pathways for ammonia nitrogen and heavy metals in rare earth mining areas. Bacterial genera that promote plant nitrogen fixation also occur in the area, further revealing the nitrogen cycle. This research is important for health assessment and recovery of rare earth mines.

As restricted CA-4 analogues, a novel series of [1,2,4]triazolo[1,5-a]pyrimidines possessing 3,4,5-trimethoxylphenyl groups has been achieved successfully via an efficient one-pot three-component reaction of 3-(3,4,5-trimethoxyphenyl)-1H-1,2,4-triazol-5-amine, 1,3-dicarbonyl compounds and aldehydes. Initial biological evaluation demonstrated some of target compounds displayed potent antitumor activity in vitro against three cancer cell lines. Among them, the most highly active analogue 26 inhibited the growth of HeLa, and A549 cell lines with IC50 values at 0.75, and 1.02 µM, respectively, indicating excellent selectivity over non-tumoural cell line HEK-293 (IC50 = 29.94 µM) which suggested that the target compounds might possess a high safety index. Moreover, cell cycle analysis illustrated that the analogue 26 significantly induced HeLa cells arrest in G2/M phase, meanwhile the compound could dramatically affect cell morphology and microtubule networks. In addition, compound 28 exhibited potent anti-tubulin activity with IC50 values of 9.90 µM, and molecular docking studies revealed the analogue occupied the colchicine-binding site of tubulin. These observations suggest that [1,2,4]triazolo[1,5-a]pyrimidines represent a new class of tubulin polymerization inhibitors and well worth further investigation aiming to generate potential anticancer agents.
