RESUMO

Nine commercial kits of "captured" and "indirect" format ELISA assay for the detection of specific IgM and IgG in sera of patients with measles were compared to each other. 72 sera specimens from typical medium-severity cases from a measles outbreak (2010) were collected on the 5-6th day after the rash onset. IgM was detected with "capture" tests (Vecto-Measles IgM, Vector Best, Measles IgM capture EIA, Microimmune Ltd) close to 100% of cases, irrespectively to the age and the initial vaccination status of the patients. The IgM result was negative in 23.6% by average while investigating using "indirect" format tests (Enzygnost Anti-Measles Virusll/IgM, Siemens; Anti-Measles Viruses ELISA (IgM), Eurominimum, Virion-Serion IgM (GmbH). These cases were in adults, the majority of which had 1-2 vaccinations in the past. The analysis of the presented data shows high correlation connection between the tests used and high confidence level for OD IgM and IgG of the sera of the patients with the primary and secondary immune response.

119 specimens of blood sera collected from measles cases with different vaccination history (aged 4 months to 48 years) on 5th-6th days after rash onset were Investigated using EIA. The obtained results showed that the primary immune response (PIR) was developed in 59 patients; the secondary immune response (SIR) was developed in 60 patients with a significant increase in the specific high avidity IgG (22.34 IU/ml +/- 3.2). The specific IgM were detected in 100% cases studied with capture ELISA in both previously vaccinated and unvaccinated individuals of different age. The specific IgM were detected by indirect ELISA in 100% cases in unvaccinated patients, while IgM positive sera was defined only in 23.3% of individuals with SIR. It was concluded that measles virus infection in previously vaccinated and unvaccinated adults had clinical differences. The role of patients with SIR in virus transmission was discussed.

RESUMO

33 wild poliovirus strains isolated on the territory of the former USSR were subjected to the comparative sequence analysis of the 150 bp genome fragment VP1/2A on the junction of the genome regions coding basic capsid protein VP1 and viral protease 2A. The dendrograms characterizing the similarity of nucleotide sequences revealed the existence of 3 geographical genotypes (geotypes) of wild poliovirus strains of type 1 (A, G, T) and broad geotype (C) of wild poliovirus strains of type 3. The comparison of the analyzed strains with strains circulating in the neighboring countries at the same period provided information on their genetic relationship. The data thus obtained made it possible to establish the pathways of the transmission of wild poliovirus strains in the common epidemic area of the Russia u CIS.

One hundred and fifty nucleotide-long VP1/2A junction regions were sequenced in the RNAs of 19 strains isolated in 1990-1991 from patients with paralytic poliomyelitis in different regions of the former USSR. On the basis of the alignments of these sequenced RNAs, four pairs of 19-25 base-long oligodeoxynucleotide PCR primers were designed capable of detecting polio RNAs in isolated strains and of discriminating between polio genotypes. PCR with 520 polio virus strains isolated from patients, normal subjects, and environmental objects showed 428 of these strains to be related to Sabin's vaccine strains, whereas the rest were referred to A (30), T (24), and G (1) genotypes of serotype 1 and to C-genotype (37) of serotype 3. The designed primers were highly specific and did not cross-react between themselves and with primers specific for Sabin's vaccine strains in PCR.

RESUMO

Thirty-two hantavirus strains and 8 samples of lung tissue from rodents collected in different regions of Russia have been examined by molecular biological methods. Two methodological approaches have been employed for the study of genetic relationships between the viruses: nested PCR assay and common RT-PCR with subsequent direct sequencing of 200 and 365 base pair of G2 protein encoding regions of M-segment, respectively, and the resultant sequences were compared with those of the prototype hantavirus. The study revealed a mosaic pattern of distribution of different hantavirus genotypes on the territory of Russia.

RESUMO

Eleven virus strains isolated from poliomyelitis patients in Moscow in 1973-1986 were analyzed by the method of oligonucleotide mapping of RNA. The genome of the isolates showed considerable similarity to the genomes of Sabin's vaccine strains and mainly to the vaccine strain of antigenic type 2. The conclusion was made that the sporadic cases of poliomyelitis registered in this region were etiologically linked with the vaccine strains of poliomyelitis virus. Only in one case the disease appeared in the recipient of the vaccine, in all other cases the patients were infected through contacts.

RESUMO

Oligonucleotide maps of some poliovirus type 2 strains isolated from polio cases, while being clearly related to that of the Sabin vaccine type 2 strain, exhibited, nevertheless, marked differences from the reference (vaccine) map. Several large oligonucleotides derived from 4 such strains were subjected to enzymatic sequencing. The results strongly suggest that all of them were intertypic recombinants between the Sabin strains. The 5'-parts of the genomes of these strains were derived from the type 2 vaccine whereas the 3'-parts were of type 1 (in 2 strains) or type 3 (in other 2 strains) origin.

RESUMO

Oligonucleotide mapping of the RNA genome was applied for analysis of poliovirus clinical isolates in Byelorussia in 1967-1985. All the maps have been clearly shown to resemble the reference maps of the corresponding vaccine strains. Examination of the oligonucleotide maps of poliovirus isolates provides a measure of distribution of some of the mutations over the viral genome.

RESUMO

Twelve poliovirus isolates of serotype 3 from patients with paralytic poliomyelitis have been analyzed by oligonucleotide mapping of the viral genomes. All the studied strains were isolated from patients in different regions of the Moldavian SSR in 1982. The maps of all isolates are similar but they do not practically possess any large oligonucleotides characteristic of the vaccine strain of type 3 poliovirus. It is concluded that a wild neurovirulent strain of type 3 poliovirus, that circulated in 1982 in the Moldavian SSR was the cause of paralytic poliomyelitis cases. All the studied isolates are suggested to have been derived relatively recently from the common ancestor.
