RESUMO

Chronic stress induces a variety of physiological and/or psychological abnormalities, including hyperalgesia. Researchers have discovered sex differences in the prevalence of stress-induced hyperalgesia (SIH) in recent years. Sex differences may be one of the reasons for the heterogeneity of susceptibility to stress-related diseases. In this review, the potential mechanisms of sex differences in SIH are discussed, such as hypothalamus-pituitary-adrenal axis responses, regulation of sex hormones, and immune system responses.

BACKGROUND: Cimicifugae Rhizoma, known in Chinese as Shengma, is a common medicinal material in traditional Chinese medicine (TCM), mainly used for treating wind-heat headaches, sore throat, uterine prolapse, and other diseases. OBJECTIVES: An approach using a combination of ultra-performance liquid chromatography (UPLC), MS, and multivariate chemometric methods was designed to assess the quality of Cimicifugae Rhizoma. METHODS: All materials were crushed into powder and the powdered sample was dissolved in 70% aqueous methanol for sonication. Chemometric methods, including hierarchical cluster analysis (HCA), principal component analysis (PCA), and orthogonal partial least-squares discriminant analysis (OPLS-DA), were adopted to classify and perform a comprehensive visualization study of Cimicifugae Rhizoma. The unsupervised recognition models of HCA and PCA obtained a preliminary classification and provided a basis for classification. In addition, we constructed a supervised OPLS-DA model and established a prediction set to further validate the explanatory power of the model for the variables and unknown samples. RESULTS: Exploratory research found that the samples were divided into two groups, and the differences were related to appearance traits. The correct classification of the prediction set also demonstrated a strong predictive ability of the models for new samples. Subsequently, six chemical makers were characterized by UPLC-Q-Orbitrap-MS/MS, and the content of four components was determined. The results of the content determination revealed the distribution of representative chemical markers caffeic acid, ferulic acid, isoferulic acid, and cimifugin in two classes of samples. CONCLUSIONS: This strategy can provide a reference for assessing the quality of Cimicifugae Rhizoma, which is significant for the clinical practice and QC of Cimicifugae Rhizoma. HIGHLIGHTS: The HCA, PCA and OPLS-DA models visually classify Cimicifugae Rhizoma by appearance traits and obtain the chemical markers that influence the classification. The training and prediction sets were built to demonstrate the accuracy of the classification. Advanced UPLC-Q-Orbitrap-MS/MS technology provides powerful elucidation of critical chemical markers.

Introduction: The B3 transcription factor has been identified in Arabidopsis thaliana, Oryza sativa, and Solanum lycopersicum, among other species. This family of transcription factors regulates seed growth, development, and stress. Cannabis is a valuable crop with numerous applications; however, no B3 transcription factors have been identified in this plant. Materials and Methods: The cannabis B3 gene family was identified and analyzed using bioinformatics analysis tools, such as the NCBI database, plantTFDB website, TBtools, and MEGA software. Quantitative real-time polymerase chain reaction (qRT-PCR) experiments were used to confirm its function. Results: The cannabis B3 family contains 65 members spread across 10 chromosomes. The isoelectric point ranged from 10.03 to 4.65, and the molecular weight ranged from 99,542.88 to 14,310.9 Da. Most of the members were found in the nucleus. The upstream promoter region of the gene contains a variety of cis-acting elements related to the stress response. RNA-seq data and qRT-PCR results showed that CsB3 genes were expressed differently in five organs of female Diku plants and in glandular hairs of nine distinct types of female cannabis inflorescences. Collinearity analysis revealed that there were more homologous genes between cannabis and dicotyledons than monocotyledonous plants, which was consistent with the evolutionary relationship. Conclusions: Hormones and external environmental factors might influence CsB3 expression. Furthermore, some genes such as CsB3-02, CsB3-07, CsB3-50, CsB3-62, and CsB3-65 may participate in cannabis growth and development and play a role in secondary metabolite synthesis. This study provides a solid foundation for further research into the gene function of the cannabis B3 family.

OBJECTIVE: To investigate the effect of Bazi Granules on sperm quality in male rats with oligoasthenozoospermia (OAS) induced by multi-glycosides of tripterygium wilfordii (GTW). METHODS: Thirty-six SD male rats were randomly divided into six groups of equal number: normal control, OAS model control, Wuziyanzong Pills (WYP), and low-, medium- and high-dose Bazi. The OAS model was established in the rats except those of the normal control group by intragastrical delivery of GTW at 30 mg/kg/d for 40 days. From the 41st day, the animals of the normal and OAS model control groups were fed with distilled water, those of the WYP group treated by gavage with WYP at 1.02 g/kg/d, and those of the low-, medium- and high-dose Bazi groups intragastically given Bazi Granules 3 (5.27 g/kg), 6 (10.54 g/kg) and 12 (21.08 g/kg) times, respectively, that of the human-equivalent dose. Semen parameters and the contents of superoxide dismutase (SOD) and malondialdehyde (MDA) in the testis tissue were determined after 28 days of treatment. RESULTS: After treatment, the rats of the of the high-, medium- and low-dose Bazi groups, compared with the OAS model controls, showed significant increases in sperm concentration (ï¼»1 050.71 ± 203.71ï¼½, ï¼»1 370.06 ± 166.01ï¼½ and ï¼»1 302.53 ± 476.51ï¼½ vs ï¼»617.01 ± 237.08ï¼½ ×106/ml, P < 0.05), sperm motility (ï¼»0.56 ± 0.24ï¼½%, ï¼»0.73 ± 0.14ï¼½% and ï¼»0.70 ± 0.23ï¼½% vs ï¼»0.07 ± 0.05ï¼½%, P < 0.05), sperm average path velocity (ï¼»85.71 ± 30.35ï¼½, ï¼»83.83 ± 10.31ï¼½ and ï¼»75.06 ± 19.70ï¼½ vs ï¼»43.45 ± 38.74ï¼½ µm/s, P < 0.05), sperm curvilinear velocity (ï¼»101.76 ± 23.28ï¼½, ï¼»119.60 ± 21.22ï¼½ and ï¼»102.11 ± 32.89ï¼½ vs ï¼»53.63 ± 47.91ï¼½ µm/s, P < 0.05), sperm straight line velocity (ï¼»62.75 ± 7.63ï¼½, ï¼»67.80 ± 5.05ï¼½ and ï¼»64.11 ± 12.03ï¼½ vs ï¼»40.18 ± 36.86ï¼½ µm/s, P < 0.05), and the SOD level (ï¼»380.23 ± 75.07ï¼½, ï¼»349.53 ± 97.48ï¼½ and ï¼»415.07 ± 72.01ï¼½ vs ï¼»304.62 ± 27.17ï¼½ U/mg, P < 0.05), but a remarkable decrease in the MDA level (ï¼»0.33 ± 0.16ï¼½, ï¼»0.22 ± 0.05ï¼½ and ï¼»0.34 ± 0.22ï¼½ vs ï¼»0.73 ± 0.20ï¼½ nmol/mg, P < 0.05). CONCLUSIONS: Bazi Granules can significantly improve the sperm quality of OAS rats, which may be related to its abilities of repairing oxidative stress injury and enhancing oxidation resistance.

Vascular endothelial insulin resistance (IR) is a critically initial factor in cardiocerebrovascular events resulted from diabetes and is becoming a worldwide public health issue. Thiazolidinediones (TZDs) are clinical insulin-sensitizers acting through a canonical peroxisome proliferator-activated receptor gamma (PPARγ)-dependent insulin trans-activation pathway. However, it remains elusive whether there are other mechanisms. In current study, we investigated whether TZDs improve endothelial IR induced by high glucose concentration or hyperglycemia via a non-canonical PPARγ-dependent nuclear factor-kappa B (NF-κB) trans-repression pathway. Our results showed that pre-treatment with TZDs dramatically decrease the susceptibility of endothelial cell to IR, while post-treatment notably improve the endothelial IR both in vitro and in vivo. Moreover, TZDs substantially increase the levels of endothelial nitric oxide synthase (eNOS) and inhibitory κB alpha (IκBα), whereas decrease those of the phosphorylated inhibitory κB kinase alpha/beta (phosphor-IKKα/ß) and the cytokines including tumor necrosis factor alpha (TNFα), interleukin-6 (IL-6), soluble intercellular adhesion molecule-1 (sICAM-1) and soluble vascular cellular adhesion molecule-1 (sVCAM-1), suggesting that TZDs act indeed through a PPARγ-dependent NF-κB trans-repression pathway. These findings highlighted a non-canonical mechanism for TZDs to ameliorate endothelial IR which might provide a potential strategy to prevent and treat the diabetic vascular complications clinically.
