RESUMO

The tendency of global urban expansion to be slope climbing has partly become possible with scarce cropland resources in plains. However, the scientific understanding of the quantity, intensity, pattern, and effect of the slope climbing of urban expansion (SCE) is minimal globally. In this study, we have attempted to quantify and evaluate global SCE from Suomi National Polar-orbiting Partnership (SNPP)-Visible Infrared Imaging Radiometer Suite (VIIRS)-like data and other auxiliary data. Results revealed that global SCE areas unevenly increased from 22,760 km2 to 90,720 km2 from 2000 to 2020, with an annual growth rate of 21.72%, in which low-environment cost type areas increased from 21,550 km2 to 84,010 km2 while high-environment cost type (HEC) areas increased from 1210 km2 to 6710 km2. One remarkable phenomenon is that China's SCE areas in 2020 were more than 11 times those in 2000. In addition, global SCE intensity increased by about 3.4-fold from 2000 to 2020 and the rapid growth of HEC intensity is concentrated in Asia and North America. SCE is mostly affected by urban population growth and terrain. Economic development also promotes its development to a certain extent. We also noted that global SCE potentially made a considerable contribution to saved cropland, saving about 46,747 km2 with a theoretical increased grain yield of 25,020 × 103 t. Our study provides timely and transparent monitoring of global SCE and offers new insights into sustainable urban development.

Land urbanization (LU) and population urbanization (PU) maintain the nature of spatiotemporal heterogeneity in China. As a municipality directly administered by the central government in the mode of "large cities and large rural areas", Chongqing's urbanization process is the epitome of China's urbanization process. This paper examines the spatiotemporal variability of LU and PU in Chongqing on the basis of nighttime light data, the elasticity coefficient of the coupling relationship, and GWR. The results show that (1) the urban land and urban population in Chongqing grew notably from 2008 to 2018, with average annual growth rates of 9.4% and 2.3%, respectively. (2) The coupling coordination coefficient of LU and PU in Chongqing was 0.24, and the total number of districts and counties with uncoordinated development increased, but the overall uncoordinated situation gradually improved over the period. (3) The influence of PU on LU in each district and county increased year by year, and it showed a decreasing trend from southwest to northeast in Chongqing, which indicates that LU was increasingly adapted to the construction needs of PU. The gap between LU and PU widened due to the household registration system, land fiscal policies and other policies. After the reform of the household registration system and the adjustment of new pilot policies targeting the construction of new-type urbanization, the coupling relationship between LU and PU was gradually improving to the coordinated mode. The findings indicate that Chinese urban areas should adhere to the principle of new-type urbanization construction and carry out scientific land planning strategies, strictly controlling land expansion to promote the reasonable development of population growth.

Neural cell adhesion molecule (NCAM) is involved in cell multi-directional differentiation, but its role in osteoblast differentiation is still poorly understood. In the present study, we investigated whether and how NCAM regulates osteoblastic differentiation. We found that NCAM silencing inhibited osteoblast differentiation in pre-osteoblastic MC3T3-E1 cells. The function of NCAM was further confirmed in NCAM-deficient mesenchymal stem cells (MSCs), which also had a phenotype with reduced osteoblastic potential. Moreover, NCAM silencing induced decrease of Wnt/ß-catenin and Akt activation. The Wnt inhibitor blocked osteoblast differentiation, and the Wnt activator recovered osteoblast differentiation in NCAM-silenced MC3T3-E1 cells. We lastly demonstrated that osteoblast differentiation of MC3T3-E1 cells was inhibited by the PI3K-Akt inhibitor. In conclusion, these results demonstrate that NCAM silencing inhibited osteoblastic differentiation through inactivation of Wnt/ß-catenin and PI3K-Akt signaling pathways.

BACKGROUND: Microbial metabolism confounds antibiotic efficacy. However, information regarding effect of metabolism on cephalosporin antibiotics-mediated killing and Vibrio spp is largely absence, although the drugs are widely used in clinic and the bacteria are pathogens to both human and aquaculture animals. PURPOSE: This study explores the metabolome of cephalosporin antibiotic-resistant Vibrio alginolyticus and analyzes the role of bacterial metabolism in drug and multidrug-resistance. RESULTS: The metabolomes of isogenic ceftazidime-resistant V. alginolyticus (VA-RCAZ) and ceftazidime-sensitive V. alginolyticus (VA-S) were analyzed using gas chromatography -mass spectrometry. The metabolome of VA-RCAZ is characterized by inefficient respiration, an inefficient pyruvate cycle (P cycle), increased biosynthesis of fatty acids and decreased membrane proton motive force. This hypothesis was confirmed by the fact that furfural and malonate, inhibitors of pyruvate dehydrogenase and succinate dehydrogenase (P cycle enzymes), respectively, increased resistance of VA-RCAZ to antibiotics, while exposure to triclosan, to inhibit biosynthesis of fatty acids, decreased resistance. CONCLUSION: These results contribute to our understanding of mechanisms of bacterial antibiotic-resistance and may lead to more effective approaches to treat, manage or prevent infections caused by antibiotic-resistant pathogens including those of the Vibrio species.

The emergence of multidrug-resistant bacteria presents a severe threat to public health and causes extensive losses in livestock husbandry and aquaculture. Effective strategies to control such infections are in high demand. Enhancing host immunity is an ideal strategy with fewer side effects than antibiotics. To explore metabolite candidates, we applied a metabolomics approach to investigate the metabolic profiles of mice after Klebsiella pneumoniae infection. Compared with the mice that died from K. pneumoniae infection, mice that survived the infection displayed elevated levels of l-valine. Our analysis showed that l-valine increased macrophage phagocytosis, thereby reducing the load of pathogens; this effect was not only limited to K. pneumoniae but also included Escherichia coli clinical isolates in infected tissues. Two mechanisms are involved in this process: l-valine activating the PI3K/Akt1 pathway and promoting NO production through the inhibition of arginase activity. The NO precursor l-arginine is necessary for l-valine-stimulated macrophage phagocytosis. The valine-arginine combination therapy effectively killed K. pneumoniae and exerted similar effects in other Gram-negative (E. coli and Pseudomonas aeruginosa) and Gram-positive (Staphylococcus aureus) bacteria. Our study extends the role of metabolism in innate immunity and develops the possibility of employing the metabolic modulator-mediated innate immunity as a therapy for bacterial infections.

We have recently reported that the survival of tilapia, Oreochromis niloticus, during Edwardsiella tarda infection is tightly associated with their metabolome, where the survived O. niloticus has distinct metabolomic profile to dying O. niloticus. Glucose is the key metabolite to distinguish the survival- and dying-metabolome. More importantly, exogenous administration of glucose to the fish greatly enhances their survival for the infection, indicating the functional roles of glucose in metabolome repurposing, known as reprogramming metabolomics. However, the underlying information for the reprogramming is not yet available. Here, GC/MS based metabolomics is used to understand the mechanisms by which how exogenous glucose elevates O. niloticus, anti-infectious ability to E. tarda. Results showed that exogenous glucose promotes stearic acid and palmitic acid biosynthesis but attenuates TCA cycle to potentiate O. niloticus against bacterial infection, which is confirmed by the fact that exogenous stearic acid increases immune protection in O. niloticus against E. tarda infection in a manner of Mx protein. These results indicate that exogenous glucose reprograms O. niloticus anti-infective metabolome that characterizes elevation of stearic acid and palmitic acid and attenuation of the TCA cycle. Therefore, our results proposed a novel mechanism that glucose promotes unsaturated fatty acid biosynthesis to cope with infection, thereby highlighting a potential way of enhancing fish immunity in aquaculture.
