RESUMO

Porphyrin-based porous organic polymer (POP) with uniformly immobilized platinum nanoparticles (Pt NPs) were designed and synthesized, and it was demonstrated that such nanocomposites (Pt/POP) have oxidase-like activity. Surprisingly, Hg2+ significantly enhanced the oxidase-like activity of Pt/POP. The enhancement was attributed to the capture of Hg2+ by the thioether group in Pt/POP and the subsequent redox reaction of Hg2+ with Pt NPs, accelerating the electron transfer. In the presence of Hg2+, Pt/POP catalyzed the colorless 3,3',5,5'-tetramethylbenzidine (TMB) to turn blue rapidly and changed its absorbance at 652 nm. Based on this, a fast-response colorimetric sensor was constructed for the sensitive detection of Hg2+ with a linear range of 0.2-50 µM and a detection limit of 36.5 nM. Importantly, Pt/POP can be used as an adsorbent for the efficient removal of Hg2+ with a removal efficiency as high as 99.4%. This work provides a valuable strategy for colorimetric detection and efficient removal of Hg2+.

Multistimuli responsiveness and programmable shape recovery are crucial for soft actuators in soft robotics, electronics, and wearables. However, existing strategies for actuation cannot attain power-free shape retention after removing the external energy supply. Here, a self-assembled density deposition method was developed to fabricate an electrothermal-NIR-magnetic triple-response actuator which was composed of cellulose nanofiber/poly(vinyl alcohol)/liquid metal (CNF/PVA/LM) and magnetic polydimethylsiloxane (MPDMS) layer. Interestingly, the large deformation can be controllably fixed and the temporary configuration will be programmable recovered under a magnetic field due to the thermal-plastic transferring behavior of the CNF/PVA/LM. Rolling robot prepared based on soft actuators exhibits good ability to avoid obstacles. In addition, the object handling and programmable release capabilities of the carrier robots demonstrate that this actuation approach will contribute to a better understanding of how to more rationally utilize various stimuli for application purposes.

The proven high carcinogenicity to humans and high destructive force to the environment determine the extreme urgency of eliminating hypertoxic Cr(VI) in water bodies. Herein, a route of room temperature synthesis and secondary grafting was proposed to fabricate graphene oxide-based nanoadsorbent co-functionalized with polydopamine and branched polyethyleneimine (GOPP) to remove Cr(VI). The flexible decoration of polydopamine and polyethyleneimine on GO flakes could gradually enhance the amount of N-containing functional groups and realize selective removal of Cr(VI) with the maximum experimental adsorption capacity of 564.7 mg/g, displaying a significantly high separation factor against alkali metal, alkaline earth metal, and other transition metal ions. Various combination mechanisms, such as electrostatic attraction, reduction, complexation, and hydrogen bonding, were demonstrated to be involved in the adsorption process of Cr(VI) by XPS, ESP, and DFT calculations. And the interaction energies of the five protonated configurations of primary amine, tertiary amine, secondary amine, imine, and secondary amine on the ring with HCrO4- were: -22.66, -12.08, -24.92, -24.26, -27.64 kcal/mol. In the actual industrial wastewater study, a Cr(VI) removal rate of 85.8% was realized. This work provided a viable idea for the elimination of Cr(VI) and was expected to be applied in the field of wastewater treatment.

A hollow porphyrin-based porous organic polymer (H-Fe-POP) was prepared for rapid and sensitive colorimetric determination of Cr(VI), which exhibited excellent dual enzyme-like activities, including oxidase-like and peroxidase-like activities. Due to the specific binding of 8-hydroxyquinoline (8-HQ) to Cr(VI), 3,3',5,5'-tetramethylbenzidine (TMB) was liberated, and TMB was oxidized to blue ox-TMB catalyzed by H-Fe-POP. Based on the excellent oxidase-like activity of H-Fe-POP, an ultra-fast colorimetric platform for the detection of Cr(VI) was constructed, allowing the quantification of Cr(VI) in the range 2-130 µM within 30 s with a detection limit of 0.23 µM. Importantly, the sensor can accurately determine Cr(VI) in industrial wastewater, indicating its high potential for environmental monitoring.

BACKGROUND & AIMS: Human neutrophil peptides (HNP)-1, -2 and -3 are the most abundant proteins in neutrophil azurophilic granules and are rapidly released via neutrophil degranulation upon activation. The aims of our study were to assess the role of HNP1-3 as biomarkers of disease severity in patients with decompensated cirrhosis and their value in predicting short-term mortality. METHODS: In this study, 451 patients with acutely decompensated cirrhosis (AD) were enrolled at the two medical centres. Overall, 281 patients were enrolled as the training cohort from October 2015 to April 2019, and 170 patients were enrolled as the validation cohort from June 2020 to February 2021. Plasma HNP1-3 levels were measured using enzyme-linked immunosorbent assay (ELISA). RESULTS: Plasma HNP1-3 increased stepwise with disease severity (compensated cirrhosis: 0.3 (0.2-0.4); AD without acute-on-chronic liver failure (ACLF): 1.9 (1.3-4.8); ACLF-1: 2.3 (1.8-6.1); ACLF-2: 5.6 (2.9-12.3); ACLF-3: 10.3 (5.7-17.2) ng/ml). From the multivariate Cox regression analysis, HNP1-3 emerged as independent predictors of mortality at 30 and 90 days. Similar results were observed in the subgroup analysis. On ROC analysis, plasma HNP1-3 showed better predictive accuracy for 30- and 90-day mortality (area under the receiver operating characteristic (AUROC) of 0.850 and 0.885, respectively) than the neutrophil-to-lymphocyte ratio (NLR) and similar accuracy as end-stage liver disease (MELD: 0.881 and 0.874) and chronic liver failure-sequential organ failure (CLIF-SOFA: 0.887 and 0.878). CONCLUSIONS: Plasma HNP1-3 levels were closely associated with disease severity and might be used to identify patients with AD at high risk of short-term mortality.

Soft magnetic actuators with programmable structure design and controllable deformation ability based on 3D printing technology have attracted extensive attention. In this paper, a novel 3D printing strategy is developed to manufacture the ultraflexible magnetic actuator, in which the printed material is composed of magnetic particles and thermoplastic rubber materials. Different from the traditional fused deposition printing, this printing strategy introduces screw extrusion technology to the heating components of the printer to overcome the problem of filament buckling in the flexible material. Thus, the tensile modulus of the printed products can be reduced to as low as ≈2 MPa. Based on the above method, biomimetic magnetic actuators of the sucker and the pump are constructed for adhering and releasing object and pumping liquid. The contraction performance of the magnetic actuator is studied via a series of experiments and the magnetic field-induced deformation is analyzed by the multiphysics-based finite element model. This work proves that ultraflexible magnetic actuators fabricated by this 3D printing strategy show broad prospects in the fields of soft robotics and bionics.

Background and Aims: Patients with decompensated HBV-related liver cirrhosis (HBV D-LC) showed compromised immune responses, which manifested as a proneness to develop infections and hyporesponsiveness to vaccines, resulting in accelerated disease progression. The alterations in T cell-dependent B cell responses in this pathophysiological process were not well understood. This study aimed to investigate T cell-dependent B cell responses in this process and discuss the mechanism from the perspective of metabolism. Methods: Changes in phenotypes and subsets of peripheral B cells between HBV D-LC patients and healthy controls (HCs) were compared by flow cytometry. Isolated B cells were activated by coculture with circulating T follicular (cTfh) cells. After coculture, the frequencies of plasmablasts and plasma cells and immunoglobin levels were analyzed. Oxidative phosphorylation (OXPHOS) and glycolysis were analyzed by a Seahorse analyzer. Mitochondrial function and the AKT/mTOR pathway were analyzed by flow cytometry. Results: The proliferation and differentiation capacities of B cells after T cell stimulation were impaired in D-LC. Furthermore, we found that B cells from D-LC patients showed reductions in OXPHOS and glycolysis after activation, which may result from reduced glucose uptake, mitochondrial dysfunction and attenuated activation of the AKT/mTOR pathway. Conclusions: B cells from HBV D-LC patients showed dysfunctional energy metabolism after T cell-dependent activation. Understanding the regulations of B cell metabolic pathway and their changes may provide a new direction to rescue B cell hyporesponsiveness in patients with HBV D-LC, preventing these patients be infected and improving sensitivity to vaccines.
