RESUMO

Wheat is the major staple food in Western Europe and an important source of energy, protein, dietary fibre, minerals, B vitamins and phytochemicals. Plant breeders have been immensely successful in increasing yields to feed the growing global population. However, concerns have been expressed that the focus on increasing yield and processing quality has resulted in reduced contents of components that contribute to human health and increases in adverse reactions. We review the evidence for this, based largely on studies in our own laboratories of sets of wheats bred and grown between the 18th century and modern times. With the exception of decreased contents of mineral micronutrients, there is no clear evidence that intensive breeding has resulted in decreases in beneficial components or increases in proteins which trigger adverse responses. In fact, a recent study of historic and modern wheats from the UK showed increases in the contents of dietary fibre components and a decreased content of asparagine in white flour, indicating increased benefits for health.

Increasing the dietary fiber of staple foods such as bread is an attractive way to promote healthy eating in a large part of the population, where dietary fiber consumption is reportedly below the recommended values. However, many consumers prefer white breads, which are typically low in dietary fiber. In this work, white bread was made from two wheat cultivars with differing fiber contents. The resulting breads showed similar quality parameters (volume, specific volume, firmness, inner structure characteristics) with any differences maintained below 7%. Bread digestibility was evaluated using a novel dynamic in vitro digestion model. Reduced digestion rates of 30% were estimated for the high-fiber white bread compared to that in the control. Overall, this work demonstrates the potential to produce healthy, high-fiber white breads that are acceptable to consumers, with a reduced rate of starch digestion, by exploiting a genetic variation in the dietary fiber content of wheat cultivars.

Wheat is the staple food crop in temperate countries and increasingly consumed in developing countries, displacing traditional foods. However, wheat products are typically low in bioavailable iron and zinc, contributing to deficiencies in these micronutrients in countries where wheat is consumed as a staple food. Two factors contribute to the low contents of bioavailable iron and zinc in wheat: the low concentrations of these minerals in white flour, which is most widely consumed, and the presence of phytates in mineral-rich bran fractions. Although high zinc types of wheat have been developed by conventional plant breeding (biofortification), this approach has failed for iron. However, studies in wheat and other cereals have shown that transgenic (also known as genetically modified; GM) strategies can be used to increase the contents of iron and zinc in white flour, by converting the starchy endosperm tissue into a 'sink' for minerals. Although such strategies currently have low acceptability, greater understanding of the mechanisms which control the transport and deposition of iron and zinc in the developing grain should allow similar effects to be achieved by exploiting naturally induced genetic variation. When combined with conventional biofortification and innovative processing, this approach should provide increased mineral bioavailability in a range of wheat products, from white flour to wholemeal.

1. The aim of this study was to examine chemical composition, quality characteristics, apparent metabolisable energy (AME) and nutrient utilisation of wheat samples currently available to the UK poultry industry and their effect on broiler growth performance. 2. Seventeen current UK wheat samples were used to formulate 17 diets, all of which included 670 g/kg of each wheat sample and 330 g/kg of a balancer feed. Eight hundred 1-day-old male Ross 308 broilers were allocated randomly to 160 raised floor pens. Each diet was replicated eight times, fed ad libitum from 0 to 21d age in a randomised complete block design. Excreta were quantitatively collected during the last 3 days for AME determination. 3. The content of protein, ash and gross energy (GE) ranged from 97 to 143 g/kg DM, 12.8 to 19.6 g/kg DM and 17.81 to 18.24 MJ/kg DM, respectively. The amount of starch and total non-starch polysaccharides (NSP) ranged from 671 to 728 and 80.1 to 98.2 g/kg DM, respectively. The quality characteristics of wheat samples were in the expected range. 4. There were differences (P < 0.05) in AME and N-corrected AME (AMEn) of wheat samples. The AME of the wheat had a maximum range of 1.13 MJ/kg DM between samples. Dry matter retention (DMR) and fat digestibility (FD) were significantly different (P < 0.05) between wheat samples. 5. The daily feed intake (FI) and weight gain (WG) of broilers fed two wheat samples were significantly (P < 0.05) lower as compared to other samples and their low FI and WG were not related to their chemical composition and quality characteristics. 6. The ash content of wheat samples was negatively associated with AMEn (r = - 0.489, P < 0.05). The coefficient of FD was positively related to AMEn (r = 0.552, P < 0.05). 7. Chemical composition and quality characteristics of the wheat did not relate (P > 0.05) to FI and WG of broilers. There was no relationship between growth performance of broilers and AMEn of the wheat samples.

RESUMO

Polysaccharides derived from plant foods are major components of the human diet, with limited contributions of related components from fungal and algal sources. In particular, starch and other storage carbohydrates are the major sources of energy in all diets, while cell wall polysaccharides are the major components of dietary fiber. We review the role of these components in the human diet, including their structure and distribution, their modification during food processing and effects on functional properties, their behavior in the gastrointestinal tract, and their contribution to healthy diets.

Different F5 recombinant inbred lines from the cross Yumai 34×Ukrainka were grown in replicated trials on a single site in one harvest year at Rothamsted Research. A total of 10 samples from those lines were harvested and used in a broiler experiment. Twenty nutritionally complete meal-form diets that had 630 g/kg of wheat with different amounts of pentosan, with and without exogenous xylanase supplementation, were used to compare broiler growth performance and determine apparent metabolizable energy corrected for N retention (AMEn). We examined the relationship between the nutritive value of the wheat samples and their chemical compositions and results of quality tests. The amounts of total and water soluble pentosans in wheat samples ranged from 36.7 to 48.0 g/kg DM, and 6.7 to 11.6 g/kg DM, respectively. The mean crude oil and protein contents of the wheat samples were 10.5 and 143.9 g/kg DM, respectively. The average determined value for the kinematic viscosity was 0.0018 mPa.s, and 2.1 mPa.s for the dynamic viscosity. The AMEn of the wheat-based diets had a maximum range of 0.47 MJ/kg DM within the ten wheat samples that were tested. Xylanase supplementation improved (P<0.05) dietary AMEn, dry matter, and fat digestibility coefficients. There was a positive (P<0.05) relationship between in vitro kinematic viscosity of the wheat samples and the total pentosan content. There was a negative relationship between the total pentosan content in the wheat and broiler growth performance. An increase by 10 g of pentosan per kg of wheat reduced (P<0.001) daily feed intake and weight gain by 2.9 g and 3.5 g, respectively. The study shows that the feeding quality of wheat samples can be predicted by their total pentosan content. Supplementary xylanase improved energy and nutrient availability of all wheat samples that was independent of differences in pentosan content.

A combination of enzyme mapping, FT-IR microscopy and NMR spectroscopy was used to study temporal and spatial aspects of endosperm cell wall synthesis and deposition in developing grain of bread wheat cv. Hereward. This confirmed previous reports that changes in the proportions of the two major groups of cell wall polysaccharides occur, with beta-glucan accumulating earlier in development than arabinoxylan. Changes in the structure of the arabinoxylan occurred, with decreased proportions of disubstituted xylose residues and increased proportions of monosubstituted xylose residues. These are likely to result, at least in part, from arabinoxylan restructuring catalysed by enzymes such as arabinoxylan arabinofurano hydrolase and lead to changes in cell wall mechanical properties which may be required to withstand stresses during grain maturation and desiccation.

RESUMO

Although studies have shown percutaneous dilational tracheostomy to be a safe and cost-effective alternative to conventional surgical tracheostomy, there are inherent risks and complications. We report an incident occurring during percutaneous dilational tracheostomy using the Portex technique, in which a significant cuff leak occurring on insertion of the tracheostomy tube necessitated an immediate tube change. During this latter procedure, using a fresh Portex kit and guidewire, the guidewire introducer became dislodged from the guidewire assembly and inadvertently impacted in the trachea. Routine bronchoscopy identified the hazard and the foreign body was successfully removed via the bronchoscope with no adverse sequelae.

The plant hormones gibberellin and abscisic acid regulate gene expression, secretion and cell death in aleurone. The emerging picture is of gibberellin perception at the plasma membrane whereas abscisic acid acts at both the plasma membrane and in the cytoplasm - although gibberellin and abscisic acid receptors have yet to be identified. A range of downstream-signalling components and events has been implicated in gibberellin and abscisic acid signalling in aleurone. These include the Galpha subunit of a heterotrimeric G protein, a transient elevation in cGMP, Ca2+-dependent and Ca2+-independent events in the cytoplasm, reversible protein phosphory-lation, and several promoter cis-elements and transcription factors, including GAMYB. In parallel, molecular genetic studies on mutants of Arabidopsis that show defects in responses to these hormones have identified components of gibberellin and abscisic acid signalling. These two approaches are yielding results that raise the possibility that specific gibberellin and abscisic acid signalling components perform similar functions in aleurone and other tissues.

Two polypeptides of M(r) 68 kDa and 18 kDa were gibberellin (GA)-photoaffinity labelled in vitro in plasma membrane preparations from oat (Avena sativa L.) aleurone and from leaves and stems of wild-type and GA-sensitivity mutants of different species. Labelling of these polypeptides could be competed by biologically active, but not by inactive, GAs, indicating the likely biological significance of these interactions. On 2-dimensional gels the radiolabelled polypeptides were each resolved as one intensely labelled low abundance spot with a slightly lower pl form adjacent to it. There was a strong pH dependency for both labelling events, which correlated well with pH values at which GA are known to be most biologically active. A semi-dwarf GA-sensitivity mutant of sweet pea (Lathyrus odoratus L.), lb, showed reduced photoaffinity labelling of both polypeptides compared with the wild type, Lb. In the GA-insensitive Arabidopsis thaliana mutant gai, the level of labelling was the same as in wild type, GAI. This is the first report of GA-binding proteins in plant plasma membranes. Some preliminary sequence data are given for one of the labelled polypeptides. We discuss these mutants and consider their possible roles in GA perception or action.

The role of heterotrimeric G proteins in gibberellin (GA) induction of a-amylase gene expression was examined in wild oat aleurone protoplasts. Mas7, a cationic amphiphilic tetradecapeptide that stimulates GDP/GTP exchange by heterotrimeric G proteins, specifically induced alpha-amylase gene expression and enzyme secretion in a very similar manner to GA1. In addition, Mas7 stimulated expression of an alpha-Amy2/54:GUS promoter and reporter construct in transformed protoplasts. Both Mas7 and GA1 induction of alpha-amylase mRNA were insensitive to pertussis toxin. Hydrolysis-resistant nucleotides were introduced into aleurone protoplasts during transfection with reporter gene constructs. GDP-beta-S, which inhibits GDP/GTP exchange by heterotrimeric G proteins, completely prevented GA1 induction of alpha-Amy2/54:GUS expression, whereas GTP-gamma-S, which activates heterotrimeric G proteins, stimulated expression very slightly. Novel cDNA sequences from Galpha and Gbeta subunits were cloned from wild oat aleurone cells. By using RNA gel blot analysis, we found that the transcripts were expressed at a low level. Heterotrimeric G proteins have been implicated in several events during plant growth and development, and these data suggest that they may be involved in GA regulation of alpha-amylase gene expression in aleurone.

In a community survey of 953 adults we identified 40 who reported having had psoriasis. Eight of these cases were subsequently documented from physicians' records. alpha 1-Antitrypsin (alpha 1-AT) phenotyping identified 35 MZ individuals, 4 (11.4%) of whom reported psoriasis. Among the 918 non-MZ individuals 36 (3.9%) reported psoriasis, yielding a relative incidence of 3.2 (p less than 0.05). This is consistent with previous reports suggesting an association between moderate alpha 1-AT deficiency and psoriasis.

Among 114 men of Pi type or MZ, 42% (17/40) of those of Pi type MZ reported having been told by a physician that they had had pneumonia at least once whereas only 18% (13/74) of those of Pi type M did so (p less than 0.05). This significant difference could not be accounted for by differences in age, smoking status, occupation or baseline lung function (FEV1). Examination of medical records of those reporting pneumonia for the preceding 20 years provided confirmation of the diagnosis of pneumonia and support for the association; 12% (5/40) of the MZ group but only 3% (2/74) of the M group had confirmed episodes of pneumonia during the preceding 20 years. Since alpha 1-antitrypsin has been implicated as a factor in inflammatory response, reduced levels may permit the development of more frequent and/or more serious episodes of pneumonia.
