RESUMO

Fluorescent copper nanoparticles (CuNPs) was synthesized by one-step chemical reduction method using ascorbic acid (AA) and copper sulfate (CuSO4⋅5H2O) as raw materials, which had good water solubility and fluorescence properties. A green, simple and safe CuNPs@Fe2+ fluorescence probe was developed for the detection of hydrogen peroxide and glucose using Fe2+ as a bridge. The prepared CuNPs could obtain the maximum fluorescence emission wavelength at 440 nm when the excitation wavelength was 360 nm. The average particle size of CuNPs was 10 nm, which had good photobleach resistance, stability and salt tolerance. The fluorescence intensity was quenched due to electron transfer (ET) process when hydrogen peroxide was added to CuNPs@Fe2+ system. This result was mainly because Fenton reaction occured between hydrogen peroxide and Fe2+, producing hydroxyl free radicals (OH) and Fe3+. Since glucose could be catalyzed by specific glucose oxidase (GOX) to produce H2O2 and corresponding oxidation products, the quantitative analysis of glucose was realized when glucose oxidase was introduced into the CuNPs@Fe2+ sensor system. Therefore, a novel CuNPs@Fe2+ fluorescent probe sensor study was constructed to further achieve quantitative detection of H2O2 and glucose. Under the optimized experimental conditions, the linear ranges for H2O2 and glucose were 28.219-171.562 µM and 1.237-75.771 µM, respectively. And the detection limits for H2O2 and glucose were 7.169 µM and 0.540 µM, respectively. In addition, the mechanism of fluorescence probe quenching caused by the interaction between H2O2 and CuNPs@Fe2+ was also discussed. The proposed sensing system had been applied successfully to the detection of glucose in human serum samples.

In this study, 5,10,15,20-(4-sulphonatophenyl)porphyrin (TPPS4 ) was selected as a fluorescent probe due to its excellent characteristics including high quantum yield, good water solubility, and exceptional biocompatibility. With an excitation wavelength set at 515 nm, the optimal fluorescence emission wavelength for TPPS4 was measured at 642 nm. At this moment, the fluorescence signal of TPPS4 pink solution was in the 'ON' state. The fluorescence intensity of TPPS4 was quenched when ascorbic acid (AA) was introduced, which was due to the electron transfer quenching effect between AA and TPPS4 . The colour of the corresponding solution changed from pink to green, and the fluorescence signal was in the 'OFF' state. When HPO4 2- was further introduced into the TPPS4 -AA system, the quenched fluorescence intensity of TPPS4 was recovered due to the unique interaction between HPO4 2- and AA. At this time, the colour of the corresponding solution changed from green to red, and the fluorescence signal was in the 'ON' state. Therefore, an 'ON-OFF-ON' signal-switchable fluorescent probe was constructed based on TPPS4 to detect HPO4 2- . The results showed that the linear range of HPO4 2- was 4.0 × 10-9 to 1.7 × 10-6  M, and the detection limit was 1.3 × 10-9  M (S/N = 3). The sensing system exhibited high accuracy and sensitivity, and it could be used successfully to detect HPO4 2- in real samples.

BACKGROUND: Fallopia aubertii (L. Henry) Holub is a perennial semi-shrub with both ornamental and medicinal value. The mitochondrial genomes of plants contain valuable genetic traits that can be utilized for the exploitation of genetic resources. The parsing of F. aubertii mitochondrial genome can provide insight into the role of mitochondria in plant growth and development, metabolism regulation, evolution, and response to environmental stress. METHODS: In this study, we sequenced the mitochondrial genome of F. aubertii using the Illumina NovaSeq 6000 platform and Nanopore platform. We conducted a comprehensive analysis of the mitochondrial genome of F. aubertii, which involved examining various aspects such as gene composition, repetitive sequences, RNA editing sites, phylogeny, and organelle genome homology. To achieve this, we employed several bioinformatics methods including sequence alignment analysis, repetitive sequence analysis, phylogeny analysis, and more. RESULTS: The mitochondrial genome of F. aubertii has 64 genes, including 34 protein-coding genes (PCGs), three rRNAs, and 27 tRNAs. There were 77 short tandem repeat sequences detected in the mitochondrial genome, five tandem repeat sequences identified by Tandem Repeats Finder (TRF), and 50 scattered repeat sequences observed, including 22 forward repeat sequences and 28 palindrome repeat sequences. A total of 367 RNA coding sites were predicted in PCGs, with the highest number (33) found within ccmB. Ka/Ks values estimated for mitochondrial genes of F. aubertii and three closely related species representing Caryophyllales were less than 1 for most of the genes. The maximum likelihood evolutionary tree showed that F. aubertii and Nepenthes ×ventrata are most closely related. CONCLUSIONS: In this study, we obtained basic information on the mitochondrial genome of F. aubertii and this study investigated repeat sequences and homologous segments, predicted RNA editing sites, and utilized the Ka/Ks ratio to estimate the selection pressure on mitochondrial genes of F. aubertii. We also discussed the systematic evolutionary position of F. aubertii based on mitochondrial genome sequences. Our study revealed variations in the sequence and structure of mitochondrial genomes in Caryophyllales. These findings are of great significance for identifying and improving valuable plant traits and serve as a reference for future molecular studies of F. aubertii.

Cr3+ is one of the most essential trace elements in living organisms and plays a vital role in human metabolism. However, both deficiency and excess intake of Cr3+ can be harmful to the human body. Therefore, the quantitative determination of Cr3+ is of great significance in the field of life science. Based on this, in this study, a g-CNQDs@p-acetaminophenol fluorescence sensing system was developed for the quantitative detection of Cr3+ in actual complex samples. G-CNQDs were synthesized with sodium citrate and urea as precursors. The fluorescence signal was enhanced by the synergistic effect between p-acetaminophenol (APAP) and g-CNQDs. The fluorescence quenching phenomenon can be produced when Cr3+ is introduced into the fluorescence-enhanced g-CNQDs@p-acetaminophenol system. An "on-off" fluorescence sensing system was constructed based on g-CNQDs@p-acetaminophenol for the quantitative detection of Cr3+. The experimental data showed a wide linear region in the concentration range of 0.64-63.0 µM, and the detection limit was as low as 0.23 µM. The construction of the sensor system broadens the research field for the practical application of Cr3+.

In this study, the F-CuNPs were synthesized by a modified liquid-phase chemical reduction method. Throughout the preparation process, anhydrous copper sulfate was used as the copper source, and ascorbic acid in the NaOH solution served as the reducing and protective agent. Förster resonance energy transfer (FRET) may exist between F-CuNPs and vitamin B2 due to the large spectral overlap between the fluorescence emission spectra of F-CuNPs and the UV-vis absorption spectra of vitamin B2. Therefore, the detection of vitamin B2 was designed based on a FRET system between F-CuNPs and vitamin B2. With S2- into the F-CuNPs&VB2 system, the fluorescence intensity of vitamin B2 was quenched, while the fluorescence intensity of F-CuNPs was almost unchanged. There may be a specific reaction between S2- and vitamin B2. Therefore, the research system can be further used to detect S2- based on ratiometric fluorescent probe. The research findings show that the linear range of vitamin B2 was 0.51 nM-34.64 nM with a detection limit of 0.25 nM (S/N = 3), the linear range of S2- was 0.64 µM-60.00 µM with a detection limit of 0.32 µM (S/N = 3). Furthermore, the simultaneous fluorescent sensing system has high sensitivity and selectivity. Therefore, this system was designed and successfully used to detect the content of vitamin B2 and S2- in actual samples to find a new effective method to detect analytes.

The pig is an excellent animal model for simulating human physiology and a major animal for meat production and xenotransplantation. Therefore, researching porcine embryonic development is crucial for studying human reproductive diseases and improving litter size in commercial pigs. Embryonic development in pigs occurs under a complex regulatory mechanism, in which epigenetic regulatory mechanisms play an essential role. Recently, studies on the effects of epigenetic modifications on embryonic development have been conducted at different developmental stages and in different cell lines. Increasing evidence suggests that a certain amount of crosstalk exists between different epigenetic modifications. This review describes four regulatory mechanisms of epigenetics involved in porcine embryonic development: DNA methylation, histone modification, non-coding RNA function, and chromatin accessibility, and explores the possible crosstalk between them.

Fast and efficient information processing and encryption, including writing, reading, and encryption memory, is essential for upcoming terahertz (THz) communications and information encryption. Here, we demonstrate a THz multi-level, nonvolatile, optically rewritable memory and encryption memory based on chalcogenide phase-change materials, Ge2Sb2Te5 (GST). By tuning the laser fluence irradiated on GST, we experimentally achieve multiple intermediate states and large-area amorphization with a diameter of centimeter-level in the THz regime. Our memory unit features a high operating speed of up to 4 ns, excellent reproducibility, and long-term stability. Utilizing this approach, hexadecimal coding information memories are implemented, and multiple writing-erasing tests are successfully carried out in the same active area. Finally, terahertz photoprint memory is demonstrated, verifying the feasibility of lithography-free devices. The demonstration suggests a practical way to protect and store information and paves a new avenue toward nonvolatile active THz devices.

The amygdala is one of the most common origins of seizures, and the amygdala mouse model is essential for the illustration of epilepsy. However, few studies have described the experimental protocol in detail. This paper illustrates the whole process of amygdala electrical kindling epilepsy model making, with the introduction of a method of bipolar electrode fabrication. This electrode can both stimulate and record, reducing brain injury caused by implanting separate electrodes for stimulation and recording. For long-term electroencephalogram (EEG) recording purposes, slip rings were used to eliminate the record interruption caused by cable tangles and falling off. After periodic stimulation (60 Hz, 1 s every 15 min) of the basolateral amygdala (AP: 1.67 mm, L: 2.7 mm, V: 4.9 mm) for 19.83 ± 5.742 times, full kindling was observed in six mice (defined as induction of three continuous grade V episodes classified by Racine's scale). An intracranial EEG was recorded throughout the entire kindling process, and an epileptic discharge in the amygdala lasting 20-70 s was observed after kindling. Therefore, this is a robust protocol for modeling epilepsy originating from the amygdala, and the method is suitable for revealing the role of the amygdala in temporal lobe epilepsy. This research contributes to future studies on the mechanisms of mesial temporal lobe epilepsy and novel antiepileptogenic drugs.

Chinese Tibetan tea made from the tender leaves of Malus transitoria is a widely consumed health drink, but there are few reports on its chemical composition and biological activity. In this study, we found that a 50% ethanol extract of M. transitoria had good antioxidant and α-glucosidase inhibitory activities in vitro. Guided by in vitro bioassays, chromatographic separation and purification were conducted, and the most active fraction in M. transitoria was determined. UPLC-Orbitrap-MS/MS was used to further quickly and comprehensively characterize the chemical composition. Library searches, MS/MS fragmentation patterns of two isolated reference compounds, and bibliography were used to annotate 81 compounds, of which 2 were new compounds, and 79 were identified from M. transitoria for the first time. This study provides a scientific basis for the development of antioxidant and anti-diabetic functional foods from M. transitoria.

In this paper, 5,10,15,20-(4-sulphonatophenyl) porphyrin (TSPP) was synthesized by a facile route and used as a fluorescent probe to construct a sensor system based on the high water solubility and high quantum yield. It was found that when riboflavin (RF) was introduced into the TSPP solution, the fluorescence intensity of TSPP decreased for the peaks at 645 nm and 700 nm based on the principle of the electrostatic attractions and hydrophobic interactions between TSPP and riboflavin. When the fluorescence emission peak of riboflavin appeared at 550 nm, the fluorescence sensor system changed from the "on" state to the "off" state. When sulfur ions (S2-) were further introduced into the TSPP-riboflavin system, the fluorescence intensity of riboflavin was further decreased based on the specific reaction between S2- and riboflavin. However, the fluorescence signal of TSPP was restored and the fluorescence sensing system changed from the "off" state to the "on" state. Therefore, TSPP was used as a fluorescent probe to construct an "on-off-on" fluorescent sensing system, the linear range of S2- detected by this system is 5.0 × 10-9 to 3.6 × 10-5 M, and the detection limit (LOD) is 1.1 × 10-9 M. The sensing system has higher accuracy and sensitivity, and it can be successfully used in the sensing of S2- in real samples.

Fallopia aubertii (L.Henry) Holub (Polygonaceae), a woody plant with a voluble stem, is used as a folk herbal medicines for the treatment of gout, fever and pneumonia. To better understand the molecular genetics of F. aubertii, its complete chloroplast genome was sequenced and annotated. The assembled chloroplast genome is a circular 160,951 bp sequence consisting of large single copy (87,279 bp) and small single copy (13,394 bp) regions, separated by two inverted repeat regions (30,860 bp each). The genome contains 131 genes including 86 protein-coding, 37 tRNA and 8 tRNA genes. Phylogenetic analysis based on the complete chloroplast genome showed that F. aubertii is more closely related to M. australis than to F. sachalinensis, which exhibited a polyphyletic relationship with respect to F. aubertii. These results require further analyses. This study provides additional data for reconstructing species relationships in Fallopia.

RESUMO

In terahertz (THz) technologies, generation and manipulation of THz waves are two key processes usually implemented by different device modules. Integrating THz generation and manipulation into a single compact device will advance the applications of THz technologies in various fields. Here, we demonstrate a hybrid nonlinear plasmonic metasurface incorporating an epsilon-near-zero (ENZ) indium tin oxide (ITO) layer to seamlessly combine efficient generation and manipulation of THz waves across a wide frequency band. The coupling between the plasmonic resonance of the metasurface and the ENZ mode of the ITO thin film enhances the THz conversion efficiency by more than 4 orders of magnitude. Meanwhile, such a hybrid device is capable of shaping the polarization and wavefront of the emitted THz beam via the engineered nonlinear Pancharatnam-Berry (PB) phases of the plasmonic meta-atoms. The presented hybrid nonlinear metasurface opens a new avenue toward miniaturized integrated THz devices and systems with advanced functionalities.

Broadband light sources emitting in the terahertz spectral range are highly desired for applications such as noninvasive imaging and spectroscopy. Conventionally, THz pulses are generated by optical rectification in bulk nonlinear crystals with millimetre thickness, with the bandwidth limited by the phase-matching condition. Here we demonstrate broadband THz emission via surface optical rectification from a simple, commercially available 19 nm-thick indium tin oxide (ITO) thin film. We show an enhancement of the generated THz signal when the pump laser is tuned around the epsilon-near-zero (ENZ) region of ITO due to the pump laser field enhancement associated with the ENZ effect. The bandwidth of the THz signal generated from the ITO film can be over 3 THz, unrestricted by the phase-matching condition. This work offers a new possibility for broadband THz generation in a subwavelength thin film made of an ENZ material, with emerging physics not found in existing nonlinear crystals.

Fallopia aubertii (L. Henry) Holub is a traditional Tibetan medicine to treat gout, but the research on its chemical composition is limited, probably due to the complex purification process. In this study, the chemical constituents of F. aubertii were isolated and identified by medium pressure chromatography and high-pressure preparative liquid chromatography. The general separation process was as follows: first, the extraction part of ethyl acetate collected seven components according to the chromatographic peaks of the sample through the medium pressure chromatographic separation system. Next, the separation effects of Fr2 on five different preparation columns were compared. A reversed-phase chromatographic column with hydrophilic groups was selected for separation. Finally, the principal component compound (gallic acid) in Fr2 was isolated and identified. The results show that the combination of medium pressure chromatographic separation gel column and reversed phase chromatographic column with hydrophilic groups is an effective separation method for the separation of gallic acid from F. aubertii.

Strongly confined surface waves can be achieved on periodically structured metal surfaces and are known as spoof surface plasmon polaritons (SPPs). In this work, several terahertz SPP devices based on curved waveguides are demonstrated. The transmittance and bending loss of 90-degree curved spoof SPP waveguides with a radius of curvature ranging from 200 to 2300 µm are investigated to identify the regime for high transmission. A commutator is designed and experimentally demonstrated. Furthermore, coupling equations are derived and verified for efficient coupling between bend-straight waveguides and between bend-bend waveguides. The results will be of great value for future integrated terahertz plasmonic systems.

Terahertz waves have attracted considerable research interest in recent years because of their potential applications in diverse fields. As an important device to control terahertz waves, beam splitters with greater flexibility and higher degrees of freedom are highly desirable. In order to obtain higher degrees of freedom in beam splitting, 2-bit or higher-bit coding elements are usually introduced into metamaterial beam splitters based on the coding theory. In this work, a new "offset" coding scheme using only the 1-bit coding elements of "0" and "1" is presented, and the period of coding for beam splitting can be a non-integer multiple of the length of a single unit rather than only its integer multiples. Therefore, more beam-splitting degrees of freedom can be obtained, and the design strategy is experimentally verified. We believe that the new coding scheme will also be of significance in radar cross section reduction and flexible wave control.

High concentration restaurant oily wastewater from restaurants and food processing industries discharged into water environment usually results in environment pollution and inhibits the activity of microorganisms in biological wastewater treatment systems. In this study, 75 strains from oily sludge were isolated with oil degradation activity for edible oil-contained wastewater. Eight isolates were able to grow well in liquid cultures with edible oil as the sole carbon source and discovered with high efficient oil-degrading ability. Seven out of eight isolates were identified as Acinetobacter and one isolate as Kluyvera cryocrescens, based on their 16S rRNA gene sequences. Three highly efficient oil degrading bacteria (Acinetobacter dijkshoorniae LYC46-2, Kluyvera cryocrescens LYC50-1a and Acinetobacter pittii LYC73-4b) were selected and their degradation characteristic were examined, the results showed that the three isolates were effective under pH range from 7.0 to 10.0, and temperature from 25 to 35 °C. For degradation of 2-4% (v/v) of vegetable oil, > 85% degradation percentage were obtained within 30 h. Degradation of the higher concentration oil (6-8%, v/v) result in 50-70% degradation percentage within 72 h, and the degradation percentage for the isolated strains were decreased about 50% for the degradation of 10% oil (< 45%) compared to 2% oil. Different type of oils were also tested, > 90% of degradation percentage were obtained by the three isolates, implied that these strains are capable of removing various oils efficiently. These results suggested that Acinetobacter dijkshoorniae LYC46-2, Kluyvera cryocrescens LYC50-1a and Acinetobacter pittii LYC73-4b are potential species could be efficiently used for high concentration restaurant oily wastewater treatment and might be applicable to a wastewater treatment system for the removal of oil.

OBJECTIVE: To investigate the anticancer effect of curcumin Solid Dispersions (SDs). METHODS: Curcumin SDs were prepared by patent technology. The anticancer effect of curcumin SDs were investigated by vivo and vitro tests of SCG-7901, BEL-7402, S-180 and Ehrlich ascites tumor models. RESULTS: The results showed that Curcumin SDs had markedly anticancer effect and could improve the anticancer effect of cisplatin. CONCLUSION: Curcumin SDs could be developed into one kind of adjuvant drug for anticancer, as it has markedly anticancer effect, and could improve the anticancer effects of cisplatin.

A simple and rapid method for simultaneous separation and determination of xanthones in Swertia mussotii and Swertia franchetiana by high performance liquid chromatography has been established. The analysis was performed on a Kromasil C18 column (250 mm x 4.60 mm i.d., 5 microm) (at 20 degrees C) eluted with methanol and 0.1% aqueous phosphoric acid (73: 27 in volume ratio) as mobile phase at a flow rate of 1.0 mL/min and with UV detection at 260 nm. Results showed that xanthones were separated successfully from each other and from other interfering components. There was a good linear relationship between the content of component and its peak area for each xanthone, with the correlation coefficients of 0.999 2 - 0.999 9. The convenient method can be used for quantitative analysis of xanthones.
