BACKGROUND: This study aimed to evaluate the effectiveness and safety of recombinant human endostatin (Rh-endostatin) plus programmed cell death 1 (PD-1) inhibitors and chemotherapy as first-line treatment for advanced or metastatic non-small cell lung cancer (NSCLC) in a real-world setting. METHODS: This was a retrospective study on patients with EGFR/ALK-negative, advanced or metastatic NSCLC. Patients received Rh-endostatin plus PD-1 inhibitors and chemotherapy every three weeks for 4 to 6 cycles. The primary endpoint was progression-free survival (PFS), and the secondary endpoints were objective response rate (ORR), disease control rate (DCR), overall survival (OS), and safety. RESULTS: A total of 68 patients were included in this retrospective analysis. As of data cutoff (December 13, 2022), the median follow-up of 21.4 months (interquartile range [IQR], 8.3-44.4 months). The median PFS and OS was 22.0 (95% confidence interval [CI]: 16.6-27.4) and 31.0 months (95% CI: 23.4-not evaluable [NE]), respectively. The ORR was 72.06% (95% CI: 59.85-82.27%), and DCR was 95.59% (95% CI: 87.64-99.08%). Patients with stage IIIB/IIIC NSCLC had significantly longer median PFS (23.4 vs. 13.2 months), longer median OS (not reached vs. 18.0 months), and higher ORR (89.2% vs. 51.6%) than those with stage IV NSCLC (all p ≤ 0.001). The ORR was higher in patients with high PD-L1 expression (tumor proportion score [TPS] ≥ 50%) than in those with low PD-L1 expression or positive PD-L1 expression (75% vs. 50%, p = 0.025). All patients experienced treatment-related adverse events (TRAEs), and ≥ grade 3 TRAEs occurred in 16 (23.53%) patients. CONCLUSIONS: Rh-endostatin combined with PD-1 inhibitors plus chemotherapy as first-line treatment yielded favorable effectiveness with a manageable profile in patients with advanced or metastatic NSCLC, representing a promising treatment modality.

Alzheimer's disease (AD) is a progressive and fatal neurodegenerative disease. The prevalent features of AD pathogenesis are the appearance of ß-amyloid (Aß) plaques and neurofibrillary tangles, which cause microglial activation, synaptic deficiency, and neuronal loss. Microglia accompanies AD pathological processes and is also linked to cognitive deficits. Purinergic signaling has been shown to play a complex and tight interplay with the chemotaxis, phagocytosis, and production of pro-inflammatory factors in microglia, which is an important mechanism for regulating microglia activation. Here, we review recent evidence for interactions between AD, microglia, and purinergic signaling and find that the purinergic P2 receptors pertinently expressed on microglia are the ionotropic receptors P2X4 and P2X7, and the subtypes of P2YRs expressed by microglia are metabotropic receptors P2Y2, P2Y6, P2Y12, and P2Y13. The adenosine P1 receptors expressed in microglia include A1R, A2AR, and A2BR. Among them, the activation of P2X4, P2X7, and adenosine A1, A2A receptors expressed in microglia can aggravate the pathological process of AD, whereas P2Y2, P2Y6, P2Y12, and P2Y13 receptors expressed by microglia can induce neuroprotective effects. However, A1R activation also has a strong neuroprotective effect and has a significant anti-inflammatory effect in chronic neuroinflammation. These receptors regulate a variety of pathophysiological processes in AD, including APP processing, Aß production, tau phosphorylation, neuroinflammation, synaptic dysfunction, and mitochondrial dysfunction. This review also provides key pharmacological advances in purinergic signaling receptors.

BACKGROUND: The incidence of dyslipidemia increases after menopause. Electroacupuncture (EA) has been recommended for menopause-related disease. However, the positive effect on lipid metabolism disorders is still unclear. OBJECTIVES: To investigate the underlying mechanism of EA treatment on lipid metabolism disorders through ONT full-length transcriptome sequencing Methods: Adult female SD rats were randomly divided into Ctrl, sham operation+high-fat feed(Sham+HFD), Ovariectomized+high-fat feed (OVX+HFD), Ovariectomized+high-fat feed + Atorvastatin (OVX+HFD+ATO) and OVX+HFD+EA groups. Periovarian adipose tissue around the bilateral ovaries of rats in the Sham+HFD group was resected. Rats in the OVX+HFD, OVX+HFD+ATO and OVX+HFD+EA groups were subjected to bilateral oophorectomy to prepare the ovariectomized rat model. Treatment was applied to rats in the OVX+HFD+EA group. ST36, PC6, SP6, BL18 and ST40 were the selected acupoints. Daily food intake and body weights of rats were recorded. The samples were collected 30 days after treatment. The serum levels of total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein (HDL-C) were detected to assess the improvement of lipid metabolism disorders. HE and oil red O staining were used to stain the liver tissues. Total RNA was extracted from liver tissues, and its transcriptional changes were determined by high-throughput sequencing. Additionally, RTÁqPCR and immunofluorescence staining were used to verify the crucial signal pathway screened by the ONT fullÁlength transcriptome sequencing. RESULTS: EA treatment resulted in a lowered weight of perirenal fat and liver and a significant improvement in the color of the liver. In addition, EA could improve the lipid profile and hepatic steatosis in OVX+HFD rats. According to fullÁlength transcriptome sequencing, 2292 genes showed differential expression in the OVX+HFD group; of these, 1121 were upregulated and 1171 down-regulated. 609 DEGs were found in the OVX+HFD+EA group compared to the OVX+HFD group; 235 up-regulated and 374 down-regulated. We also found that 77 genes are significantly upregulated after EA intervention through Venn map analysis (including Agtr1a, Pdia3, etc.), which may be the targeted genes for EA treatment of lipid metabolism disorders. Finally, we verified the expression of Pdia3, Perk and Qrich1 levels in liver tissues. HFD feeding could increase the expression of Pdia3 and its downstream signal pathways molecular Perk and Qrich1. But these effects were reversed by EA treatment, the results demonstrated that the expression of pdia3, Perk, as well as Qrich1 of OVX+HFD rats had a decreasing trend after EA treatment. CONCLUSIONS: EA could ameliorate lipid metabolic disorder in OVX+HFD rats. The Pdia3/Perk/Qrich1 signal pathway may play crucial roles in the improvement of lipid metabolism disorder of OVX+HFD rats after EA treatment.

Background/purpose: History of periodontitis is a well-documented risk indicator of peri-implantitis. However, the influence of severity of periodontitis is still unclear, especially for severe periodontitis. This study was aimed to investigate the prevalence of peri-implant disease and analyze the risk indicators in patients with treated severe periodontitis. Materials and methods: A total of 182 implants from 88 patients (44 males and 44 females) with severe periodontitis with a mean fellow-up period of 76.5 months were enrolled in this study. Patient and implant information, and periodontal and peri-implant conditions were collected to evaluate the prevalence of peri-implant disease and risk indicators. Results: The prevalence of peri-implantitis was 9.1% and 6.6% at the patient-level and implant-level. The prevalence of peri-implant mucositis was 76.1% and 51.1% at the patient-level and implant-level. Risk indicators of peri-implantitis included older age (OR: 1.132), poor proximal cleaning habits (OR: 14.218), implants in anterior area (OR: 10.36), poor periodontal disease control (OR: 12.76), high peri-implant plaque index (OR: 4.27), and keratinized tissue width (KTW)<2 mm (OR: 19.203). Conclusion: Implants in patients with severe periodontitis after periodontal treatment and maintenance show a low prevalence (9.1%) of peri-implantitis and a relatively high prevalence (76.2%) of peri-implant mucositis. Patient age, peri-implant proximal cleaning habits, implant position, periodontal disease control, peri-implant plaque index, and KTW are associated with prevalence of peri-implantitis.

Background/purpose: Excessive host immune response is thought to be an important cause of periodontal tissue damage during periodontitis. The potent chemotaxis produced by locally released chemokines is the key signal to trigger this response. Here, we aimed to investigate the expression of CXC chemokine receptor 1 (CXCR1), and chemokines interleukin-8 (IL-8) and pro-platelet basic protein (PPBP) in human inflammatory gingival tissues compared with healthy tissues. Materials and methods: A total of 54 human gingival tissues, 27 healthy and 27 inflammatory samples, were collected. Fifteen specimens of each group were employed for quantitative reverse transcription polymerase chain reaction to determine the mRNA levels of CXCR1, IL-8, and PPBP. Six samples of each group were used for Western blotting to investigate the protein expression of CXCR1 and for enzyme-linked immunosorbent assay to evaluate the protein levels of IL-8 and PPBP, respectively. Results: The mRNA levels of chemokine receptor CXCR1, chemokine IL-8, and PPBP in inflammatory gingival tissues were significantly higher than those in healthy controls (P < 0.05). The protein levels of CXCR1, IL-8, and PPBP in inflammatory gingival tissues were also significantly higher than those in healthy gingival tissues (P < 0.05). Conclusion: When compared to healthy gingival tissues, the expression of CXCR1, IL-8, and PPBP in inflammatory gingival tissues is higher.

Inflammatory pain is a commonly observed clinical symptom in a range of acute and chronic diseases. However, the mechanism of inflammatory pain is far from clear yet. Rab11a, a small molecule guanosine triphosphate enzyme, is reported to regulate orofacial inflammatory pain in our previous works. However, the mechanism of Rab11a's involvement in the regulation of inflammatory pain remains obscure. Here, we aim to elucidate the potential mechanisms through which Rab11a contributes to the development of inflammatory pain in the spinal level. It's shown that neurons, rather than glial cells, were the primary cell type expressing Rab11a in the spinal dorsal horn (SDH). After intra-plantar injection of CFA, both the number of Fos/Rab11a-immunopositive neurons and the expression of Rab11a were increased. Administration of Rab11a-shRNA into the SDH resulted in significantly analgesic effect in mice with CFA injection. Application of Rab11a-shRNA also reduced the NMDA receptor-mediated excitatory post-synaptic current (EPSC) and the spike number of neurons in lamina II of the SDH in mice with CFA injection, without affecting the presynaptic glutamate release and the postsynaptic AMPA receptor-mediated EPSC. Our results thus suggest that the enhanced expression of neuronal Rab11a may be important for the process of inflammatory pain in mice with CFA injection, which is likely mediated by Rab11a's potentiation of the competence of post-synaptic NMDAR and spiking of SDH neurons.

Objective: The occurrence of breast cancer-related lymphedema (BCRL) in postoperative breast cancer survivors is described and the independent risk factors of BCRL are analyzed. A BCRL nomogram prediction model is constructed, and its effectiveness is evaluated to screen out high-risk patients with BCRL. Methods: A univariate analysis was carried out to determine the risk factors possibly related to BCRL, and a logistic regression analysis was utilized to determine the independent risk factors related to BCRL. A BCRL nomogram prediction model was built, and a nomogram was drawn by R software v4.1.0. The area under the curve (AUC) of the receiver operating characteristic (ROC) and the Hosmer-Lemeshow test were used to evaluate the efficacy of the constructed model to assess its clinical application value. Results: The risk factors independently associated with BCRL were body mass index (BMI), handedness on the operation side, no BCRL-related rehabilitation plan, axillary lymph node dissection (ALND), taxane-based chemotherapy, and radiotherapy (all p < 0.05). The BCRL nomogram prediction model was built on this basis, and the results of the efficacy evaluation showed a good fit: AUC = 0.952 (95% confidence interval: 0.930-0.973) for the ROC and χ2 = 6.963, p = 0.540 for the Hosmer-Lemeshow test. Conclusions: The risk factors for BCRL included higher BMI, handedness on the operation side, no BCRL-related rehabilitation plan, ALND, taxane-based chemotherapy, and radiotherapy. In addition, the BCRL nomogram prediction model accurately calculated the risk of possible BCRL among breast cancer survivors and effectively screened for high-risk patients with BCRL. Therefore, this prediction model can provide a basis for rehabilitation physicians and therapists to formulate early and individualized prevention and treatment programs.

Traffic intersections are important scenes that can be seen almost everywhere in the traffic system. Currently, most simulation methods perform well at highways and urban traffic networks. In intersection scenarios, the challenge lies in the lack of clearly defined lanes, where agents with various motion plannings converge in the central area from different directions. Traditional model-based methods are difficult to drive agents to move realistically at intersections without enough predefined lanes, while data-driven methods often require a large amount of high-quality input data. Simultaneously, tedious parameter tuning is inevitable involved to obtain the desired simulation results. In this paper, we present a novel adaptive and planning-aware hybrid-driven method (TraInterSim) to simulate traffic intersection scenarios. Our hybrid-driven method combines an optimization-based data-driven scheme with a velocity continuity model. It guides the agent's movements using real-world data and can generate those behaviors not present in the input data. Our optimization method fully considers velocity continuity, desired speed, direction guidance, and planning-aware collision avoidance. Agents can perceive others' motion plannings and relative distances to avoid possible collisions. To preserve the individual flexibility of different agents, the parameters in our method are automatically adjusted during the simulation. TraInterSim can generate realistic behaviors of heterogeneous agents in different traffic intersection scenarios in interactive rates. Through extensive experiments as well as user studies, we validate the effectiveness and rationality of the proposed simulation method.

Neurokinin 3 receptor (NK3R) is a tachykinin receptor essential for the hypothalamic-pituitary-gonadal axis. The endogenous peptide agonist neurokinin B (NKB) preferentially activates NK3R, while substance P (SP) binds preferentially to NK1R. In addition, the SP analogue senktide more potently activates NK3R than NKB and SP. However, the mechanisms of preferential binding of peptide and NK3R activation remain elusive. Herein, we determined the cryogenic electron microscopy (cryo-EM) structures of the NK3R-Gq complex bound to NKB, SP and senktide. The three NK3R-Gq/peptide complexes utilize a class of noncanonical receptor activation mechanisms. Combining the structural analysis and functional assay illustrated that the consensus C-termini of the three peptide agonists share a conserved binding mode to NK3R, while the divergent N-termini of the peptides confer the preferential binding of the agonist to NK3R. In addition, the specific interactions between the N-terminus of senktide and the N-terminus and extracellular loops (ECL2 and ECL3) of NK3R lead to the improved activation displayed by senktide compared to SP and NKB. These findings pave the way to understand tachykinin receptor subtype selectivity and provide ideas to rationally develop drugs targeting NK3R.

Pedestrian trajectory prediction is an important technique of autonomous driving. In order to accurately predict the reasonable future trajectory of pedestrians, it is inevitable to consider social interactions among pedestrians and the influence of surrounding scene simultaneously, which can fully represent the complex behavior information and ensure the rationality of predicted trajectories obeyed realistic rules. In this article, we propose one new prediction model named social soft attention graph convolution network (SSAGCN), which aims to simultaneously handle social interactions among pedestrians and scene interactions between pedestrians and environments. In detail, when modeling social interaction, we propose a new social soft attention function, which fully considers various interaction factors among pedestrians. Also, it can distinguish the influence of pedestrians around the agent based on different factors under various situations. For the scene interaction, we propose one new sequential scene sharing mechanism. The influence of the scene on one agent at each moment can be shared with other neighbors through social soft attention; therefore, the influence of the scene is expanded both in spatial and temporal dimensions. With the help of these improvements, we successfully obtain socially and physically acceptable predicted trajectories. The experiments on public available datasets prove the effectiveness of SSAGCN and have achieved state-of-the-art results. The project code is available at.

Emotion categories configure the basic semantic knowledge of the human cognitive structure. Previous studies with people with Williams syndrome (WS) investigated their ability to process basic emotions and the dimensions of emotional valences. However, little is known about the categorization of emotions from the subordinate perspective of lexical words in people with WS. In this study, emotion priming was used as the research paradigm. Three types of emotional valence were used as stimuli: positive, neutral, and negative. Each emotional valence was used as a prime matched to a target in one of these same three types of emotional valence. All participants were asked to judge whether the prime and the target were matched in their emotional valence. People with WS (n = 14, 11M/3F, CA = 10.49, and MA = 6.57) showed priming patterns for emotion valences like those of the typically developing controls. When positive primes were presented, accuracy was higher for positive and negative targets than neutral targets. When neutral primes were presented, accuracy was highest for negative targets. When negative primes were presented, accuracy was the lowest for negative targets. All participants showed high priming accuracy for positive emotions; however, they confused neutral with positive targets. A negative priming effect was observed when negative primes preceded negative targets. Considering previous findings that people with WS show developmental delays in the basic emotions of anger and surprise, this study concludes that people with WS responded least accurately to the classification of emotional valence. The findings regarding the categorization of emotions in people with WS not only advance our understanding of their emotion knowledge and socioemotional cognition but also confirm the superficial enrichment of lexical semantics with weak conceptual change in people with WS. This weakness may result in impaired contextual integration in people with WS.

Weakly supervised object detection (WSOD) has attracted more and more attention since it only uses image-level labels and can save huge annotation costs. Most of the WSOD methods use Multiple Instance Learning (MIL) as their basic framework, which regard it as an instance classification problem. However, these methods based on MIL tend to only converge on the most discriminative regions of different instances, rather than their corresponding complete regions, that is, insufficient integrity. Inspired by the human habit of observing things, we propose a new method by comparing the initial proposals and the extended ones to optimize those initial proposals. Specifically, we propose one new strategy for WSOD by involving contrastive proposal extension (CPE), which consists of multiple directional contrastive proposal extensions (D-CPEs), and each D-CPE contains LSTM-based encoders and dual-stream decoders. Firstly, the boundary of initial proposals in MIL is extended to different positions according to well-designed sequential order. Then, the CPE compares the extended proposal and the initial one by extracting the feature semantics of them using the encoders, and calculates the integrity of the initial proposal to optimize its score. These contrastive contextual semantics will guide the basic WSOD to suppress bad proposals and improve the scores of good ones. In addition, a simple dual-stream network is designed as the decoder to constrain the temporal coding of LSTM and improve the performance of WSOD further. Experiments on PASCAL VOC 2007, VOC 2012 and MS-COCO datasets show that our method has achieved the state-of-the-art results.

Underwater wireless optical communication (UWOC) has attracted much attention recently due to its superiority of high transmission speed. In many UWOC applications, to establish communication links easier, the receiver has quite a wide field. However, the performance of the receiving field is affected by watertight encapsulation in practice, especially when the optical window is produced with plain glass. To study such an influence, in this paper, we first establish a theoretical model based on the Monte Carlo method. Then, we analyze the influence with different structure parameters of watertight encapsulation. The simulation results show that to reduce such an impact, the optical window should be thinner, the detector photosurface and optical window surface should be larger, and the space between the detector and optical window should be smaller. In the ideal situation, the largest workable receiving field is about 96° for UWOC. In other situations, the workable receiving field would be smaller. The simulation method and results presented in this paper are pragmatic and useful to UWOC receiver design.

Underwater wireless optical communication (UWOC) has attracted much attention recently, thanks to its high transmission speed. In many UWOC applications, the transmitter is based on light emitting diodes (LEDs). This is because it has up to a 120° divergence angle, which is helpful with establishing the communication link more easily. However, the light field of such a transmitter is affected by watertight encapsulation in practice. In this paper, we establish a theoretical model based on the Monte Carlo method to study this influence when the watertight optical window is based on plain glass. The results show that both the divergence angle and transmittance become smaller because of the optical window. In order to reduce this influence, the illumination surface, distance from the light source to the optical window, and thickness of the optical window should be smaller, while the optical window surface should be larger. In the ideal situation, the largest divergence angle is 94.4°, which is defined with full width at half-maximum (FWHM). The maximum transmittance is related to the optical materials of the window, while it is 93.54% for quartz glass. When light is modulated with on-off keying (OOK) format, if the signal-to-noise ratio (SNR) is larger than 40 at 0° radiation angle, the UWOC system could work when the angle is up to ±48.7∘. While the SNR reduces, the workable angle range decreases. The simulation model presented in this paper is convenient to analyze the influence of a watertight optical window with plain glass. The simulation results presented in this paper are useful to the UWOC transmitter design.

An efficient synthesis method will allow a large number of tetramic acid analogues to be synthesized for property and potency optimization. In this study, a facile and efficient method was described for the synthesis of 3-acyltetramic acids. The synthesis was accomplished mainly via (1) mild intramolecular cyclization and (2) the formation of ß-ketoamides between nucleophiles and acyl Meldrum's acids. 3-Acyltetramic acid exhibited phytotoxicity against Echinochloa crusgalli and Portulaca oleracea. At a dosage of 750 g ha-1, 6k and 6a showed high herbicidal activity against E. crusgalli, Digitaria sanguinalis and P. oleracea, Amaranthus retroflexus, respectively. 6k inhibited the synthesis of endogenous abscisic acid, thus seedling germination and plant growth. The incorporation of various acyl Meldrum's acids and amino acid esters was applicable to the parallel synthesis of 3-acyltetramic acids. The mode of action and herbicidal activity indicate that 3-tetramic acid had good herbicidal performance and was a promising herbicide candidate. This study will provide a reference for novel herbicide development.

Aphasia is a common consequence of stroke and repetitive transcranial magnetic stimulation (rTMS) may be a promising brain stimulation technique in the treatment of aphasia. However, there are few reports about the therapeutic effect of rTMS for Broca's area in patients with sensory aphasia. This study reported one stroke patient with sensory aphasia who received 6 treatment sessions of low-frequency rTMS before speech and language therapy. The target area was the Broca mirror area in the right hemisphere. After treatment, the auditory comprehension of the patient improved from 46 to 112, the naming improved from 18 to 32, and the AQ improved from 34.2 to 42.6. However, the level of functional language, spontaneous speech and repetition did not show obvious improvement.

Flavonoids can sensitize and quench the photolysis of pesticides such as profenofos in surface water. Profenofos has been frequently detected in surface and underground water. The present study investigated the photolysis of profenofos under various conditions, including natural and artificial light illumination, with and without cyanidin-3-O-glucoside (Cy3G) and in pure and surface water. The degradation half-lives of profenofos in distilled water with 10 equivalents Cy3G of profenofos were 21.7 min, 9.5 h, 12.5 h and 180 h under high-pressure mercury light, UV, Xenon lamp and solar irradiation, respectively, while those without Cy3G were 8.1 min, 6.1 h, 8.2 h and 89.9 h, respectively. The photolysis rate of profenofos under sunlight and artificial light was reduced by 1.5-2.7 times due to Cy3G, compared to the Cy3G-free control. Under sunlight irradiation, the effects of Cy3G on profenofos photolysis were larger than those under high-pressure mercury lamp irradiation. Cy3G also significantly reduced the photolysis rate of profenofos under different pH conditions and in natural water. In addition, Cy3G exhibited a significant capacity of scavenging hydroxyl radicals and quenching 1O2 in water. The effect of Cy3G on profenofos photolysis was demonstrated through their interrelations in the natural environment. These findings can help understanding of the effect of flavonoids on profenofos photolysis and are of significance for predicting the degradation kinetics of profenofos and accurately assessing its potential biological impacts.

BACKGROUND: Emotional recognition is a key aspect of mentalizing other people's minds. People with Williams syndrome (WS) are reported to be hypersocial and empathetic; however, few studies have investigated their emotion processing ability. AIMS: This study aimed to examine emotion knowledge in people with WS and to further understand their emotion-related mentalizing ability. METHODS AND PROCEDURES: Six basic emotions (happiness, sadness, anger, fear, disgust, and surprise) were tested by narrating scenarios targeting specific emotions. Participants with WS (n = 16, 8F/8 M, CA = 8.46, MA = 5.27) listened to each scenario and pointed to an emoticon displayed on a card. Correct percentages of each target emotion and replacement by other emotions were analyzed. Comparison groups comprised mental age-matched controls (n = 16, 8F/8 M, CA = 5.26) and chronological age-matched controls (n = 16, 8 F/8 M, CA = 8.31), sixth graders (n = 20, 10F/10M, CA = 12.04), and college students (n = 20, 10F/10M, CA = 20.03). All were Chinese-speaking participants with right handedness. OUTCOMES AND RESULTS: People with WS were delayed compared to the mental age-matched group but differed from the chronological age-matched group in emotion processing. The 6th graders showed different patterns from college students, suggesting that development of emotion processing takes time. CONCLUSIONS AND IMPLICATIONS: These findings suggest (1) a gap in the maturation of emotion knowledge between people with WS and typically developing controls, (2) a nonequivalent chronological emotion-related mentalizing ability in people with WS, and (3) developmental changes in emotion processing from childhood to adulthood.
