RESUMO

Activities of glutathione-dependent enzymes and glucose-6-phosphate dehydrogenase (GPDH) were studied in blood of the patients with lymphosarcoma (LS). The activity of glutathione reductase (GR), glutathione dehydroascorbate reductase (GDAR), gamma-glutamyltranspeptidase (GGT) and G6PDH in plasma, leucocytes, lymphocytes and erythrocytes of peripheral blood in 30 patients (42-56 years) with LS and in 20 healthy have been determined with spectrophotometric methods (Humalyzer 2000 DE). Leucocytes and lymphocytes were separated from blood using Boyum method. Spearmen method used for correlative analysis. The levels of enzymes activity and results of correlative analysis showed an imbalance of antioxidative system defense and metabolic disturbances in patients with LS. The strong functional interrelation was estimated only between GR and G6PDH in the patients' lymphocytes (r=+0,716; p<0.0005). Interrelation was found between the levels of activity of antioxidative enzymes and activity (stage) of pathologic process, this may be used as the additional biochemical test for differential diagnostics of LS and estimation of the cells proliferative activity.

RESUMO

Polymorphisms c.202G > A of prothrombin F2 gene, c.1691G > A of coagulation factor V F5 gene, c.675delinsG of plasminogen activator1 gene, and (-5)T > C Kozak gene of thrombocytic receptor were studied in the Russian and Moldavian ethnic groups. We have found no association between these polymorphisms and the risk of ischemic stroke development in both ethnic groups. No association was revealed between the risk of stroke development and various combinations of the single nucleotide polymorphisms examined in the sample of ischemic stroke patients from Russia.

RESUMO

The authors made a comparative analysis of the activity of antioxidative enzymes and a search for a possible relationship between the activity of glutathione reductase and that of glucose-6-phosphate dehydrogenase in the blood and saliva of patients with breast tumors with varying activity of the pathological process.

Lipid peroxidation (LPO) processes are enhanced and metabolism is disturbed in patients with lymphosarcoma (LS) (non-Hodgkin's lymphoma). The blood enzymatic redox system was analyzed in patients with LS of two types: lymphoblastic LS (LB LS) and prolymphocytic LS (PL LS). The activities of glutathione reductase (GR), glutathione peroxidase (GP), glutathione dehydroascorbate reductase (GDAR), gamma-glutamyl transpeptidase (GGT), and glucose-6-phosphate dehydrogenase (G6PDH) were spectrophotometrically (Humalyzer 2000, DE) determined in the peripheral plasma, white blood cells, lymphocytes, and red blood cells of 32 aged 42-57 years who had LS and 25 healthy individuals. Peripheral blood lymphocytes and leukocytes were obtained by the method developed by A. Böyum. A search for a correlation was made by the Spearman method. The activities of the enzymes and the data of the correlation analysis suggested antioxidant defense system imbalance and metabolic disturbances in patients with LS. Close functional correlations between GR and GP, GR and G6PDH persisted in patients with both types of the disease. Functional relationships between GR and GDAR remained only in patients with PL LS (r = 0.946; p < 0.001). That between GR and GGT was impaired in patients with LS. A correlation between the activity of antioxidant enzymes and the proliferative activity of blood cells was found in patients with LB LS, which may be used as an additional biochemical test in the differential diagnosis of LS.

The activities of antioxidative enzymes (glutathione reductase, glutathione S-transferase) and content of reduced glutathione (GSH), thiocyanate (SCN) and protein were determined in saliva of patients with parodontitis treated with traditional and complex therapy, which additionally included the antihomotoxic preparations Traumeel S ointment, Coenzyme compositum or Lymphomyosot. Inflammation process led to the metabolic disturbances and imbalance of the antioxidative defense system in the patients with parodontitis. The results suggest that complex therapy with the antihomotoxic preparations restored imbalance of the antioxidative defense and was more effective than the traditional therapy alone in the patients with parodontitis. Analysis of interrelation between salivary parameters in patients with parodontitis indicated positive correlation before and after the complex therapy (as an exception there was lack of correlation between content of protein and tiocyanate in the saliva of patients before the beginning of the therapeutic course). So these results reflect activityof pathological process and antioxidant defense imbalance in saliva of patients with parodontitis and may be a basis for recommendation of employment of the complex antihomotoxic therapy as the initial stage of pathological process.

Forty-five patients aged 20-47 years who had mild, moderate, or severe periodontitis and 32 healthy individuals (a control group) were studied during 10-15-day treatment with traditional therapy and combined therapy including the traditional approach and the antihomotoxic agent Traumeel S ointment as a supplement. Increased free radical generation and lipid peroxidation were considered to play an important role in the pathogenesis of periodontitis. Salivary indices are a reflection of a patient's metabolic state and have clinical diagnostic values in patients with oral tissue inflammation. The activities of antioxidative enzymes (glutathione reductase, glutathione-S-transferase, glucose-6-phosphate dehydrogenase) and the content of reduced glutathione (GSH) were determined in the saliva of patients with periodontitis during traditional and complex (traditional + Traumeel S) therapies. Inflammation led to metabolic disturbances and antioxidative defense system imbalance in patients with periodontitis. The findings suggest that the complex therapy with Traumeel S restored antioxidative defense balance and it was more effective than the traditional therapy in patients with periodontitis. An analysis showed a direct correlation between the activity of antioxidative enzymes and clinical characteristics of the disease. These results reflect the activity of a pathological process and the imbalance of antioxidative defense in patients with periodontitis.

Fluorosis caused by long-term intake of high fluoride levels is characterized by clinical bone and tooth manifestations. The adverse impact of high fluoride intake was also observed in soft tissues. Although fluorosis is irreversible it could be prevented by appropriate and timely interventions through the understanding of the process at biochemical and molecular levels. Increased production of reactive oxygen forms and lipid peroxidation are considered to play an important role in the pathogenesis of chronic fluoride toxicity. Saliva is a biological fluid of the human organism may be a reflection of the metabolic state. Salivary indices are clinical diagnostic indicators. The purpose of this investigation was to comparatively study the salivary antioxidative defense system, including glutathione, glutathione reductase, glutathione-S-transferase, and glucose-6-phosphate dehydrogenase in adult patients with fluorosis before and after complex antioxidative therapy. Analysis indicated that there was a negative correlation between the level of glutathione and the clinical characteristics of the disease in patients with fluorosis. There was a direct relationship between the activity of glutathione-S-transferase and the clinical manifestations in the patients. These results reflected dose-dependent fluoride intoxication and metabolic imbalance. The imbalanced salivary antioxidative defense system was in part corrected by complex antioxidative therapy.

The aim of this investigation was to examine glutathione metabolism (concentration of reduced glutathione, the activity of glutathione reductase, glutathione peroxidase, glutathione-S-transferase, gamma-glutamyl transferase) in the tissues of the brain hemispheres in different periods after complicated trauma (V. Kulagin's model). The obtained results show that the trauma causes compression of the soft tissues and therefore induces noticeable changes in glutathione metabolism in the tissues of the brain hemispheres: diminution of the reduced glutathione content early after trauma and its increase in the late period; high activity of glutathione reductase and gamma-glutamyl n-transferase; activation of glutathione-S-transferase after the trauma when peroxidation was the most active.
