PURPOSE: To investigate the rate of bacterial contamination of semitendinosus (ST) tendons during graft harvest in anterior cruciate ligament reconstruction (ACLR), in order to precisely specify the underlying pathogens and obtain data on their susceptibility to potential antibiotics. METHODS: In a prospective study, a total of 59 consecutive patients undergoing primary ACLR were recruited from one centre. No patient had history of previous surgery to the knee or showed clinical signs of infection. Four tissue samples of harvested ST tendons for anterior cruciate ligament (ACL) autografts (case group; ST) were examined for evidence of bacterial colonisation and compared to four tissue samples of the native ACL as negative controls (control group; ACL). Three of the respective samples were subjected to cultural microbiological examination and one to 16S rRNA-PCR. Antibiotic susceptibility testing was performed for each pathogen that was identified. RESULTS: A total of 342 samples were analysed by culture. Significantly more patients showed a positive culture of the ST (33.9%; n = 20/59) compared to 3.4% of patients (n = 2/59) with positive culturing of the ACL (p < 0.0001). Including 16S rRNA-PCR, in a total of 42.4% (25/59) of patients, bacteria were detected in at least one ST sample either by PCR and/or culture. All species found (n = 33) belong to the typical skin flora with Staphylococcus epidermidis (39.4%; n = 13/33) being the most common species, followed by Staphylococcus capitis (24.2%; n = 8/33). All tested isolates (n = 29) were susceptible to vancomycin (29/29, 100%), 69% (n = 20/29) to oxacillin and 65.5% (n = 19/29) to clindamycin. CONCLUSION: ST autografts for ACLR were commonly contaminated with skin commensal bacteria during harvest. One-third of the isolates showed resistance to typical perioperative intravenous antibiotics, whereas all isolates were sensitive to vancomycin. Therefore, routine prophylactic decontamination of all hamstring autografts before implantation should be recommended, preferably with topical vancomycin. LEVEL OF EVIDENCE: Level III.

RESUMO

The advent of X-ray free-electron lasers (XFELs) has revolutionized fundamental science, from atomic to condensed matter physics, from chemistry to biology, giving researchers access to X-rays with unprecedented brightness, coherence and pulse duration. All XFEL facilities built until recently provided X-ray pulses at a relatively low repetition rate, with limited data statistics. Here, results from the first megahertz-repetition-rate X-ray scattering experiments at the Spectroscopy and Coherent Scattering (SCS) instrument of the European XFEL are presented. The experimental capabilities that the SCS instrument offers, resulting from the operation at megahertz repetition rates and the availability of the novel DSSC 2D imaging detector, are illustrated. Time-resolved magnetic X-ray scattering and holographic imaging experiments in solid state samples were chosen as representative, providing an ideal test-bed for operation at megahertz rates. Our results are relevant and applicable to any other non-destructive XFEL experiments in the soft X-ray range.

While stripe phases with broken rotational symmetry of charge density are known to emerge in doped strongly correlated perovskites, the dynamics and heterogeneity of spatial ordering remain elusive. Here we shed light on the temperature dependent lattice motion and the spatial nanoscale phase separation of charge density wave order in the archetypal striped phase in La2-xSrxNiO4+y (LSNO) perovskite using X-ray photon correlation spectroscopy (XPCS) joint with scanning micro X-ray diffraction (SµXRD). While it is known that the CDW in 1/8 doped cuprates shows a remarkable stability we report the CDW motion dynamics by XPCS in nickelates with an anomalous quantum glass regime at low temperature, T < 65 K, and the expected thermal melting at higher temperature 65 < T < 120 K. The nanoscale CDW puddles with a shorter correlation length are more mobile than CDW puddles with a longer correlation length. The direct imaging of nanoscale spatial inhomogeneity of CDW by scanning micro X-ray diffraction (SµXRD) shows a nanoscale landscape of percolating short range dynamic CDW puddles competing with large quasi-static CDW puddles giving rise to a novel form of nanoscale phase separation of the incommensurate stripes order landscape.

Three-dimensional photon correlation spectroscopy (3D PCS) is a well-known technique developed to suppress multiple scattering contributions in correlation functions, which are inevitably involved when an optical laser is employed to investigate dynamics in a turbid system. Here, we demonstrate a proof-of-principle study of 3D PCS in the hard X-ray regime. We employ an X-ray optical cross-correlator to measure the dynamics of silica colloidal nanoparticles dispersed in polypropylene glycol. The obtained cross correlation functions show very good agreement with auto-correlation measurements. This demonstration provides the foundation for X-ray speckle-based studies of very densely packed soft matter systems.

We study the temporal stability of stripe-type spin order in a layered nickelate with x-ray photon correlation spectroscopy and observe fluctuations on timescales of tens of minutes over a wide temperature range. These fluctuations show an anomalous temperature dependence: they slow down at intermediate temperatures and speed up on both heating and cooling. This behavior appears to be directly connected with spatial correlations: stripes fluctuate slowly when stripe correlation lengths are large and become faster when spatial correlations decrease. A low-temperature decay of nickelate stripe correlations, reminiscent of what occurs in cuprates as a result of a competition between stripes and superconductivity, hence occurs via loss of both spatial and temporal correlations.

Polymer production is a major source of greenhouse gas (GHG) emissions. To reduce GHG emissions, the polymer industry needs to shift towards renewable carbon feedstocks such as biomass and CO2. Both feedstocks have been shown to reduce GHG emissions in polymer production, however often at the expense of increased utilization of the limited resources biomass and renewable electricity. Here, we explore synergetic effects between biomass and CO2 utilization to reduce both GHG emissions and renewable resource use. For this purpose, we use life cycle assessment (LCA) to quantify the environmental benefits of the combined utilization of biomass and CO2 in the polyurethane supply chain. Our results show that the combined utilization reduces GHG emissions by 13% more than the individual utilization of either biomass or CO2. The synergies between bio- and CO2-based production save about 25% of the limited resources biomass and renewable electricity. The synergistic use of biomass and CO2 also reduces burden shifting from climate change to other environmental impacts, e.g., metal depletion or land use. Our results show how the combined utilization of biomass and CO2 in polymer supply chains reduces both GHG emissions and resource use by exploiting synergies between the feedstocks.

The cement industry emits 7% of the global anthropogenic greenhouse gas (GHG) emissions. Reducing the GHG emissions of the cement industry is challenging since cement production stoichiometrically generates CO2 during calcination of limestone. In this work, we propose a pathway towards a carbon-neutral cement industry using CO2 mineralization. CO2 mineralization converts CO2 into a thermodynamically stable solid and byproducts that can potentially substitute cement. Hence, CO2 mineralization could reduce the carbon footprint of the cement industry via two mechanisms: (1) capturing and storing CO2 from the flue gas of the cement plant, and (2) reducing clinker usage by substituting cement. However, CO2 mineralization also generates GHG emissions due to the energy required for overcoming the slow reaction kinetics. We, therefore, analyze the carbon footprint of the combined CO2 mineralization and cement production based on life cycle assessment. Our results show that combined CO2 mineralization and cement production using today's energy mix could reduce the carbon footprint of the cement industry by 44% or even up to 85% considering the theoretical potential. Low-carbon energy or higher blending of mineralization products in cement could enable production of carbon-neutral blended cement. With direct air capture, the blended cement could even become carbon-negative. Thus, our results suggest that developing processes and products for combined CO2 mineralization and cement production could transform the cement industry from an unavoidable CO2 source to a CO2 sink.

Direct 2D spatial-coherence measurements are increasingly gaining importance at synchrotron beamlines, especially due to present and future upgrades of synchrotron facilities to diffraction-limited storage rings. We present a method to determine the 2D spatial coherence of synchrotron radiation in a direct and particularly simple way by using the Fourier-analysis method in conjunction with curved gratings. Direct photon-beam monitoring provided by a curved grating circumvents the otherwise necessary separate determination of the illuminating intensity distribution required for the Fourier-analysis method. Hence, combining these two methods allows for time-resolved spatial-coherence measurements. As a consequence, spatial-coherence degradation effects caused by beamline optics vibrations, which is one of the key issues of state-of-the-art X-ray imaging and scattering beamlines, can be identified and analyzed.

Despite intensive studies in the past decades, the local structure of disordered matter remains widely unknown. We show the results of a coherent x-ray scattering study revealing higher-order correlations in dense colloidal hard-sphere systems in the vicinity of their crystallization and glass transition. With increasing volume fraction, we observe a strong increase in correlations at both medium-range and next-neighbor distances in the supercooled state, both invisible to conventional scattering techniques. Next-neighbor correlations are indicative of ordered precursor clusters preceding crystallization. Furthermore, the increase in such correlations is accompanied by a marked slowing down of the dynamics, proving experimentally a direct relation between orientational order and sample dynamics in a soft matter system. In contrast, correlations continuously increase for nonequilibrated, glassy samples, suggesting that orientational order is reached before the sample slows down to reach (quasi-)equilibrium.

We present a systematic 2D spatial-coherence analysis of the soft-X-ray beamline P04 at PETRA III for various beamline configurations. The influence of two different beam-defining apertures on the spatial coherence properties of the beam is discussed and optimal conditions for coherence-based experiments are found. A significant degradation of the spatial coherence in the vertical direction has been measured and sources of this degradation are identified and discussed. The Fourier-analysis method, which gives fast and simple access to the 2D spatial coherence function of the X-ray beam, is used for the experiment. Here, we exploit the charge scattering of a disordered nanodot sample allowing the use of arbitrary X-ray photon energies with this method.

The results of an X-ray cross-correlation analysis (XCCA) study on hard-sphere colloidal crystals and glasses are presented. The article shows that cross-correlation functions can be used to extract structural information beyond the static structure factor in such systems. In particular, the powder average can be overcome by accessing the crystals' unit-cell structure. In this case, the results suggest that the crystal is of face-centered cubic type. It is demonstrated that XCCA is a valuable tool for X-ray crystallography, in particular for studies on colloidal systems. These are typically characterized by a rather poor crystalline quality due to size polydispersity and limitations in experimental resolution because of the small q values probed. Furthermore, nontrivial correlations are observed that allow a more detailed insight into crystal structures beyond conventional crystallography, especially to extend knowledge in structure formation processes and phase transitions.

We present a method to determine the two-dimensional spatial coherence of synchrotron radiation in the soft X-ray regime by analyzing the Fourier transform of the magnetic speckle pattern from a ferromagnetic film in a multidomain state. To corroborate the results, a Young's double-pinhole experiment has been performed. The transverse coherence lengths in vertical and horizontal direction of both approaches are in a good agreement. The method presented here is simple and gives a direct access to the coherence properties of synchrotron radiation without nanostructured test objects.

Capture and utilization of CO2 as alternative carbon feedstock for fuels, chemicals, and materials aims at reducing greenhouse gas emissions and fossil resource use. For capture of CO2, a large variety of CO2 sources exists. Since they emit much more CO2 than the expected demand for CO2 utilization, the environmentally most favorable CO2 sources should be selected. For this purpose, we introduce the environmental-merit-order (EMO) curve to rank CO2 sources according to their environmental impacts over the available CO2 supply. To determine the environmental impacts of CO2 capture, compression and transport, we conducted a comprehensive literature study for the energy demands of CO2 supply, and constructed a database for CO2 sources in Europe. Mapping these CO2 sources reveals that CO2 transport distances are usually small. Thus, neglecting transport in a first step, we find that environmental impacts are minimized by capturing CO2 first from chemical plants and natural gas processing, then from paper mills, power plants, and iron and steel plants. In a second step, we computed regional EMO curves considering transport and country-specific impacts for energy supply. Building upon regional EMO curves, we identify favorable locations for CO2 utilization with lowest environmental impacts of CO2 supply, so-called CO2 oases.

Does the excitation of ultrafast magnetization require direct interaction between the photons of the optical pump pulse and the magnetic layer? Here, we demonstrate unambiguously that this is not the case. For this we have studied the magnetization dynamics of a ferromagnetic cobalt/palladium multilayer capped by an IR-opaque aluminum layer. Upon excitation with an intense femtosecond-short IR laser pulse, the film exhibits the classical ultrafast demagnetization phenomenon although only a negligible number of IR photons penetrate the aluminum layer. In comparison with an uncapped cobalt/palladium reference film, the initial demagnetization of the capped film occurs with a delayed onset and at a slower rate. Both observations are qualitatively in line with energy transport from the aluminum layer into the underlying magnetic film by the excited, hot electrons of the aluminum film. Our data thus confirm recent theoretical predictions.

The success of allogeneic hematopoietic cell transplantation is limited by acute graft-versus-host disease (GvHD), a severe complication accompanied by high mortality rates. Yet, the molecular mechanisms initiating this disease remain poorly defined. In this study, we show that, after conditioning therapy, intestinal commensal bacteria and the damage-associated molecular pattern uric acid contribute to Nlrp3 inflammasome-mediated IL-1ß production and that gastrointestinal decontamination and uric acid depletion reduced GvHD severity. Early blockade of IL-1ß or genetic deficiency of the IL-1 receptor in dendritic cells (DCs) and T cells improved survival. The Nlrp3 inflammasome components Nlrp3 and Asc, which are required for pro-IL-1ß cleavage, were critical for the full manifestation of GvHD. In transplanted mice, IL-1ß originated from multiple intestinal cell compartments and exerted its effects on DCs and T cells, the latter being preferentially skewed toward Th17. Compatible with these mouse data, increased levels of active caspase-1 and IL-1ß were found in circulating leukocytes and intestinal GvHD lesions of patients. Thus, the identification of a crucial role for the Nlrp3 inflammasome sheds new light on the pathogenesis of GvHD and opens a potential new avenue for the targeted therapy of this severe complication.

Rodlike hematite particles in suspension align perpendicular to an external magnetic field due to a negative anisotropy of their magnetic susceptibility Δχ. The diffusion tensor consists of two principal constants D(∥) and D(⊥) for the diffusion parallel and perpendicular to the long particle axis. X-ray photon correlation spectroscopy is capable of probing the diffusive motion in optically opaque suspensions of rodlike hematite particles parallel to the direction of the scattering vector Q. Choosing Q parallel or perpendicular to the direction of an external magnetic field H the direction dependent intermediate scattering function is measured by means of x-ray photon correlation spectroscopy. From the intermediate scattering function in both directions the principal diffusion constants D(∥) and D(⊥) are determined. The ratio D(∥)/D(⊥) increases with increasing aspect ratio of the particles and can be described via a rescaled theoretical approach for prolate ellipsoids of revolution.

RESUMO

We present the first single-shot images of ferromagnetic, nanoscale spin order taken with femtosecond x-ray pulses. X-ray-induced electron and spin dynamics can be outrun with pulses shorter than 80 fs in the investigated fluence regime, and no permanent aftereffects in the samples are observed below a fluence of 25 mJ/cm(2). Employing resonant spatially muliplexed x-ray holography results in a low imaging threshold of 5 mJ/cm(2). Our results open new ways to combine ultrafast laser spectroscopy with sequential snapshot imaging on a single sample, generating a movie of excited state dynamics.
