RESUMO

Mangrove plants, which inhabit and form sensitive ecosystems in the intertidal zones of tropical and subtropical coastlines, though vulnerable to petroleum pollution, still maintain their growth under oil contamination. To elucidate the molecular response of mangrove plants to crude oil-sediment mixture, seeds of Avicennia marina were planted and grown on 0, 2.5, 5.0, 7.5 and10 % (w/w) oil-contaminated soil. Plant biomass was highly affected from 3.05 ± 0.28 (Control) to 0.50 ± .07 (10 %) and from 3.47 ± 0.12 to 1.88 ± 0.08 in 2 and 4 months old plants respectively. The expression analysis of 11genes belonging to detoxification pathways in the roots and leaves of 2 and 4 month-old plants was evaluated by qRT-PCR. Our results showed changes in expression levels of Fe-SOD, Mn-SOD, CAT, PRX, PPOs, GSTs, and NAP2 whose products are involved in reactive oxygen species (ROS) and xenobiotic detoxification. PPOA showed the highest expression induction of 43 ± 1.15, followed by CAT (12.61 ± 3.25) and PPOB (6.38 ± 1.34) in leaves of 2 months old seedlings grown on 7.5, 10 and 7.5 % oil contaminated soil respectively. PPOA (39.23 ± 2.1), PRX (32.13 ± 1.2) as well as PPOB (26.11 ± 1.3) showed the highest expression induction in leaves of 4 months old plants grown in 2.5 % oil contaminated soil. Our data indicated that PPOA can be a good biomarker candidate gene for long term exposure to oil contamination in A. marina.

RESUMO

Manipulation of cell culture media by elicitors is one of most important strategies to inducing secondary metabolism for the production of valuable metabolites. In this investigation, inducing effect of chitosan on physiological, biochemical, and molecular parameters were investigated in cell suspension cultures of Scrophularia striata Boiss. The results showed that chitosan concentration and time of elicitation are determinants of the effectiveness of the elicitor. Accumulation of aromatic amino acids (phenylalanine [Phe] and tyrosine [Tyr]), phenylpropanoid compounds (phenolic acids [PAs] and echinacoside [ECH]), hydrogen peroxide (H2O2) production, phenylalanine ammonia-lyase (PAL) activity and gene expression, and antioxidant enzymes (superoxide dismutase [SOD], peroxidase [POX], catalase [CAT]) activities were altered by changing the exposure time of elicitation. Results showed that, upon elicitation with chitosan, oxidative events were induced, antioxidant responses of S. striata cells were boosted through enhanced activity of an effective series of scavenging enzymes (SOD, CAT, and POX), and biosynthesis of non-enzymatic antioxidants (ECH and PAs [cinnamic, p-coumaric and, caffeic acids]). The increase in amino acid content and PAL activity at early days of exposure to chitosan was related with rises in phenolic compounds. These results provide evidence that chitosan by up-regulation of PAL gene differentially improves the production of phenylpropanoid compounds, which are of medical commercial value with good biotechnological prospects.

RESUMO

Oil contamination of soil limits plants' access to water and nutrients. Leucanthemum vulgare colonized by mycorrhizae could provide an effective tool in remedying oil contamination. Seeds of L. vulgare were planted in pots containing soil mixed with petroleum at 0, 2.5, 5, 7.5, and 10% w/w and propagules of mycorrhizal fungi. Plants were grown under ambient conditions for 16 weeks. Seed germination data were collected weekly for three weeks. Mycorrhizal percentage, spore counts, length and weight of roots and shoots were determined after harvesting. Results showed significant differences in seed germination rates between oil-treated, mycorrhizal and non-mycorrhizal plants. The overall germination rate was greater at 7.5% w/w crude oil contamination (p = 0.05) in mycorrhizal and non-mycorrhizal pots with significant differences between their respective Root:Shoot ratios (both length and weight). Results of this research showed L. vulgare could be germinated and grown in crude oil contaminated soils and could be used to augment plant establishment as part of phytoremediation practices.

RESUMO

The effects of Piriformospora indica and Glomus mosseae on some growth and physiological parameters, and cadmium (Cd) amounts in Triticum aestivum cv. Sardari39 plants under the toxic levels of Cd were investigated. The experiment was carried out including four treatments (P. indica inoculation, G. mosseae inoculation, co-inoculation of G. mosseae and P. indica, and non-inoculation), each having four Cd concentrations (0, 0.3, 0.6 and 0.9 mM Cd). In non-inoculated plants, by increasing Cd concentration in the soil, growth parameters, chlorophyll content, performance index (PI) and Fv/Fm were reduced, whereas root and shoot Cd accumulation were increased. The colonized plants with P. indica and G. mosseae (in solitary or together) had higher growth, chlorophyll content and PI compared to control. In the presence of P. indica, the Cd concentration was increased in roots, whereas it was decreased in shoots under higher soil Cd concentrations. The presence of G. mosseae decreased root Cd concentration under lower Cd concentration in the soil. Results from this study indicate that P. indica inoculation yielded greater readings of growth parameters, chlorophyll content, PI, root colonization and also reduced shoot Cd content compared to G. mosseae inoculation.
