Long-term monoculture may affect soil environment biodiversity. An example of such a plant is radish (Raphanus sativus var. sativus), an economically important crop in Poland, a quick-growing vegetable with intensified harvest throughout the season. The aim of this study was to determine changes in biodiversity of soil under radish cultivation and to compare the research methods applied. The monoculture practice affected soil pH, but the organic carbon content remained stable. 16S RNA-seq analysis revealed changes in soil microbial population, with the dominant phyla Proteobacteria (37.3%), Acidobacteria (19%), and Actinobacteria (16%), and the dominant taxa Gaiella (1.59%), Devosia (1.51%) and Nocardioides (1.43%). These changes have not fully expressed in the number of culturable microorganisms, where only fungal abundance changed significantly. However, the physiological state of microbial cells (λ) indicated that oligotrophs and copiotrophs were in a vegetative (λ > 3.0) state at the beginning of the season and fungi at the end of the year. Changes in the biodiversity of soil microorganisms were visualised using Community Level Physiological Profiling, where an oscillation in Average Well Colour Development (OD560 = 0.78-1.48) was observed in successive months of radish culture, with biodiversity indices (Shannon and Substance richness) remaining similar. The greatest variation in the influence of monoculture practice on soil factors was observed for the soil enzymes activities (for dehydrogenase and peroxidase activities - 0.5 µg TPF/h/g DW and 1.5 µmolPYGL/h/g DW respectively). Alkaline phosphatases predominated among this group of enzymes, and the activity of carbon metabolism enzymes decreased over the season, except for invertases, where an increase in activity of up to 50 µg Glc/h/g DW was observed. All the parameters studied indicated changes in the soil environment. Nevertheless the microbial community remains stable during the whole experiment returning to equilibrium in a quite short time after changing conditions.

RESUMO

Endometrial cancer (EC) is one of the most common malignant tumors among women in the 21st century, whose mortality rate is increasing every year. Currently, the diagnosis of EC is possible only after a biopsy. However, it is necessary to find a new biomarker that will help in both the diagnosis and treatment of EC in a non-invasive way. Circular RNAs (circRNAs) are small, covalently closed spherical and stable long non-coding RNAs (lncRNAs) molecules, which are abundant in both body fluids and human tissues and are expressed in various ways. Considering the new molecular classification of EC, many studies have appeared, describing new insights into the functions and mechanisms of circRNAs in EC. In this review article, we focused on the problem of EC and the molecular aspects of its division, as well as the biogenesis, functions, and diagnostic and clinical significance of circRNAs in EC.

Siderophores are molecules that exhibit a high specificity for iron (Fe), and their synthesis is induced by a deficiency of bioavailable Fe. Complexes of Fe-siderophore are formed extracellularly and diffuse through porins across membranes into bacterial cells. Siderophores can bind heavy metals facilitating their influx into cells via the same mechanism. The aim of the studies was to determine the ability of siderophore-producing bacteria isolated from soils in the north-west part of Wedel Jarlsberg Land (Spitsbergen) to chelate non-Fe metals (Al, Cd, Co, Cu, Hg, Mn, Sn, and Zn). Specially modified blue agar plates were used, where Fe was substituted by Al, Cd, Co, Cu, Hg, Mn, Sn, or Zn in metal-chrome azurol S (CAS) complex, which retained the blue color. It has been proven that 31 out of 33 strains were capable of producing siderophores that bind to Fe, as well as other metals. Siderophores from Pantoea sp. 24 bound only Fe and Zn, and O. anthropi 55 did not produce any siderophores in pure culture. The average efficiency of Cd, Co, Cu, Mn, Sn, and Zn chelation was either comparable or higher than that of Fe, while Al and Hg showed significantly lower efficiency. Siderophores produced by S. maltophilia 54, P. luteola 27, P. luteola 46, and P. putida 49 exhibited the highest non-Fe metal chelation activity. It can be concluded that the siderophores of these bacteria may constitute an integral part of the metal bioleaching preparation, and this fact will be the subject of further research.

OBJECTIVE: Both animal and human studies, though limited, showed that multi-strain probiotic supplementation may reduce the number of seizures and/or seizure severity. Here, we evaluated the effect of a single strain probiotic supplementation on seizure susceptibility, antiseizure efficacy of sodium valproate, and several behavioral parameters in mice. METHODS: Lactobacillus helveticus R0052 was given orally for 28 days. Its influence on seizure thresholds was evaluated in the ivPTZ- and electrically-induced seizure tests. The effect on the antiseizure potency of valproate was assessed in the scPTZ test. We also investigated the effects of probiotic supplementation on anxiety-related behavior (in the elevated plus maze and light/dark box tests), motor coordination (in the accelerating rotarod test), neuromuscular strength (in the grip-strength test), and spontaneous locomotor activity. Serum and brain concentrations of valproate as well as cecal contents of SCFAs and lactate were determined using HPLC method. RESULTS: L. helveticus R0052 significantly increased the threshold for the 6 Hz-induced psychomotor seizure. There was also a slight increase in the threshold for myoclonic and clonic seizure in the ivPTZ test. L. helveticus R0052 did not affect the threshold for tonic seizures both in the maximal electroshock- and ivPTZ-induced seizure tests. No changes in the antiseizure potency of valproate against the PTZ-induced seizures were reported. Interestingly, L. helveticus R0052 increased valproate concentration in serum, but not in the brain. Moreover, L. helveticus R0052 did not produce any significant effects on anxiety-related behavior, motor coordination, neuromuscular strength, and locomotor activity. L. helveticus R0052 supplementation resulted in increased concentrations of total SCFAs, acetate, and butyrate. CONCLUSIONS: Altogether, this study shows that a single-strain probiotic - L. helveticus R0052 may decrease seizure susceptibility and this effect can be mediated, at least in part, by increased production of SCFAs. In addition, L. helveticus R0052 may affect bioavailability of valproate, which warrants further investigations.

RATIONALE: The gut microbiota may play an important role in the development and functioning of the mammalian central nervous system. The assumption of the experiment was to prove that the use of probiotic bacterial strains in the diet of mice modifies the expression of brain proteins involved in metabolic and immunological processes. OBJECTIVES AND RESULTS: Albino Swiss mice were administered with Bifidobacterium longum Rosell®-175 or Lactobacillus rhamnosus JB-1 every 24 h for 28 days. Protein maps were prepared from hippocampal homogenates of euthanized mice. Selected proteins that were statistically significant were purified and concentrated and identified using MALDI-TOF mass spectrometry. Among the analysed samples, 13 proteins were identified. The mean volumes of calcyon, secreted frizzled-associated protein 3, and catalase in the hippocampus of mice from both experimental groups were statistically significantly higher than in the control group. In mice supplemented with Lactobacillus rhamnosus JB-1, a lower mean volume of fragrance binding protein 2, shadow of prion protein, and glycine receptor α4 subunit was observed compared to the control. CONCLUSION: The psychobiotics Bifidobacterium longum Rosell®-175 and Lactobacillus rhamnosus JB-1enhances expression of proteins involved in the activation and maturation of nerve cells, as well as myelination and homeostatic regulation of neurogenesis in mice. The tested psychobiotics cause a decrease in the expression of proteins associated with CNS development and in synaptic transmission, thereby reducing the capacity for communication between nerve cells. The results of the study indicate that psychobiotic bacteria can be used in auxiliary treatment of neurological disorders.

Salvia miltiorrhiza synthesises tanshinones with multidirectional therapeutic effects. These compounds have a complex biosynthetic pathway, whose first rate limiting enzyme is 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGR). In the present study, a new 1646 bp fragment of the S. miltiorrhiza HMGR4 gene consisting of a promoter, 5' untranslated region and part of a coding sequence was isolated and characterised in silico using bioinformatics tools. The results indicate the presence of a TATA box, tandem repeat and pyrimidine-rich sequence, and the absence of CpG islands. The sequence was rich in motifs recognised by specific transcription factors sensitive mainly to light, salicylic acid, bacterial infection and auxins; it also demonstrated many binding sites for microRNAs. Moreover, our results suggest that HMGR4 expression is possibly regulated during flowering, embryogenesis, organogenesis and the circadian rhythm. The obtained data were verified by comparison with microarray co-expression results obtained for Arabidopsis thaliana. Alignment of the isolated HMGR4 sequence with other plant HMGRs indicated the presence of many common binding sites for transcription factors, including conserved ones. Our findings provide valuable information for understanding the mechanisms that direct transcription of the S. miltiorrhiza HMGR4 gene.

Salvia miltiorrhiza is a medicinal plant that synthesises biologically-active tanshinones with numerous therapeutic properties. An important rate-limiting enzyme in the biosynthesis of their precursors is 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGR). This study presents the organ-specific expression profile of the S. miltiorrhiza HMGR4 gene and its sensitivity to potential regulators, viz. gibberellic acid (GA3), indole-3-acetic acid (IAA) and salicylic acid (SA). In addition, it demonstrates the importance of the HMGR4 gene, the hormone used, the plant organ, and the culture environment for the biosynthesis of tanshinones. HMGR4 overexpression was found to significantly boost the accumulation of dihydrotanshinone I (DHTI), cryptotanshinone (CT), tanshinone I (TI) and tanshinone IIA (TIIA) in roots by 0.44 to 5.39 mg/g dry weight (DW), as well as TIIA in stems and leaves. S. miltiorrhiza roots cultivated in soil demonstrated higher concentrations of the examined metabolites than those grown in vitro. GA3 caused a considerable increase in the quantity of CT (by 794.2 µg/g DW) and TIIA (by 88.1 µg/g DW) in roots. In turn, IAA significantly inhibited the biosynthesis of the studied tanshinones in root material.

The effectiveness of phytoremediation is closely related to the various interactions between pollutants, soil particles, rhizosphere microorganisms, and plants. Therefore, the object of current study was a cadmium-tolerant bacterium isolated from the rye rhizosphere, with a high degree of genetic similarity to the genus Chryseobacterium. Chryseobacterium sp. DEMBc1 was able to grow with 36 different BiologGN2 carbon sources and show the adaptation to stress factors such as Cd (100 µg ml-1), low temperature (8 °C), and salinity (2% NaCl). Furthermore, it was shown that DEMBc1 had the characteristics of plant growth-promoting microorganisms: it was able to produce ammonia, indole acetic acid, 1-aminocyclopropane-1-carboxylic acid deaminase, and siderophores, as well as solubilize Ca3(PO4)3. After inoculation with DEMBc1, a significant decrease in the concentration of Cd was observed in the roots of Festuca ovina grown in Cd-polluted soil, compared to the non-inoculated Cd-polluted soil. It was also noticed that DEMBc1 produced a large amount of extracellular polymeric substances that were significantly higher than the cellular biomass. These polymers can form a barrier to reduce the translocation of Cd from the growth medium to the plant roots. According to the current study, DEMBc1 has a stabilizing potential and can decrease the mobility of Cd in the F. ovina rhizosphere, bioaccumulate metals in plant tissues, and effectively improve the bioavailability of nutrients, especially Fe, N, and P in a polluted environments.

The aim of the study was to compare two sources of tannins on fatty acids (FA) composition in rumen. Treatments were (g tannins/kg diet as-feed-basis) as follows: (1) no supplemental tannin addition (CON), (2) addition of 13 g of oak bark extract (OAK), and (3) 4 g of tannic acid (TAN). The basal diet contained 55:45 forage to concentrate ratio. Net consumption of tannins (g/d) was 4 g for both tannins sources. The study was performed on three Polish Mountain ewes fitted with rumen cannulas, and was divided into three experimental periods (I, II, and III). Both sampling time and animal diet had a significant effect on FA profile in the rumen fluid. In general, FA concentrations were higher before feeding in comparison to samples collected 2 and 4 h after feeding. In terms of dietary effect, it was shown that TAN addition had a greater influence on FA profile in the ruminal fluid than the OAK diet. Briefly, in the TAN group significantly increased concentrations of C18:2 c9c12 (linoleic acid, LA) 8 h after feeding (vs. control, CON and OAK), C18:3 c9c12c15 (α-linolenic acid, LNA) 4 h after feeding (vs. OAK), C20:3 n-6 before feeding (vs. CON), C20:4 before feeding (vs. CON and OAK) and 8 h after feeding (vs. OAK) were recorded. In contrast, OAK addition significantly reduced C20:3 n-6 concentration 2 h after feeding (vs. CON). In conclusion, increased concentrations of both LA and LNA in the rumen indicated that supplemental tannic acid may inhibit the initial stage of FA biohydrogenation in the rumen.

The presented study describes the regulation of the promoter region of the Salvia miltiorrhiza kaurene synthase-like gene (SmKSL) by ethylene and yeast extract. The isolated fragment is 897 bp and is composed of a promoter (763 bp), 5'UTR (109 bp), and a short CDS (25 bp). The initial in silico analysis revealed the presence of numerous putative cis-active sites for trans-factors responding to different stress conditions. However, this study examines the influence of ethylene and yeast extract on SmKSL gene expression and tanshinone biosynthesis regulation. The results of 72h RT-PCR indicate an antagonistic interaction between ethylene, provided as ethephon (0.05, 0.10, 0.25, and 0.50 mM), and yeast extract (0.5%) on SmKSL gene expression in callus cultures of S. miltiorrhiza. A similar antagonistic effect was observed on total tanshinone concentration for up to 60 days. Ethylene provided as ethephon (0.05, 0.10, 0.25, and 0.50 mM) is a weak inducer of total tanshinone biosynthesis, increasing them only up to the maximum value of 0.67 ± 0.04 mg g-1 DW (60-day induction with 0.50 mM ethephon). Among the tanshinones elicited by ethephon, cryptotanshinone (52.21%) dominates, followed by dihydrotanshinone (45.00%) and tanshinone IIA (3.79%). In contrast, the 0.5% yeast extract strongly increases the total tanshinone concentration up to a maximum value of 13.30 ± 1.09 mg g-1 DW, observed after 50 days of induction. Yeast extract and ethylene appear to activate different fragments of the tanshinone biosynthesis route; hence the primary tanshinones induced by yeast extract were cryptotanshinone (81.42%), followed by dihydrotanshinone (17.06%) and tanshinone IIA (1.52%).

OBJECTIVE: The aim of the study was to compare the effect of two plant additives, rich in polyphenolic compounds, supplemented to sheep diets on microorganisms and carbohydrate fermentation in rumen. METHODS: In the experiment, 6 ewes of the Polish Mountain breed were fitted with ruminal cannulas. Sheep were divided into three feeding groups. The study was performed in a crossover design of two animals in each group, with three experimental periods (n = 6 per each group). The animals were fed a control diet (CON) or additionally received 3 g of dry and milled lingonberry leaves (VVI) or oak bark (QUE). Additionally, plant material was analyzed for tannins concentration. RESULTS: Regardless of sampling time, QUE diet increased the number of total protozoa, as well as Entodinium spp., Diplodinium spp. and Isotrichidae family, while decreased bacterial mass. In turn, a reduced number of Diplodinium spp. and increased Ophryoscolex spp. population were noted in VVI fed sheep. During whole sampling time (0, 2, 4, and 8 h), the number of protozoa in ruminal fluid of QUE sheep was gradually reduced as opposed to animals receiving CON and VVI diet, where rapid shifts in the protozoa number were observed. Moreover, supplementing sheep with QUE diet increased molar proportions of butyrate and isoacids in ruminal fluid. Unfortunately, none of the tested additives affected gas production. CONCLUSION: The addition of VVI or QUE in a small dose to sheep diets differently affected rumen microorganisms and fermentation parameters, probably because of various contribution of catechins in tested plant materials. However, it is stated that QUE diet seems to create more favorable conditions for growth and development of ciliates. Nonetheless, the results of the present study showed that VVI and QUE additives could serve as potential natural modulators of microorganism populations and, consequently, carbohydrate digestion in ruminants.

The purpose of this study was to compare the effect of the presence of protozoa in the rumen of wild roe deer (Capreolus capreolus) on the bacteria composition and digestion rate of the main carbohydrates of forage. The research material involved rumen content and rumen fluid, which were collected in the autumn-winter season, from eight adult males of roe deer with an average body mass of 22.6 kg. The microscopic analysis demonstrated that there were only protozoa in 50% of the animals sampled. Qualitative analysis revealed the presence of protozoa belonging to the genus Entodinium. The density of protozoal population varied from 6.5 to 38.7 × 105/mL rumen fluid. The analysis of bacteria composition indicated that protozoa did not have an effect on bacterial diversity. Furthermore, the results of hydrolytic activity revealed that the fastest digestion of carbohydrates was for pectin, while the slowest was inulin. The pH and redox potential in the rumen varied from 5.9 to 6.1 and from -248.1 to -251.1 mV, respectively. In summary, the presence of protozoa in the rumen of wild roe deer does not have an effect on the bacterial population and has no effect on the digestion rate of carbohydrates in the rumen.

SIGNIFICANCE: Optical coherence tomography speckle is used here in an unconventional way as the indirect source of information on tissue microstructure. The study reveals that the corneal speckle of glaucoma suspects has a similar relationship between the parameters of scattering exhibited in glaucoma patients. PURPOSE: The purpose of this study was to evaluate the potential of optical coherence tomography imaging of the cornea in early diagnosis of glaucoma. METHODS: Corneas of 64 subjects of three groups, including 18 primary open-angle glaucoma patients, 24 glaucoma suspects with normal levels of IOP and uncompromised visual field, and 22 age-matched controls, were imaged with spectral optical coherence tomography. A central region of interest in each optical coherence tomography image visualizing the stroma was automatically extracted, and the intensity data within that region were fit with the generalized gamma distribution. Its parameters describe the scatterer cross section and scatterer density and indirectly assess corneal microstructure. In addition, subjects underwent measurements of IOP, visual field, Heidelberg Retinal Tomography, retinal nerve fiber layer thickness, and biometry. Group means of all parameters were tested for equality. Within each group of subjects, correlation was evaluated between the statistical parameters of the corneal speckle. RESULTS: Glaucoma suspects had statistically significantly different IOP, visual field parameters, mean retinal fiber layer thickness, and central corneal thickness from those of glaucoma patients but not from those of the control group. The parameters of the corneal speckle were not significantly different between the groups, but they showed a markedly higher and statistically significant coefficient of determination for glaucoma patients and suspects than that for the control group (0.927 and 0.707 vs. 0.336). CONCLUSIONS: The results indicate that glaucoma suspects have similar relationship between the corneal scatterer cross section and scatterer density to that exhibited in the glaucoma patients but markedly different from that of healthy controls.

Both hormonal balance and plant growth may be shaped by microorganisms synthesizing phytohormones, regulating its synthesis in the plant and inducing plant resistance by releasing elicitors from cell walls (CW) by degrading enzymes (CWDE). It was shown that the Trichoderma DEMTkZ3A0 strain, isolated from a healthy rye rhizosphere, colonized the rhizoplane of wheat seedlings and root border cells (RBC) and caused approximately 40% increase of stem weight. The strain inhibited (in over 90%) the growth of polyphagous Fusarium spp. (F. culmorum, F. oxysporum, F. graminearum) phytopathogens through a mechanism of mycoparasitism. Chitinolytic and glucanolytic activity, strongly stimulated by CW of F. culmorum in the DEMTkZ3A0 liquid culture, is most likely responsible for the lysis of hyphae and macroconidia of phytopathogenic Fusarium spp. as well as the release of plant resistance elicitors. In DEMTkZ3A0 inoculated plants, an increase in the activity of the six tested plant resistance markers and a decrease in the concentration of indoleacetic acid (IAA) auxin were noted. IAA and gibberellic acid (GA) but also the 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase (ACCD) enzyme regulating ethylene production by plant were synthesized by DEMTkZ3A0 in the liquid culture. IAA synthesis was dependent on tryptophan and negatively correlated with temperature, whereas GA synthesis was positively correlated with the biomass and temperature.

The objective of the study was to assess the physiological diversity and metabolic activity of the soil bacterial communities inhabiting Spitsbergen soils in search of bacterial abilities facilitating plant growth promotion. In the soil, the total number of culturable microorganisms, the number of their individual physiological groups (including Siderophore Synthesizing; SSB and Phosphate Solubilizing Bacteria; PSB), the dehydrogenase (DH) activity, and the ability to utilize sources of C, N, P (EcoPlate) were analysed. In bacterial isolates, siderophores production, ACC (1-aminocyclopropane-1-carboxylate) deaminase (ACCD) activity, IAA (indole-3-acetic acid) synthesis were examined. The isolates were applied to the seeds of Phaseolus coccineus regarding their germination and root length. The results showed differences between copio- and oligotrophic bacteria. A usually high number of SSB was accompanied by the raised number of PSB. A bigger number of SSB was connected with low values of Fe in the soil. High DH activity was assisted by greater number of copio- and oligotrophic bacteria, raised average well color development value, and N and C contents in the soil. Germination index was more alike relative seed germination than relative root growth. IAA concentration and ACCD activity were conversely related. Synthesis of siderophores was matched with ACCD activity and its high level was combined with elevated germination index. In spite of different localization of soil samples, some isolates proved similar traits of activity. Distinct affiliation of isolates and their various localizations were displayed. Among all isolates tested, some possessed one main trait of activity, but most of them had two or more significant features for potential plant growth stimulation. These isolates could be an important source of useful bacteria.

OBJECTIVE: The purpose of this study was to evaluate the effect of rapeseed and linseed oil supplementations on performance and meat quality of lambs. METHODS: The experiment was conducted on 18 growing (100-day-old) lambs of 19.7±1.9 kg live weight, assigned to 3 groups of 6 animals each. Control lambs were fed meadow hay and concentrate alone. Experimental animals additionally received rapeseed or linseed oils at a dose of 50 g/d. The lambs were slaughtered at an average body weight of 35.7±0.5 kg. RESULTS: The dressing percentage was higher in lambs fed rapeseed oil. Total saturated fatty acids (SFA) and C15:0, C16:0, C17:0, C21:0, C24:0 were lower in longissimus dorsi muscle (MLD) in lambs fed linseed oil. Supplementation of diet with linseed oil decreased concentrations of total monounsaturated fatty acids and C16:1, C17:1, C18:1 cis-9 in MLD. The concentrations of n-3 polyunsaturated fatty acids (PUFA) and C18:3 n-3, C20:5 n-3 in MLD were higher in lambs fed linseed oil than in other groups. Oils supplementation to diets resulted in increased concentration of C22:6 n-3 in MLD. The inclusion of linseed oil into the diet increased the contents of total PUFA, n-3 PUFA and C18:3 n-3, C20:5 n-3, C22:6 n-3 in semitendinosus muscle in comparison to control. A tendency towards a lower n:6/n:3 ratio in MLD was observed when lambs were supplemented linseed oil. CONCLUSION: The supplementation of linseed oil to diets seems to reduce the concentration of SFA and increase the concentration of n-3 PUFA. The n-6/n-3 ratio is an important nutritional factor, and its value has been favorably decreased below 2, thereby achieving an important target related to human health. Due to these changes carcass fatty acid profile was improved, and so enhanced lamb meat healthy properties.

The aim of this study was to compare the effect of rapeseed and linseed oils added to the diet on pancreatic enzyme activities in sheep. The experiment was conducted on six adult sheep with a catheter introduced into the common bile-pancreatic duct and a T-cannula into the duodenum. The animals were divided into three groups, two sheep in each. A twice-replicated 3 × 3 Latin square was used in the experimental design. The sheep were fed meadow hay and the concentrate alone or the same ration supplemented with rapeseed or linseed oils at a dose of 5% of the basal diet. After adaptation period, a mixture of pancreatic and bile juice was collected for three consecutive days. The secretion of bile-pancreatic juice showed an increasing trend in sheep fed rapeseed and linseed oils (69.5 and 68.5 ml/hr respectively) in comparison with control ones (59.8 ml/hr). Lipase and trypsin activities were significantly increased when sheep were fed diets with rapeseed or linseed oils (175 and 21.6 or 179 and 23.2 U/L respectively) in comparison with those fed control diet (128 and 13.1 U/L respectively). It was concluded that oil as a dietary supplement can mainly modify the secretion of bile-pancreatic juice and enzymatic activity of the pancreas and also might affect animal production.

OBJECTIVE: Aim: To assess the prevalence of metabolic syndrome (MS) components in overweight or obese children and adolescents, as well as analyze the risk factors of its occurrence. PATIENTS AND METHODS: Material and methods: The study was conducted in a group of 70 children and adolescents aged 5-18 hospitalized in the Department of Gastroenterology, Allergology and Pediatrics, Polish Mother's Memorial Hospital - Research Institute in Lodz (Poland) based on the results of medical histories, physical examination, biochemical investigation, and calculation of the Homeostatic Model Assessment of Insulin Resistance (HOMA-IR) index. RESULTS: Results: MS was diagnosed in 14 children (20%). The most common abnormalities besides obesity included: decreased High Density Lipoprotein Cholesterol (HDL-C) levels (n=13, 92.9%), increased triglycerides (TG) concentrations (n=10, 71.4%) and arterial hypertension (n=10, 71,4%). Among all the children, insulin resistance was diagnosed in 29 subjects (41.4%). The results of univariate logistic regression showed that the occurrence of lipid disorders, obesity, hypertension and diabetes in their parents, as well as the duration of pregnancy, birth weight, or breastfeeding were not associated with the risk of MS development in the subjects (p>0.05). However, in the study group, 92.9% of subjects had one or more particular risk factor for MS development. CONCLUSION: Conclusions: Besides visceral obesity, lipid disorders were the most frequently observed components of MS in the subjects analyzed, which may have prognostic significance. The occurrence of one or more MS risk factors in almost all of the children studied indicates the increased risk of cardiovascular diseases in the studied group in the future.

RESUMO

The complete exploration of the regulation of gene expression remains one of the top-priority goals for researchers. As the regulation is mainly controlled at the level of transcription by promoters, study on promoters and findings are of great importance. This review summarizes forty selected databases that centralize experimental and theoretical knowledge regarding the organization of promoters, interacting transcription factors (TFs) and microRNAs (miRNAs) in many eukaryotic and prokaryotic species. The presented databases offer researchers valuable support in elucidating the regulation of gene transcription.

PURPOSE: Tonometry is widely used as the main screening tool supporting glaucoma diagnosis. Still, its accuracy could be improved if full knowledge about the variation of the corneal biomechanical properties was available. In this study, Optical Coherence Tomography (OCT) speckle statistics are used to infer the organisation of the corneal micro-structure and hence, to analyse its influence on intraocular pressure (IOP) measurements. METHODS: Fifty-six subjects were recruited for this prospective study. Macro and micro-structural corneal parameters as well as subject age were considered. Macro-structural analysis included the parameters that are associated with the ocular anatomy, such as central corneal thickness (CCT), corneal radius, axial length, anterior chamber depth and white-to-white corneal diameter. Micro-structural parameters which included OCT speckle statistics were related to the internal organisation of the corneal tissue and its physiological changes during lifetime. The corneal speckle obtained from OCT was modelled with the Generalised Gamma (GG) distribution that is characterised with a scale parameter and two shape parameters. RESULTS: In macro-structure analysis, only CCT showed a statistically significant correlation with IOP (R2 = 0.25, p<0.001). The scale parameter and the ratio of the shape parameters of GG distribution showed statistically significant correlation with IOP (R2 = 0.19, p<0.001 and R2 = 0.17, p<0.001, respectively). For the studied group, a weak, although significant correlation was found between age and IOP (R2 = 0.053, p = 0.04). Forward stepwise regression showed that CCT and the scale parameter of the Generalised Gamma distribution can be combined in a regression model (R2 = 0.39, p<0.001) to study the role of the corneal structure on IOP. CONCLUSIONS: We show, for the first time, that corneal micro-structure influences the IOP measurements obtained from noncontact tonometry. OCT speckle statistics can be employed to learn about the corneal micro-structure and hence, to further calibrate the IOP measurements.
