RESUMO

We evaluated the expression of the Fas receptor gene in Vero cells infected with the Lederle vaccine strain of canine distemper virus using RT-PCR. Vero cells were plated, and after being grown for 24 h in MEM with 5% FBS, 80-90% confluent monolayer cultures were infected with the virus. The cells were harvested at 3, 6, 9, and 15 h post-infection. Uninfected Vero cells were used as a control. Total RNA was isolated from Vero cells using 1 mL Trizol(®) LS, and RT was performed using 2 µg total RNA. Primer pairs for RT-PCR amplification for the canine distemper virus nucleocapsid gene, the S26 reference gene, and the Vero rFas gene were used to analyze expression in Vero cells. RT-PCR results revealed virus activity at 3, 6, 9, and 15 h in the virus-infected Vero cells. The S26 housekeeping gene was amplified in virus infected and control samples. However, expression of the cell death receptor Fas was detected in Vero cells only at 15 h post-infection. We suggest that the Lederle vaccine induces apoptosis by Fas receptor signaling, possibly through caspase-8 signaling rather than through mitochondrial signaling in the infected cells.

RESUMO

The use of an external transverse magnetic field to trigger and to control electron self-injection in laser- and particle-beam driven wakefield accelerators is examined analytically and through full-scale particle-in-cell simulations. A magnetic field can relax the injection threshold and can be used to control main output beam features such as charge, energy, and transverse dynamics in the ion channel associated with the plasma blowout. It is shown that this mechanism could be studied using state-of-the-art magnetic fields in next generation plasma accelerator experiments.

RESUMO

The concepts of matched-beam, self-guided laser propagation and ionization-induced injection have been combined to accelerate electrons up to 1.45 GeV energy in a laser wakefield accelerator. From the spatial and spectral content of the laser light exiting the plasma, we infer that the 60 fs, 110 TW laser pulse is guided and excites a wake over the entire 1.3 cm length of the gas cell at densities below 1.5 × 10(18) cm(-3). High-energy electrons are observed only when small (3%) amounts of CO2 gas are added to the He gas. Computer simulations confirm that it is the K-shell electrons of oxygen that are ionized and injected into the wake and accelerated to beyond 1 GeV energy.

RESUMO

A method, which utilizes the large difference in ionization potentials between successive ionization states of trace atoms, for injecting electrons into a laser-driven wakefield is presented. Here a mixture of helium and trace amounts of nitrogen gas was used. Electrons from the K shell of nitrogen were tunnel ionized near the peak of the laser pulse and were injected into and trapped by the wake created by electrons from majority helium atoms and the L shell of nitrogen. The spectrum of the accelerated electrons, the threshold intensity at which trapping occurs, the forward transmitted laser spectrum, and the beam divergence are all consistent with this injection process. The experimental measurements are supported by theory and 3D OSIRIS simulations.

RESUMO

We investigate the influence of a tilted laser-pulse-intensity front on laser-wakefield acceleration. Such asymmetric light pulses may be exploited to obtain control over the electron-bunch-pointing direction and in our case allowed for reproducible electron-beam steering in an all-optical way within an 8 mrad opening window with respect to the initial laser axis. We also discovered evidence of collective electron-betatron oscillations due to off-axis electron injection into the wakefield induced by a pulse-front tilt. These findings are supported by 3D particle-in-cell simulations.

RESUMO

The acceleration of electrons to approximately 0.8 GeV has been observed in a self-injecting laser wakefield accelerator driven at a plasma density of 5.5x10(18) cm(-3) by a 10 J, 55 fs, 800 nm laser pulse in the blowout regime. The laser pulse is found to be self-guided for 1 cm (>10zR), by measurement of a single filament containing >30% of the initial laser energy at this distance. Three-dimensional particle in cell simulations show that the intensity within the guided filament is amplified beyond its initial focused value to a normalized vector potential of a0>6, thus driving a highly nonlinear plasma wave.

RESUMO

A laser wakefield acceleration study has been performed in the matched, self-guided, blowout regime producing 720 +/- 50 MeV quasimonoenergetic electrons with a divergence Deltatheta_{FWHM} of 2.85 +/- 0.15 mrad using a 10 J, 60 fs 0.8 microm laser. While maintaining a nearly constant plasma density (3 x 10{18} cm{-3}), the energy gain increased from 75 to 720 MeV when the plasma length was increased from 3 to 8 mm. Absolute charge measurements indicate that self-injection of electrons occurs when the laser power P exceeds 3 times the critical power P{cr} for relativistic self-focusing and saturates around 100 pC for P/P{cr} > 5. The results are compared with both analytical scalings and full 3D particle-in-cell simulations.

Lentinula edodes, commonly called shiitake, is considered a choice edible mushroom with exotic taste and medicinal quality. L. edodes grows very well and produces a range of enzymes when cultivated on eucalyptus residues. Development of appropriate experimental procedures for recovery and determination of enzymes became a widely important cash crop. In this work, enzymes produced by L. edodes were extracted using different pH buffer and determined regarding peroxidases and proteases. Lignin peroxidase (LiP) was not detected in the extracts based on veratryl alcohol or azure B oxidation. Proteases were very low while Mn-peroxidases (MnP) predominated. The optimal pH for MnP recovery was 5.0, under agitation at 25 degrees C. The oxidation of phenol red decreased after dark-colored small compounds or ions were eliminated by dialysis. The extract of L. edodes contained components of high molecular weight, such as proteases or high polyphenol, that could be involved in the LiP inactivation. L. edodes sample previously submitted to dialysis was also joined to LiP of Phanerochaete chrysosporium and a total inhibition of LiP was observed.

With previously defined criteria of inclusion and previous consent, twenty six consecutive patients (19 to 64 years old), with upper digestive symptoms, were submitted to endoscopy, with biopsy, constant of eight samples of the antropyloric region (four of the anterior aspect and four of the posterior aspect). Two samples were been for culture; two for free urease test, two for smears; all gathered in adequate transport medium over refrigeration. Two samples immersed in formaline to 10% for histopathologic exam. 25/26 (96%) of the patients showed infection by H. pylori by means of one or more methods utilized. In 16/26 (61%), alterations were observed in endoscopy (gastric inflammation in eleven, peptic ulcer in two and ulcer scars in three cases). Of the patients with endoscopic gastric inflammation, presented positive as well as all (100%) bearers of scar or peptic ulcer. A close relationship was observed between the presence of H. pylori and chronic gastric inflammation 24/25 (96%). Histopathologic slices stained by hematoxilin-eosin was the test of highest diagnostic sensitivity 24/25 (96%), followed by urease test 23%25 (92%), stained smears 19/25 (76%) and culture 18/25 (72%). Our conclusion is that the prevalence of gastric infection for H. pylori in symptomatic patients is high, correlated with chronic gastric inflammation and ulcers. Histopathologic slices stained by hematoxilin-eosin and free urease test are the most sensitive indicators of the presence of H. pylori. This study should proceed for further elucidation of questions realised and include a control group of symptomatics individuals paired for sex and age.
