BACKGROUND: Enterotoxigenic E. coli (ETEC) significantly contribute to diarrhea in piglets and weaners. The smallholder pig producers in Uganda identified diarrhea as one of the major problems especially in piglets. The aim of this study was to; i) characterize the virulence factors of E. coli strains isolated from diarrheic and non-diarrheic suckling piglets and weaners from smallholder herds in northern and eastern Uganda and ii) identify and describe the post-mortem picture of ETEC infection in severely diarrheic piglets. Rectal swab samples were collected from 83 piglets and weaners in 20 herds and isolated E. coli were characterized by PCR, serotyping and hemolysis. RESULTS: The E. coli strains carried genes for the heat stable toxins STa, STb and EAST1 and adhesins F4 and AIDA-I. The genes for the heat labile toxin LT and adhesins F5, F6, F18 and F41 were not detected in any of the E. coli isolates. Where the serogroup could be identified, E. coli isolates from the same diarrheic pig belonged to the same serogroup. The prevalence of EAST1, STb, Stx2e, STa, AIDA-I, and F4 in the E. coli isolates from suckling piglets and weaners (diarrheic and non-diarrheic combined) was 29, 26.5, 2.4, 1.2, 16, and 8.4 %, respectively. However the prevalence of F4 and AIDA-I in E. coli from diarrheic suckling piglets alone was 22.2 and 20 %, respectively. There was no significant difference in the prevalence of the individual virulence factors in E. coli from the diarrheic and non-diarrheic pigs (p > 0.05). The main ETEC strains isolated from diarrheic and non-diarrheic pigs included F4/STb/EAST1 (7.2 %), F4/STb (1.2 %), AIDA/STb/EAST1 (8 %) and AIDA/STb (8 %). At post-mortem, two diarrheic suckling piglets carrying ETEC showed intact intestinal villi, enterocytes and brush border but with a layer of cells attached to the brush border, suggestive of ETEC infections. CONCLUSION: This study has shown that the F4 fimbriae is the most predominant in E. coli from diarrheic piglets in the study area and therefore an F4-based vaccine should be considered one of the preventive measures for controlling ETEC infections in the piglets in northern and eastern Uganda.

BACKGROUND: The quality of the floor is essential to the welfare of piglets as abrasions often are recorded in newborn piglets, and such lesions may lead to lameness. Apart from animal suffering, lameness contributes to losses in form of dead piglets, decreased growth, and increased use of antibiotics and manual labour. METHODS: In a herd with three different farrowing systems, 37 litters (390 piglets) were studied until the age of 3 weeks with respect to presence of skin wounds and abrasions. Lameness was registered until the age of 7 weeks. Eight lame piglets were sacrificed before medical treatment and subjected to necropsy including histopathological and microbiological examinations. Isolates of streptococci, staphylococci and E. coli were tested with respect to antimicrobial resistance. Mastitis was observed in ten sows. RESULTS: The most severe abrasions at carpus and soles were seen in the system with a new solid concrete floor with a slatted floor over the dunging area. The lowest magnitude was observed in the deep litter system with peat. Sole bruising was more common in the systems with concrete floor compared to the deep litter system with peat, and the differce in prevalence was significant at all examination days. The lesions decreased with time and about 75% of the treatments for lameness were performed during the first three weeks of life. The overall prevalence of lameness was highest in the system with new solid concrete floor with a slatted floor over the dunging area (9.4%) followed by the old solid concrete floor (7.5%). A lower (p < 0.05) prevalence was seen in the deep litters system with peat (3.3%). No significant relationship between mastitis and abrasions or lameness in the offspring was observed. CONCLUSION: There were large differences in the prevalence of abrasions and lameness between the floor types. The deep litter system with peat provided a soft and good floor for piglets. The overall prevalence of lameness was only diagnosed in every fourth litter in that system compared to in every second litter in the systems with concrete floor. In contrast, the incidence of mastitis in the sows during the first week after farrowing was higher than in the systems with concrete floor.

BACKGROUND: Oedema disease is a severe disease, mainly affecting recently weaned pigs. It is caused by E. coli strains that express fimbriae F18 and produce verotoxin 2e, mainly belonging to serotype O138, O139 or O141. The aim of this study was to compare E. coli isolates within these serotypes with respect to diversity. METHODS: Faecal E. coli strains belonging to serotypes O138, O139 and O141 isolated during the period 1994-1998 from Swedish pigs aged less than 12 weeks were compared using a biochemical fingerprinting system. Aiming to compare the results obtained over time, also strains isolated during 1964-67 and 1975-80 were included in the study. The study comprised 129, 263 and 95 isolates of E. coli serotype O138, O139 and O141, respectively. RESULTS: Biochemical phenotypes (BPTs) were defined. At each sampling occasion each herd could only contribute with one isolate per BPT. Consequently, all but one of identical BPTs identified at a specific sampling occasion was omitted. The final number of isolates from 1994-98 that was compared included 64, 182 and 41 isolates of serotypes O138, O139 and O141, respectively. Within each serotype, the dominating BPT included over 65% of the compared isolates, demonstrating a large dominance of one BPT per serotype. These dominating BPTs were also demonstrated in the material from the 1960ies and the 1970ies. Still, the presence of other common BPTs (especially within serotype O138 and O139) demonstrated a certain variation within serotype. In a herd severely affected by oedema disease, E. coli serotype O139 was easily demonstrated in diseased pigs but only rarely in apparently healthy weaners CONCLUSION: The results obtained demonstrate the presence of dominating BPTs within the oedema disease inducing serotypes. A stability of these BPTs over time was observed, presumably at least partly due to a never-ending access to naïve pigs. Still, the presence of other common BPTs indicates a variation over time, which visualises the importance of monitoring for this. Such studies should focus on pigs affected by oedema disease, because oedema disease inducing strains of E. coli were only rarely demonstrated in healthy pigs in a herd affected by oedema disease.
