RESUMO

A combined resistivity and hard x-ray diffraction study of superconductivity and charge ordering in Ir Ir1-xPtxTe2, as a function of Pt substitution and externally applied hydrostatic pressure, is presented. Experiments are focused on samples near the critical composition x c ~ 0.045 where competition and switching between charge order and superconductivity is established. We show that charge order as a function of pressure in Ir0.95Pt0.05Te2 is preempted - and hence triggered - by a structural transition. Charge ordering appears uniaxially along the short crystallographic (1, 0, 1) domain axis with a (1/5, 0, 1/5) modulation. Based on these results we draw a charge-order phase diagram and discuss the relation between stripe ordering and superconductivity.

UNLABELLED: Biovolatilization and bioaccumulation capabilities of different elements by microscopic filamentous fungus Scopulariopsis brevicaulis were observed. Accumulation of As(III), As(V), Se(IV), Se(VI), Sb(III), Sb(V), Te(IV), Te(VI), Hg(II), Tl(I) and Bi(III) by S. brevicaulis was quantified by analysing the amount of elements in biomass of the fungus using ICP AAS. The highest amounts of bioaccumulated metal(loid)s were obtained as follows: Bi(III) > Te(IV) > Hg(II) > Se(IV) > Te(VI) > Sb(III) at different initial contents, with Bi(III) accumulation approximately 87%. The highest percentages of volatilization were found using Hg(II) (50%) and Se(IV) (46·5%); it was also demonstrated with all studied elements. This proved the biovolatilization ability of microscopic fungi under aerobic conditions. The highest removed amount was observed using Hg(II) (95·30%), and more than 80% of Se(IV), Te(IV), Bi(III) and Hg(II) was removed by bioaccumulation and biovolatilization, which implies the possibilities of use of these processes for bioremediations. There were reported significant differences between bioaccumulation and biovolatilization of almost all applied metal(loid)s if valence is mentioned. SIGNIFICANCE AND IMPACT OF THE STUDY: Microbial accumulation and volatilization are natural processes involved in biogeochemical cycles of elements. Despite their impact on mobility, bioavailability and toxicity of various metal(loid)s, only few papers deal with these processes under aerobic conditions with microscopic fungi. Thus, the proving of ability of microscopic fungus Scopulariopsis brevicaulis to accumulate and transform metals and metalloids by methylation or alkylation and quantification of these processes were demonstrated. The results can provide basic information on natural elements cycling and background for more specific studies focusing, for example, on application of these processes in mitigation of metal(loid) contamination.

BACKGROUND: There is increased worldwide concern about the consequences of folic acid and vitamin B12 deficiencies on health, which include megaloblastic anemia, neural tube defects and cardiovascular disease. OBJECTIVE: This study intended to determine the prevalence of folic acid and vitamin B12 deficiencies in vulnerable groups in labor and poor socioeconomic strata of the Venezuelan population. METHODS: A total of 5658 serum samples were processed to determine folic acid and vitamin B12 concentrations. The study involved three surveys performed during 2001-2002 and included infants, children, adolescents and pregnant women from labor and poor socioeconomic strata of the population. The method used was a radio immunoassay designed for the simultaneous measurement of serum folic acid and vitamin B12. RESULTS: The prevalence of folic acid deficiency was higher than 30% for all groups studied, reaching 81.79% in adolescents. Vitamin B12 deficiency was 11.4% in samples collected nationwide, but there was also a similar prevalence of high serum levels. The prevalence of folic acid and vitamin B12 deficiencies in pregnant women reached 36.32 and 61.34%, respectively. CONCLUSION: This work shows that there is a high prevalence of folic acid deficiency, especially in women of reproductive age, pregnant adolescents and in the whole population studied in Vargas state. This situation requires immediate intervention as supplementation or food fortification programs.

An optimised BCR three steps sequential extraction procedure (BCR SEP) and several single extractions with KCl, NH(4)Cl, Na(4)P(2)O(7) and 0.5 mol L(-1) HCl were used for the fractionation of Al, Cu, Fe, Mn, Pb and Zn in CRMs and in samples from a mining area with sulphidic deposits. A good interlaboratory comparability was obtained for Cu, Pb and Zn in CRM 483, CRM 701, SRM 2710 and SRM 2711 by BCR SEP. The reliability of the results obtained is also very satisfactory. Some differences were found between our results and the indicative data for Al and Fe fractionation. However, serious discrepancies were found for Mn, not only for individual steps of the fractionation, but for the data obtained overall (sum of 1-3 steps), and for the total concentration as well. Our results could be utilized as a contribution to the existing indicative values for CRM 483, SRM 2710 and SRM 2711 for interlaboratory study. Moreover, data for the fractionation of elements mentioned above for CRM 701 are first presented here.A high correlation between 0.5 mol L(-1) of HCl-extractable amounts of the elements studied, and the sum of the three steps of BCR SEP in acid sulphatic weathering products and naturally acidified soils was established, which allows us to suggest this rapid and cost-effective single extraction procedure as a valuable tool in contamination assessment.

Los cultivos de gliomas malignos se han realizado con el objeto de conocer su biología, estudiar nuevas drogas antineoplásicas y determinar la sensibilidad diferencial a citostáticos. Catorce pacientes con diagnósticos clínico y radiológico sugestivos de tumor cerebral maligno fueron seleccionados de acuerdo a criterios específicos, lo que incluyó la evaluación según el esquema de Karnofsky, el cual determina el estado funcional independiente del paciente. Se realizó la resección quirúrgica, se efectuó el estudio histopatológico y se tomó una muestra del tumor para cultivo en monocapas. La confluencia celular se alcanzó entre 15 y 45 días y los cultivos fueron expuestos a los citostáticos (1,3-bis(2-cloroetil)-1- nitrosouréa, cisplatino o vincristina por una hora. Se determinó la integridad de la membrana mediante la exclusión del colorante azúl de Tripán. Se encontraron diferencias estadísticamente significativas entre las drogas utilizadas y entre los tumores, incluso con el mismo diagnóstico histopatológico, lo cual indica la existencia de resistencia diferencial de las células tumorales a los citostáticos utilizados. Este estudio constituye un ensayo que sustenta la realización de evaluaciones a largo plazo con el fin de emplear una quimioterapia específica en pacientes en los que probablemente se combine ésta con la radioterapia

The amino acid taurine plays an important trophic role during development and regeneration of the central nervous system. Other amino acid systems, such as those for glutamate and gamma-aminobutyric acid (GABA), are modified during the same physiological and pathological processes. After crushing the optic nerve, goldfish retinal explants were plated in the absence and in the presence of different amino acids and amino acid receptor agonists. The length and the density of the neurites were measured at 5 days in culture. Taurine increased the length and the density of neurites. Glutamate and glycine increased them at low concentration, but were inhibitors at higher concentration. The combination of N-methyl-D-aspartate (NMDA) and glycine produced a greater inhibitory effect than NMDA alone. NMDA or alpha-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) added simultaneously with taurine impaired the stimulatory effect of the latter. GABA stimulated the emission of neurites in a concentration dependent manner. Hypotaurine also elevated the length of neurites, but cysteinsesulfinic acid did not produce a significant effect. The concentrations of taurine, glutamate and GABA were determined by HPLC with fluorescent detection in the retina of goldfish at various days post-crushing the optic nerve. The levels of taurine were significantly increased at 48h after the crush, and were elevated up to 20 days. Glutamate level decreased after the lesion of the optic nerve and was still low at 20 days. GABA concentration was not significantly different from the control. The interaction of these amino acids during the regenerative period, especially the balance between taurine and glutamate, may be a determinant in restoring vision after the crush.

The sulfur amino acid taurine and the indoleamine serotonin increases and decreases, respectively, the outgrowth from goldfish retinal explants. Taurine seems to be acting, at least partially, through an increase in calcium fluxes, and the serotonin-inhibiting effect appears to be mediated by serotonin1A receptors and cAMP. Isolated cells of postcrush goldfish retina and of retina from 5-day-old rats were cultured in the presence of taurine or serotonin. In the goldfish, the classical morphology of postcrush ganglion cells was observed. An antibody against the glycoprotein Thy-1 labelled three types of cells in the cultures of goldfish retina. The number of cells outgrowing and the length of the main neurite was measured at 5 days in culture in both species. The number of cells presenting neurites was increased in the goldfish retina by the addition of taurine, and decreased by serotonin. However, the length of the neurites was unaffected by the addition of the modulators. In the rat, only a slight decrease in the number of cells outgrowing was observed in the presence of serotonin. The incorporation of [3H]thymidine was not modified after 5 days in culture in the presence of taurine or serotonin, either in the goldfish or in the rat retina. The antibody Thy 1.1 can label retinal cells of the goldfish in vitro, one of them being ganglion cells. The trophic effect exerted by taurine in the postcrush goldfish retina needs the integrity of the tissue favoring the interaction of cells and factors, because outgrowth increases in retinal explants, but not in isolated cells.

Serotonin is present in the retina of many species, in which plays roles as a neurotransmitter, as a modulator of regeneration, and as the precursor of melatonin. The turnover of serotonin in the goldfish retina is modified by the lesion of the optic nerve and, in postcrush goldfish retinal explants, serotonin inhibits the outgrowth. In the present study, the modification of the serotonergic system of the retina induced by the process of regeneration was explored. The addition of the precursor of serotonin, 5-hydroxytryptophan, to retinal explants, increased the levels of serotonin in a concentration-dependent manner. The concentration of serotonin differentially increased in control and postcrush explants cultured in the presence of 5-hydroxytryptophan for various periods of time, indicating a greater accumulation of the indoleamine at early periods of time in the control than in the postcrush tissue culture. This observation, together with the fact that serotonin concentration in postcrush retina cultured in the absence of 5-hydroxytryptophan and exposed to the precursor for 60 min increased less than in the control, indicates a saturation of the serotonergic system produced by the lesion. The addition of imipramine or citalopram, serotonin uptake blockers, did not significantly change the concentration of serotonin in the cultures, thus, the elevation of serotonin accumulation, especially in the post-crush tissue, might not be due to the transport from the medium. The intraocular injection of 5-hydroxytryptophan after the crush of the optic nerve resulted in a decrease in the outgrowth of retinal explants, supporting the in vivo role of serotonin during the regenerating process in situ. The lesion of the optic nerve did not affect the specific cells, since the number of serotonin-immunoreactive neurons in the retina were not modified by the crush. Taken together, retinal serotonin system is regulated after producing a lesion of the optic nerve, a modulation which has been demonstrated in vivo and in vitro. Thus, there is a reciprocal interaction, since serotonin influences outgrowth in the postcrush retina and the serotonergic system is modulated by the crush, indicating a mechanism of feed-back regulation.

The aim of this work was to study the prevalence, behavior and opinions towards smoking in a sample of 288 physicians from Santiago, stratified according to sex and specialty. The WHO Europe questionnaire was applied and smoker was defined as the person that consumed one or more cigarettes per day. Thirty six percent of the sample (40.2% of men and 23.8% of women) smoked an average of 9 cigarettes per day, compared to the figure of 28.3% obtained in a similar study in 1983. Fifty percent of smokers felt that they will not quit. No differences in knowledge about problems associated to smoking or forbidding smoking in hospitals, were observed between smokers and non-smokers. A majority of smokers did not smoke in front of their patients and did not allow smoking in their private clinics. It is concluded that, although a high proportion of physicians continue to smoke, their opinions and behavior will support smoking cessation efforts.

The regeneration of explants prepared from goldfish retinas with a prior crush of the optic nerve is stimulated by the sulphur amino acid, taurine. Serotonin has been reported to modify survival, proliferation, and outgrowth of nervous tissue. In the present work we evaluated the effect of serotonin and some serotonergic agonists on the neuritic outgrowth from goldfish retinal explants. Serotonin, its precursor, 5-hydroxytryptophan, and the 5HT1A receptor agonists, 8-hydroxy-2-(di-n-propylamino)tetralin and buspirone, inhibited the outgrowth. The blockers of serotonin uptake, imipramine and citalopram, were also inhibitors of neurite sprouting. Imipramine favoured the inhibitory effect of serotonin at 10 days in culture. The concentration of serotonin and its metabolite, 5-hydroxyindoleacetic acid, decreased in the retina at 3 and 5 days after the crush of the optic nerve. Serotonin levels started to recover after 5 days post-lesion, and the metabolite also increased. This indicates that the lesion increases the turnover rate of serotonin and this may be related to its role in regeneration. Serotonin concentration was elevated by the intraocular administration of its precursor, 5-hydroxytryptophan, indicating that the capacity for synthesis was preserved after the crush, but that it was smaller in the post-lesioned retinas. The trophic effect of taurine was impaired by a low concentration of serotonin, probably by opposing the final effect on growth via different targets. These results support a role of serotonin in the regeneration of goldfish retina probably through 5HT1A receptors.

A possible mechanism of action of taurine as a trophic substance was studied in goldfish retina by investigating the effect of extracellular and intracellular calcium chelators on in vitro outgrowth, and the effect of taurine on calcium influx into postcrush retinal cells in culture. The amino acid stimulated the outgrowth from goldfish retinal explants, an effect that was blocked by EGTA and 1,2-bis (o-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid methyl ester (BAPTA). The influx of calcium into cultured cells from postcrush retina was increased by taurine by day 5 in culture, but not by day 10, supporting previous results indicating a critical period in which taurine stimulates outgrowth. The present results suggest that taurine partially exerts its regenerative effect on postcrush retinal explants by increasing calcium influx.

The transport system of taurine was investigated in fragments of goldfish retina prepared from the total tissue and from concentric regions: center and periphery. A high-affinity, saturable, sodium-dependent system was demonstrated in the three types of fragments. The Km for one-site analysis was similar in the two regions and the total retina. The analysis for two sites revealed a significant higher Km for the high-affinity site in fragments from the central region. The maximal uptake rate was higher in the central zone than in the total retina or the periphery. The Hill slopes obtained from saturation experiments of fragments of total retina, center, and periphery were similar to one other and near to 1. The slope of the time course uptake was intermediate for total retina and higher in the center than in the periphery. Hypotaurine and beta-alanine were found to inhibit taurine uptake, but GABA was a weak inhibitor. The values of Ki for hypotaurine by one- and two-site analysis were lower in the central region. The disruption of photoreceptors by shaking did not modify significantly the uptake of the amino acid. Remotion of endogenous taurine by dialysis of central and peripheral fragments increased the uptake in the periphery, but not in the center. The differences observed among the three samples revealed less affinity and high capacity for taurine uptake in the center, plus a higher sensitivity of inhibition. In addition, the peripheral zone had a greater affinity for taurine, and the maximal velocity of the entrance seems to be inhibited by the higher concentration of the amino acid in this zone. These observations may reflect differences between proliferating and non-proliferating regions of the retina (i.e., periphery and center).

Retinal growth from explants attached to different substrates was evaluated in the absence and the presence of various concentrations of taurine. Fiber thickness was higher in poly-L-lysine, collagen and laminin than in the other substrates tested. Explants on wheatgerm, soybean, and red kidney bean lectins did not show emission of neurites at 5 days in culture; growth on laminin was very poor at this period of time. By day 10 nerve growth index was possible to evaluate on soybean and laminin. Outgrowth from retinal explants on wheatgerm lectin was only observed in the presence of taurine. The percentage of growing explants on laminin was increased by the amino acid. Emission of neurites from explants on concanavalin A and lentil lectin was constant from 5 to 10 days in culture and was not accelerated or increased by taurine. Growth on poly-L-lysine and collagen was stimulated by the addition of taurine in a concentration-dependent manner. The amino acid stimulates outgrowth from explants attached to peanut lectin in a bell-shaped concentration dependency similar to that observed on poly-L-lysine. The effect of taurine on retinal explants is substrate-dependent, as an indication of the interaction of soluble molecular components plus charge and structure of the extracellular matrix in order to modulate the outgrowth from regenerating tissue.

Teleost and amphibian retina grows throughout life at the circumferential margin distal to the center. Retinas of two teleosts, goldfish and Eugerres plumieri; and two mammals, rat and rabbit were dissected into concentric regions; center and periphery; or center, intermediate and periphery. The concentration of glutamic acid, taurine, and gamma-aminobutyric acid was determined in these areas. A non-significant difference was found between the center and periphery of retinas dissected into two regions. By dissecting the teleost retina in three concentric regions a progressive increase of gamma-aminobutyric acid and taurine for center to periphery was observed, which was statistically significant for taurine. Rat and rabbit retina did not present this difference. The concentration of these amino acids was also determined in two concentric regions of goldfish retina 24 hr after crush of the optic nerve. Taurine content significantly increased in the center of the regenerating retina, but not in the periphery. Outgrowth was measured in explants prepared from central and peripheral retina in the presence of exogenous taurine. The amino acid produced an increase of the nerve growth index of central explants, but inhibited the growth from peripheral explants. This report presents a correlation between taurine concentration in a region of the retina that could be considered as embryonic tissue, suggesting that this amino acid plays a role in the formation of new cells. Moreover, the results from post-crush and cultured retina, with respect to taurine concentration or regenerating effect, suggest a saturation of the taurine-enriched zone.

Explants from retinas from which the optic nerve was previously crushed exhibited neuritic outgrowth after 4-5 days in culture, and up to 10 days in the presence of the same nutrient medium. Intact retinas did not exhibit vigorous outgrowth. Taurine added in the medium to post-crush explants accelerated the emission of neurites in 1-2 days, and increased the nerve growth index in a concentration-dependent manner up to 4 mM. Greater concentration did not produce any effect on the nerve growth index. The effect of taurine was produced between 0 and 5 days in culture; from day 5 to day 10 explants did not exhibit further growth in the presence of the amino acid. However, post-crush control explants exhibited growth between 5 and 10 days. Taurine effect is blocked in the absence of fetal calf serum, and is reduced in the presence of dialysed serum. Providing new medium on days 1 and 2, but not later, elevated the nerve growth index with respect to explants regenerating in the same medium since day 0. In addition, taurine supplied at various days after plating produced the same pattern of growth as that observed by adding fresh medium, but the nerve growth index was statistically greater in the presence of the amino acid. The mechanisms for these observations remain to be elucidated.
