RESUMO

An in situ wavelength calibration system for the X-ray Imaging Crystal Spectrometer (XICS) on W7-X has been developed to provide routine calibration between plasma shots. XICS is able to determine plasma flow profiles by measuring the Doppler shift of x-ray line emission from highly charged impurity species. A novel design is described that uses an x-ray tube with a cadmium anode placed in front of the diffracting spherically bent crystal. This arrangement provides calibration lines over the full detector extent for both the Ar16+ and Ar17+/Fe24+ spectrometer channels. This calibration system can provide a relative wavelength accuracy of 3 × 10-7 Å across the full spatial extent of the detector, which corresponds to 50 m/s in the W7-X system. An absolute wavelength calibration of 1 × 10-5 Å is expected, corresponding to 1 km/s, based on the current known accuracy of the calibration wavelength and the achievable measurement of the absolute positioning of the hardware. This calibration system can be used to independently calibrate XICS systems on both stellarators and tokamaks, without the need for special plasma conditions often used for calibration, such as locked modes on tokamaks. Experimental and simulated results are shown along with expected results, and the complete design of the calibration hardware that is to be installed in the W7-X XICS system.

Rice is the most important staple food for half of the world's population, but also accounts for about 10% of all anthropogenic CH4 emissions. In spite of a wealth of information on the mechanistic basis and the importance of the rice plant in mediating these emissions, the significance of root exudation for CH4 emissions and the processes that determine root exudation are not well understood. Root exudates derive from photosynthate allocated to the root and subjected to root anabolic and catabolic processes. Key processes in roots that determine the extent of root exudation and, hence, CH4 emission from rice agriculture, include (i) deviation of metabolites from root anabolic and catabolic pathways facilitating root exudation, but also (ii) xylem loading and transport of potential root exudates for reallocation to the leaves, and (iii) xylem loading of sucrose in roots for its transport into reproductive organs, both suppressing root exudation. These processes are modulated by plant development and metabolic requirements resulting from different functions of root exudation. In the present report the interplay of root exudation, CH4 emission and yield are discussed.

BACKGROUND: Lower-limb discrepancy following total hip arthroplasty is the third-most common reason for patient dissatisfaction in orthopaedic surgery. Therefore, accurate planning and evaluation methods are mandatory. The main aim of this study was to evaluate the reliability of the EOS™ system by establishing and comparing the reproducibility of lower-limb automatic and manual 3D measurements. We hypothesized that the reproducibility of the lower-limb measurements is similar regardless of the method used and with an agreement higher than 0.95 for the length parameters. MATERIALS AND METHODS: This study utilized an EOS radiological database of 112 patients. Two independent observers performed two rounds of lower-limb measurements twice, either in manual 3D or automatic 3D mode. The intra- and inter-observer reproducibility was evaluated by the calculation of the intra-class coefficient for each measurement method. The methods were then compared. RESULTS: The intra- and inter-observer reproducibility for length measurements found with the manual and automatic 3D methods was always > 0.98. There was no significant difference in the reproducibility between the two measurement modes, with the exception of the offset, hip-knee-shaft, and neck-shaft angles. CONCLUSION: Our results indicate a very good reproducibility of EOS™ length measurement, regardless of the method used. Automated 3D mode is preferred for the collection of angular and offset measurements. Furthermore, manual mode measurements are not affected by surgical history. Level of evidence IV.

AIMS: It is not clear whether anterior knee pain and osteoarthritis (OA) of the patellofemoral joint (PFJ) are contraindications to medial unicompartmental knee arthroplasty (UKA). Our aim was to investigate the long-term outcome of a consecutive series of patients, some of whom had anterior knee pain and PFJ OA managed with UKA. PATIENTS AND METHODS: We assessed the ten-year functional outcomes and 15-year implant survival of 805 knees (677 patients) following medial mobile-bearing UKA. The intra-operative status of the PFJ was documented and, with the exception of bone loss with grooving to the lateral side, neither the clinical or radiological state of the PFJ nor the presence of anterior knee pain were considered a contraindication. The impact of radiographic findings and anterior knee pain was studied in a subgroup of 100 knees (91 patients). RESULTS: There was no relationship between functional outcomes, at a mean of ten years, or 15-year implant survival, and pre-operative anterior knee pain, or the presence or degree of cartilage loss documented intra-operatively at the medial patella or trochlea, or radiographic evidence of OA in the medial side of the PFJ. In 6% of cases there was full thickness cartilage loss on the lateral side of the patella. In these cases, the overall ten-year function and 15-year survival was similar to those without cartilage loss; however they had slightly more difficulty with descending stairs. Radiographic signs of OA seen in the lateral part of the PFJ were not associated with a definite compromise in functional outcome or implant survival. CONCLUSION: Severe damage to the lateral side of the PFJ with bone loss and grooving remains a contraindication to mobile-bearing UKA. Less severe damage to the lateral side of the PFJ and damage to the medial side, however severe, does not compromise the overall function or survival, so should not be considered to be a contraindication. However, if a patient does have full thickness cartilage loss on the lateral side of the PFJ they may have a slight compromise in their ability to descend stairs. Pre-operative anterior knee pain also does not compromise the functional outcome or survival and should not be considered to be a contraindication. Cite this article: Bone Joint J 2017;99-B:632-9.

Antigen-mediated cross-linking of Immunoglobulin E (IgE) bound to mast cells/basophils via FcɛRI, the high affinity IgE Fc-receptor, is a well-known trigger of allergy. In humans, but not mice, dendritic cells (DCs) also express FcɛRI that is constitutively occupied with IgE. In contrast to mast cells/basophils, the consequences of IgE/FcɛRI signals for DC function remain poorly understood. We show that humanized mice that express FcɛRI on DCs carry IgE like non-allergic humans and do not develop spontaneous allergies. Antigen-specific IgE/FcɛRI cross-linking fails to induce maturation or production of inflammatory mediators in human DCs and FcɛRI-humanized DCs. Furthermore, conferring expression of FcɛRI to DCs decreases the severity of food allergy and asthma in disease-relevant models suggesting anti-inflammatory IgE/FcɛRI signals. Consistent with the improved clinical parameters in vivo, antigen-specific IgE/FcɛRI cross-linking on papain or lipopolysaccharide-stimulated DCs inhibits the production of pro-inflammatory cytokines and chemokines. Migration assays confirm that the IgE-dependent decrease in cytokine production results in diminished recruitment of mast cell progenitors; providing a mechanistic explanation for the reduced mast cell-dependent allergic phenotype observed in FcɛRI-humanized mice. Our study demonstrates a novel immune regulatory function of IgE and proposes that DC-intrinsic IgE signals serve as a feedback mechanism to restrain allergic tissue inflammation.

A multi-camera soft x-ray diagnostic has been developed to measure the equilibrium electron temperature profile and temperature fluctuations due to magnetohydrodynamic activity on the Compact Toroidal Hybrid experiment. The diagnostic consists of three separate cameras each employing two 20-channel diode arrays that view the same plasma region through different beryllium filter thicknesses of 1.8 µm and 3.0 µm allowing electron temperature measurements between 50 eV and 200 eV. The Compact Toroidal Hybrid is a five-field period current-carrying stellarator, in which the presence of plasma current strongly modifies the rotational transform and degree of asymmetry of the equilibrium. Details of the soft x-ray emission, effects of plasma asymmetry, and impurity line radiation on the design and measurement of the two-color diagnostic are discussed. Preliminary estimates of the temperature perturbation due to sawtooth oscillations observed in these hybrid discharges are given.

A new Thomson scattering system using standard commercially available components has been designed for the non-axisymmetric plasmas of the Compact Toroidal Hybrid (CTH). The beam, generated by a frequency doubled Continuum PL DLS 2 J Nd:YAG laser, is passed vertically through an entrance Brewster window and an aperturing baffle system to minimize the stray laser light that could enter the collection optics. The beam line has been designed with an 8 m propagation distance to the mid-plane of the CTH device with the beam diameter kept less than 3 mm inside the plasma volume. The beam exits the vacuum system through another Brewster window and enters a beam dump, again to minimize the stray light in the vacuum chamber. Light collection, spectral processing, and signal detection are accomplished with an f/#∼ 1 aspheric lens, a commercially available Holospec f/1.8 spectrometer, and an Andor iStar DH740-18U-C3 image intensified camera. Spectral rejection of stray laser light, if needed, can be performed with the use of an optional interference filter at the spectrometer input. The system has been developed for initial single point measurements of plasmas with core electron temperatures of approximately 20-300 eV and densities of 5 × 10(18) to 5 × 10(19) m(-3) dependent upon operational scenario.

A linear array of 16 Hall effect sensors has been developed to directly measure the poloidal magnetic field inside the boundary of a non-axisymmetric hybrid torsatron/tokamak plasma. The array consists of miniature gallium arsenide Hall sensor elements mounted 8 mm apart on a narrow, rotatable printed circuit board inserted into a re-entrant stainless steel tube sheathed in boron nitride. The sensors are calibrated on the bench and in situ to provide accurate local measurements of the magnetic field to aid in reconstructing the equilibrium plasma current density profiles in fully three-dimensional plasmas. Calibrations show that the sensor sensitivities agree with the nominal manufacturers specifications of 1.46 V/T. Poloidal fields measured with the Hall sensor array are found to be within 5% of poloidal fields modeled with a Biot-Savart code.

Neuronal circuitries in the hippocampus are involved in navigation and memory and are controlled by major networks of GABAergic interneurons. Parvalbumin (PV)-expressing interneurons in the dentate gyrus (DG) are identified as fast-spiking cells, playing a crucial role in network oscillation and synchrony. The inhibitory modulation of these interneurons is thought to be mediated mainly through GABAA receptors, the major inhibitory neurotransmitter receptors in the brain. Here we show that all PV-positive interneurons in the granular/subgranular layer (GL/SGL) of the mouse DG express high levels of the GABAA receptor δ subunit. PV-containing interneurons in the hilus and the molecular layer, however, express the δ subunit to a lower extent. Only 8% of the somatostatin-containing interneurons express the δ subunit, whereas calbindin- or calretinin-containing interneurons in the DG seem not to express the GABAA receptor δ subunit at all. Hence, these cells receive a GABAergic control different from that of PV-containing interneurons in the GL/SGL. Experiments investigating a possible co-expression of GABAA receptor α1, α2, α3, α4, α5, ß1, ß2, ß3, or γ2 subunits with PV and δ subunits indicated that α1 and ß2 subunits are co-expressed with δ subunits along the extrasynaptic membranes of PV-interneurons. These results suggest a robust tonic GABAergic control of PV-containing interneurons in the GL/SGL of the DG via δ subunit-containing receptors. Our data are important for better understanding of the neuronal circuitries in the DG and the role of specific cell types under pathological conditions.

A unique in situ calibration technique has been used to spatially calibrate and characterize the extensive new magnetic diagnostic set and close-fitting conducting wall of the High Beta Tokamak-Extended Pulse (HBT-EP) experiment. A new set of 216 Mirnov coils has recently been installed inside the vacuum chamber of the device for high-resolution measurements of magnetohydrodynamic phenomena including the effects of eddy currents in the nearby conducting wall. The spatial positions of these sensors are calibrated by energizing several large in situ calibration coils in turn, and using measurements of the magnetic fields produced by the various coils to solve for each sensor's position. Since the calibration coils are built near the nominal location of the plasma current centroid, the technique is referred to as an "artificial plasma" calibration. The fitting procedure for the sensor positions is described, and results of the spatial calibration are compared with those based on metrology. The time response of the sensors is compared with the evolution of the artificial plasma current to deduce the eddy current contribution to each signal. This is compared with simulations using the VALEN electromagnetic code, and the modeled copper thickness profiles of the HBT-EP conducting wall are adjusted to better match experimental measurements of the eddy current decay. Finally, the multiple coils of the artificial plasma system are also used to directly calibrate a non-uniformly wound Fourier Rogowski coil on HBT-EP.

A three-channel 1 mm wave interferometer has been designed, assembled, and installed on the Compact Toroidal Hybrid torsatron (CTH). The interferometer design makes novel use of a subharmonic mixer for detection, which simplifies alignment. It employs a single electronically tunable source that is repetitively chirped using a sawtooth waveform of frequency up to 1 MHz. The 15.25 GHz drive oscillator is multiplied in two stages to 122 GHz before a final doubler stage brings it to 244 GHz. Local oscillator (LO) power at 122 GHz is directed through waveguide to the LO input of the subharmonic mixer of each viewing chord, simplifying alignment. Phase detection is performed by directly digitizing the amplified mixer outputs at 50 MHz and processing them with a software algorithm. Initial measurements made with the central chord of the new interferometer agree with those from the existing 4 mm system at low densities. The 1 mm system performs well in current-driven discharges reaching densities over 10(19) m(-3), whereas the lower frequency interferometer is found to be less reliable due to loss of fringes. This is a critical improvement for experiments studying the onset, avoidance, and vacuum magnetic transform dependence of disruptions in the CTH device.

Single-antigen bead (SAB) testing permits reassessment of immunologic risk for kidney transplantation. Traditionally, high panel reactive antibody (PRA), retransplant and deceased donor (DD) grafts have been associated with increased risk. We hypothesized that this risk was likely mediated by (unrecognized) donor-specific antibody (DSA). We grouped 587 kidney transplants using clinical history and single-antigen bead (SAB) testing of day of transplant serum as (1) unsensitized; PRA = 0 (n = 178), (2) third-party sensitized; no DSA (n = 363) or (3) donor sensitized; with DSA (n = 46), and studied rejection rates, death-censored graft survival (DCGS) and risk factors for rejection. Antibody-mediated rejection (AMR) rates were increased with DSA (p < 0.0001), but not with panel reactive antibody (PRA) in the absence of DSA. Cell-mediated rejection (CMR) rates were increased with DSA (p < 0.005); with a trend to increased rates when PRA>0 in the absence of DSA (p = 0.08). Multivariate analyses showed risk factors for AMR were DSA, worse HLA matching, and female gender; for CMR: DSA, PRA>0 and worse HLA matching. AMR and CMR were associated with decreased DCGS. The presence of DSA is an important predictor of rejection risk, in contrast to traditional risk factors. Further development of immunosuppressive protocols will be facilitated by stratification of rejection risk by donor sensitization.

The Thomson scattering diagnostic on the High Beta Tokamak-Extended Pulse (HBT-EP) is routinely used to measure electron temperature and density during plasma discharges. Avalanche photodiodes in a five-channel interference filter polychromator measure scattered light from a 6 ns, 800 mJ, 1064 nm Nd:YAG laser pulse. A low cost, high-power spatial filter was designed, tested, and added to the laser beamline in order to reduce stray laser light to levels which are acceptable for accurate Rayleigh calibration. A detailed analysis of the spatial filter design and performance is given. The spatial filter can be easily implemented in an existing Thomson scattering system without the need to disturb the vacuum chamber or significantly change the beamline. Although apertures in the spatial filter suffer substantial damage from the focused beam, with proper design they can last long enough to permit absolute calibration.

BACKGROUND: Metastasis is associated with poor prognosis for melanoma. The formation of metastases is a multi-step process, in which cancer cells can subsequently acquire the potential to intravasate into the blood or lymph vessels, disseminate through the circulation, extravasate through the endothelium and invade the connective tissue. There is increasing evidence that chemokines have a pivotal role in the dissemination and establishment of melanoma metastasis. METHODS: We isolated melanoma cells from melanoma metastasis and performed different migration assays and transendothelial resistance measurements of endothelial monolayers co-cultured with melanoma cells, in order to monitor barrier function and diapedesis and confirmed these results by confocal microscopy. RESULTS: We observed that tumour endothelial cells (ECs) secrete high levels of CXCL9 in all, and CXCL10 in most melanoma metastases. Migration studies revealed that low concentrations of these chemokines induce chemotaxis, whereas high concentrations induce spontaneous migration of melanoma cells (chemokinesis/chemorepulsion) and the disruption of the endothelial barrier, resulting in an accelerated transendothelial migration (TEM). Addition of anti-CXCL9 or anti-CXCR3 antibodies to the co-cultures delayed the TEM of melanoma cells. CONCLUSION: Our data represent novel mechanisms by which tumour cells in melanoma metastases might use the chemokine-expressing endothelium to leave the tumour and eventually to form additional metastases at distinct sites.

We compared the development of sensitivity to first- versus second-order global motion in 5-year-olds (n=24) and adults (n=24) tested at three displacements (0.1, 0.5 and 1.0 degrees). Sensitivity was measured with Random-Gabor Kinematograms (RGKs) formed with luminance-modulated (first-order) or contrast-modulated (second-order) concentric Gabor patterns. Five-year-olds were less sensitive than adults to the direction of both first- and second-order global motion at every displacement tested. In addition, the immaturity was smallest at the smallest displacement, which required the least spatial integration, and smaller for first-order than for second-order global motion at the middle displacement. The findings suggest that the development of sensitivity to global motion is limited by the development of spatial integration and by different rates of development of sensitivity to first- versus second-order signals.

The objectives of the present study were to assess how the stability of the emulsion recovered from aqueous extraction processing of soybeans was affected by characteristics of the starting material and extraction and demulsification conditions. Adding endopeptidase Protex 6L during enzyme-assisted aqueous extraction processing (EAEP) of extruded soybean flakes was vital to obtaining emulsions that were easily demulsified with enzymes. Adding salt (up to 1.5 mM NaCl or MgCl(2)) during extraction and storing extruded flakes before extraction at 4 and 30 degrees C for up to 3 months did not affect the stabilities of emulsions recovered from EAEP of soy flour, flakes and extruded flakes. After demulsification, highest free oil yield was obtained with EAEP of extruded flakes, followed by flour and then flakes. The same protease used for the extraction step was used to demulsify the EAEP cream emulsion from extruded full-fat soy flakes at concentrations ranging from 0.03% to 2.50% w/w, incubation times ranging from 2 to 90 min, and temperatures of 25, 50 or 65 degrees C. Highest free oil recoveries were achieved at high enzyme concentrations, mild temperatures, and short incubation times. Both the nature of enzyme (i.e., protease and phospholipase), added alone or as a cocktail, concentration of enzymes (0.5% vs. 2.5%) and incubation time (1 vs. 3 h), use during the extraction step, and nature of enzyme added for demulsifying affected free oil yield. The free oil recovered from EAEP of extruded flakes contained less phosphorus compared with conventional hexane-extracted oil. The present study identified conditions rendering the emulsion less stable, which is critical to increasing free oil yield recovered during EAEP of soybeans, an environmentally friendly alternative processing method to hexane extraction.

Here, we measured sensitivity to global form, global motion, and biological motion in a sample of 23, five- to nine-year-old children born at <32 weeks gestation, and in 20 full-term controls matched to the clinical sample in age, socioeconomic status, and estimated Verbal IQ. As a group, premature children showed reduced sensitivity, relative to controls, on all three tasks (F>4.1, p<0.05). By computing a deficit score for each task (the ratio between a premature child's threshold and the mean threshold for three age-matched controls) we were able to compare performance across tasks directly. Mean deficit scores were significantly greater than 1 (indicating some level of impairment) for biological motion and global motion (ps<0.03). In contrast, the mean deficit score for global form was not significantly different from 1 (indicating no impairment, relative to age-matched control children). Rates of impairment (deficit score>or=2) were four times higher for global motion than for global form (p<0.04); rates of impairment on the biological motion task fell at an intermediate level. In agreement with previous studies, we find impairments in the processing of global motion (Atkinson & Braddick; MacKay et al.) and of biological motion (Pavlova et al.), which are larger than the impairments in the processing of global form (Atkinson & Braddick). In addition, we show that the impairments are not correlated with each other. The differential vulnerability that we observed across tasks could not be accounted for by stereoacuity deficits, amblyopia, or attentional problems. We suspect, instead, that it reflects the fact that these forms of visual processing develop at different rates, and may be differentially vulnerable to early brain injury or atypical neurodevelopment [c.f., Atkinson, J. & Braddick, O. RESUMO

A feedback system for controlling external, long-wavelength magnetohydrodynamic activity is described. The system is comprised of a network of localized magnetic pickup and control coils driven by four independent, low-latency field-programable gate array controllers. The control algorithm incorporates digital spatial filtering to resolve low mode number activity, temporal filtering to correct for frequency-dependent amplitude and phase transfer effects in the control hardware, and a Kalman filter to distinguish the unstable plasma mode from noise.

Adults' expertise in recognizing facial identity involves encoding subtle differences among faces in the shape of individual facial features (featural processing) and in the spacing among features (a type of configural processing called sensitivity to second-order relations). We used fMRI to investigate the neural mechanisms that differentiate these two types of processing. Participants made same/different judgments about pairs of faces that differed only in the shape of the eyes and mouth, with minimal differences in spacing (featural blocks), or pairs of faces that had identical features but differed in the positions of those features (spacing blocks). From a localizer scan with faces, objects, and houses, we identified regions with comparatively more activity for faces, including the fusiform face area (FFA) in the right fusiform gyrus, other extrastriate regions, and prefrontal cortices. Contrasts between the featural and spacing conditions revealed distributed patterns of activity differentiating the two conditions. A region of the right fusiform gyrus (near but not overlapping the localized FFA) showed greater activity during the spacing task, along with multiple areas of right frontal cortex, whereas left prefrontal activity increased for featural processing. These patterns of activity were not related to differences in performance between the two tasks. The results indicate that the processing of facial features is distinct from the processing of second-order relations in faces, and that these functions are mediated by separate and lateralized networks involving the right fusiform gyrus, although the FFA as defined from a localizer scan is not differentially involved.

This study evaluated the impact of premature birth on the development of local and global motion processing in a group of very low birthweight (<1500 g), 5- to 8-year-old children. Sensitivity to first- and second-order local motion stimuli and coherence thresholds for global motion in random dot kinematograms were measured. Relative to full-term controls, premature children showed deficits on all three aspects of motion processing. These problems could not be accounted for by stereo deficits, amblyopia, or attentional problems. A history of mild retinopathy of prematurity and/or intraventricular hemorrhage increased risk, but deficits were observed in some children with no apparent ocular or cerebral pathology. It is important to note that, despite the observed group differences, individual profiles of performance did vary; the results suggest that these three forms of motion processing may involve separate neural mechanisms. These findings serve to increase our understanding of the organization and functional development of motion-processing subsystems in humans, and of the impact of prematurity and associated complications on visual development.
