RESUMO

Exposure to less-hygienic conditions during early childhood has been associated with stimulation and development of the immune system. A recent study indicated that exposure of piglets to soil-borne microbes during lactation was related with modulation of gut microbiota and immune function. To identify the potential molecular mechanisms and pathways impacted by early-life topsoil exposure, we analyzed the messenger RNA (mRNA) and micro-RNA (miRNA) expression in peripheral blood mononuclear cells (PBMCs) from these piglets. Total RNA was extracted from the PBMCs of piglets exposed to topsoil only from d 4-d 21 of life (mRNA n = 6; miRNA n = 5) or unexposed control pigs (mRNA n = 6; miRNA n = 8) at 11, 20, and 56 days of age. Small RNA and mRNA were sequenced with 50-bp single-end reads using Illumina chemistry. Sequence data were quality checked with FASTQC software and aligned to the Sscrofa 11.1 genome with the STAR aligner for mRNA and mirDeep2 for miRNA. Differential expression (DE) analysis was performed using PROC Glimmix of SAS to evaluate changes in expression due to topsoil exposure over time with genes declared DE at a false discovery rate (FDR) of q < 0.10. A total of 138 mRNA and 21 miRNAs were identified as DE for the treatment by age interaction. Ontology enrichment analysis of DE mRNA revealed Gene ontology (GO) terms directly involved in the connection between T-cell and antigen-presenting cells that are associated with T-cell activation. Key regulatory genes identified include PTPRJ, ITGB3, TRBV30, CD3D, mir-143, mir-29, and mir-148a. While these results require validation, this study provides data supporting the hypothesis that less-hygienic environments during early life may contribute to the development of the immune system.

The objectives of this study were to elucidate the effects of a dirty environment and a challenge plus associated environmental contamination on pig growth performance, diet utilization efficiency, and gas emissions (CO, NH, CH, NO, and HS) from stored manure. Twenty-four weaned barrows, aged 31 d at initiation of the trial, were randomly allotted to 3 different treatments in a completely randomized design. Treatments were: pigs housed in cages with manure removed and cages washed daily (Clean); pigs housed in cages sprayed daily with manure slurry mixtures (Dirty); or pigs challenged with Typhimurium DT104 and housed in cages that were not washed, but manure was removed daily ( challenge). Rectal temperature, body weight, daily feed intake, manure output, manure composition, and gas emissions from stored manure were measured throughout the 24-d animal phase. The Dirty and challenge treatments were statistically compared to the Clean treatment to evaluate individual effects. Dirty housing tended to decrease ADG from d 1 to 24 ( = 0.06) but there were no other effects on pig performance compared with the Clean treatment. In contrast, a challenge was associated with a marked reduction in each of the measured indicators of pig performance. challenge increased the carbon to nitrogen ratio, ether extract, and lignin concentrations in excreted manure ( = 0.02, 0.01, 0.003, respectively), and increased manure and head space temperatures in manure tanks ( < 0.0001). Gas emissions from stored manure of pigs on the Dirty or treatments were increased for each of the measured gases as compared to the Clean treatment ( < 0.01) when expressed per unit of BW gain. When gas emissions from manure of pigs housed in the Dirty treatment were expressed per unit of manure volatile solids (VS), they were increased for NH, CH, and HS ( < 0.02). challenge was associated with increased emissions of CO, and NO and decreased emissions of HS per kilogram manure VS compared to the Clean treatment ( = 0.06, 0.03, 0.04, respectively). Collectively, these results indicated that a challenge and associated housing contamination caused depressed growth rate and increased manure gas emissions, while exposure to a Dirty environment slightly reduced growth performance and clearly increased manure gas emissions per unit of BW gain as compared to Clean control.

Two experiments were conducted to determine the effect of feeding supplemental -Arg during late pregnancy on piglet birth weight and preweaning performance. In Exp. 1, 97 gilts and sows were allotted (gestation d 93) to receive a control diet (CON; 19.8 g standardized ileal digestible [SID] Arg/d) or the CON + 1.0% -Arg (ARG; 46.6 g SID Arg/d). Gilts and sows were weighed on gestation d 93 and 110, 48 h after farrowing, and at weaning. Data, including number born alive, number weaned, individual birth and weaning weight, and placenta weight, were recorded. Blood samples were collected on d 93 and 110 and analyzed for plasma IGF-1, insulin, and blood urea nitrogen concentration. In a second experiment, 383 sows in a commercial research farm were allotted to receive CON or ARG. An -Arg premix was provided daily by top dress beginning at gestation d 81 (±0.1 d) and fed for an average of 35 d (±0.2 d). Sows received 2.73 kg feed/d with CON sows provided 17 g SID Arg/d and ARG sows receiving a total of 44 g SID Arg/d. Litter birth weight was recorded and average birth weight was computed. In a subset of 82 sows, individual birth weights were recorded. In Exp. 1, there was a tendency for greater late pregnancy maternal BW gain ( = 0.06) in ARG compared with CON. A tendency for a parity × treatment interaction was observed for late pregnancy BW gain, with first litter sows fed ARG gaining the most, gilts fed ARG intermediate, and all other treatments gaining the least ( = 0.10). No differences between treatment groups were observed for maternal plasma IGF-1, insulin, and urea nitrogen and in progeny performance to weaning ( > 0.28). In Exp. 2, piglet birth weight was more effectively tested because of the large number of multiparous sows involved. There was a tendency for individual birth weight to decline in ARG compared with CON ( < 0.08), but birth weight distribution between <0.80 and >2.8 kg was similar. No other differences were observed ( > 0.18). In conclusion, late pregnancy supplementation with -Arg had no effect on number of pigs born alive, piglet birth weight, or lactation performance.

Progeny of GPK-35 females mated to PIC 380 boars were blocked by initial BW, and within the 9 blocks, pens of pigs (3 gilts and 3 barrows/pen) were randomly assigned to dietary treatments where CP of finisher-I, -II, and -III diets was 1) 16.04, 14.55, and 16.23%, respectively (Ctrl); 2) 14.76, 13.48, and 15.27%, respectively (ILE); 3) 14.26, 12.78, and 14.28%, respectively (VAL); or 4) 12.65, 12.38, and 13.32%, respectively (NoSBM). All finisher-III diets included 10 mg/kg of ractopamine hydrochloride (RAC) and a Lys:ME ratio of 2.79 g/Mcal. At slaughter, HCW and Fat-O-Meat'er data were recorded before carcasses were subjected to a rapid chilling process. A subsample of whole hams (2/pen) and whole loins (2/pen) were transported under refrigeration to the University of Arkansas. Hams were dissected with a knife into lean, fat, and bone, and 2.5-cm-thick chops from the semimembranosus (SM) and the LM were used to measure fresh pork quality characteristics. Both ADG and G:F decreased (linear, = 0.05) as CP decreased in finisher-I diets, whereas ADFI was reduced (linear, = 0.01) in response to decreasing CP in finisher-II diets. When RAC was included in the finisher-III diets, ADFI and BW decreased (linear, ≤ 0.03) with decreasing CP, and pigs fed the ILE diet had greater (cubic, < 0.01) G:F than pigs fed the Ctrl and VAL diets. Across the entire finishing period, ADG and ADFI decreased (linear, = 0.01) in response to reductions in dietary CP. Conversely, reducing CP in finisher diets did not ( ≥ 0.13) affect carcass yield, fat depth, LM depth, or calculated fat-free lean yield, and dietary CP content did not ( ≥ 0.09) alter the lean, fat, or bone composition of fresh hams. Moreover, there was no effect of dietary CP on the visual and instrumental color or firmness of the LM ( ≥ 0.06) or SM ( ≥ 0.12). However, there were linear increases in LM marbling scores ( = 0.02) and intramuscular fat content ( = 0.03) as CP was reduced in the finisher diets. Although reducing dietary CP decreased overall ADG and ADFI by approximately 6.1 and 4.9%, respectively, carcass composition was not impacted by dietary CP level. More importantly, reducing dietary CP, although meeting the standard ileal digestible requirements for Lys, Thr, Trp, Met, Ile, and Val with crystalline AA, did not impact pork color or water-holding capacity and actually increased the intramuscular fat content of the LM.

A cooperative study comprising growth performance, bone mineralization, and nutrient balance experiments was conducted at 11 stations to determine the standardized total-tract digestible (STTD) P requirement of 20-kg pigs using broken-line regression analysis. Monocalcium phosphate and limestone were added to a corn-soybean meal-based diet at the expense of cornstarch to establish 6 concentrations of STTD P from 1.54 to 5.15 g/kg in increments of 0.62 g/kg at a constant Ca:total P of 1.52:1.0. Diets were fed to 936 pigs (average initial BW of 19 kg) in 240 pens for 20 replicate pens of barrows and 20 replicate pens of gilts per diet. As STTD P increased from 1.54 to 5.15 g/kg of the diet for d 0 to 14, 14 to 28, and 0 to 28, the ADG, ADFI, and G:F increased ( < 0.01). Barrows gained and ate more ( < 0.05) than gilts during d 14 to 28 and 0 to 28. There was no interaction between sex and STTD P concentration for any of the growth performance response criteria. There were both linear and quadratic increases ( < 0.05) in mineral density and content of ash, Ca, and P in the femur expressed as a percentage of dry, fat-free metacarpal as dietary STTD P increased. Furthermore, the maximum load of the femur and mineral density and content and maximum load as well as the Ca and P expressed as a percentage of metacarpal ash linearly increased ( < 0.01) with increasing dietary concentrations of STTD P. There were both linear and quadratic increases ( < 0.01) in apparent digestibility and retention of P with increasing concentrations of STTD P in the diets. Digestibility and retention of Ca linearly ( < 0.01) increased with increasing dietary concentrations of STTD P. Breakpoints determined from nonlinear broken-line regression analyses revealed estimates of 4.20 ± 0.102, 3.20 ± 0.036, or 3.87 ± 0.090 g/kg for ADG during d 0 to 14, 14 to 28, or 0 to 28, respectively. Corresponding estimates using G:F as the response criterion were 4.34 ± 0.146, 3.38 ± 0.139, or 4.08 ± 0.195 g/kg. When mineralization of the femur was used as criteria of response, estimates of STTD P requirement were 4.28, 4.28, or 4.34, g/kg for mineral density, mineral content, or maximum load, respectively. Using mineralization of the metacarpal as criteria of response, estimates of STTD P requirement ranged from 3.5 to 5.0 g/kg depending on the metacarpal response criteria. The study provided empirical estimates of STTD P requirements of 20- to 40-kg pigs.

The objectives of this study were to examine the effects of porcine reproductive and respiratory syndrome virus (PRRSV) infection and vaccination on pig growth, dietary nutrient efficiency of utilization, manure output, and emissions of CO, CH, HS, NO, and NH gases from stored manure. Forty-eight pigs, aged 21 d at the start of the study, were subjected to 1 of 4 treatment combinations arranged in a 2 × 2 factorial design with main factors of PRRSV vaccination and PRRSV infection. Body weight, ADFI, manure output, and nutrient efficiency of utilization were assessed and gas emissions from stored manure were determined daily from 50 to 78 d of age and for 24 d after completion of the animal phase. Infection with PRRSV markedly reduced final BW, ADG, and ADFI ( < 0.01) and reduced efficiencies of ADF and ether extract utilization ( = 0.05 and = 0.02, respectively) regardless of vaccination status. No significant treatment effects were found on manure output, manure pH, efficiencies of lignin utilization, and N retention. Infecting pigs with PRRSV increased daily manure CO emission per pig ( = 0.01). There was an interaction between immunization and infection for NO per pig with manure from uninfected, vaccinated pigs producing as much as the manure from infected, vaccinated pigs whereas there was a difference by PRRSV infection state for nonvaccinated pigs. There were also interactions between treatments for HS and NO emissions per kilogram of manure volatile solids excreted ( = 0.01 and = 0.0001, respectively) with the same pattern as for NO per pig; that is, the vaccinated pigs had similar rates of emission regardless of infection state. Pigs infected with PRRSV increased NO nitrogen per kilogram of total N excreted compared with noninfected groups ( = 0.03). Collectively, these results indicated that PRRSV infection caused decreased growth rates and nutrient utilization efficiency and increased gas emissions from stored manure.

Lysozyme is a 1,4-ß-N-acetylmuramidase that has antimicrobial properties. The objective of this study was to determine the effect of lysozyme and antibiotics on growth performance and immune response during an indirect immune challenge. Two replicates of 600 pigs each were weaned from the sow at 26 d of age, blocked by litter and sex, and then randomly assigned to 1 of 24 pens in either a nursery room that had been fully disinfected or a nursery room left unclean since the previous group of pigs. Within a room, pigs were randomly assigned to either control diets (2 phase nursery regime), control diets + antibiotics (chlortetracycline/tiamulin hydrogen fumarate), or control diets + lysozyme (100 mg/kg diet). Pig weights and feed disappearance were measured and blood was collected on d 0, 14, and 28 of treatment. A group of 20 pigs were killed at 24 d of age for initial body composition analysis and 10 pigs of median weight were killed per diet room combination for body composition analysis after 28 d of treatment. Control + antibiotics and control + lysozyme-fed pigs grew at a faster rate for the 28-d study compared to control pigs (318 ± 14,320 ± 15 vs. 288 ± 15 g/d, respectively; P < 0.05), regardless of nursery environment (P > 0.05). The indirect immune challenge did not alter growth performance from d 0 to 14 of treatment but decreased ADG from d 14 to 28 of the study (415 ± 15 vs. 445 ± 13 g/d ;: P < 0.05). Feed intake was not altered by the nursery environment (P > 0.61) or dietary treatments (P > 0.10), but feed efficiency was worsened by the indirect immune challenge (P < 0.05) and improved by both control + antibiotics and control + lysozyme diets (P < 0.01). The immune challenge did not alter nutrient accretion (P > 0.25), but both control + antibiotics and control + lysozyme pigs had decreased accretion of whole-body lipid (P < 0.01) and increased accretion of protein (P < 0.09). Blood levels of tumor necrosis factor-α (TNF-α; P < 0.01), haptoglobin (P < 0.09), and C-reactive protein (CRP; P < 0.01) were higher due to the indirect immune challenge compared to pigs reared in the clean nursery (P < 0.05). In addition, pigs consuming antibiotics or lysozyme had lower TNF-α, haptoglobin, and CRP compared to control pigs, regardless of nursery environment (P < 0.04). Thus, lysozyme is a suitable alternative to antibiotics in swine nursery diets, and lysozyme ameliorates the effects of a chronic indirect immune challenge.

Collaborative studies comprising growth performance and metabolism experiments were conducted to reevaluate growth performance and nutrient and energy utilization responses of nursery pigs to dietary lipid sources and levels. Two lipid sources (soybean oil or tallow) were included at the rate of 1%, 3%, or 5% at the expense of cornstarch in a 2 × 3 factorial arrangement for 6 diets, and Lys was added to maintain a constant calorie:Lys ratio. A growth performance experiment (d 0 to 7, d 7 to 21, and d 21 to 35 postweaning) involving a total of 822 crossbred pigs was conducted at 9 research stations. Each station contributed 2 to 7 replicate pens with 4 to 6 pigs per pen, resulting in a total of 28 replicate pens per diet. The initial BW ranged from 5.9 to 7.3 kg, and the final BW ranged from 20.9 to 28.9 kg. Diets fed during d 7 to 21 and d 21 to 35 were evaluated with 120 barrows in 2 metabolism experiments with 10 individually fed barrows per diet to determine digestibility and retention of nutrients and energy. Lipid source and the interaction between lipid level and source did not affect growth performance. Lipid did not affect growth performance from d 0 to 7. There was a quadratic decrease (P < 0.05) in ADG with increasing level of added lipid only during d 7 to 21 postweaning. The ADFI was decreased (P < 0.01) both during d 7 to 21 and d 21 to 35 postweaning with increasing level of lipid supplementation. A linear improvement (P < 0.001) in G:F with increasing level of lipid supplementation occurred during both d 21 to 35 and d 0 to 35 postweaning. There was a linear increase in DE, ME, and nitrogen-corrected ME (MEn) of diets fed during d 7 to 21 postweaning with increasing lipid level regardless of lipid source. For diets fed during d 21 to 35 postweaning, there were interactions (P < 0.05) between lipid source and level for digestibility of DM, N, lipid, and energy and for DE, ME, and MEn of diets. The improved feed efficiency was, perhaps, partly attributable to the increase in ME content of the diets with added lipids. There were interactions (P < 0.05) between lipid source and level for efficiency of energy use for BW gain for diets fed during d 21 to 35. The efficiencies of use of DE, ME, and NE for BW gain were not affected in pigs fed diets containing increasing levels of soybean oil; however, there were linear increases in these response criteria for pigs fed diets containing increasing levels of tallow. These results indicate that the energy values of soybean oil from the 2012 NRC study used in formulating these diets were fairly accurate.

Crossbred pigs (n = 216) were used to test the effects of phase-feeding beef tallow (BT) and yellow grease (YGr) on live performance, carcass characteristics, and fatty acid composition of subcutaneous (s.c.) fat depots of growing-finishing swine. Pigs were blocked by initial BW (26.0 ± 5.3 kg) before allotment to pens (6 pigs/pen), and pens (6 pens/block) were assigned randomly to 1 of 6 dietary treatments: 1) corn-soybean meal-based grower and finisher diets formulated with 4.7% YGr fed during all 5 feeding phases (YG15), 2) corn-soybean meal-based diets formulated with 5.0% BT fed during all 5 phases (BT15), 3) diets containing 5.0% BT fed during the first 2 phases and diets with 4.7% YGr fed the last 3 phases (YG345), 4) diets formulated with 5.0% BT fed during first 3 phases and diets containing 4.7% YGr fed during the last 2 phases (YG45), 5) diets containing 4.7% YGr fed during the first 3 phases and diets with 5.0% BT fed during the last 2 feeding phases (BT45), or 6) diets formulated with 4.7% YGr fed during the first 2 phases and diets with 5.0% BT fed during the last 3 phases (BT345). Overall performance was similar (P ≥ 0.06) among dietary treatments; however, overall ADG and ADFI increased (quadratic, P ≤ 0.05) with increasing days fed BT, but duration of dietary YGr did not affect ADG (P ≥ 0.22) or ADFI (P ≥ 0.30). There was no (P ≥ 0.23) effect of fat inclusion on carcass characteristics, but carcass lean yield decreased (linear, P = 0.02) as duration of YGr feeding increased from 37 to 103 d. Proportions of back fat SFA (quadratic, P = 0.03) and jowl fat MUFA (linear, P = 0.02) increased as the time fed BT increased from 47 to 103 d. Conversely, PUFA content of both s.c. fat depots increased (linear, P < 0.01) with increasing durations of dietary YGr. Moreover, quadratic slopes for linolenic (18:2n-6) and linolenic (18:3n-3) acids in jowl fat differed (P ≤ 0.05) between BT and YGr durations, indicating that the percentages of 18:2n-6 and 18:3n-3 increased at a greater rate with increasing time fed YGr than the rate of decreases in 18:2n-6 and 18:3n-3 associated with increasing time fed BT. Results of this study confirm that the fatty acid composition of s.c. fat depots are similar to that of the dietary fat source fed during the last 2 or 3 feeding phases, yet deposition rates of specific fatty acids appear to be dependent on the length of time pigs are fed a specific fat source.

Crossbred pigs (n = 216) were used to test the effects of phase-feeding beef tallow (BT) and yellow grease (YGr) on fresh belly and bacon quality characteristics of growing-finishing swine fed dried distillers grains with solubles (DDGS). Pigs were blocked by initial BW (26.0 ± 5.3 kg) before allotment to pens (6 pigs/pen), and pens (6 pens/block) were assigned randomly to 1 of 6 dietary treatments: 1) corn-soybean meal-based grower and finisher diets formulated with 4.7% YGr fed during all 5 feeding phases (YG15); 2) corn-soybean meal-based diets formulated with 5.0% BT fed during all 5 phases (BT15); 3) diets containing 5.0% BT fed during the first 2 phases and diets with 4.7% YGr fed the last 3 phases (YG345); 4) diets formulated with 5.0% BT fed during first 3 phases and diets containing 4.7% YGr fed during the last 2 phases (YG45); 5) diets containing 4.7% YGr fed during the first 3 phases and diets with 5.0% BT fed during the last 2 feeding phases (BT45); or 6) diets formulated with 4.7% YGr fed during the first 2 phases and diets with 5.0% BT fed during the last 3 phases (BT345). All dietary treatments were formulated with 30% dried distillers grains with solubles (DDGS) during the first 3 phases, 15% DDGS in the fourth phase, and no DDGS during the last phase. Fresh belly quality data were collected on the left-side bellies, whereas bacon from the right-side bellies was prepared under commercial processing conditions. Additionally, USDA-certified No. 1 slices were collected for cooking characteristics and sensory panel evaluations. Bellies from the YG15-fed pigs were softer (P ≤ 0.05) than bellies from BT15-fed pigs; however, instrumentally measured belly firmness was not (P ≥ 0.06) different among treatments. Concentrations of palmitic, stearic, and oleic acids, as well as all SFA and all MUFA, were greater (P < 0.01) in bellies from BT15- than YG15-fed pigs. In contrast, proportions of linoleic acid, all PUFA, and iodine value were greater (P < 0.01) in belly fat from YG15-fed pigs in comparison with BT15-fed pigs. Yield of commercially processed bacon (P ≥ 0.06), mechanical bacon tenderness (P ≥ 0.69), and bacon palatability attributes (P ≥ 0.55) were not affected by the dietary treatments. Thus, results of this study indicated that phase-feeding BT to pigs fed diets formulated with DDGS produced minor improvements in fresh belly firmness due to greater proportions of SFA but had no effect on yields of commercially processed bacon or bacon quality characteristics.

Lysozyme is a 1,4-ß-N-acetylmuramidase that has antimicrobial properties. The objective of this experiment was to determine the effect of a purified granulated lysozyme, compared with antibiotics, on growth performance, small intestinal morphology, and Campylobacter shedding in 10-d-old pigs. Forty-eight pigs (n = 16 per treatment), with an initial BW of 4.0 ± 0.1 kg (P > 0.40), were weaned at 10 d of age, blocked by litter and sex, and assigned to pens (8 pigs/pen). Each block was randomly assigned to consume 1 of 3 liquid dietary treatments for 14 d: a control diet, the control diet + lysozyme (100 mg/kg of diet), or the control diet + antibiotics (neomycin and oxytetracycline, 16 mg/kg of diet). Pigs were weighed and blood was sampled on d 0, 7, and 14. Blood was analyzed for plasma urea N and IgA. After 14 d of treatment, pigs were killed and samples of the jejunum and ileum were collected and fixed to measure villus height and crypt depth. Rectal swabs were taken on d 0, 7, and 14 of treatment, and samples of ileal and cecal contents were taken at d 14 of treatment to determine the presence of Campylobacter. Pigs consuming lysozyme and antibiotics gained BW at a faster rate than did control pigs over the course of the study (402 ± 12 and 422 ± 14 g/d, respectively, vs. 364 ± 14 g/d; P < 0.02), resulting in heavier ending BW (9.9 ± 0.3, 9.9 ± 0.3, and 9.0 ± 0.2 kg for pigs in the lysozyme, antibiotic, and control groups, respectively; P < 0.03). Immunoglobulin A decreased and plasma urea N increased over the course of the study (P < 0.1), regardless of dietary treatment (P > 0.6). Crypt depth was increased in pigs fed lysozyme- and antibiotic-treated diets, compared with pigs fed the control diet, in both the jejunum (60.0 ± 2.8 and 62.2 ± 3.0 µm, respectively, vs. 50.7 ± 3.1 µm; P < 0.03) and ileum (76.0 ± 7.5 and 72.2 ± 5.0 µm, respectively, vs. 52.4 ± 3.5 µm; P < 0.02). Villus height did not differ in the jejunum (P > 0.2) but was increased in the ileum of pigs consuming the lysozyme- and antibiotic-treated diets, compared with pigs fed the control diet (312 ± 20 and 314 ± 10 µm, respectively, vs. 263 ± 15 µm; P < 0.4). Small intestinal total mucosa and mucosal protein concentrations, as well as disaccharidase-specific activities, were not altered by lysozyme or antibiotics (P > 0.05). Campylobacter was detected in 27% of control samples but in only 5% of samples from pigs fed antibiotics and 8% of samples from pigs fed lysozyme (P < 0.01). Thus, granulated lysozyme is a suitable alternative to antibiotics for 10-d-old pigs consuming manufactured liquid diets.

Early colonisation in the gastrointestinal tract by commensal microbes influences the progressive development and maturity of digestive and immune system functionality in the neonate. Application of strategically selected direct-fed microbials to neonatal pigs may provide an opportunity to dictate a portion of the intestinal microbial community and exert a beneficial influence on these developmental processes. Experiments were conducted to determine the effects of early administration of Lactobacillus brevis strain 1E1 to neonatal piglets (n=224) via a milk supplement system on gastrointestinal microbial counts, villous architecture, and immune cell phenotypes during the lactation phase and after weaning. Pigs administered the direct-fed microbial had lower Escherichia coli counts in the jejunum and ileum (P<0.05), and lower coliform counts in the jejunum compared to unsupplemented pigs (P<0.05). The villous height:crypt depth ratio was greater in the ileum at 9 days of age when pigs were provided L. brevis 1E1 compared to unsupplemented pigs (P<0.05), as well as in the duodenum of pigs supplemented with L. brevis 1E1 at 22 days of age (P<0.05). The number of leukocytes expressing CD2 (P<0.05), CD4 (P=0.07) and MHC-II (P=0.07) was lower in the jejunum of pigs administered L. brevis 1E1 compared to unsupplemented pigs, however direct-fed microbial treatment had no effect on the number of leukocytes expressing CD8, CD25 or SWC3. These data demonstrate that early colonisation of the porcine gastrointestinal tract with L. brevis strain 1E1 during the lactation phase influences the progression of intestinal structure, immune system development, and pathogen establishment, indicating a relationship between early microbial colonisation and development of intestinal maturity and integrity.

Crossbred pigs (n = 216) were used to test the effect of supplemental L-carnitine (CARN) on the fatty acid composition and quality characteristics of fresh pork bellies from pigs fed diets formulated with different inclusion levels of corn oil. Pigs were blocked by BW (43.6 ± 1.0 kg) and allotted randomly to pens of 6 pigs within blocks. Then, within blocks, pens were assigned randomly to 1 of 6 dietary treatments in a 2 × 3 factorial arrangement, with either 0 or 100 mg/kg of supplemental CARN and 3 dietary inclusion levels (0, 2, or 4%) of corn oil (CO). When the lightest block weighed 125.0 kg, all pigs were slaughtered, and left-side bellies were captured during carcass fabrication for quality data collection. Fresh pork bellies were evaluated for length, width, thickness, and firmness (bar-suspension and Instron-compression methods) before a 2.5-cm-wide strip of belly was removed and subsequently dissected into subcutaneous fat, primary lean (latissimus dorsi), secondary lean (cutaneous trunci), and intermuscular fat for fatty acid composition determination. Although belly length, width, and thickness of fresh pork bellies were not affected by CARN (P ≥ 0.128) or CO (P ≥ 0.073), belly firmness decreased linearly (P < 0.001) with increasing dietary CO, but there was no (P ≥ 0.137) effect of CARN on any belly firmness measure. Dietary CARN increased (P < 0.05) the proportion of total SFA in the intermuscular fat layer, increased (P < 0.05) the proportion of total MUFA in the primary and secondary lean layers, and decreased (P < 0.05) the proportion of total PUFA in the intermuscular fat and secondary lean layers of pork bellies. Moreover, the SFA and MUFA compositions decreased linearly (P < 0.001) with increasing dietary CO, and the rate of the decrease in SFA composition was greater (P < 0.001) in the fat layers than the lean layers. Conversely, the PUFA content increased linearly (P < 0.001) with increasing dietary CO, and the rate of the increase in PUFA was greater (P < 0.001) in the fat than the lean layers, and greater (P = 0.022) in the primary than secondary lean layer. Results from this study would indicate that differences in the amount and rate of fatty acid deposition associated with feeding increased amounts of CO, along with moisture differences among the belly layers, combine to negatively affect fresh pork belly firmness.

Crossbred pigs (n=288) were used to test the interactive effects of dietary fat source and slaughter weight on live performance, carcass traits, and fatty acid composition of the LM. Pigs were blocked by initial BW, and, within each of 9 blocks, pens (8 pigs/pen) were randomly assigned to either control corn-soybean meal grower and finisher diets devoid of added fat (Ctrl) or diets formulated with 5% beef tallow (BT), poultry fat (PF), or soybean oil (SBO). Immediately after treatment allotment, as well as at mean block BW of 45.5, 68.1, 90.9, and 113.6 kg, 1 pig was randomly selected from each pen, slaughtered, and allowed to chill for 48 h at 1 degrees C. Backfat was measured on the right sides, and a sample of the LM was removed for fatty acid composition analysis. Regardless of source, inclusion of fat in swine diets did not (P >or= 0.349) affect ADG, ADFI, or G:F. Furthermore, carcasses from pigs fed diets formulated with 5% fat had greater (P=0.013) average backfat depths than those from pigs fed the Ctrl diet. Body weight, carcass weight, and backfat depths increased (P<0.001) as slaughter weight increased from 28.1 to 113.6 kg. The proportion of SFA in the LM increased (P<0.001) with increasing slaughter weight from 28.1 to 68.1 kg, but SFA percentages were similar between 68.1 and 113.6 kg, and pigs fed the Ctrl diet had greater (P=0.032) proportions of SFA than pigs fed the SBO and PF diets. Moreover, the proportion of all MUFA increased (P<0.001) by 9.4 percentage units from 28.1 to 113.6 kg; however, only pigs fed the SBO diet had reduced (P=0.004) MUFA percentages than those fed the Ctrl, BT, and PF diets. Even though the proportion of PUFA in the LM decreased with increasing slaughter weight, pigs fed SBO had greater PUFA percentages, a greater PUFA-to-SFA ratio, and greater iodine values than pigs fed all other dietary treatments when slaughtered at BW of 45.5 kg or greater (fat source x slaughter weight, P < 0.001). Results of this study indicate that fat source had little to no impact on live pig performance, but feeding a polyunsaturated fat source altered the fatty acid profile of the LM within the first 17.4 kg of BW gain; more specifically, including 5% SBO in swine diets could lead to economical ramifications associated with soft pork or fat.

Crossbred pigs (n=288) were used to test the interactive effects of dietary fat source and slaughter weight on the fatty acid composition of subcutaneous fat. Pigs were blocked by initial BW (28.1 kg), and, within blocks, pens (8 pigs/pen) were randomly assigned to either grower and finisher diets devoid of added fat (Ctrl) or diets formulated with 5% beef tallow (BT), poultry fat (PF), or soybean oil (SBO). Immediately after treatment allotment, as well as at mean block BW of 45.5, 68.1, 90.9, and 113.6 kg, 1 pig was randomly selected from each pen, slaughtered, and, within 1 h postmortem, samples of backfat were removed from each carcass between the 4th and 8th thoracic vertebra and separated into the inner, middle, and outer layers for fatty acid composition analysis. During the first 17.4 kg of BW gain, percentages of all SFA increased by more than 4% in subcutaneous fat of pigs fed the Ctrl and BT diets, but decreased by 4.4 and 7.7% in pigs fed the PF and SBO diets, respectively (fat source x slaughter weight, P<0.001). Proportions of all MUFA in subcutaneous fat from BT-fed pigs increased by 6.1% during the first 17.4 kg of BW gain, but MUFA percentages in SBO-fed pigs decreased by 9.1% between 28.1 and 45.5 kg (fat source x slaughter weight, P<0.001). Conversely, percentages of all PUFA from SBO-fed pigs increased by 39.9%, whereas PUFA concentrations in BT-fed pigs decreased by 12.6% as slaughter weight increased from 28.1 to 45.5 kg (fat source x slaughter weight, P<0.001). Resultant iodine values (IV) of subcutaneous fat from SBO-fed pigs increased (P<0.05) from 73.5 to 85.2 within the first 17.4 kg of BW gain, and remained elevated above those of their contemporaries fed the Ctrl, BT, or PF diets at each subsequent slaughter weight (fat source x slaughter weight, P<0.001). The inner backfat layer had the greatest (P<0.05) proportions of all SFA and the least (P<0.05) proportions of all PUFA, whereas the outer layer had the least (P<0.05) percentages of all SFA but the greatest (P<0.05) percentages of all MUFA. Even though the middle and outer subcutaneous fat layers had similar (P>0.05) PUFA percentages, the greatest (P<0.05) and least (P<0.05) IV were in the outer and middle layers, respectively. As expected, the fat source included in swine diets was responsible for the fatty acid compositional changes in subcutaneous fat, yet the results of this study indicate that feeding 5% SBO dramatically increased the polyunsaturation of subcutaneous fat within the first 17.4 kg of BW gain, with backfat IV exceeding 80 thereafter.

Crossbred pigs (n=288) were used to test the interactive effects of dietary fat source and slaughter weight on dissected carcass composition and fatty acid composition of composite carcass samples. Pigs were blocked by initial BW, and within each of 9 blocks, pens (8 pigs/pen) were randomly assigned to either control corn-soybean meal grower and finisher diets (Ctrl) or diets formulated with 5% beef tallow (BT), poultry fat (PF), or soybean oil (SBO). Immediately after treatment allotment, as well as at mean block BW of 45.5, 68.1, 90.9, and 113.6 kg, 1 pig was randomly selected from each pen and slaughtered, and primal cuts from right carcass sides were dissected into muscle, fat, bone, and skin components. Muscle and fat tissues were then ground, and random composite samples were collected from each carcass for fatty acid composition analysis. Fat source did not alter pork primal cut yields (P >or= 0.294), nor were the percentages of carcass muscle (P=0.213), fat (P=0.502), and bone (P=0.551) affected by dietary fat source. Conversely, percentages of the whole shoulder and ham decreased linearly (P<0.001), and the percentages of loin and belly increased (P<0.001) linearly with increasing slaughter weight. Moreover, linear decreases (P<0.001) in carcass muscle, bone, and skin, as well as a linear increase (P<0.05) in carcass fat, were observed as slaughter weight increased from 28.1 to 113.6 kg. Composite samples from pigs fed the BT or Ctrl diets had greater (P<0.05) proportions of SFA, particularly oleic and stearic acids, than those from pigs fed the PF and SBO diets when slaughtered at 45.5, 68.1, and 90.9 kg (fat source x slaughter weight, P<0.001). Percentages of MUFA (including palmitoleic, oleic, and cis-vaccenic acids) decreased (P<0.05), and percentages of all PUFA, especially linoleic and linolenic acids, and iodine values increased (P<0.05) in samples from SBO-fed pigs as slaughter weight increased from 28.1 to 113.6 kg (fat source x slaughter weight, P<0.001). Dietary fat source did not affect carcass composition; however, including 5% SBO in swine diets increased the polyunsaturation of pork, which could lead to economic ramifications associated with soft pork and pork fat.

Crossbred pigs (n = 216) were used to test the interactive effect, if any, of ractopamine (RAC) and dietary fat source on the performance of finishing pigs, pork carcass characteristics, and quality of LM chops during 5 d of simulated retail display (2.6 degrees C and 1,600 lx warm-white fluorescent lighting). Pigs were blocked by BW and allotted randomly to pens (6 pigs/pen), and, after receiving a common diet devoid of RAC for 2 wk, pens within blocks were assigned randomly to 1 of 4 diets in a 2 x 2 factorial arrangement, with 5% fat [beef tallow (BT) vs. soybean oil (SBO)] and RAC (0 vs. 10 mg/kg). Diets were formulated to contain 3.1 g of lysine/Mcal of ME and 3.48 Mcal/kg of ME. Across the entire 35-d trial, pigs fed RAC had greater (P < 0.01) ADG and G:F, but RAC did not affect (P = 0.09) ADFI; however, performance was not affected (P >or= 0.07) by dietary fat source. A cooperative regional research study using 194 sows, from which data were collected from 381 litters, was conducted at 3 research stations to determine the effects of added psyllium (a concentrated fiber source) or soybean hulls to gestation diets on reproductive performance of sows and preweaning performance of their pigs. Primiparous and multiparous sows were allotted to the 3 treatments of control (corn and soybean meal-based), 0.30% psyllium, or 20% soybean hulls. Sows fed the control and 0.30% psyllium diets were provided 1.82 kg/d, and sows fed the 20% soybean hulls diet were provided 2.0 kg/d to equalize ME, Lys, Ca, P, and vitamin and trace mineral intake. Treatments 1 to 3 had 130, 130, and 121 litters per treatment from 64, 64, and 63 sows, respectively. Gestating sows fed psyllium had a greater (P < 0.01 to 0.10) d 110 gestation, farrowing, weaning, and 17 d postpartum BW and gestation ADG compared with sows fed soybean hulls. Sows fed psyllium also had a greater (P < 0.10 and 0.08) d 110 gestation BW and gestation ADG than the control sows. Sows fed soybean hulls had a reduced (P < 0.06) farrowing BW compared with the control sows. Sows fed psyllium weaned lighter (P < 0.09) pigs than sows fed the control diet. Litter size was not affected (P > 0.10) by diet. Sows fed psyllium had a reduced (P < 0.03) feed intake compared with sows fed soybean hulls for d 5 to 7 postpartum, and sows fed the control diet were intermediate. Fecal scores (1 to 5 with 1 = dry and 5 = watery) were greater (P < 0.001) and DM content was less (P < 0.001 to 0.01) in the feces of sows fed soybean hulls compared with sows fed psyllium or the control diet on d 112 of gestation and d 4 postpartum. Fecal scores were greater (P < 0.10) and fecal DM content was less (P < 0.02) in sows fed psyllium compared with sows fed the control diet only on d 4 postpartum. In summary, sows fed soybean hulls during gestation had reduced BW compared with sows fed the control diets. In contrast, sows fed psyllium had an increased BW.

A direct-fed microbial (DFM) based on a combination of Bacillus organisms specifically selected to increase the manure decomposition process was evaluated to determine its efficacy for improving growth performance and manure dissolution time. Three experiments involving 336 crossbred barrows and gilts were conducted to determine the effect of the Bacillus-based direct-fed microbial on growth performance and pen cleaning time. In each experiment, 2 dietary treatments (0 and 0.05% DFM) were fed during the growing-finishing period throughout the experiment, such that the DFM provided 1.47 x 10(8) cfu of Bacillus organisms per gram of supplement. Data from the 3 experiments were combined for analysis, such that there were 28 pens representing each of the 2 treatments. Pigs were weighed and feed intake was determined at the initiation and termination of each phase (starter, grower, and finisher). At the end of Exp. 1 and 3, pen cleaning time was determined by measuring the time required for each pen to be scraped and washed with a high-pressure sprayer. Additionally, 2 solid manure mat samples weighing approximately 4 g each were collected from solid manure buildup in each pen (16 pens/treatment), and the time required to completely disperse each manure mat sample was determined. Gain:feed improved (P < 0.05) in pigs fed Bacillus compared with the control diet during the finisher phase and throughout the combined growing-finishing period. The time required to dissolve the manure mat was reduced (P < 0.01) in samples collected from pens containing pigs fed Bacillus compared with samples from control pens. An additional evaluation was conducted in a commercial swine production facility using statistical process control analysis. Statistical process control analysis determined that supplementation with Bacillus increased the expected mean for ADG and decreased the expected mean for death loss percentage. Supplementation with a DFM composed of specifically selected Bacillus organisms improved G:F and decreased the time required to disperse a swine manure mat sample in a controlled study conducted at swine research facilities. Furthermore, when evaluated in a commercial swine production facility, the Bacillus-based DFM improved ADG and reduced mortality of pigs during the growing-finishing period.

Crossbred pigs (n = 216) were used to test the interaction, if any, of ractopamine (RAC) and dietary fat source on the characteristics of fresh pork bellies. Pigs were blocked by BW (77.6 +/- 6.5 kg) and allotted randomly to pens (6 pigs/pen). After receiving a common diet devoid of RAC for 2 wk, pens within blocks were assigned randomly to 1 of 4 treatments arranged in a 2 x 2 factorial design, with 5% fat (beef tallow vs. soybean oil) and RAC (0 vs. 10 mg/kg). At the conclusion of the 35-d feeding period, pigs were slaughtered at a commercial pork packing plant (average BW of 108.8 +/- 0.6 kg), and fresh bellies were captured during carcass fabrication. Neither RAC (P = 0.362) nor fat source (P = 0.247) affected belly thickness. Subjective (bar-suspension) or objective (compression test) measures of belly firmness were not (P > or = 0.148) affected by the inclusion of RAC in the diet; however, bellies from pigs fed soybean oil (SBO) were softer than those from pigs fed beef tallow (BT), as indicated by perpendicular (P < or = 0.005) and parallel (P < 0.001) suspensions. Moreover, bellies from BT-fed pigs required more (P = 0.096) force to compress 50% of their thickness than bellies from SBO-fed pigs (52.29 vs. 43.51 kg). Color (L*, a*, and b* values) of the belly lean and fat was not (P > or = 0.131) affected by RAC, and lean color was similar (P > or = 0.262) between fat sources; however, belly fat from BT-fed pigs was lighter (P = 0.030) and redder (P = 0.013) in color than belly fat from SBO-fed pigs. Bellies of SBO-fed pigs had greater (P < 0.001) proportions of PUFA and lower (P < 0.001) proportions of SFA and MUFA than belly fat from pigs fed BT. Regardless of the RAC inclusion level, PUFA:SFA and iodine values were lower in belly fat from pigs fed BT than SBO; however, within SBO-fed pigs, PUFA:SFA and iodine values were further increased by feeding RAC (RAC x fat source, P < 0.001). As expected, dietary fat source altered the fatty acid composition of fresh pork bellies, which subsequently impacted fresh belly firmness. Interestingly, including RAC in swine finishing diets exacerbated the effect of feeding SBO on pork fat polyunsaturation.
