RESUMO

Zooplankton biomass data have been collected in Australian waters since the 1930s, yet most datasets have been unavailable to the research community. We have searched archives, scanned the primary and grey literature, and contacted researchers, to collate 49187 records of marine zooplankton biomass from waters around Australia (0-60°S, 110-160°E). Many of these datasets are relatively small, but when combined, they provide >85 years of zooplankton biomass data for Australian waters from 1932 to the present. Data have been standardised and all available metadata included. We have lodged this dataset with the Australian Ocean Data Network, allowing full public access. The Australian Zooplankton Biomass Database will be valuable for global change studies, research assessing trophic linkages, and for initialising and assessing biogeochemical and ecosystem models of lower trophic levels.

Koalas are an iconic Australian marsupial undergoing precipitous population reduction in South-East Queensland from complex interacting threats. To investigate the causes of death and the interaction of comorbidities with demography in South-East Queensland koalas, a large scale, high-throughput prospective necropsy survey was conducted spanning 2013-2016. During this period, 519 necropsies were conducted in 155 young/subadult koalas, 235 mature, 119 old koalas and 10 of unknown age. Similar numbers of males and females were assessed. Trauma and infectious disease at were the most common single diagnoses. However, comorbidity was frequent, including multicentric infection or infectious disease in combination with trauma or senescence. Female koalas had proportionally more reproductive chlamydiosis compared to males in which the ocular and urinary systems were more commonly affected. Comorbidity and disease were strongly associated with poor body condition, and trauma was associated with good body condition. Animals affected by motor vehicle trauma were often in better body condition than those affected by animal attack, tree fall or other causes of trauma. This study identified a higher frequency of infections and comorbidity then previously reported, confirming the complex nature of interacting threats to the koala population.

Macrofibrils, the main structural features within the cortical cells of mammalian hair shafts, are long composite bundles of keratin intermediate filaments (KIFs) embedded in a matrix of keratin-associated proteins. The KIFs can be helically arranged around the macrofibril central axis, making a cylinder within which KIF helical angle relative to macrofibril axis increases approximately linearly from macrofibril centre to edge. Mesophase-based self-assembly has been implicated in the early formation of macrofibrils, which first appear as liquid-crystal tactoids in the bulb of hair follicles. Formation appears to be driven initially by interactions between pre-keratinized KIFs. Differences in the nature of these KIF-KIF interactions could result in all macrofibrils being internally twisted in a single handedness, or a 50:50 mixture of handedness within each cortical cell. We data-mined 41 electron tomograms containing three-dimensional macrofibril data from previously published studies of hair and wool. In all 644 macrofibrils examined we found that within each tomogram all macrofibrils had the same handedness. We concluded that earlier reports of left- and right-handed macrofibrils were due to artefacts of imaging or data processing. A handedness marker was used to confirm (using re-imaged sections from earlier studies) that, in both human and sheep, all macrofibrils are left-handed around the macrofibril axis. We conclude that this state is universal within mammalian hair. This also supports the conclusion that the origin of macrofibril twist is the expression of chiral twisting forces between adjacent KIFs, rather than mesophase splay and bending forces relaxing to twisting forces acting within a confined space.

OBJECTIVE: Compare the use of four techniques to measure body temperature in koalas: intraperitoneal (thermal data logger and temperature sensitive radio transmitter), rectal (certified thermometer), tympanic (infrared thermometer), and hind foot (infrared camera). METHODS: The body temperature data collected concurrently from the intraperitoneal loggers were used as the benchmark in the analyses. RESULTS: The rectal, foot and tympanic methods consistently recorded lower body temperature when compared with the benchmark. There was a strong positive relationship (R2 = 0.79) between logger and rectal measurements, but no significant relationship between logger and foot or logger and tympanic measurements. CONCLUSION: Rectal measurements can be used to record internal body temperature, with the caveat that such measurements will generally register a temperature approximately 0.25°C lower than the actual intraperitoneal temperature.

RESUMO

Macrofibrils are the main structural component of the hair cortex, and are a composite material in which trichokeratin intermediate filaments (IFs) are arranged as organised arrays embedded in a matrix composed of keratin-associated proteins (KAPs) and keratin head groups. Various architecture of macrofibrils is possible, with many having a central core around which IFs are helically arranged, an organisation most accurately described as a double-twist arrangement. In this chapter we describe the architecture of macrofibrils and then cover their formation, with most of the material focusing on the theory that the initial stages of macrofibril formation are as liquid crystals.

BACKGROUND: The northern hairy-nosed wombat (Lasiorhinus krefftii) is critically endangered, with only 200 individuals remaining in the wild. Individuals are rarely available for detailed pathological assessment and identification of disease threats to individuals is critically important to species conservation. CASE REPORT: Two male northern hairy-nosed wombats, part of the Richard Underwood Nature Refuge population, were presented for necropsy, 5 months apart. They were found to have succumbed to adiaspiromycosis caused by the fungus Emmonsia parva. Pathological presentations were of severe pulmonary oedema and fibrosis, and pleuritis, respectively. Characteristic fungal adiaspores were noted on histopathological examination. The wombats had concurrent variably severe ectoparasite and endoparasite burdens. CONCLUSION: These are the first reported cases of adiaspiromycosis in northern hairy-nosed wombats and the organism was associated with significant pathological changes. The rarity and the logistical challenges of presenting northern hairy-nosed wombats for pathological assessment are a challenge to identifying disease threats in this critically endangered species.

Discrepancies between bioavailable nitrogen (N) concentrations and phytoplankton growth rates in the oligotrophic waters of the Great Barrier Reef (GBR) suggest that undetermined N sources must play a significant role in supporting primary productivity. One such source could be biological dinitrogen (N2) fixation through the activity of "diazotrophic" bacterioplankton. Here, we investigated N2 fixation and diazotroph community composition over 10° S of latitude within GBR surface waters. Qualitative N2 fixation rates were found to be variable across the GBR but were relatively high in coastal, inner and outer GBR waters, reaching 68 nmol L-1 d-1. Diazotroph assemblages, identified by amplicon sequencing of the nifH gene, were dominated by the cyanobacterium Trichodesmium erythraeum, γ-proteobacteria from the Gamma A clade, and δ-proteobacterial phylotypes related to sulfate-reducing genera. However, diazotroph communities exhibited significant spatial heterogeneity, correlated with shifts in dissolved inorganic nutrient concentrations. Specifically, heterotrophic diazotrophs generally increased in relative abundance with increasing concentrations of phosphate and N, while Trichodesmium was proportionally more abundant when concentrations of these nutrients were low. This study provides the first in-depth characterization of diazotroph community composition and N2 fixation dynamics within the oligotrophic, N-limited surface waters of the GBR. Our observations highlight the need to re-evaluate N cycling dynamics within oligotrophic coral reef systems, to include diverse N2 fixing assemblages as a potentially significant source of dissolved N within the water column.

CASE REPORT: Cystic urolithiasis is the most common form of urolithiasis reported in horses. In contrast to the gelding or stallion, clinical disease is less common in the mare and manual removal techniques are possible because of their shorter, wider urethral anatomy. However, these manual removal techniques can be traumatic and are limited by the size of the urolith. This report describes the use of a commercial laparoscopic specimen pouch to remove a cystic urolith in a mare. CONCLUSION: This approach may allow the extraction of larger uroliths per urethra in the mare, while affording some protection to the mucosa of the bladder neck and urethra.

BACKGROUND & OBJECTIVES: The present study addresses gaps in knowledge regarding the association between trauma memory processes and posttraumatic stress responses in youth. Our primary goal was to explore the relative contribution of perceptions of trauma memory quality versus narrative trauma memory characteristics to explain overall adjustment. METHODS: Children (N = 67) were interviewed within four weeks (T1) of an injury leading to hospital treatment and then again eight weeks later (T2). In each interview, the child told a trauma narrative (which were later coded), and answered the Trauma Memory Quality Questionnaire (Meiser-Stedman, Smith, Yule, & Dalgleish, 2007a), a self-report measure indexing the sensory, fragmented, and disorganised characteristics of trauma memory. They then completed measures of Acute Stress Disorder (ASD) symptoms and associated psychopathology at T1 and measures of Posttraumatic Stress (PTS) symptoms and associated psychopathology at T2. RESULTS: Self-reported trauma memory characteristics predicted ASD symptoms cross-sectionally at T1 and PTS symptoms prospectively over time. At both time points, self-reported trauma memory characteristics accounted for all of the unique variance in symptoms initially explained by narrative characteristics. A reduction in self-report ratings, but not the hypothesised narrative features (e.g., disorganised or lexical elements of the narrative), significantly predicted a reduction in PTS symptoms over time. LIMITATIONS: The small sample size and the absence of a within-subjects narrative control were the main limitations of the study. CONCLUSIONS: These findings underscore the importance of self-reported trauma memory characteristics to the aetiology of PTSD.

OBJECTIVE: To investigate the comparative efficacy of BioRelease Deslorelin® (BRD) and Ovuplant® for induction of ovulation in cyclic mares in Australia. METHODS: Ovarian follicular activity of 60 mares for a total of 95 cycles was monitored by ultrasonography until they developed a follicle ≥30 mm and a uterine oedema pattern of 3. Mares were then randomly allocated to one of three treatment groups: (1) treatment with 1.25 mg BRD, (2) a single Ovuplant pellet or (3) 1 mL compound sodium lactate control. Follicular activity was monitored with ultrasonography every 12 h until ovulation was detected or for at least 5 days post treatment. The injection site on each mare was monitored for reaction for a minimum of 5 days post treatment. RESULTS: There was no difference in the percentage of mares ovulating within 48 h when treated with BRD (93.75%) compared with Ovuplant (87.09%). Treatment with both ovulating agents significantly decreased the time to ovulation compared with control mares (P < 0.00005). More mares had injection site reactions with Ovuplant (64.5%) treatment compared with BRD (15.6%) or control mares (0%) (P < 0.00005). CONCLUSIONS: Treatment of mares with 1.25 mg BRD when there is a follicle ≥30 mm and uterine oedema pattern of 3 is as effective as treatment with Ovuplant.

Monstrilloid copepods collected during the past two decades from zooplankton surveys in reef and coastal areas of Australia were analyzed. A first contribution included the taxonomic analysis of three genera of the Monstrilloida, Monstrillopsis Sars, 1921, Maemonstrilla Grygier & Ohtsuka, 2008, and the newly described Australomonstrillopsis Suárez-Morales & McKinnon, 2014. In this document a taxonomic analysis of the species belonging to the genus Cymbasoma Thompson, 1888 is provided. A total of 28 species were found, most of them being undescribed. Seventeen species were described based on females only and eight on male specimens while three species were described from both sexes. Males of Australian species of Cymbasoma are distinguished by details of the genital complex, body size and proportions, ornamentation and processes of the cephalic region, number of caudal setae, and the characteristic structure or ornamentation of the genital lappets. Two main groups of males were distinguished on the basis of the number of caudal setae (3 or 4). As for the females, 20 of the 25 new species of Cymbasoma have fifth legs with an unarmed inner lobe and three setae on the outer lobe; one of these species (C. jinigudira sp. nov.) belongs to the C. longispinosum species-group (sensu Üstün et al. 2014). Another group, consisting of five species, has only two setae on the outer (exopodal) lobe. There were no Australian species of Cymbasoma with a single lobe. A species group, named after C. agoense, is proposed to include species sharing a globose body and a female fifth leg with a large endopodal lobe and an outer (exopodal) lobe with two setae. The females of the new species of Cymbasoma from Australia can be distinguished from their known congeners by unique combinations of characters including the type of body ornamentation, body size and shape, antennule armature and proportions, the presence of distinctive features of the legs 1-4, the presence/absence of processes on the genital compound somite, and the presence/absence of a constriction of the anal somite. We report the occurrence of two previously described species, C. agoense Sekiguchi, 1982 from Japan and C. bali Desai & Krishnaswamy, 1962 from India in Australian waters. Considering the addition of the 25 new species here described, the number of nominal species of the genus is now 66. A key to the Australian species of Cymbasoma (males and females) and a map showing their occurrence in Australia are also provided.

RESUMO

There have been many individual phytoplankton datasets collected across Australia since the mid 1900s, but most are unavailable to the research community. We have searched archives, contacted researchers, and scanned the primary and grey literature to collate 3,621,847 records of marine phytoplankton species from Australian waters from 1844 to the present. Many of these are small datasets collected for local questions, but combined they provide over 170 years of data on phytoplankton communities in Australian waters. Units and taxonomy have been standardised, obviously erroneous data removed, and all metadata included. We have lodged this dataset with the Australian Ocean Data Network (http://portal.aodn.org.au/) allowing public access. The Australian Phytoplankton Database will be invaluable for global change studies, as it allows analysis of ecological indicators of climate change and eutrophication (e.g., changes in distribution; diatom:dinoflagellate ratios). In addition, the standardised conversion of abundance records to biomass provides modellers with quantifiable data to initialise and validate ecosystem models of lower marine trophic levels.

OBJECTIVE: In the light of the high prevalence of physical comorbidities in people with psychotic illness, there is a need to explore modifiable risk factors that may contribute to this disease burden. The benefits of physical activity to both physical and mental health have been well established. We aimed to examine the prevalence and correlates of physical activity in a national sample of adults living with psychotic illness. METHODS: Physical activity was assessed in 1801 people using the International Physical Activity Questionnaire. Participants were dichotomised into low and moderate-high physical activity groups and associations between physical activity and a range of sociodemographic, clinical and physical comorbidity variables were examined using logistic regression. RESULTS: More than half the participants were categorised in the moderate-high physical activity group with nearly half of the sample engaged in physical activity every day. There were significant associations between low physical activity and older age, unemployment, educational non-participation, antipsychotic medication use, social dysfunction, self-reported loneliness and obesity. However, there was no significant association between physical activity and sex, psychosis type, illness duration, physical comorbidity or negative symptoms. CONCLUSION: The findings from this study may inform future interventions designed to increase physical activity in people with psychotic illness.

BACKGROUND: Under predicted climate change scenarios, koala distribution in Australia is expected to be adversely affected. Recent studies have attempted to identify suitable habitat, based on models of bioclimatic regions, but to more accurately reflect the thermal tolerance and behavioural adaptations of the various regional populations, the koala's response to periods of heat stress will need to be investigated at the individual animal level. OBJECTIVE: To explore the safety and suitability of temperature-sensitive intra-abdominal implants for monitoring core body temperature in the koala. METHODS: A temperature-sensitive radio transmitter and thermal iButton data-logger, waxed together as a package, were surgically implanted into the abdominal cavity of four captive koalas. In one animal the implant was tethered and in the other three, it was left free-floating. RESULTS: After 3 months, the implants were removed and all four koalas recovered without complications. The tethering of the package in the one koala resulted in minor inflammation and adhesion, so this practice was subsequently abandoned. The free-floating deployments were complication-free and revealed a diurnal body temperature rhythm, with daily ranges of 0.4-2.8°C. The minimum recorded body temperature was 34.2°C and the maximum was 37.7°C. The difference in the readings obtained from the transmitters and iButtons never exceeded 0.3°C. CONCLUSIONS: The suitability of the surgical approach was confirmed, from both the animal welfare and data collection points of view.

Coral reefs are in decline worldwide due to a combination of local and global causes. Over 40% of the recent coral loss on Australia's Great Barrier Reef (GBR) has been attributed to outbreaks of the coral-eating Crown-of-Thorns Seastar (CoTS). Testing of the hypotheses explaining these outbreaks is hampered by an inability to investigate the spatio-temporal distribution of larvae because they resemble other planktotrophic echinoderm larvae. We developed a genetic marker and tested it on 48 plankton samples collected during the 2014 spawning season in the northern GBR, and verified the method by PCR amplification of single larva. Surprisingly, most samples collected contained CoTS larvae. Larvae were detected 100 km south of current outbreaks of adult seastars, highlighting the potential for rapid expansion of the outbreak. A minimum estimate suggested that larvae numbers in the outbreak area (>10(10)) are about 4 orders of magnitude higher than adults (~10(6)) in the same area, implying that attempts to halt outbreaks by removing adults may be futile.

The specific activity of aminoacyl-tRNA synthetases (spAARS), an index of growth rate, and of the electron transport system (spETS), an index of respiration, was measured in three size fractions (73-150 µm, >150 µm and >350 µm) of zooplankton during five cruises to tropical coastal waters of the Kimberley coast (North West Australia) and four cruises to waters of the Great Barrier Reef (GBR; North East Australia). The N-specific biomass of plankton was 3-4-fold higher in the Kimberley than on the GBR in all 3 size classes: Kimberley 1.27, 3.63, 1.94 mg m-3; GBR 0.36, 0.88 and 0.58 mg m-3 in the 73-150 µm, >150 µm and >350 µm size classes, respectively. Similarly, spAARS activity in the Kimberley was greater than that of the GBR: 88.4, 132.2, and 147.6 nmol PPi hr-1 mg protein -1 in the Kimberley compared with 71.7, 82.0 and 83.8 nmol PPi hr-1 mg protein -1 in the GBR, for the 73-150 µm, >150 µm and >350 µm size classes, respectively. Specific ETS activity showed similar differences in scale between the two coasts: 184.6, 148.8 and 92.2 µL O2 hr-1 mg protein-1 in the Kimberley, against 86.5, 88.3 and 71.3 µL O2 hr-1 mg protein-1 in the GBR. On the basis of these measurements, we calculated that >150 µm zooplankton grazing accounted for 7% of primary production in the Kimberley and 8% in GBR waters. Area-specific respiration by >73 µm zooplankton was 7-fold higher in the Kimberley than on the GBR and production by >150 µm zooplankton was of the order of 278 mg C m-2 d-1 in the Kimberley and 42 mg C m-2 d-1 on the GBR. We hypothesize that the much stronger physical forcing on the North West shelf is the principal driver of higher rates in the west than in the east of the continent.

Although Chlamydia causes disease of the urethra and prostate of male koalas, its impact on the testis and epididymis has not been examined. This study describes chronic-active and granulomatous orchitis and epididymitis with interstitial fibrosis associated with infection by Chlamydia pecorum in 2 of 18 adult male koalas being euthanized at a koala hospital, 8 of which also had chlamydial prostatitis. By immunohistochemistry and transmission electron microscopy, chlamydial inclusions were demonstrated within Sertoli cells directly associated with mild inflammation surrounding intact seminiferous and epididymal tubules, marked pyogranulomatous inflammation around disrupted tubules, replacement of tubules by interstitial fibrosis, and aspermia. The presence of C. pecorum but not Chlamydia pneumoniae was detected by quantitative polymerase chain reaction of formalin-fixed tissues of the left and right testes and right epididymis in 1 animal. This is the first report of orchitis and epididymitis in a koala infected with C. pecorum.

Monstrilloid copepods were collected during zooplankton surveys in reef and coastal areas of Australia. Representatives of all four genera of the Monstrilloida (Monstrilla Dana, Monstrillopsis Sars, Cymbasoma Thompson, and Maemonstrilla Grygier & Ohtsuka) were recorded. In this contribution a taxonomic analysis of specimens belonging to the latter two genera is provided, and a new genus described. The genus Monstrillopsis was represented exclusively by male specimens, on the basis of which three new species are described: Mon. hastata sp. nov., Mon. boonwurrungorum sp. nov., and Mon. nanus sp. nov. These are distinguished from each other and previously described species of this genus by details of the genital complex (or genital apparatus), body size, ornamentation of the cephalic surface, number of caudal setae, and characteristic modifications of the fifth antennular segment. All have distinctive characters not associated with sexual modifications, which will ease the task of matching females collected in future studies. Australomonstrillopsis gen. nov. is proposed to accommodate a male specimen with a unique combination of characters including massively developed caudal rami, cephalic perioral protuberances, and absence of an inner seta on the first exopodal segment of legs 1-4, among other characters. The new genus is monotypic and contains A. crassicaudata sp. nov. Three of the four new species of Maemonstrilla (Mae. ohtsukai sp. nov., Mae. hoi sp. nov., and Mae. protuberans sp. nov.) belong to the Mae. hyottoko species group, and the remaining one, Mae. crenulata sp. nov., belongs to the Mae. turgida group. Each of the new species of Maemonstrilla from Australia can be distinguished from its known congeners by a unique combination of characters including the type of body reticulation, body size, antennule and body proportions, distinctive characters of the swimming legs, details of the antennular armature, and the presence/absence of a posteroventral process on the genital compound somite. With the addition of the four new species of Monstrillopsis and the four of Maemonstrilla described herein, the number of species in these genera has increased to 13 and 11 species, respectively. In no case did congeneric species co-occur, hinting that there may be a rich species diversity yet to be discovered within the Australian Monstrilloida.
