The present study was designed to evaluate equine blastocyst re-expansion rate, quality, and sex following perforation of the blastocoel, collection of blastocoel fluid (BF), and PCR amplification of free DNA. Experiment 1 tested the feasibility of the BF sample collection with a hand-held, small-gauged needle (26g) and subsequent PCR amplification of the TSP-Y gene for males and AMEL-Y gene for males and AMEL-X gene for females. Experiment 2 tested the application of the technique. Equine embryos were collected via uterine flushes 8d after ovulation. Thereafter, embryos (n = 19) were initially assessed and transferred to a 50 µL droplet of holding medium in which the blastocoel was manually perforated as in Experiment 1. Within 1 min of detecting a diameter decrease or collapse, the entire volume of each droplet of medium was collected and stored at -20 °C until PCR. In Experiment 1, amplification of the TSP-Y gene was positive for males at 60% (9/15) and negative for females at 40% (6/15). In Experiment 2, a total of 42 embryos were randomly assigned to a collapsed embryo (CE) or intact embryo (IE) groups and stored at room temperature (RT, 25 °C) or cold temperature (CT, 5 °C) for 24h as follows: 1) CERT, n = 11; 2) CECT n = 11; 3) IERT, n = 10; and 4) IECT, n = 10. After 24h, embryo diameter and quality were reassessed. For all collapsed embryos (n = 19), blastocoel fluid was subjected to double PCR amplification of the TSPY gene with blood from adult male and female horses as controls. Positive gene amplification indicated 57.9% (11/19) of embryos were male and negative amplification indicated 31.6% (6/19) of embryos were female. Relative to the least diameter (0%) after perforation of collapsed embryos or fullest diameter (100%) of intact embryos at T0, percentage change in diameter and quality Grade 1 or 2 embryos after 24h of storage for all groups were, respectively: 31.2% and 54% for CERT group, 28.2% and 0% for CECT group, 25.9% and 100% for IERT group, 4.3% and 80% for IECT group, respectively. Thus, needle-induced leakage and collapse of the blastocoel at T0 resulted in a high rate of blastocyst re-expansion (69%) with many embryos (54%) achieving good quality at T24 with potential for transfer as either male or female embryos. For both collapsed and intact embryos, it was observed that storage for 24h at room temperature (25 °C) was associated with improved embryo growth and morphological quality compared to storage at cold temperature (5 °C).

Ozone (O3 ) therapy has been used to improve peripheral tissue oxygenation in humans and domestic animals. The goal of the present study was to characterize histological changes in the endometria of healthy equines following tissue exposure to gas mixtures enriched with different concentrations of O3 . Cycling mares without endometrial degeneration were divided into three groups according to treatment (n = 9 mares/group). The uteri from the O3 , ½O3 and control groups were insufflated for 3 min with gas containing 42, 21 and 0 µg O3 ml-1 , respectively. Treatments were performed every three days from D0 to D6. Endometrial samples were collected immediately before the first treatment and 24 hr after the last treatment. The following nine histological parameters were evaluated: (i) the number of endometrial blood vessels, (ii) endometrial vascular degree (EVD), (iii) increase rate of blood vessels, (iv) increase rate of EVD, (v) glandular total area, (vi) glandular lumen area, (vii) intraglandular secretion area, (viii) glandular epithelial height and (ix) luminal epithelial height. In the O3 group, a positive effect from treatment (p < .01) was detected for all vascular parameters (i, ii, iii and iv), glandular total area, intraglandular secretion area and glandular epithelial height. Compared to the control group, the ½O3 group had greater (p < .01) EVD (84.1 ± 12%) and a higher increase rate of blood vessels (151.9 ± 47.1%). Uterine insufflation with low or intermediate concentrations of the O2 -O3 gas mixture induced endometrial angiogenesis. Morphometry, but not morphology, of the endometrial glands was affected by local O3 therapy. These findings would be of great significance for the development of new therapies for infertility in mares.

Aiming to investigate the effects of different hormonal protocols on the endometrium morphometry of anestrous mares, 26 animals were assigned to four different treatment groups: (1) EB2.5LAP4: single dose of 2.5 mg of estradiol benzoate (EB); (2) EB5LAP4: 5 mg of EB in 2 consecutive days; (3) EB10LAP4: 10 mg of EB in three consecutive days, considering that all EB-treated groups received a single dose of 1,500 mg of long-acting progesterone (LA P4) after the single/last EB dose; and (4) LAP4: only 1,500 mg of LA P4. Results were also compared with those found in cyclic mares (control group). Endometrial biopsies were collected before and after the hormonal treatments in anestrous mares, and during estrus and at 5 days after ovulation in cyclic mares (D5). Samples were prepared for histological and histomorphometric analysis. Tissue sections were examined to determine luminal epithelium height (LEH), glandular epithelium height (GEH), endometrial thickness (ET), and glandular density (GD). Similar morphometric changes were observed after EB and P4 were administered to groups EB5LAP4 and EB10LAP4. Five days after LA P4 administration (D5), all the assessed variables were similar between all EB-treated groups. In addition, all variables of the EB-treated groups were similar to the control group on D5. Although most of the LAP4 group variables on D5 were similar to the EB-treated groups (except GD), reduced GD and GEH were found when compared with the control group, demonstrating the importance of estradiol priming before P4 on glandular activity and density.

The present study characterized the luteal status and the dynamic of the conceptus during the first 20 days of gestation in mares with different ages and degrees of endometrial degeneration. Total area of the corpus luteum (CL), luteal vascularity, CL area with blood signals, progesterone concentrations (P4), embryonic vesicle diameter, number of embryonic location changes, embryonic fixation position and uterine contractility were evaluated. In Experiment 1, mares ≤6 years of age (Young group, 5.6 ± 0.2 years, n = 7 mares) and mares ≥15 years of age (Old group, 17.2 ± 0.9 years, n = 6 mares) were used to investigate the effect of age. In Experiment 2, the luteal and embryonic parameters were compared between mares with minimal (Mild group, endometrial category I, n = 9 mares) and severe (Severe group, endometrial category III, n = 7 mares) endometrial degeneration. The Old and Severe groups had greater (p ≤ 0.04) total CL area and reduced luteal vascularity (p ≤ 0.04) than the Young and Mild groups, respectively. However, P4 levels and CL area with blood signals were similar (p ≥ 0.8) between the groups. A negative effect of age (p < 0.01), but not of endometrial degeneration (p = 0.6), was found for the embryonic vesicle diameter. The conceptus mobility was high (p > 0.1) until day 14 of gestation in the Severe group, while a reduced number of changes of the embryo location was detected earlier (p < 0.05) in the Old group. In conclusion, the newly formed CL of aged mares and mares with severe endometrial degeneration suffered a structural remodelling to safeguard the local blood supply and the continuous P4 output during early gestation. Moreover, an earlier reduction of the embryonic mobility and a delayed development of the conceptus were associated with advanced age, regardless of the degree of endometrial degeneration.

The present study evaluated the development of equine conceptus transferred to non-cyclic recipient mares treated with altrenogest. Twenty four mares were used as embryo recipients during the ovulatory phase (Control group; n=8) or anestrus/transitional phases (Altrenogest group; n=16) and were evaluated by transrectal ultrasonography, at five day intervals, to monitor the development of embryonic vesicles from 15 to 45 days of pregnancy and embryo proper/fetus from 20 to 60 days. Embryonic vesicle's features such as shape, embryo location within the vesicle and umbilical cord development were similar between cyclic and non-cyclic recipient mares. The embryonic vesicle and embryo proper/fetus growth was significant (P<0.05) between 15 and 60 days of gestation in Altrenogest and Control groups, except for days 20 to 30, where embryonic vesicle growth decrease was observed (P>0.05). The embryonic vesicle and embryo proper/fetus growth was similar (P>0.05) when gestational days were compared between groups. The similarity in conceptus growth between cyclic and non-cyclic recipient mares during early pregnancy indicates that the uterine environment of non-cyclic recipient mares treated with progestins provides similar conditions for the development of transferred embryos.


O presente trabalho avaliou o desenvolvimento de conceptos equinos transferidos para éguas receptoras acíclicas, tratadas com altrenogest. Vinte e quatro receptoras gestantes por transferência de embrião durante a fase cíclica (grupo Controle; n=8) ou acíclica (grupo Altrenogest; n=16) foram avaliadas por meio de ultrassonografia transretal, a cada cinco dias, para monitorar o desenvolvimento da vesícula embrionária entre 15 e 45 dias de gestação e embrião/feto dos 20 aos 60 dias. As características da vesícula embrionária quanto ao formato, localização do embrião dentro da vesícula e formação do cordão umbilical foram similares em receptoras cíclicas e acíclicas. O crescimento da vesícula embrionária e embrião/feto foi significativo (P<0,05) entre 15 e 60 dias de gestação nos grupos Altrenogest e Controle, com exceção dos dias 20 a 30, em que redução no crescimento da vesícula embrionária foi observada (P>0,05). O crescimento da vesícula embrionária e embrião/feto também foi similar (P>0,05) entre grupos nos diferentes dias de gestação. A similaridade no crescimento do concepto entre éguas receptoras cíclicas e acíclicas durante o início da gestação indica que o ambiente uterino de receptoras acíclicas, mantidas com progestágeno, oferece condições similares para o desenvolvimento de um embrião transferido.

The primary objective of this study was to examine the follicular and ovulatory responses following treatment with pFSH in association with ablation-induced or spontaneous follicular wave emergence or follicle deviation during diestrus in crossbred (Mangalarga×Arabian) and Brazilian Warmblood mares with a propensity for spontaneous multiple ovulations; secondary considerations were given to the collection of embryos. In Experiment 1, crossbred mares were administered (im) saline (control, n=7) or pFSH (25mg) when the largest follicle of the ablation-induced follicular wave reached ≥13mm (n=7) or ≥20mm (n=7) or, after pre-treatment ovulation (Day 0) on Day 6 (n=7). In Experiment 2, crossbred mares were administered (im) saline (control, n=10) or a larger dose of pFSH (50mg, n=7) when the largest follicle of the ablation-induced follicular wave reached ≥13mm. In Experiment 3, Brazilian Warmblood mares were administered (im) saline (control, n=7), pFSH (25mg, n=7 or 50mg, n=5) or EPE (12.5mg, n=7) as a positive control on Day 6. Ultrasonic technology was used to ablate all follicles ≥8mm and to monitor follicular development and detect ovulation. Treatment with pFSH or EPE was done twice daily until the largest follicle reached ≥32mm; thereafter, hCG (2500IU) was administered (iv) when the largest follicle reached ≥35mm. Artificial insemination was done 12h after hCG and embryo collections were done 8 d after post-treatment ovulations. In Experiments 1 and 2, treatment of crossbred mares with pFSH post-ablation in association with the expected time of wave emergence or follicle deviation did not (P>0.05) enhance the follicular or ovulatory responses or collection of embryos compared to controls. In Experiment 3, although the enhanced ovulatory response of mares to EPE at the expected time of spontaneous wave emergence was not different (P>0.05) from controls, it was greater (P<0.05) than the response to pFSH. In conclusion, the novelty of using follicle ablation prior to pFSH treatment at the time of wave emergence or follicle deviation did not enhance the follicular or ovulatory responses or collection of embryos to treatment in crossbred mares. In addition, the hypothesis that Brazilian Warmblood mares with a greater propensity for spontaneous multiple ovulations are as responsive to pFSH compared to EPE was not supported. Thus, the combined experimental results of the present study continue to support the general consensus that pFSH is relatively ineffective for follicular superstimulation/superovulation in mares.

Os programas comerciais de transferência de embriões em éguas existem por mais de três décadas e são hoje uma das biotécnias mais utilizadas na reprodução assistida de equinos. O exame ultrassonográfico nos períodos pré e pós-cobertura de doadoras de embriões, assim como a avaliação de receptoras no momento da inovulação é de vital importância para o êxito de um programa de transferência de embriões. A ultrassonografia Doppler é uma técnica não-invasiva que permite a avaliação em tempo real da hemodinâmica do trato reprodutivo de animais de grande porte. Por fornecer detalhes anatômicos e informações imediatas sobre a fisiologia do fluxo sanguíneo de tecidos e órgãos, o exame doppler permite a avaliação do potencial ovulatório de folículos e do status funcional de corpo lúteo e útero, além de ser uma técnica auxiliar para o diagnóstico de distúrbios na hemodinâmica do sistema reprodutivo. Alterações na perfusão sanguínea dos futuros folículos dominantes são detectáveis em diferentes fases de seu desenvolvimento, como na divergência folicular e no período pré-ovulatório. A partir dessas informações, é possível determinar o princípio da atividade sexual e o momento ideal para o início de tratamentos superovulatórios e indutores de ovulação, assim como o momento mais apropriado para a realização de coberturas de éguas doadoras de embriões. A avaliação através do modo-Doppler do corpo lúteo e útero de éguas receptoras é também um instrumento auxiliar para a seleção de animais com perfil sérico de progesterona e ambiente uterino adequados para a sobrevivência do embrião e manutenção da gestação. Essa técnica pode ser útil ainda ao se avaliar a interação concepto-maternal. Apesar da aplicabilidade da tecnologia doppler dentro de programas de transferência de embriões, novos estudos visando a determinar padrões de normalidade e posterior caracterização de distúrbios de fluxo sanguíneo de trato reprodutivo ainda se fazem necessários.


Embryo transfer in mares has been commercially used for more than three decades and it is one of most frequently applied biotechnologies on equine reproduction. Ultrasonic doppler exam of donors during the pre and post-breeding and recipients evaluation at the embryo transfer moment is essential for embryo transfer program success. Doppler ultrasonography is a non-invasive technic that allows real-time evaluation of the reproductive system hemodynamics in large animal. For providing anatomical details and blood-flow physiologic information of vessels and tissues, doppler exam can be used for ovulation prognostication, and to evaluate uterine and luteal functional status. Additionally, it can serve as a diagnostic aid of hemodynamic disturbance in reproductive system. Vascularity changes of future ovulatory follicles precede the diameter deviation. Based on this, it is possible to estimate the beginning of the breeding season and the best moment to initiate superovulatory treatments. Moreover, follicular blood flow can be used to decide the most appropriated moment for ovulation induction and breeding of donors mares. Uterine and luteal evaluation using Doppler-mode in mares can be useful to select embryo recipients with satisfactory progesterone production and adequate uterine vascularity for embryo development and maintenance of pregnancy. The maternal recognition of pregnancy also can be evaluated. Although the applicability of the Doppler technology in embryo transfer programs, additional studies are necessary to determine the standards of normality and to characterize different blood-flow disturbances of the reproductive system.

The present study was conducted in order to verify the efficacy of lower doses and alternative routes of a prostaglandin F2alpha analogue, luprostiol (PGF), for the induction of luteolysis and the precipitation of estrus in nonlactating Nelore cows (Bos taurus indicus). A conventional dose (15 mg) of PGF was compared to doses lower than the conventional dose, which ranges from 10 to 50%, that were administered intramuscularly (IM), intravulvosubmucosally (IVSM), or in the Bai-hui acupuncture site located within the lumbosacral area. The cows were administered PGF 8 day after estrus in the presence of a corpus luteum, and randomly assigned to the following groups: G1 (positive control), 15 mg, IM (n = 23); G2, 7.5 mg, IM (n = 23); G3, 3.75 mg, IM (n = 24); G4, 7.5 mg, IVSM (n = 25); G5, 3.75 mg, Bai-hui acupoint (n = 24); and G6, 1.5 mg, Bai-hui acupoint (n = 25). The results indicated that 50% of a conventional dose of PGF (7.5 mg) resulted in a complete luteal regression (plasma progesterone <1 ng/ml) at Hour 48, and hastened estrus, regardless of whether or not PGF was administered IM or IVSM. Comparatively, 10 or 25% of the conventional dose, even when administered to the Bai-hui acupoint, resulted in an initial reduction in the concentration of progesterone at Hour 24, followed by an increase observed at Hour 48. In conclusion, 25% of a conventional PGF dose administered via the Bai-hui acupoint proved inadequate to induce a complete luteal regression, whereas 50% of a conventional dose administered IM or IVSM was found to be the minimal dose required to induce effectively a complete luteal regression, and to precipitate the onset of estrus in nonlactating Nelore cows.

O presente trabalho deve como objetivo testar uma alternativa para a avaliaçäo da morfológia espermática proposta por PAPA et al. (1989), em funçäo da dificuldade de obtençäo e alto custo do corante amarelo metacromo. No Laboratório de Andrologia da F.M.V.Z. - UNESP - Botucatu foram feitos 20 esfregaços de amostras de sêmen descongelados das espécies bovina, caprina, eqüina, canina e coradas pela técnica anteriormente proposta, substituindo-se a soluçäo do amarelo metacromo glicerinada pela soluçäo de rosa bengala glicerinada (20 ml de soluçäo mäe de rosa bengala citado por MIES FILHO (1987) + 55 ml de álcool metílico + 5 ml de tampäo sorensen + 20 ml de glicerina). A análise dos esfregaços de sêmen corados com a substituiçäo proposta permitiu perfeita visualizaçäo do acrossomo, bem como das demais estruturas do espermatozóides e manteve o padräo de coloraçäo da morfologia espermática do sêmen descongelado
