RESUMO

Understanding the role of cysteine-rich receptor-like kinases (CRKs) in plant defense mechanisms is crucial for enhancing wheat resistance to leaf rust fungus infection. Here, we identified and verified 164 members of the CRK gene family using the Triticum aestivum reference version 2 collected from the international wheat genome sequencing consortium (IWGSC). The proteins exhibited characteristic features of CRKs, including the presence of signal peptides, cysteine-rich/stress antifungal/DUF26 domains, transmembrane domains, and Pkinase domains. Phylogenetic analysis revealed extensive diversification within the wheat CRK gene family, indicating the development of distinct specific functional roles to wheat plants. When studying the expression of the CRK gene family in near-isogenic lines (NILs) carrying Lr57- and Lr14a-resistant genes, Puccinia triticina, the causal agent of leaf rust fungus, triggered temporal gene expression dynamics. The upregulation of specific CRK genes in the resistant interaction indicated their potential role in enhancing wheat resistance to leaf rust, while contrasting gene expression patterns in the susceptible interaction highlighted potential susceptibility associated CRK genes. The study uncovered certain CRK genes that exhibited expression upregulation upon leaf rust infection and the Lr14a-resistant gene. The findings suggest that targeting CRKs may present a promising strategy for improving wheat resistance to rust diseases.

In the present study, an experiment was carried out on the postharvest of cucumber fruit during a 14-day shelf life. The aim was to assess the impact of calcium nanoparticles (CaNPs) blended with different concentrations of salicylic acid (SA) on the shelf life of cucumbers during the seasons of 2018 and 2019. The investigation further monitored the influences of CaNPs-SA on some physical properties of cucumber, including the percentage weight loss, color, and fruit firmness. In addition, chemical properties, such as total soluble solids (SSC%), total acidity (TA%), total soluble sugars, and chlorophyll pigmentation of the fruit skin, were assessed during a 14-day shelf lifeCell wall degradation enzymes (CWEAs) such as polygalacturonase (PG), cel-lulase (CEL), xylanase (XYL), and pectinase (PT) were also researched. In addition, the generation rates of H2O2 and O2•- were calculated, as well as the reduction of DPPH. The lipid peroxidation (malondialdehyde, MDA) and cell membrane permeability (IL%) of cell wall composites were also determined. CaNPs-SA at 2 mM suppressed CWEAs, preserved fruit quality, reduced weight loss throughout the shelf-life period, and reduced the percent leakage value. At this concentration, we also found the lowest levels of MDA and the highest levels of DPPH.

This work aimed to co-digest various wastes to assess the best combination of all mixing ratio, also at choosing the best ratio between untreated primary sludge (UPS) singly from two sources, (South valley University (SUPS) and Abu tesht wastewater station (AUPS) and raw chicken manure (RCM) and comparing the results in either case. The co-digestions of untreated primary sludge from Abu tesht wastewater treatment stations with different levels of raw chicken manure (0:100, 10:90, 30:70, 50:50, 90:10, and 100:0) to obtain the best mixtures. Also, co-digestion of untreated primary sludge from south valley university with different levels of raw chicken manure at the same ratios, to obtain the best mixtures. Batch digestion tests were applied in 2.5 L digester with a working volume of 2.0 L. The samples in triplicates were separately loaded into the digesters locally fabricated and kept for 20 days as a retention period and diluted with the same amount of water. Mesophilic under 35 °C was adopted for untreated primary sludge as well as mixtures with raw chicken manure based on total solids (TS) and volatile solid (VS) proportions. The average biogas yields from AUPS/RCM mixture obtained ranged from 8570 to 5600 ml, by the following descending order, 10: 90 > 90:10 and so on >100:0, and the average biogas yields from SUPS/RCM obtained ranged from 6330 to 5635 ml, in the order of 90: 10 > 10:90 and so on >100:0. The results showed highest biogas yield from AUPS/RCM and SUPS/RCM mixtures with mixing ratio of 10:90 and 90:10, respectively, however, the lowest biogas production detected in separate digestion of AUPS and SUPS. The results indicated that co-digestion between the sludge and raw chicken manure could increase total biogas production volume, enhance sludge treatment process, and produce eco-friendly sludge because of co-digestion process than separate processing of each feedstock.

This paper presents a multiphysical numerical study of a photothermal therapy performed on a numerical phantom of a mouse head containing a glioblastoma. The study has been designed to be as realistic as possible. Heat diffusion simulations were performed on the phantom to understand the temperature evolution in the mouse head and therefore in the glioblastoma. The thermal dose has been calculated and lesions caused by heat are shown. The thermal damage on the tumor has also been quantified. To improve the effectiveness of the therapy, the photoabsorber's concentration was increased locally, at the tumor site, to mimic the effect of using absorbing contrast agents such as nanoparticles. Photoacoustic simulations were performed in order to monitor temperature in the phantom: as the Grüneisen parameter changes with the temperature, the photoacoustic signal undergoes changes that can be linked to temperature evolution. These photoacoustic simulations were performed at different instants during the therapy and the evolution of the photoacoustic signal as a function of the spatio-temporal distribution of the temperature in the phantom was observed and quantified. We have developed in this paper a numerical tool that can be used to help defining key parameters of a photothermal therapy.

Background Acute upper gastrointestinal (GIT) bleeding is a common medical emergency clinically manifested by hematemesis and/or melena. This study aimed to elucidate the roles of Helicobacter pylori and the platelet-spleen ratio as risk factors for variceal bleeding in patients with portal hypertension secondary to liver cirrhosis. Methods The study was conducted on 200 patients with liver cirrhosis of various etiologies who were divided into two groups: group 1 included 100 patients with liver cirrhosis and portal hypertension with or without a history of upper GIT bleeding, and group 2 included 100 patients with liver cirrhosis without portal hypertension. Upper GIT endoscopy was performed, and biopsy samples were taken from the gastric antral mucosa for rapid urease testing. The platelet-spleen diameter ratio was calculated for all patients. Results In group 1, most patients who had a history of variceal bleeding were H. pylori-negative whereas most patients without a history of variceal bleeding were H. pylori-positive, implying that H. pylori may play a significant role as a protective factor against variceal bleeding. The calculated odds ratio for the rapid urease test was low (0.851), whereas the calculated odds ratio for the platelet-spleen diameter ratio was higher (9.766) than that for the rapid urease test. Thus, the rapid urease test plays a significantly higher role than the platelet-spleen ratio as a risk factor for bleeding (p-value = 0.001). Conclusions H. pylori has a more significant relationship with upper GIT bleeding than the platelet-spleen diameter ratio.

Profitable production of rabbit meat requires comfortable environment for expression of good quality traits embedded in the genetic components of rabbits. One of the major problems facing rabbit production is heat stress, especially in hot regions of the world. This is further aggravated by global warming being experienced as a result of climate change. Rabbits have no or few sweat glands, complicating the problem of heat dissipation, thereby predisposing the animals to heat stress (HS). Heat stress adversely affect welfare and adaptation, feed consumption and utilization, immunity and health status, growth, reproduction, and milk production in rabbits. Rabbits are homoeothermic animals. They should be capable of body temperature regulation within a narrow range. Thermoregulation is extremely poor in rabbits because of lack of sweat glands. Rabbits are highly sensitive to high ambient temperature. The presence of thick insulator fur on the skin further impedes heat loss in rabbits. This review summarizes data available literatures in the last two decades (2000-2020). Short compilation on management techniques adopted in rearing rabbits under hot conditions is included.

The current work investigated the influences of different light sources on growth performance, carcass characteristics, blood biochemicals, physiological status and health aspects. One hundred and twenty growing California rabbits were assigned into four empirical collections in an experiment of completely randomized design. Rabbits of the first collection were reared under incandescent light bulbs (control), while the second, third and fourth collections were reared under fluorescent, saving light and LED light bulbs, respectively. The results indicated that rabbits reared under incandescent and LED had the heaviest body weight at 16-weeks-old. Rabbits reared under LED recorded the greatest gain during 14-16 weeks of age and 6-16 weeks of age. The lowest feed conversion ratio was observed in rabbits reared under LED lighting, during 14-16 weeks of age, and during the whole period without significant variations. Fat percentage (%) was low in rabbits exposed to fluorescent and LED. Rabbits grown under LED showed the lowest albumin/globulin and the highest glucose concentration. Rabbits kept under LED showed the lowest rectal temperature, mortality and health risks %. Conclusively, LED lighting source might be appropriate for alteration of incandescent and fluorescent light sources in rabbit farms to improve growth performance without compromising rabbit's carcass and welfare.

INTRODUCTION: Fibrosis is an inevitable complication of chronic hepatitis C virus (HCV) infection. Direct acting antivirals (DAAs) radically treated HCV and were suggested to ameliorate fibrosis. Silymarin (a natural herbal remedy) was proposed to further decrease hepatic inflammation and fibrosis. Consequently, serial monitoring of liver fibrosis status by different biomarkers is needed. AIM OF THE STUDY: To assess hyaluronic acid (HA) as a potential marker of fibrosis regression after DAAs in chronic HCV patients; in addition, to evaluate silymarin as an agent that, beside DAAs, could further improve fibrosis. MATERIAL AND METHODS: Two groups were included (150 patients each). Group 1 received DAAs only, while group 2 received DAAs followed by silymarin. Hyaluronic acid and FIB4 score were assessed at baseline before treatment and 1 year after inclusion in the study. RESULTS: We found that DAA therapy alone or in combination with silymarin resulted in a significant reduction in serum HA level. However, the latter case showed a statistically significantly greater reduction (p = 0.034). Mean ±SD of serum HA level was 211.8 ±179.9 and 143.3 ±123.9 µg/l before and one year after inclusion respectively in group 1 (p = 0.001) and also, its level decreased significantly in group 2 from 188.3 ±211.8 µg/l before receiving DAAs to 126.4 ±136.9 µg/l at one year after inclusion (p = 0.001). There was no significant difference between the 2 studied groups as regards FIB-4 at 1 year after inclusion (p = 0.103). CONCLUSIONS: Hyaluronic acid might be a sensitive marker for monitoring fibrosis regression in treated chronic HCV patients. Adding silymarin to treatment protocols could ameliorate the fibrosis status.

Gene therapy represents a powerful therapeutic tool to treat diseased tissues and provide a durable and effective correction. The central nervous system (CNS) is the target of many gene therapy protocols, but its high complexity makes it one of the most difficult organs to reach, in part due to the blood-brain barrier that protects it from external threats. Focused ultrasound (FUS) coupled with microbubbles appears as a technological breakthrough to deliver therapeutic agents into the CNS. While most studies focus on a specific targeted area of the brain, the present work proposes to permeabilize the entire brain for gene therapy in several pathologies. Our results show that, after i.v. administration and FUS sonication in a raster scan manner, a self-complementary AAV9-CMV-GFP vector strongly and safely infected the whole brain of mice. An increase in vector DNA (19.8 times), GFP mRNA (16.4 times), and GFP protein levels (17.4 times) was measured in whole brain extracts of FUS-treated GFP injected mice compared to non-FUS GFP injected mice. In addition to this increase in GFP levels, on average, a 7.3-fold increase of infected cells in the cortex, hippocampus, and striatum was observed. No side effects were detected in the brain of treated mice. The combining of FUS and AAV-based gene delivery represents a significant improvement in the treatment of neurological genetic diseases.

Glioblastoma multiforme (GBM) is one of the most common and aggressive malignant primary brain tumors in adults. The treatment of GBM is limited by the blood-brain barrier (BBB), which limits the diffusion of appropriate concentrations of therapeutic agents at the tumor site. Among experimental therapies, photo-thermal therapy (PTT) mediated by nanoparticles is a promising strategy. To propose a preclinical versatile research instrument for the development of new PTT for GBM, a multipurpose integrated preclinical device was developed. The setup is able to perform: i) BBB permeabilization by focused ultrasound sonication (FUS); ii) PTT with continuous wave laser; iii) in situ temperature monitoring with photo-acoustic (PA) measurements. In vivo preliminary subcutaneous and transcranial experiments were conducted on healthy or tumor-bearing mice. Transcranial FUS-induced BBB permeabilization was validated using single photon emission computed tomography (SPECT) imaging. PTT capacities were monitored by PA thermometry, and are illustrated through subcutaneous and transcranial in vivo experiments. The results show the therapeutic possibilities and ergonomy of such integrated device as a tool for the validation of future treatments.

Owing to strong plasmonic absorption and excellent biocompatibility, gold nanostructures are among best candidates for photoacoustic bioimaging and photothermal therapy, but such applications require ultrapure Au-based nanoformulations of complex geometry (core-shells, nanorods) in order to shift the absorption band toward the region of relative tissue transparency (650-1000 nm). Here, we present a methodology for the fabrication of Si@Au core-satellite nanostructures, comprising of a Si core covered with small Au nanoparticles (NP), based on laser ablative synthesis of Si and Au NPs in water/ethanol solutions, followed by a chemical modification of the Si NPs by 3-aminopropyltrimethoxysilane (APTMS) and their subsequent decoration by the Au NPs. We show that the formed core-satellites have a red-shifted plasmonic absorption feature compared to that of pure Au NPs (520 nm), with the position of the peak depending on APTMS amount, water-ethanol solvent percentage and Si-Au volume ratio. As an example, even relatively small 40-nm core-satellites (34 nm Si core + 4 nm Au shell) provided a much red shifted peak centered around 610 nm and having a large tail over 700 nm. The generation of the plasmonic peak is confirmed by modeling of Si@Au core-shells of relevant parameters via Mie theory. Being relatively small and exempt of any toxic impurity due to ultraclean laser synthesis, the Si@Au core-satellites promise a major advancement of imaging and phototherapy modalities based on plasmonic properties of nanomaterials.

In the present study, 200 Brown commercial egg-type layers (60 wk old) were used to study the effects of different levels of ecofriendly synthesis of calcium (Ca) nanoparticles (0.0, 0.50, 1.0, and 1.5 g/kg diet) with biocompatible Sargassum latifolium algae extract (SL-CaNps) on exterior egg quality traits, electronic microscopic view of eggshells, Ca and phosphorus (P) retention, serum Ca and P concentrations, and the histology of the uterus. Hens fed with dietary SL-CaNps powder had higher egg weight and shell weight % values than those of the control group. All SL-CaNps treatment groups had the greatest values of shell weight per unit surface area and shell thickness. Dietary supplementation of SL-CaNps at graded levels up to 1.5 g/kg diet had higher serum Ca and inorganic P levels than that of the control. Laying hens fed with SL-CaNps-added diets had beneficial effects on shell ultrastructure in terms of well-developed palisade and mammillary layers. The numbers of apical cells along the branched tubular gland were greater in SL-CaNps-treated groups than those of control. Conclusively, supplementing SL-CaNps powder up to 1.5 g/kg to the diet of laying hens improved eggshell thickness, shell weight% and shell weight per unit surface and has no adverse effect on their eggshell quality or electronic microscopic view of their eggshell.

Lighting regimes are possibly used as a strategy to improve rabbit production in tropical regions; on this background, this study was conducted to assess the possible role of different light (L):dark (D) regimes in improving growing New Zealand white rabbit's performance. Sixty-four growing rabbits were randomly divided into four groups. Rabbits of the first group (C) were exposed to 12 h of light (from 10.00 to 22.00 h). Rabbits of the second group (T1) were exposed to 12 h of light (from 22.00 to 10.00 h). The third group (T2) was exposed to 8 h of light (from 10.00 to 18.00 h), while the fourth group (T3) was exposed to 8 h of light (from 22.00 to 06.00 h). Temperature-humidity index values ranged between 27.00 and 28.87, indicating exposure of growing rabbits to very severe heat stress. Body weight (BW), body weight gain (BWG), feed consumption (FC), feed conversion ratio (FCR), carcass traits, blood parameters, and some physiological (rectum temperature, respiration rate) and health aspects (morbidity, mortality, and health risks %) were investigated. Rabbits of T1 showed the highest (P < 0.05) averages of BW and BWG in comparison with their counterparts. There were no significant differences in FC; however, rabbits of T1 had the lowest value of FCR at marketing. The percentages of dressed carcass, liver, and kidney in T1 and T3 were the highest. Non-significant changes were observed among the groups in most of the metabolic and hematology parameters of blood, except for ALT and N/L ratio. Further, rabbits of T1 showed the lowest rectal temperature and respiration rate in comparison with their counterparts. Conclusively, the utilization of photoperiod of afternoon program for over a short period of time in rabbitry brought in beneficial effects on growth performance and other productive and physiological parameters of growing rabbits.

The adverse influences of elevated ambient temperatures during the summer season on the rabbit industry have received increased global attention. Therefore, this study intended to compare the potential effects of nano-selenium (nano-Se) synthesized by biological (BIO) and chemical (CH) methods on growth performance, carcass variables, serum metabolites, and inflammatory cytokines responses of growing rabbits in the summer season. Two hundred and fifty weaned rabbits (males, 35 days of age) were randomly divided into five treatment groups of 50 rabbits each (each group had five replicates with ten male rabbits). Treatment groups were fed a control diet and four controlled diets supplemented with nano-Se synthesized by biological method (BIO25 and BIO50, with a 25 and 50 mg of nano-Se/kg diet, respectively) and chemical method (CH25 and CH50, with a 25 and 50 mg of nano-Se/kg diet, respectively) for eight weeks. During 11 to 13 weeks of age, a gradual enhancement in live body weight (LBW), feed intake (FI) and feed conversion ratio (FCR) was noticed with BIO25 and BIO50 treatments compared to those in the other groups. The carcass percentage was significantly higher (p < 0.01) for animals fed with BIO25 than the other groups. The other organ functions were significantly higher (p < 0.01) in heat-stressed groups compared to that of nano-Se groups. Increasing the level of only BIO from a 25 to a 50 mg/kg diet gave more improvement in the studied parameters. Additionally, the concentrations of serum urea, triglycerides (TG), and glutamyl transferase (GGT) were lower (p < 0.01) in both treated and untreated groups. Likewise, the supplementation with nano-Se (BIO25, BIO50, or CH25) significantly improved the antioxidant indices and inflammatory cytokines responses as indicated from serum metabolites. Based on the study results, nano-Se especially synthesized by the biological method at diet levels of 25 or 50 mg/kg improved the growth performance, kidney and liver functions, carcass traits, antioxidants indices, and inflammatory cytokines of growing rabbits during thermal stress.

Unsuitable pH is a major limiting factor for all organisms, and a low pH can lead to organism death. Late embryogenesis abundant (LEA) peptides confer tolerance to abiotic stresses including salinity, drought, high and low temperature, and ultraviolet radiation same as the LEA proteins from which they originate. In this study, LEA peptides derived from group 3 LEA proteins of Polypedilum vanderplanki were used to enhance low pH tolerance. Recombinant Escherichia coli BL21 (DE3) cells expressing the five designed LEA peptides were grown at pH 4, 3, and 2. The transformants showed higher growth capacity at low pH as compared to control cells. These results indicate that LEA peptide could prevent E. coli cell death under low pH conditions.

AIM OF THE STUDY: Chronic hepatitis B virus (HBV) infection is one of the major health problems worldwide. Use of non-invasive tests for assessment of hepatic fibrosis such as the FIB-4 index could be used to avoid liver biopsy. Another promising noninvasive test, FIB-5, could also be used to detect significant hepatic fibrosis. The aim of the study was to compare the use of FIB-5 and FIB-4 as noninvasive markers to assess chronic HBV-related hepatic fibrosis. MATERIAL AND METHODS: This study was done on 176 chronic HBV patients who underwent liver biopsy. Grading and staging of liver fibrosis was done according to the METAVIR scoring system. FIB-5 and FIB-4 scores were calculated for all patients. RESULTS: As regards FIB-4 for differentiation between non-significant fibrosis (group I) and significant fibrosis (group II), at a cutoff level of 1.28 with positive predictive value (PPV) 41.4% and specificity 48% while at a cutoff level of 7.08 with PPV 98.8% and specificity 98% for FIB-5. CONCLUSIONS: As regards both scores, the FIB-5 score was more specific than FIB-4 for diagnosing significant from nonsignificant hepatic fibrosis in patients with chronic HBV infection.

BACKGROUND AND AIM: Upper gastrointestinal bleeding (UGIB) is a serious complication of portal hypertension in cirrhotic patients. The objective of this study is to identify the risk factors for morbidity and mortality occurring after an UGIB attack. METHODS: A total of 1097 UGIB attacks in 690 patients with liver cirrhosis were studied. Their clinical, laboratory, and endoscopic data were reviewed. RESULTS: Mean age 53.2 ± 10.6 (20-90) years, 78% men and the main cause of liver disease was hepatitis C (94.9%). Complications occurred after 467 attacks (42.6%): hepatic encephalopathy 31.4%, spontaneous bacterial peritonitis 18%, renal impairment 13.2%, and re-bleeding in 7.8%, while 199 patients (18.1%) died. Complications followed 78.4% of bleeding from gastric varices, 75% of post-interventional ulcers, 10.8% of peptic ulcers, and 5.9% of telangiectasias. By univariate analysis: packed red blood cells units transfused, transaminases, Child-Pugh (CP), model of end-stage liver disease (MELD), and albumin-bilirubin (ALBI) scores, beside the presence of hepatocellular carcinoma (HCC), previous hemorrhage in the previous 6 months, and the source of bleeding, were associated with occurrence of complications. By multivariate analysis, independent predictors of complications were CP, MELD, and ALBI scores (odds ratio, 95% confidence interval: 5.63, 3.55-8.93; 1.15, 1.11-1.19; and 2.11, 1.4-3.19, respectively) beside the presence of HCC (4.89, 2.48-9.64). Mortality predictors were packed red blood cells units transfused (1.11, 1.01-1.24), CP (5.1, 1.42-18.25) MELD (1.27, 1.21-1.32) scores, and presence of HCC (6.62, 2.93-14.95). CONCLUSION: High CP, MELD, and ALBI scores beside the presence of HCC could predict poor outcome of UGIB. In the absence of these risk factors, early discharge could be considered if the source of bleeding is peptic ulcer or telangiectasia.

Exhibiting a red-shifted absorption/scattering feature compared to conventional plasmonic metals, titanium nitride nanoparticles (TiN NPs) look as very promising candidates for biomedical applications, but these applications are still underexplored despite the presence of extensive data for conventional plasmonic counterparts. Here, we report the fabrication of ultrapure, size-tunable TiN NPs by methods of femtosecond laser ablation in liquids and their biological testing. We show that TiN NPs demonstrate strong and broad plasmonic peak around 640-700 nm with a significant tail over 800 nm even for small NPs sizes (<7 nm). In vitro tests of laser-synthesized TiN NPs on cellular models evidence their low cytotoxicity and excellent cell uptake. We finally demonstrate a strong photothermal therapy effect on U87-MG cancer cell cultures using TiN NPs as sensitizers of local hyperthermia under near-infrared laser excitation. Based on absorption band in the region of relative tissue transparency and acceptable biocompatibility, laser-synthesized TiN NPs promise the advancement of biomedical modalities employing plasmonic effects, including absorption/scattering contrast imaging, photothermal therapy, photoacoustic imaging and SERS.

RESUMO

Near-infrared (NIR) azacalixphyrins bearing aryl substituents strongly impacting the physico-chemical properties of the macrocycles were designed, enabling hyperchromic and bathochromic shifts of the absorption compared to their N-alkylated analogues. This engineering enhances the photoacoustic response under NIR excitation, making azacalixphyrins promising organic contrast agents that reach the 800-1000 nm range.

This paper reports a methodology for the non-destructive ultrasonic evaluation of welds, based on probing, residue back-focusing and topological energy calculation using an enhanced (focused) adjoint method. The proposed method combines the advantages of time reversal to compensate for the cumulative distorsions experienced by a wave propagating in a heterogeneous medium, and topological imaging to highlight the defect location. The synergistic effect of this combined approach makes it possible to detect anomalies in the most efficient way. The method paves the way towards a matched-insonification imaging of anomalies in anisotropic media.
