RESUMO

A family of novel binary insecticidal crystal proteins, with activity against western corn rootworm, Diabrotica virgifera virgifera LeConte, was identified from Bacillus thuringiensis Berliner. A binary insecticidal crystal protein (bICP) from B. thuringiensis strain PS149B1 is composed of a 14-kDa protein (Cry34Abl) and a 44-kDaprotein (Cry35Ab1). These proteins have been co-expressed in transgenic maize plants, Zea mays L., and effectively control western corn rootworm larvae under field conditions. Laboratory experiments were conducted to better understand the contribution of each component protein to the in vivo activity of the bICP. The 14-kDa protein is active alone against southern corn rootworm, Diabrotica undecimpunctata howardi Barber, and was synergized by the 44-kDa protein. In mixtures, the concentration of the 14-kDa protein had a greater impact on efficacy than the 44-kDa component. Although both proteins are clearly required for maximal insecticidal activity, laboratory results did not support the formation of a stable, fixed-ratio complex of the two component proteins.

Evidence for the rapid metabolic turnover of leaf monoterpenes is a significant component of theories regarding the evolution and metabolic cost of plant chemical defenses. We re-examined whether monoterpenes are continuously synthesized and lost in intact peppermint plants, and demonstrate that the rapid monoterpene turnover previously observed using detached stems does not occur in intact plants. The apparent artifactual nature of rapid monoterpene turnover in peppermint suggests that a re-evaluation of the rates of metabolic turnover of plant defenses is needed before accurate hypotheses regarding the cost of plant chemical defense can be proposed.

Rosettes of Heterotheca subaxillaris were grown at four levels of nitrate. Individual leaf volatile mono- and sesquiterpene content, leaf nitrogen content, and root and shoot dry weight were measured on individual leaves every two weeks for 18 weeks. Rosettes with the highest nitrate availability had 2.2-fold greater leaf nitrogen levels compared to plants with the lowest availability. As nitrate availability became increasingly limited, carbon allocation to both volatile leaf terpenes and root growht increased. Leaf mono- and sesquiterpene content was greatest in the young leaves of individuals growing at the lowest nitrate availability conditions. Higher levels of carbon-based herbivore-deterring chemicals in nitrate-limited plants may increase net productivity through retention of nitrogen that would otherwise be lost to herbivory.
