RESUMO

Sulfanyl radical addition-addition-cyclization (SRAAC) of unbranched diynes proceeded smoothly to give cyclized exo-olefins, while the sulfanyl radical addition-cyclization-addition (SRACA) of diynes having a quaternary carbon gave cyclized endo-olefins. This method was successfully applied to the synthesis of A-ring fragment of 1alpha,25-dihydroxyvitamin D3.

RESUMO

A combination of sulfanyl radical addition-cyclization of dienes connected with hydroximates and subsequent conversion of the resulting cyclic hydroximate to the lactones provides a novel method for the construction of alpha,beta-disubstituted gamma-lactones. Upon treatment with thiophenol in the presence of AIBN, dienes connected with hydroximates smoothly underwent sulfanyl radical addition-cyclization to give cyclic cis- and trans-hydroximates. Hydrolysis of cyclic hydroximates gave the desired cis- and trans-lactones in high yield. This method was successfully applied to the practical synthesis of (+/-)-oxo-parabenzlactone.

Plasma apo E-rich HDL was studied in regard to its quantity and chemical composition in the members of a family with cholesteryl ester transfer activity deficiency, exhibiting familial hyperalphalipoproteinemia. The approach involved a simple precipitation method established in our laboratory. Serum apo E-rich HDL concentrations for two homozygous members were elevated up to 66 and 60 mg/dl in terms of cholesterol (normal, 6.7 +/- 2.3 mg/dl, n = 38), and to 9.4 and 10.8 mg/dl in terms of apo E (normal, 2.6 +/- 1.5 mg/dl, n = 38). The cholesterol/apo E ratio (mole/mole) of apo E-rich HDL was higher in two homozygotes (669 and 531) than in two cholestatic patients with elevated apo E-rich HDL (268 and 149) and in normal subjects (242 +/- 115, n = 38). Chromatographic studies of the serum from a homozygote showed enlargement of all HDL subclasses and apo E in the larger HDL subclass. These facts indicate that the increase of apo E-rich HDL in this disease occurs secondarily to the enlargement of HDL particles, which require substances to cover their cores, having expanded due to the accumulation of cholesteryl ester. The sera from the homozygotes gave HDL cholesterol concentrations which were remarkably discrepant among commercial precipitating reagents, because of the difference in recovery of apo E-rich HDL with these reagents.

The propositus was a 43-year-old Japanese male with a plasma total cholesterol (chol) level of 252 mg/dl and a high density lipoprotein (HDL)-chol of 169 mg/dl. His brother also had a markedly higher HDL-chol level of 149 mg/dl. In addition, his mother, sister and all 3 children had higher HDL-chol levels of 75-91 mg/dl. These data suggest that the propositus and his brother were homozygous for familial hyperalphalipoproteinemia (FHALP), whereas his mother, sister and 3 children were heterozygous for FHALP. None had any clinical signs of atherosclerosis. The propositus and his brother (homozygous FHALP) also showed markedly higher levels of apo AI (greater than or equal to 190 mg/dl) and E (greater than 16 mg/dl). Ultracentrifugal analysis disclosed an increase of HDL2-chol in the propositus. Cholesteryl ester transfer activity (CETA) was completely absent in the propositus (0.0% transfer/5 microliters/18 hr) and his brother (0.3% transfer/5 microliters/18 hr). It is concluded that this case is a family of homozygous FHALP probably caused by complete deficiency of CETA.

Solid phase fluoroimmunoassay of serum 11-deoxycortisol (17,21-dihydroxy-4-pregnene-3,20-dione) was established using fluorescein isothiocyanate-labelled 11-deoxycortisol and anti-11-deoxycortisol antibody-conjugated polyacrylamide beads. 21-Amino-17-hydroxyprogesterone (21-amino-17-hydroxy-4-pregnene-3,20-dione) was synthesized as a useful derivative for preparing the fluorescent dye conjugate. Serum 11-deoxycortisol was measured with this assay system after extraction and purification by Sephadex LH-20 column chromatography. The minimal amount of 11-deoxycortisol detected was 40 pg/tube and the measurable range was from 0.04 to 5.0 microgram/dl. Intra- and inter-assay coefficients of variation were 8.3% (n=6) and 9.8% (n=5), respectively. 11-deoxycortisol values determined by the present assay correlated well with those determined by radioimmunoassay. The present assay is particularly suitable for estimating the conditions of the pituitary and adrenocortical functions.

In order to compare the deformation of the castings in the firing process with the results of the previous paper, 1/10-tapered ring type thimble crowns were prepared using nine Au-Pt-Pd ternary alloys of the same composition as in the previous paper. The deformation in the firing process was determined by the shoulder gap change measurement. Conclusions are summarized as follows. 1) The correlation between the thermal expansion hysteresis H and the shoulder gap change K is highly significant (r= 0.96, phie=7),and it can be represented by the following equation: K(mm)=0.15 + 1.22 H(%) 2) The gap change increases with increase of Pt content. 3) Most of the deformation occurs in degassing treatment, while in the other firing process the deformation scarcely occurs.
