RESUMO

Plastic ingestion in birds is a widespread phenomenon of increasing concern. However, little is known about how exposure to microplastics (MP) affects the health of birds. In other organisms, MP exposure alters lipid metabolism and composition. If also true in birds, this could affect their fitness, especially since birds heavily rely on lipids during migration and egg production. Therefore, the aim of this study was to determine if ingestion of MP (polypropylene and polyethylene collected in nature) in two size ranges, large (3 mm) and small (<125 µm), affects lipid metabolism in the Japanese quail (Coturnix japonica). We orally exposed 55 one-week-old quail during 5 weeks to a total of 600 mg of MP in sizes of either large, small, or a mixture of both. After the exposure period, females fed small MP had higher liver masses compared to control females (on average ± SD, 8.95 ± 2.3 g vs. 6.34 ± 1.0 g), while liver lipid content did not differ in either males or females. The levels of monounsaturated fatty acids were lower in females exposed to large MP and the mixture of both MP sizes compared to controls. Females exposed to MP also had different levels of oleic- (18:1) and palmitoleic (16:1) acid compared to controls dependent on MP size. Exposure to small MP increased levels of palmitic- (16:0) and palmitoleic (16:1) acid in both males and females suggesting a possible increase in de novo fatty acid synthesis. Taken together, our results point towards a sex-specific sensitivity to MP as well as size-dependent MP effects on lipid metabolism in birds. Disruption of fatty acid composition could affect important life stages in female birds, such as migration and egg-laying. We stress the importance of further research focused on determining the mechanisms of action of MP on lipid metabolism.

Plastic pollution can pose a threat to birds. Yet, little is known about the sublethal effects of ingested microplastics (MP), and the effects of MP < 1 mm in birds remain unknown. This study therefore aimed at evaluating the toxicity of environmentally relevant polypropylene and polyethylene particles collected in the Norwegian coast in growing Japanese quail (Coturnix japonica). Birds were orally exposed to 600 mg MP over 5 weeks, covering small (<125 µm) and large (3 mm) MP, both separately and in a mixture. We evaluated multiple sublethal endpoints in quail, including oxidative stress, cytokine levels, blood-biochemical parameters, and reproductive hormones in blood, as well as body mass. Exposure to small MP significantly induced the activities of the antioxidant enzymes catalase, glutathione-S-transferase, and glutathione peroxidase. Exposure to large MP increased the levels of aspartate aminotransferase (liver parameter) and decreased 17ß-estradiol levels in females. Body mass was not directly affected by MP ingestion; however, quail exposed to small MP and a mixture of large and small MP had a different growth rate compared to control quail. Our study used similar levels of MP as ingested by wild birds and demonstrated size-dependent effects of MP that can result in sublethal effects in avifauna.

The occurrence of organohalogenated compounds (OHCs) in wildlife has received considerable attention over the last decades. Among the matrices used for OHCs biomonitoring, feathers are particularly useful as they can be collected in a minimally or non-invasive manner. In this study, concentrations of various legacy OHCs -polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs) and polybrominated diphenyl ethers (PBDEs)-, as well as emerging OHCs -per- and polyfluoroalkyl substances (PFAS) and organophosphate ester flame retardants (OPEs)- were determined in feathers of 72 Eurasian eagle-owls (Bubo bubo) from Norway, with the goal of studying spatiotemporal variation using a non-invasive approach. Molted feathers were collected at nest sites from northern, central and southern Norway across four summers (2013-2016). Additionally, two museum-archived feathers from 1979 to 1989 were included. Stable carbon (δ13C) and nitrogen isotopes (δ15N) were used as dietary proxies. In total, 11 PFAS (sum range 8.25-215.90 ng g-1), 15 PCBs (4.19-430.01 ng g-1), 6 OCPs (1.48-220.94 ng g-1), 5 PBDEs (0.21-5.32 ng g-1) and 3 OPEs (4.49-222.21 ng g-1) were quantified. While we observed large variation in the values of both stable isotopes, suggesting a diverse diet of the eagle-owls, only δ13C seemed to explain variation in PFAS concentrations. Geographic area and year were influential factors for δ15N and δ13C. Considerable spatial variation was observed in PFAS levels, with the southern area showing higher levels compared to northern and central Norway. For the rest of OHCs, we observed between-year variations; sum concentrations of PCBs, OCPs, PBDEs and OPEs reached a maximum in 2015 and 2016. Concentrations from 1979 to 1989 were within the ranges observed between 2013 and 2016. Overall, our data indicate high levels of legacy and emerging OHCs in a top predator in Norway, further highlighting the risk posed by OHCs to wildlife.

Lead contamination is a widely recognised conservation problem for raptors worldwide. There are a number of studies in individual raptor species but those data have not been systematically evaluated to understand raptor-wide lead exposure and effects at a pan-European scale. To critically assess the extent of this problem, we performed a systematic review compiling all published data on lead in raptors (1983-2019) and, through a meta-analysis, determined if there was evidence for differences in exposure across feeding traits, geographical regions, between hunting and non-hunting periods, and changes over time. We also reviewed the impact of lead on raptors and the likely main source of exposure. We examined 114 studies that were unevenly distributed in terms of time of publication and the countries in which studies were performed. Peer-reviewed articles reported data for 39 raptor species but very few species were widely monitored across Europe. Obligate (vultures) and facultative scavengers (golden eagle, common buzzard and white-tailed sea eagle) accumulated the highest lead concentrations in tissues and generally were the species most at risk of lead poisoning. We found no evidence of a spatial or decadal trend in lead residues, but we demonstrated that high lead blood levels relate to hunting season. Exposure at levels associated with both subclinical and lethal effects is common and lead from rifle bullets and shot is often the likely source of exposure. Overall, our review illustrates the high incidence and ubiquity of lead contamination in raptors in Europe. However, we did not find studies that related exposure to quantitative impacts on European raptor populations nor detailed studies on the impact of mitigation measures. Such information is urgently needed and requires a more harmonised approach to quantifying lead contamination and effects in raptors across Europe.

Persistent organic pollutants (POPs) are still globally distributed and some have been shown to interact with the endocrine system of birds. However, the relationship between POPs and the stress response mediated by the hypothalamic-pituitary-adrenal (HPA) axis is still poorly understood. Raising concerns are now focused on the toxic properties of emergent organophosphate ester flame retardants (OPEs), but whether OPEs interact with the HPA axis response has not yet been investigated. We measured corticosterone concentrations in feathers (CORTf) as a long-term biomarker of the bird HPA axis response and we investigated their relationship with POP and OPE concentrations in down feathers of nestling cinereous vultures (Aegypius monachus). We also examined whether high contaminant burden and high CORTf concentrations impacted the duration of chick development. The most predominant compounds were the following: p,p'-DDE (3.28 ±â€¯0.26 ng g-1 dw) > γ-HCH (0.78 ±â€¯0.09 ng g-1 dw) > BDE-99 (0.73 ±â€¯0.09 ng g-1 dw) > CB-153 (0.67 ±â€¯0.04 ng g-1 dw). The most persistent POP compounds (CB-170, -177, -180, -183, -187, -194 and p,p'-DDE) were associated (P = 0.02) with high concentrations of CORTf (range: 0.55-6.09 pg mm-1), while no relationship was found when OPEs were tested (P > 0.05). Later egg-laying was positively associated to high levels of CORTf (P = 0.02) and reduced duration of chick development (P < 0.001), suggesting a beneficial effect of the HPA axis response on the growth of the chicks. In addition, males with high concentrations of the most persistent POP compounds tended to show a reduced duration of the nestling period (P = 0.05) and an equal fledging success than chicks with lower levels. These findings suggest that POPs, but not OPEs, may interact with the HPA axis response of chicks, although levels were not high enough to cause detrimental consequences.

Life history theory states that individual fitness in one stage of life is conditioned by what occurred in previous stages. In migratory species, reproductive effort during breeding has often been found to influence body condition, molt schedule, self-provisioning and migration of individuals in subsequent seasons (i.e., carryover effects of breeding). However, there is a current uncertainty in understanding how long-distance migrants trade off among such energy-demanding activities (i.e., breeding, molting and migrating). To provide evidence to the field, we experimentally reduced the parental effort of a long-lived Procellariform, the Cory's shearwater (Calonectris borealis), by inducing failure at the incubation stage. Treatment and control birds were tracked during their subsequent migration by means of light-level and immersion loggers and sampled for six specific feathers (molted at different periods along the migratory cycle) upon the recovery of the loggers 1 yr later. Feathers were used to perform stable isotope analysis (SIA) and determine corticosterone levels (CORT). By these means, we evaluated the effect of breeding effort on migratory strategy, at-sea activity patterns, molt patterns, and levels of stress experienced by birds along the non-breeding period. We did not detect any difference between birds in the induced failure group and successful breeders in terms of spatio-temporal distribution: all birds shared common foraging areas throughout the study period and the timing of major phenological events did not differ. Failed birds significantly advanced their molt, as revealed by SIA and flying activity patterns. The stress levels of failed birds, inferred through CORT concentrations in feathers, were found to be consistently lower than in successful breeders, through the end of the breeding to the non-breeding period. Thus, we provide robust evidence that the costs of reproduction can be physiologically mediated from the breeding to the non-breeding period through molting schedules and CORT levels. However, we failed to detect clear effects on migratory behavior or subsequent breeding success, suggesting that costs of breeding in long-lived species may be rapidly buffered during the post-breeding period, as would be expected from life history theory.

Persistent organic pollutants (POPs) have the potential to impair the endocrine regulation of organisms and alter their ability to respond to environmental changes. We studied whether polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) affected the endocrine regulation of free-living and captive red kites (Milvus milvus) through studying the dynamics of corticosterone (CORT) and dehydroepiandrosterone (DHEA). We sampled migratory free-living kites coming from northern Europe and captive kites born in a rehabilitation center in Spain. We used body feathers from the interscapular region as a minimally-invasive and integrative matrix. The most abundant compound detected in free-living kites was 4,4'-dichlorodiphenyldichloroethylene (4,4'-DDE; 6.10 ±â€¯1.56 ng g-1 dw feather) followed by CB-153 (3.10 ±â€¯0.63 ng g-1 dw feather) and CB-180 (2.43 ±â€¯1.08 ng g-1 dw feather). In captive kites, the most abundant compounds were 4,4'-dichlorodyphenyltrichloroethane (4,4'-DDT; 2.38 ±â€¯1.30 ng g-1 dw feather), CB-153 (2.15 ±â€¯0.47 ng g-1 dw feather) and hexachlorobenzene (HCB; 2.03 ±â€¯0.45 ng g-1 dw feather) at similar concentrations. Free-living kites showed higher levels of 4,4'-DDE and CB-180 in comparison to captive kites. Age influenced HCB and CB-101 levels, whereas body mass was inversely related to CB-180 and 4,4'-DDT. Interestingly, captive kites showed a ratio DDT/DDE higher than 1 suggesting a relatively recent exposure of DDT, in contrast to free-living kites. Regarding hormonal levels, free-living kites showed higher levels of CORT (3.30 ±â€¯0.22 pg mm-1 feather) than captive (2.40 ±â€¯0.16 pg mm-1 feather), reflecting higher allostatic load. In addition, a positive association between PCBs and DDTs and adrenal hormones was found in free-living kites, suggesting an increase of CORT as a response of the endocrine system to cope with stressors and a subsequent elevation of DHEA to ameliorate the potential negative effects that high CORT levels could cause to the organism.

Raptor feathers have been increasingly used to assess pollutants in ecotoxicological monitoring studies. However, the suitability of down feathers to detect pollutants has not yet been investigated. In this study, concentrations of persistent organic pollutants (POPs) and organophosphate ester flame retardants (OPEs) were assessed in down and juvenile contour feathers of Spanish cinereous vulture (Aegypius monachus) nestlings (circa 73 days old) and contaminant concentrations were compared between both types of feathers from the same individuals. Concentrations of polychlorinated biphenyls (PCBs: 1.30-6.16 ng g-1 dw feather), polybrominated diphenyl ethers (PBDEs: 0.23-1.35 ng g-1 dw feather), p,p'-dichlorodiphenyldichloroethylene (pp-DDE: 0.09-6.10 ng g-1 dw feather) and tris (1-chloro-2-propyl) phosphate (TCiPP: 0.86-48.96 ng g-1 dw feather) were significantly higher in down than in contour feathers. In contrast, contour feathers showed higher levels of the more volatile POP, lindane (0.25-3.12 ng g-1 dw feather). Concentrations of hexachlorobenzene (HCB) and OPEs (except TCiPP) were similar between the two types of feathers. By showing high accumulation of the most persistent POPs investigated, down feathers presented a contamination profile similar to that previously described in raptor eggs. As these feathers grow during the first days of a vulture chicks life, they probably reflect the contaminant burden of the chick due to maternal transfer to the egg. Overall, the present study provides the first indication that down feathers may be useful for biomonitoring studies. Further research is needed to confirm whether nestling down feathers reflect the concentrations in the egg.

Concentrations of different steroid hormones have been used in cows as a measure of adrenal or gonadal activity and, thus, as indicators of stress or reproductive state. Detecting cortisol and progesterone in cow hair provides a long-term integrative value of retrospective adrenal or gonadal/placental activity, respectively. Current techniques for steroid detection require a hormone-extraction procedure that involves time, several types of equipment, management of reagents, and some assay procedures (which can also be time-consuming and can destroy the samples). In contrast, near-infrared reflectance spectroscopy (NIRS) is a multi-component predictor technique, characterized as rapid, nondestructive for the sample, and reagent-free. However, as a predictor technique, NIRS needs to be calibrated and validated for each matrix, hormone, and species. The main objective of this study was to evaluate the predictive value of the NIRS technique for hair cortisol and progesterone quantification in cows by using specific enzyme immunoassay as a reference method. Hair samples from 52 adult Friesian lactating cows from a commercial dairy farm were used. Reflectance spectra of hair samples were determined with a NIR reflectance spectrophotometer before and after trimming them. Although similar results were obtained, a slightly better relationship between the reference data and NIRS predicted values was found using trimmed samples. Near infrared reflectance spectroscopy demonstrated its ability to predict cortisol and progesterone concentrations with certain accuracy (R2 = 0.90 for cortisol and R2 = 0.87 for progesterone). Although NIRS is far from being a complete alternative to current methodologies, the proposed equations can offer screening capability. Considering the advantages of both fields, our results open the possibility for future work on the combination of hair steroid measurement and NIRS methodology.

OBJECTIVES: To develop and validate an instrument to measure health-related quality of life (HRQOL) specific to patients with allergic rhinitis (AR) and primarily for use in Spanish and Spanish-speaking populations. METHODS: An initial item pool was generated from literature review, focus groups with AR patients, and consultations with clinical experts. Item reduction was performed using clinimetric and psychometric approaches after administration of the item pool to 400 AR patients. The resulting instrument's internal consistency, test-retest (2-4 weeks) reliability, known groups and convergent validity, and sensitivity to change were tested in a longitudinal, observational, multicenter study in 210 AR patients who also completed the Rhinoconjunctivitis Quality of Life Questionnaire (RQLQ). RESULTS: The new questionnaire took a mean (SD) of 7.1 (5.4) minutes to answer. Floor and ceiling effects were less than 15% on all dimensions. Cronbach's alpha values and intraclass correlation coefficient values for six of the sevendimensions and the overall score exceeded 0.70. Statistically significant differences (P < 0.01) were observed on all ESPRINT-28 dimensions and the overall score between patients with mild (mean overall score 1.97, SD 0.99), moderate (mean overall score 2.78, SD 0.88), and severe AR (mean overall score 3.89, SD 0.87). Patients with persistent AR had worse scores (P < 0.05) on all dimensions than patients with intermittent AR. Correlations between the ESPRINT-28 and the RQLQ were generally as expected. Effect sizes for score changes between the two study visits ranged from 0.96 to 1.76 for individual dimensions and the overall score. CONCLUSIONS: This new, Spanish-developed instrument to measure HRQOL in AR patients has shown good reliability, validity, and sensitivity to change. It has also proved easy to use and administer.

BACKGROUND: Allergic Rhinitis and its Impact on Asthma (ARIA) differentiates mild from moderate/severe patients on the basis of 4 severity items. The high prevalence of moderate/severe patients suggests the need to differentiate between them. OBJECTIVES: To identify the categorization that maximizes discrimination between moderate and severe allergic rhinitis (AR) by using ARIA guidelines. METHODS: Observational, cross-sectional study. Clinical characteristics, nasal symptoms (Total Symptom Score 4), and health-related quality of life (HRQL; Rhinoconjunctivitis Quality of Life Questionnaire and Short Form 12) were assessed. The association of severity items (sleep, daily activities/sport, work/school, and troublesome symptoms) with symptoms and HRQL was analyzed using linear regression models. ANOVA and effect sizes were used to assess differences in symptoms and HRQL among groups defined by the number of affected ARIA items. RESULTS: Nontreated patients (N = 141) with moderate/severe AR were studied. All severity items showed a similar independent association with symptoms and HRQL scores, and there were no interaction effects, indicating that categorization of patients into moderate and severe could be based only on the number of items affected. Effect sizes were highest between patients with 4 affected ARIA items and those with 3, 2, or 1 affected item (effect sizes greater than 0.8 in all comparisons using Rhinoconjunctivitis Quality of Life Questionnaire and Short Form 12 Physical Composite Summary, and greater than 0.5 using the Total Symptom Score 4; P < .001). CONCLUSION: Using ARIA severity items, the criterion that best discriminates AR severity is considering moderate those with 1 to 3 affected items and severe those with 4. CLINICAL IMPLICATIONS: Discrimination between patients with moderate and severe AR should help to obtain homogeneous populations for both research and clinical purposes.
