BACKGROUND: In recent decades, consumption of simple sugars has increased dramatically, which contributes to health problems including insulin resistance and obesity. In this study, we investigated the effects of high concentrations of white sugar (WS) and brown sugar (BS) on serum concentration of brain-derived neurotrophic factor (BDNF), insulin resistance, and body weight in albino rats. METHODS: Twenty-four male Wistar rats were randomly divided into three groups: control, a group treated with 15% WS, and a group treated with 15% BS. Rats were given WS and BS by gavage (daily) for 42 days. At the end of the intervention period, serum level of BDNF, insulin resistance, and body weight were measured. RESULTS: Body weight and insulin resistance were significantly increased and serum BDNF level was decreased in both WS and BS groups compared to the control group (P<0.05). In the WS-treated rats, the amount of changes in the insulin resistance, body weight, and serum BDNF level was greater compared to that in BS-treated (P< 0.05). CONCLUSION: Due to the adverse effects of consuming high levels of WS and BS on serum level of BDNF, insulin resistance, and body weight, high intakes of these sweeteners are not recommended.

BACKGROUND AND PURPOSE: Ample evidence indicates that chronic adolescence stress is associated with an increased risk of developing neuropsychiatric disorders in adulthood. Given the importance of the effective therapeutic ways to overcome adolescent stress-related deficits, the present study investigated the effects of Spirulina platensis (SP), environmental enrichment (EE), and voluntary exercise (EX) and their combination on anxiety or depression-like behaviors, oxidative stress, and alterations of BDNF and 5HT-3 receptors in the prefrontal cortex (PFC) induced by adolescent stress in adult female rats. METHODS: During the adolescent period (PNDs30-40), rats were subjected to restraint stress. Then, the animals were subjected to SP treatment (200 mg/kg/day), EX, EE, and the combined treatments (SP+EX, and SP+EE) for 15 days between PNDs41-55. Subsequently, anxiety or depression-like behaviors, BDNF levels, oxidative stress markers and mRNA expression of BDNF and 5HT3 in the PFC were assessed. RESULTS: Stressed rats demonstrated enhanced anxiety levels and depression-like behaviors in adulthood. Regarding the oxidative stress markers, stressed rats exhibited significantly higher levels of malondialdehyde, a lipid peroxidation product, higher activities of antioxidant enzymes (glutathione peroxidase and superoxide dismutase) and significantly lower total antioxidant reactivity capacity in the PFC. Additionally, adolescent stress significantly increased 5HT3 receptor mRNA expression and decreased BDNF content and its mRNA expression in the PFC. Treatments with SP, EX, EE, and the combined interventions alleviated these deficits. CONCLUSION: Our findings indicate that appropriate interventions during the adolescent period can protect against adolescent stress-induced behavioral, and biochemical defects and oxidative stress damage in adulthood.

BACKGROUND AND PURPOSE: Type 2 diabetes mellitus (T2DM) is characterized by insulin resistance and abnormalities in insulin production. Apelin is associated with insulin resistance. According to the anti-diabetic properties of curcumin, the purpose of this study was to compare the effects of curcumin and nano-curcumin intake on insulin resistance and serum levels of fasting blood sugar (FBS), Apelin, and lipid profile (cholesterol, triglyceride, LDL, HDL and VLDL) in T2DM rats. MATERIALS AND METHODS: Forty-eight male Wistar rats were divided into six groups: Control, diabetic, diabetic treated with two doses of curcumin (100 and 200 mg/kg) and diabetic treated with two doses of nano-curcumin (100 and 200 mg/kg). Induction of T2DM was performed by intraperitoneal injection of Nicotinamide (110 mg/kg) and Streptozotocin (45 mg/kg) in the fasting state. Rats received different doses of nano-curcumin and curcumin by gavage (daily) for 28 days. At the end of the intervention period, insulin resistance and serum levels of FBS, apelin and lipid profiles were measured. RESULTS: Insulin resistance and serum levels of FBS, Apelin, cholesterol, triglycerides, LDL, and VLDL were significantly decreased in diabetic rats treated with curcumin and nano-curcumin (p<0.05) so that nano-curcumin in reducing lipid profile is more effective than curcumin (P<0.05). Serum level of HDL in nano-curcumin groups was significantly higher than diabetic and curcumin groups (p<0.05). Also, with increasing insulin resistance, serum level of apelin increased (P<0.05). CONCLUSION: The therapeutic effects of curcumin and nano-curcumin were effective in decreasing insulin resistance, serum levels of FBS, apelin and lipid profile. The dose of 100 mg/kg nano-curcumin was more effective in reducing lipid profile.

OBJECTIVE: The amygdala structural and functional abnormalities have been implicated in numerous neuropsychiatric and neurodevelopmental disorders. Given the important role of the amygdala in stress responses and the susceptibility of the females to adolescent stress, the present study investigated the beneficial effects of Spirulina platensis microalgae (SP) as a neuroprotective supplement against adolescent stress-induced oxidative stress, brain-derived neurotrophic factor (BDNF) alterations, molecular and morphological remodeling in the basolateral amygdala (BLA) of adult female rats. METHODS: During the adolescent period (PNDs 30-40) rats were subjected to restraint stress (2 h/day for 10 days). Then, the animals were subjected to 15 days treatment (PNDs 41-55) with SP (200 mg/kg/day) followed by biochemical (BDNF and stress oxidative markers), molecular (BDNF and its receptor tropomyosin receptor kinase B [TrkB] mRNA expression), and morphological (dendritic length and spines) assessments in the BLA. RESULTS: The study revealed that adolescent stress decreased BDNF levels and reduced apical dendritic length and branch points of pyramidal neurons in the BLA. In addition, chronic stress significantly increased oxidative stress parameters and decreased BDNF and TrkB mRNA expression in the BLA. Treatment with SP alleviated both biochemical, molecular, and neuroanatomical deficits that induced by adolescent stress. CONCLUSION: Our findings provide important evidence that SP as a non-pharmacological intervention during adolescent period can protect against chronic stress-induced neuroanatomical biochemical, and molecular deficits in adulthood, and thus, reduce stress-related disorders.

BACKGROUND AND PURPOSE: Natural compounds are used for prevention of inflammation. Curcumin has antioxidant and anti-inflammatory properties, and loading it into nano-phytosomes may improve its efficiency. The present study investigates the effects of curcumin and its nano-phytosome on behavioral and biochemical responses in carrageenan-induced inflammation in the mice model. METHODS: The mice were divided into six groups and received oral administration of curcumin or its nano-phytosome at a dose of 15 mg/kg for seven days before the administration of carrageenan. Acute inflammation in the mice was induced by administration of carrageenan (1%) into the subplantar region of the left paw. Antioxidant activity and behavioral responses were then evaluated. RESULTS: The results showed that the serum concentrations of antioxidant enzymes were significantly higher in the sal+sal group compared to the cara+sal group (P<0.05). Using nanophytosome, separately and in combination with indomethacin, increased the levels of antioxidant enzymes compared to the cara+sal group (P<0.05). Latency was significantly lower in the cara+sal group compared to the cara+sal group (P<0.05), but it was considerably higher in other groups, especially in the cara+nano.ph.cur+indo group (P<0.05). CONCLUSION: It can be stated that the nano-phytosome of curcumin could improve antioxidant and behavioral responses in inflamed mice.

INTRODUCTION: of the study: Post-training administration of glucocorticoids enhance memory consolidation of inhibitory avoidance learning. Given the involvement of 5-HT6 receptors in memory processing and the interaction of glucocorticoids with the brain serotonergic system in modulating memory processing, we investigated whether the effect of glucocorticoids on the consolidation of emotionally arousing training depends on hippocampal 5-HT6 receptors. METHODS: Rats were trained in an inhibitory avoidance task and immediately received the systemic injections of corticosterone (CORT) as well as the intra-hippocampal injections of 5-HT receptors agonist or antagonist. The memory retention test was done 48 hours after training and immediately after the behavioral test, the animals were sacrificed and the hippocampi (left and right) rapidly dissected out for molecular studies. RESULTS: Post-training injections of different doses of CORT (1.25, 2.5, 5, and 10 mg/kg) enhanced memory retention in a dose-dependent manner. The CORT-induced enhancement of memory consolidation was blocked by bilateral intra-hippocampal injections of 5-HT6 receptor antagonist SB271046 (5 or 10 ng/per side), but not agonist EMD386088 (5 or 10 ng/per side). Furthermore, systemic CORT reduced 5-HT6 receptor mRNA and protein expression in the hippocampus. Both doses of 5-HT6 receptor agonist and antagonist significantly enhanced and reduced the expression of the 5-HT6 receptor, respectively, and both ligands at the higher dose (10 ng) enhanced memory consolidation. Moreover, CORT injection attenuated and enhanced, respectively, the effects of agonist and antagonist on 5-HT6 receptor expression. CONCLUSION: These behavioral and molecular findings indicated an interaction between glucocorticoids and hippocampal 5-HT6 receptors in the consolidation of emotionally arousing experiences.

BACKGROUND: Using chemical agents in the treatment of diabetes mellitus type 2 may have some limitations due to frequent side effects. Some novel and natural agents may be promising alternatives in this case. This study was designed to evaluate the effects of oral Japanese sake yeast supplement, as a novel agent, on biochemical antioxidant and anti-inflammatory parameters in experimentally induced diabetic rats. MATERIALS AND METHODS: After inducing diabetes (55 mg/kg intraperitoneal injection of streptozotocin), 120 male adult Wistar rats were randomly divided into 5 groups and each group received 0 (control), 15, 30, or 45 mg/kg of sake yeast or was considered a nondiabetic control. Then, the serum levels of tumor necrosis factor-α, IL-6, C-reactive protein, malondialdehyde, glutathione, total antioxidant status, glucose, cholesterol, triglycerides, and insulin were evaluated and compared to baseline measures. RESULTS: The results showed that oral administration of sake yeast at different concentrations reduced levels of malondialdehyde, glucose, cholesterol, and triglycerides and increased levels of insulin, glutathione, and total antioxidants (P<0.05). The best responses were observed in the nondiabetic control group. CONCLUSION: Sake yeast supplement may be useful as a novel agent in the treatment of diabetes.

Objective: Due to the prevalence of stress in modern life and its impact on spatial memory, the role of inhibitory systems in brain areas such as the nucleus accumbens (NAc) in reducing stress is important. The current study aimed to examine the response of NAc shell GABAB receptors to stress and the role of intraperitoneally (i.p.) and intra-NAc injection of the GABAB receptor agonist baclofen on spatial memory impairments in stress-exposed rats. Methods: Eighty adult male Wistar rats were randomly divided into ten groups (n=8): two were control groups for intra-NAc and i.p baclofen; two groups were subjected to stress and injected with saline (baclofen vehicle); three groups were given baclofen (1, 5, and 10 µg/rat) intra-NAc 5 mins before stress was induced; and three groups received baclofen (1, 5, and 10 mg/kg/i.p.) 30 mins before being subjected to stress. Foot-shock stress was applied for 7 consecutive days. Behavioral assays using the Barnes maze were performed 24 hrs after the last baclofen injection. Results: Both the intra-NAc and the i.p administration of baclofen dose-dependently reduced escape latency and total distance and increased velocity in the treatment groups in the training trials. In the probe test, the rats that had received 5 mg/kg of baclofen had the highest target frequency, but there no significant differences were observed in velocity, duration, or distance to the target between the groups. Conclusion: According to the findings, baclofen can dose-dependently improve spatial memory, and GABAB receptor in the NAc plays an important role in spatial memory.

Metformin is a widely used drug in today's prescriptions by physicians due to its specific effects in treating and curing type II diabetes. Diabetes is a common disease that may occur throughout human life, and can increase the likelihood of the occurrence of various types of cancer, such as colon, rectum, pancreas and liver cancers, compared to non-diabetic patients. Metformin inhibits mTOR activity by activating ATM (ataxia telangiectasia mutated) and LKB1 (liver kinase B1) and then adenosine monophosphate-activated kinase (AMPK), and thus prevents protein synthesis and cell growth. Metformin can activate p53 by activating AMPK and thereby ultimately stop the cell cycle. Given the potential of metformin in the treatment of cancer, it can be used in radiotherapy, chemotherapy and to improve the response to treatment in androgen derivatives (ADT), and also, according to available evidence, metformin can also be used to prevent various types of cancers. Generally, metformin can: 1) reduce the incidence of cancers, 2) reduce the mortality from cancers, 3) increase the response to treatment in cancer cells when using radiotherapy and chemotherapy, 4) optimize tumor movement and reduce the malignancy, 5) reduce the likelihood of relapse, and 6) reduce the damaging effects of ADT. Therefore, this drug can be used as a complementary therapeutic agent for cancer treatment and prevention. In this review, we have summarized the data from various experimental and clinical studies and highlight the possible potential effects of metformin on cancer therapeutic responses.

The antidepressant-like effects of zinc (Zn) have been documented in some animal models of depression. In addition, antidepressants may reduce the abuse potential of opioids by affecting their rewarding effect. Hence, this study was performed to investigate the effect of Zn on the expression of morphine-induced conditioned place preference (CPP) in male rats. We used an unbiased CPP paradigm for investigating the effect of Zn. The intraperitoneal (i.p.) and intracerebroventricular (i.c.v.) administrations of Zn (5-20 mg/kg, i.p., and 10 nmol/rat, respectively) with or without morphine did not induce conditioned place aversion (CPA) or CPP during acquisition phase. However, the same i.p. and i.c.v. administrations of Zn induced morphine-like CPP in the expression phase. Pre-treatment with dopamine receptor antagonists (SCH23390, sulpiride, and haloperidol) and serotonin receptor antagonists (WAY100135, ketanserin, and ondansetron) reversed the enhancement effect of Zn on the expression of morphine-induced CPP (especially 20mg/kg, i.p. and 10 nmol/rat, i.c.v.). These findings suggest that acute i.p. and i.c.v administration of Zn might enhance the rewarding properties of morphine through involvement with dopaminergic and serotonergic neuronal systems.

Objective The aim of this study is to evaluate the laparoscopic method in gastric cancer staging before surgery in patients undergoing Neo-adjuvant chemotherapy. Methods This was a cross-sectional study on patients with T3-T4 and node positive non-metastatic gastric adenocarcinoma who was candidates for neo-adjuvant chemotherapy. The patients were excluded from the study when they were not in the condition to continue the study. All eligible patients underwent metastases work up by abdominal, pelvic and chest CT scans and then received neo-adjuvant chemotherapy. Pre-operative laparoscopy was performed before definitive surgery. The collected data was analyzed using SPSS software. The statistical significance, sensitivity, specificity and predictive values were calculated from this data. Results In this study, out of 50 patients, 26 (52%) were male. The mean age of the patients was 62.44 ± 12.136 years. Most tumors were located in one-third distal of stomach (21 patients, 42%). The accuracy of the laparoscopic method, to determine the degree of involvement of the lymph nodes, revealed that the method had a sensitivity of 84%, a specificity of 47.62%, a positive likelihood ratio (PLR) of 1.6, a negative likelihood ratio (NLR) of 0.33%, a positive predictive value of 65.62% and a negative predictive value of 71.43%. The accuracy of the laparoscopic method, regarding determining the presence or absence of metastasis, revealed that the method had a sensitivity, specificity, positive predictive value, and a negative predictive value of 100%. Conclusion Based on the evidence of this study, and other studies in this field, it seems that the use of a step-by-step combination methods in gastric cancer staging would be logical. Alone, no singular method would be sufficient in providing the physician with sufficient information to successfully stage the tumor and thus determine the appropriate treatment. Therefore, the combination of methods should be used which, in view of the economic constraints of health systems, would be recommended for high-risk patients for metastasis (N+/ T3-T4), diagnostic pre-operative laparoscopy.

Background It is well documented that biologically active components of microalgae can be utilized for treatment of different diseases. This study was conducted to evaluate the antihyperglycemic and antihyperlipidemic activities and weight control of Nannochloropsis oculata microalgae (NOM) in Streptozotocin-induced diabetic male rats. Methods Diabetes was induced by intraperitoneal administration of Streptozotocin (55 mg/kg). Healthy and diabetic rats were divided in to six groups. Healthy and diabetic rats orally received distilled water or NOM (10 and 20 mg/kg) for three weeks. Results Oral administration of NOM to diabetic rats significantly reduced the serum concentrations of glucose, cholesterol, triglycerides, LDL and increased the serum concentration of insulin and HDL-C (P<0.05). Treatment with NOM had no significant effect on blood parameters in healthy rats (P>0.05). Also, NOM maintained body weight in diabetic rats (P<0.05). Conclusion It can be concluded that NOM has antihyperglycemic and antihyperlipidemic activities in diabetic rats.

Chronic exposure to stress during adolescent period has been demonstrated to impair cognitive functions and the dendritic morphology of pyramidal neurons in the rat hippocampal CA3 area. The present study investigated the combined protective effects of Spirulina platensis (SP), a supplement made from blue-green algae with neuroprotective properties, voluntary exercise (EX) and environmental enrichment (EE) against cognitive deficits, alternations in hippocampal BDNF levels, and abnormal neuronal remodeling in adult female rats (PND 60) induced by exposure to chronic restraint stress during adolescent period (PND 30-40). Rats were exposed to restraint stress (2 h/day for 10 days, PND 30-40). Then, the animals were subjected to treatment with SP (200 mg/kg/day), EX, EE and the combined treatments (SP + EX, and SP + EE) between PND 41 and 55 of age. Following the interventions, spatial learning and memory, passive avoidance performance, hippocampal dendritic morphology and BDNF levels were assessed. Results showed that plasma corticosterone levels increased at PND 40 and remained elevated at PND 55 and 70 in the stressed rats. Stressed rats showed deficits in spatial learning and memory and passive avoidance performance, decreased BDNF levels in the hippocampus, and reduced apical dendritic length and branch points of the CA3 pyramidal neurons. These deficits were alleviated by the SP, EX and EE, and the combined treatments, which accompanied with a decline in serum corticosterone in stressed animals. Some treatments even enhanced cognitive functions, and BDNF levels and neuroanatomical remodeling in the hippocampus of non-stressed animals. Our findings provide important evidences that physical activity, exposure to EE, and the SP treatment during adolescent period can protect against adolescent stress induced behavioral, biochemical and neuroanatomical impairments in adulthood.

Previous studies have shown that the neuropeptide bombesin (BBS) enhances consolidation of specifically for inhibitory avoidance memory in adult rats. However, its effect on memory consolidation during premature period is not clear as well. Thus, this study evaluated the effect of BBS and its interaction with glutamatergic and dopaminergic systems on memory consolidation in rat pups. Male and female rat pups (30 days old) were trained in an inhibitory avoidance (IA) task (0.5 mA, 3 s footshock). Memory retention was tested 24 h later during which the latency to re-enter to the shock compartment was recorded. First, the effects of different doses (0.001, 0.0025, 0.005, 0.01 and 0.02 mg/kg) of BBS injected immediately following training were tested. Then, the effect of the most effective dose of BBS obtained in the previous experiment was examined in the presence of the glutamate NMDA receptor antagonist MK-801 (0.05 mg/kg), the dopamine D1 receptor antagonist SCH-23390 (0.05 mg/kg) and the dopamine D2 receptor antagonist sulpiride (20 mg/kg). Findings indicate that BBS significantly enhances memory consolidation at all tested doses in male pups and at a dose of 0.01 mg/kg in female pups. MK-801, SCH-23390 and sulpiride administration before BBS injection in individual groups significantly blocked BBS-induced memory enhancement. Our findings indicate that similar to adult rats, BBS enhances memory consolidation in developing rat. This enhancing effect is mediated, at least in part, via an interaction with glutamatergic and dopaminergic systems.

Objectives This study was conducted to evaluate the effects of oral supplementation of Spirogyra algae on oxidative damages and inflammatory responses in streptozotocin (STZ)-induced diabetic rats. Methods Diabetes was induced by administration of 55 mg/kg of streptozotocin. A total of sixty-four rats were divided into eight groups of eight rats each as follows:1) non-diabetic control; 2, 3, and 4) non-diabetic rats treated with 15, 30, and 45 mg of Spirogyra algae/kg/d; 5) control diabetic; and 6, 7, and 8) diabetic rats treated with 15, 30, and 45 mg of Spirogyra algae extract. At the end of the trial, the serum concentrations of glucose, interleukin-6 (IL-6), tumor necrosis factor-a (TNF-a), malondialdehyde (MDA), glutathione (GSH), total antioxidant status (TAS), C-reactive protein (CRP), insulin, triglycerides, and cholesterol were examined by specified procedures. Results Our findings indicated that the administration of STZ significantly increased the serum concentrations of glucose, triglycerides, cholesterol, CRP, IL-6, TNF-a, and MDA and decreased the serum levels of GSH and TAS (P<0.05) in diabetic rats. Oral administration of Spirogyra alleviated adverse effects of diabetes on oxidative stress and inflammatory factors in diabetic rats (P<0.05). Conclusion It can be stated that Spirogyra algae extract can be used for treatment of diabetes likely due to prevention of oxidative stress and alleviation of inflammation in the rat model.

BACKGROUND AND OBJECTIVE: Initial peripheral/central nerve injuries, such as chronic constriction injury (CCI)/spinal cord injury, are often compounded by secondary mechanisms, including inflammation and oxidative stress, which may lead to chronic neuropathic pain characterized by hyperalgesia or allodynia. On the other hand, exercise as a behavioral and non-pharmacological treatment has been shown to alleviate chronic neuropathic pain. Therefore, this study was conducted to examine whether or not exercise reduces neuropathic pain through modifying oxidative stress and inflammation in chronic constriction injury of the sciatic nerve. MATERIALS AND METHODS: Wistar male rats weighing 200±20 g were randomly divided into five groups (normal, sham, CCI, pre-CCI exercise, and post-CCI exercise group). Sciatic nerve of anesthetized rats was loosely ligated to induce CCI, and they were then housed in separate cages. The rats ran on treadmill at a moderate speed for 3 weeks. Mechanical allodynia and thermal hyperalgesia were determined using von Frey filament and plantar test, respectively. Tumor necrosis factor-alpha (TNF-α) assayed in the cerebrospinal fluid, malondialdehyde, and total antioxidant capacity were measured in the serum using Western blot test, thiobarbituric acid, and ferric reducing ability of plasma (FRAP), respectively. RESULTS: The mechanical allodynia (P=0.024) and thermal hyperalgesia (P=0.002) in the CCI group were higher than those in the sham group. Exercise after CCI reduced (P=0.004) mechanical allodynia and thermal hyperalgesia (P=0.025) compared with the CCI group. Moreover, the level of FRAP in the CCI group was (P=0.001) lower than that in the sham group, and post-CCI exercise reversed FRAP amount toward the control level (P=0.019). The amount of malondialdehyde did not differ between groups. Level of TNF-α increased in the CCI group (P=0.0002) compared with sham group and post-CCI exercise could reverse it toward the level of control (P=0.005). CONCLUSION: Post CCI-exercise but not pre CCI-exercise reduces CCI-induced neuropathic pain. One of the possible involved mechanisms is increasing the total antioxidant capacity and reducing the amount of TNF-α.

This study was conducted to investigate the effect of different supplementation levels of Chlorella microalgae on serum metabolites and the plasma content of minerals in laying hens reared under heat stress condition (27.5-36.7 °C, variable). A total number of 378 (40 weeks of age, with mean body weight of 1390 ± 120 g) were randomly allocated to six treatments with seven replicates. The birds were randomly assigned to 6 treatments (C, T1, T2, T3, T4, and T5) with 7 replicate cages of 9 birds. C. microalgae at the rates of 100, 200, 300, 400, and 500 ppm with water were offered to groups T1, T2, T3, T4, and T5, respectively, while group C served as a control. At 71 days of trial, blood samples (14 samples per treatment) were taken for measuring serum metabolites and at 72 days for plasma mineral analysis. The results of this experiment showed that the supplementation of 200-500 ppm C. microalgae decreased the serum content of cholesterol, triglycerides, and LDL (P < 0.05) whereas HDL content increased (P < 0.05) in the hens supplemented with C. microalgae (300 or 400 and 500 ppm). C. microalgae at rates of 300-500 ppm caused a marked (P < 0.05) increase in the plasma content of manganese or iodine and selenium but other minerals were not statistically different among treatments. Overall, from the results of the present experiment, it can be concluded that supplementation of C. microalgae at high rates was beneficial on blood parameters of laying hens reared under heat stress.
