RESUMO

Haemonchus contortus is a parasitic nematode of ruminants. Once inside its host, it is exposed to reactive oxidative species and responds by synthesising antioxidant enzymes as a defence. In Caenorhabditis elegans, antioxidant genes are regulated by the transcription factor skinhead-1 (Cel-SKN-1). However, there is little information about the function of SKN-1 in H. contortus (Hco-SKN-1). Therefore, we performed a molecular investigation to characterise Hco-SKN-1 and its putative relationship with genes encode antioxidant enzymes, namely glutathione S-transferases (Hco-GSTs, n = 3), superoxide dismutase (Hco-SOD) and catalase (Hco-CAT), which are involved in haematophagy and defence against the host. We used in silico sequence analysis of Hco-SKN-1 and Hco-GSTs to design and perform relative expression assays involving H. contortus eggs, infective larvae (L3) and adults. Furthermore, we exposed H. contortus transitional infective larvae (xL3) to erythrocytes or hydrogen peroxide (H2O2) and evaluated the relative expression of antioxidant genes at 24 or 48 h. Gene Ontology (GO) analysis revealed 31 functions associated with Hco-SKN-1 and Hco-GSTs, including stress resistance, larval development and the active immune response. Hco-GST-5957 and Hco-SOD showed the highest expression in adults, indicating a relationship with specific functions at this mature stage. xL3 exposed to erythrocytes or H2O2 showed significant upregulation of Hco-SKN-1, but it occurred after upregulation of the antioxidant genes, indicating that these genes are not regulated by Hco-SKN-1 during the blood-feeding stage. Additional investigation is necessary to understand the putative regulation of antioxidant genes by Hco-SKN-1 during the blood-feeding stage.

INTRODUCTION: Older adults constitute the most vulnerable population group to the COVID-19 pandemic. In Mexico, their biopsychosocial conditions might intensify their vulnerability. METHOD: Affiliation to health systems, health conditions and gerontological evaluation of 3,218 older adults were analyzed following the methodology of the PAHO-Mexico Health, Well-being and Aging Survey. RESULTS: 88.6 % of older adults referred being affiliated to health systems; 30.2 %, 52.4 %, 10.3 %, 4.1 % and 5.6 % referred suffering from diabetes mellitus, high blood pressure, chronic obstructive pulmonary disease, heart disease and cerebrovascular disease, respectively; 15.6 % reported urinary incontinence, and 11.3%, fecal incontinence; 12.1 % of the women referred having suffered from breast cancer at some point, and 6.3 %, cervical cancer. The habit of smoking tobacco was observed in 11.1 %, risk of malnutrition in 32.8 %, established malnutrition in 4.1 %, functional dependence for basic and instrumental activities of daily life in 16.3 % and 17.6 %, respectively. CONCLUSION: Comprehensive gerontological evaluation is essential for efficient care of older adults who suffer from COVID-19, and for adequate care of the effects or health conditions at the conclusion of the confinement imposed by the pandemic.

CONCLUSIÓN: La evaluación gerontológica integral es fundamental para la atención eficiente de los adultos mayores que padecen COVID-19 y para la adecuada atención por los efectos o condiciones de salud al terminar el confinamiento por la pandemia.

This study underscores the development of Ag hydrogel nanocomposites, as smart substrates for antibacterial uses, via innovative in situ reactive and reduction pathways. To this end, two different synthetic strategies were used. Firstly thiol-acrylate (PSA) based hydrogels were attained via thiol-ene and radical polymerization of polyethylene glycol (PEG) and polycaprolactone (PCL). As a second approach, polyurethane (PU) based hydrogels were achieved by condensation polymerization from diisocyanates and PCL and PEG diols. In fact, these syntheses rendered active three-dimensional (3D) hydrogel matrices which were used as nanoreactors for in situ reduction of AgNO3 to silver nanoparticles. A redox chemistry of stannous catalyst in PU hydrogel yielded spherical AgNPs formation, even at 4 °C in the absence of external reductant; and an appropriate thiol-functionalized polymeric network promoted spherical AgNPs well dispersed through PSA hydrogel network, after heating up the swollen hydrogel at 103 °C in the presence of citrate-reductant. Optical and swelling behaviors of both series of hydrogel nanocomposites were investigated as key factors involved in their antimicrobial efficacy over time. Lastly, in vitro antibacterial activity of Ag loaded hydrogels exposed to Pseudomona aeruginosa and Escherichia coli strains indicated a noticeable sustained inhibitory effect, especially for Ag-PU hydrogel nanocomposites with bacterial inhibition growth capabilities up to 120 h cultivation.

In this study, we investigated the effects of the aqueous extracts of Lingzhi or Reishi medicinal mushroom, Ganoderma lucidum, obtained from three localities (China; and Morelos and Michoacan, Mexico) on cervical cells transformed by human papillomavirus (HeLa and SiHa) and C-33A cancer cells. The cells were plated in DMEM medium supplemented, and were incubated in the presence of different concentrations of G. lucidum for 24 h. Cell proliferation was determined by MTT colorimetric assay and viability by trypan blue assay. Inhibitory dose was determined (IC50) of the three different extracts of G. lucidum in the culture cell lines mentioned above. The apoptosis process was confirmed by nuclear DNA fragmentation and the cell cycle was determined by flow cytometry. The results showed that aqueous extracts G. lucidum obtained from three localities produced inhibition in the proliferation of VPH transformed cells; they also induced apoptosis and cell cycle arrest in HeLa, SiHa, and C-33A cancer cells. Therefore, it was found that aqueous extracts G. lucidum obtained from three different locations produced inhibitory effect on cancer cells and may have a potential therapeutic use for the prevention and treatment of this disease.

The AKNA gene is part of the 9q32 susceptibility locus for cervical cancer. A single-nucleotide polymorphism at codon 1119 of AKNA, yields a biologically relevant amino acid change (R1119Q) at the DNA binding AT-hook motif. Genotype frequencies in 97 allele pairs were: R/R = 0.597, R/Q = 0.278, Q/Q = 0.123. Q/Q homozygosity was present in 8.33% of healthy controls, 16.67% of patients with cervical intraepithelial neoplasia and 75% of cervical cancer patients. These differences are highly significant for the presence of Q/Q in cervical cancer (p = 0.01, odds ratio 3.66, 95% confidence interval 1.35-9.94). Therefore, AKNA appears to be an important genetic factor associated with the risk cervical cancer.

INTRODUCTION: Cervical cancer development from a squamous intraepithelial lesion is thought to be favored by an impaired T cell immunity. We evaluated parameters of T cell alterations such as proliferation, cytokine, and CD3zeta expression in peripheral blood and tumor-infiltrating T lymphocytes from women with squamous intraepithelial lesions (SIL) or cervical cancer (CC). RESULTS AND DISCUSSION: T cell proliferation and cytokine messenger RNA (mRNA) expression were similar in women with SIL and healthy donors, whereas low T cell proliferation and lower mRNA expression of IL-2, IL-10 and IFN-gamma were observed in women with CC. Moreover, infiltrating cells showed marginal responses. We also found that CD3zeta mRNA expression, whose protein is required for T cell activation, correlated with a decreased proliferation in advanced stages of the disease. CONCLUSION: Experiments with T cells from healthy donors in the presence TGF-beta1 or IL-10 suggest that these cytokines have a relevant role in T cell responses during CC progression.

RESUMO

Genomics, as a scientific discipline responsible for genome maps, sequencing and functional analysis of genomes, allows for continually expanding knowledge of the structure and function of genomes. The influence of genomics on medicine generates a new perspective for how we perceive health and disease, knowing the influence of genetic variations on susceptibility to disease. In the area of public health, genetic epidemiology translates genetic knowledge into individual and public actions, evaluating the effect of the distribution of genetic determinants and their interaction with environmental factors involved in the etiology of human diseases. In addition, genomic medicine suggests new diagnostic systems, genetic associations and nutritional disorders, specific responses to diverse drugs, and the design of new drugs for susceptible groups. And yet, the greatest advances in genomic medicine in the field of health are forthcoming.

The present study was performed to determine IL-10 expression in cervical tissues in Mexican women according to the severity of the malignity and its association with HPV infection. IL-10 expression showed a clear tendency to increase during the different cervical cancer stages: 37% in LGSIL; 62% in HGSIL; and 84% in cancer. However, all the patients that expressed IL-10 were HPV positives; we found an association with HPV 16. These results suggest a clear relationship between IL-10, HPV and the stage of cervical cancer disease; this event could contribute to the immunosuppressive micro-environment in the tumor site.

Objetivo: analizar la distribución de los diferentes alelos B 27 (B 27.01-B 27.11) del complejo mayor de histocompatibilidad (CMH) en la población sana y en un grupo de pacientes con Espondiloartropatías Seronegativas (EASN) HLA B27 +. Materiales y métodos: s estudiaron 1.000 muestras de sangre provenientes de individuos normales donantes de sangre de la población general y se los comparó con 55 pacientes con diagnóstico de EASN: 33 pacientes (60 por ciento) con Espondilitis Anquilastante (EA), 12 (21.8 por ciento) con Síndrome de Reiter (SR) y 10 (18.1 por ciento) con Artropatía Psoriásica (APs). Los pacientes con EA y SR estudiados con anterioridad pertenecían a un grupo B 27 +. Se utilizaron técnicas de biología molecular PCR-SSO y por medio de sondas específicas PAN-B 27 se analizó la frecuencia de los alelos B 27.01 al B 27.11. No fue evaluada la presencia del alelo B 27.12. Resultados: de los 1000 donantes de sangre de la población general, 4.2 por ciento (42 personas) fueron B 27 + (en relación con estudios serológicos previos que mostraban un 3.5 por ciento)... (AU)

Transplantation of hematopoietic progenitor cells (HPC) from bone marrow and mobilized peripheral blood is a standard therapy in malignant and non malignant diseases. The lack of suitable donors is an important limitation. The discovery that umbilical cord blood (CB) contains high numbers of HPC that can be used as an alternative source for allogeneic stem cell transplantation led ITMO to establish BANCEL, the first Argentine and Latinoamerican experience of its kind. The blood remaining in the umbilical cord and in the placenta was requested from women who were in the last quarter of pregnancy. An informed consent together with a medical record focused on family disease was completed. Out of 65 donations, 55 (85 per cent) were collected and 51 (78 per cent) were cryopreserved. Mean collected volume was 110 ml with 68 per cent (75 ml) reduction and mean cryopreservation of 35 ml; ABO and Rh blood group systems were determined, HLA, class I, A and B loci, and class II, DR locus were typed by molecular biology methods using PCR-SSOP. Infectious disease screening was carried out for brucellosis, syphilis, Chagas, hepatitis B and C, HIV I and II, HTLV I and II, toxoplasmosis and cytomegalovirus. Two positive units for hepatitis B (anticore) and two positive units for Chagas were discarded. The quantity of total nucleated cells (TNC), CD34+ cells and the clonogenic capacity were determined twice at the collection and after the procedures of volume reduction previous to cryopreservation. A 5 per cent reduction in both TNC and CD34 cells and a 10 per cent in the colony forming units (CFU) were detected. A good correlation coefficient between TNC and CFU was obtained.

La Leucemia Mieloide Crónica (LCM) es un proceso oncohematológico caracterizado por una proliferación clonal que afecta a la célula hematopoyética primitiva. Un 95 por ciento de pacientes presenta una alteración citogenética característica conocida como Cromosoma Philadelphia (Ph1), producto de una traslocación cromosómica 9:22, que da lugar a un gen hídrido BCR/ABL. Diecinueve pacientes con LMC recibieron Trasplante Alogeneico de Médula Osea (TMO), 9 fueron de sexo femenino y 10 de sexo masculino. La média de edad fue de 32 años (rango 9-47); 15 de los pacientes estaban en fase crónica (FC) y 4 en fase acelerada (FA). Todos los pacientes al momento del diagnóstico fueron Ph1+, BCR/ABL+. El régimen de acondicionamiento consistió en Busulflán y Ciclofosfamida, con el agregado de Etoposido en los pacientes en FA. La profilaxis de EICH se efectuó con Ciclosporina A, Metotrexato y Metilprednisona en 17 pacientes y con las 2 primeras drogas en 2 pacientes. La traslocación 922 se estudió mediante técnica de RT-PCR, con empleo de las sondas NB1+, Abl3, B2A, CA3 y A2. La sensibilidad del método fue de 1 x 10(-6). De 19 pacientes que ingresaron al protocolo, 14 permanecen vivos y en remisión clínica, hematológica y citogenética (Ph, negativo); 3 pacientes fallecieron de EICH agudo, 1 de fallo de "angraftment" y 1 de síndrome urémico hemolítico. De 4 pacientes trasplantados en FA, 3 están vivos y en remisión completa. Los pacientes con LMC trasplantados presentaron una sobrevida del 74 por ciento con un seguimiento medio de 655 días. El quimerismo hematopoyético completo se demostró en 16 pacientes, mediante el estudio de 3 loci, D1S80, APO B y D17S30. No se encontró ninguna relación entre la presencia post TMO del híbrido BCR/ABL (RT.PCR) y la recaída de la enfermedad; la presencia de EICH agudo y/o crónico no tuvo influencia entre la positividad del BCR/ABL. El TMO ha demostrado se en nuestra experiencia la única alternativa terapéutica para la LMC con obtención de remisión completa, clínica, hematológica y citogenética, con una sobrevida media del 74 por ciento comparable a la de centros internacionales de trasplante.

La Leucemia Mieloide Crónica (LCM) es un proceso oncohematológico caracterizado por una proliferación clonal que afecta a la célula hematopoyética primitiva. Un 95 por ciento de pacientes presenta una alteración citogenética característica conocida como Cromosoma Philadelphia (Ph1), producto de una traslocación cromosómica 9:22, que da lugar a un gen hídrido BCR/ABL. Diecinueve pacientes con LMC recibieron Trasplante Alogeneico de Médula Osea (TMO), 9 fueron de sexo femenino y 10 de sexo masculino. La média de edad fue de 32 años (rango 9-47); 15 de los pacientes estaban en fase crónica (FC) y 4 en fase acelerada (FA). Todos los pacientes al momento del diagnóstico fueron Ph1+, BCR/ABL+. Un grupo de 19 pacientes con diagnóstico de LMC recibieron TMO en el Instituto de Trasplante de Médula Osea, Fundación Mainetti entre los años 1994 y 1997.
