RESUMO

The potential function of long non-coding RNAs in regulating neighbor protein-coding genes has attracted scientists' attention. Despite the important role of lncRNAs in biological processes, a limited number of studies focus on non-model animal lncRNAs. In this study, we used a stringent step-by-step filtering pipeline and machine learning-based tools to identify the specific Androctonus crassicauda lncRNAs and analyze the features of predicted scorpion lncRNAs. 13,401 lncRNAs were detected using pipeline in A. crassicauda transcriptome. The blast results indicated that the majority of these lncRNAs sequences (12,642) have no identifiable orthologs even in closely related species and those considered as novel lncRNAs. Compared to lncRNA prediction tools indicated that our pipeline is a helpful approach to distinguish protein-coding and non-coding transcripts from RNA sequencing data of species without reference genomes. Moreover, analyzing lncRNA characteristics in A. crassicauda uncovered that lower protein-coding potential, lower GC content, shorter transcript length, and less number of isoform per gene are outstanding features of A. crassicauda lncRNAs transcripts.

AIM: The purpose of this research was to use environmental variables for predicting the probability of Hemiscorpius lepturus existence in the provinces where situated in the west of the Zagros Mountains. MATERIALS AND METHODS: In this study, 64 occurrence records of the H. lepturus were extracted from the published documents available in electronic databases. MaxEnt model was used for predicting the ecological niches of this species. Normalized difference vegetation index (NDVI) and 19 climatic variables were used as the environmental variables affecting the distribution of this scorpion. The Jackknife test in the model was used to indicate the importance of variables to predict the probability of the presence of the studied species. The logistic threshold that was evaluated using a logistic regression algorithm showed the converting of the probability model into a binary model. The model was evaluated byarea under the curve (AUC). The probability presence map of this scorpion was then prepared in ArcGIS 10.5 Software. RESULTS: The results of the analysis showed that the most important environmental factor on the distribution of H. lepturus was the maximum temperature of the warmest month (Bio5) with a contribution rate of 43% and permutation importance of 8%. The Jackknife test revealed that NDVI did not gain any value when it used independently in the model. The logistic threshold was reported 0.255 for the maximum test sensitivity plus specificity. The AUC of the model was 0.7698, shows an acceptable value for model validity. Overall the hot spots for this toxic scorpion seem to be in Khuzestan, Lorestan, and Ilam Provinces of the studied area. CONCLUSION: Regarding our findings, MaxEnt algorithm, in combination with geographic information system contributed to revealing the effects of environmental variables on the probability of H. lepturus presence in the west of Zagros Mountains. These visualized maps as a warning alarm can be helpful to policymakers for managing, controlling, and monitoring the scorpionism in high-risk areas.

BACKGROUND: Scorpions are one of the most important medical arthropods in Iran. This study aimed to determine the fauna, spatial distribution and some morphological characteristics of these venomous arthropods in the study area. METHODS: Scorpions were collected using Ultra Violet light, rock rolling and digging methods in West Azerbaijan, East Azerbaijan, and Ardabil Provinces during 2015-2016. The specimens were preserved in 75% ethyl alcohol and transferred to the laboratory for species identification and morphological studies. RESULTS: Distribution maps were produced using ArcGIS 10.3. Totally, 368 specimens from two families of Buthidae (97.1%) and Scorpionidae (2.99%) were collected and identified as Mesobuthus eupeus (80.16%), Androctonus crassicauda (10.60%), M. caucasicus (4.89%), Hottentotta saulcyi (1.35%) and Scorpio maurus (2.99%). CONCLUSION: The presence of medically important species, including the deadly black one in northwestern Iran requires health educational and control programs for reduction of these public health problems.

INTRODUCTION: People in tropical and semi-tropical areas are in danger of scorpion sting, and this can be a serious problem for them. Mortality due to scorpion sting in the tropical and semi-tropical areas of Iran is about 75%, and this makes scorpion sting in these areas a serious medical problem. Because of this problem, our aim was to assess the epidemiological aspects of scorpion sting in Bandar Abbas, Iran, during 2009-2011. METHODS: In this cross-sectional retrospective study, epidemiologic data of 698 scorpion sting cases, who were referred to the Shahid Mohamadi Hospital of Bandar Abbas in Hormozgan Province collected from 2009 until 2011. The data included demographic and individual information, such as age, gender, geographic location, bite site, when the incident occurred, and anti-venom consumption. The required data were extracted from the patients' recorded information in the Hospital, and we recorded data in a special checklist and imported the data into the computer for statistical analysis using of SPSS software, version 21.0. Descriptive statistics, including mean, standard deviation, frequency, and percentage, were used for data analysis. RESULTS: Two hundred and sixty-one (37.4%) of the cases were urban and 437(62.6%) were rural. Males comprised 50.1% of the cases, and women comprised 49.9% (p >0.05). Twenty-five point two percent of scorpion sting cases occurred among people in the 21 to 30 age group, and there were very few cases among people in the 51 to 60 age range (p<0.05). Most of cases were recorded in April and October, and the fewest cases were recorded in July and January (p<0.05); also 32.2% of scorpion sting cases occurred after midnight and in the early morning hours. CONCLUSION: Our survey showed that there was a high incidence of scorpion stings in rural areas, among 21-30 age group, among housekeepers, and among students. These results indicate the need for public education programs and better sanitation services in the rural areas around Bandar Abbas city. Prospective studies can help to health and medicine organization for prevention and treatment of scorpion sting.

Scorpion sting is a public health problem in south and southwestern parts of Iran, with about 36,000 cases recorded annually. This study aimed to find the spatial distribution of scorpions and their stings in Bandar Abbas County. Monthly scorpion sting cases at the village level were obtained and used for mapping. Scorpions were collected from 14 collection sites using a UV lamp at night and searching under stones during the day time. During the study period, a total of 3,971 cases of scorpion sting were recorded, most of them were found in mountainous areas and affected individuals aged 25-44 yrs. In total, 18 scorpion species belonging to 10 genera were collected and identified. The peak of scorpion sting cases occurred from July to September. The northern part of the mountainous areas had a richer species composition. Hemiscorpius persicus and Hemiscorpius gaillardi were collected for the first time in the area. There were 22 scorpion species in the area across studies; among them, 10 were most dangerous. Hemiscorpius genus is the main etiologic agent in Bandar Abbas County. Mapping dangerous species allows the health system to provide relevant anti-scorpion venom serum accordingly and more cost-effectively.

BACKGROUND: Scorpion venom is important and rich source of peptides, most of which have been widely used as pharmacological tools for unraveling structure-function relationship of various ion channels. Naturally occurring toxins can be also considered as lead compounds in the development of novel drugs. OBJECTIVES: In this context, the scorpion-derived peptide neurotoxins specific to sodium channels have shown promise as potential therapeutic targets for the treatment of various human diseases. MATERIALS AND METHODS: A cDNA library from the extracted RNA was constructed using RT-PCR and semi-nested RT-PCR. DNA sequencing followed by phylogenetic analysis was applied to screen the cDNA library clones. For molecular characterization of the BMK gene we used cloning and recombinant protein expression techniques based on E.coli systems. Then we performed mice immunization and Western blot and Immunodot analyses. RESULTS: A novel BMK neurotoxin has been cloned, expressed and characterized from the Iranian scorpion M. eupeus venom. We analyzed the recombinant BMK by immunoblotting with treated antiserum. The result showed that mice antiserum can react also with scorpion crude venom, so is able to recognize native BMK toxin. CONCLUSION: The newly produced recombinant protein BMK revealed to be immunogenic. Moreover, anti-BMK antibodies produced in mice were able to recognize both the recombinant BMK neurotoxin and the one in M. eupeus crude venome. Taken together, the molecular characterization and recombinant production of the Iranian scorpion M. eupeus venom component can serve as a new probe for further studies of sodium channels function and physiology. This provides a promising perspective for the future design of selective drugs, as well as for research of antivenom production.

RESUMO

A new species of scorpions in the genus Odontobuthus (Scorpiones, Buthidae) is described from Khorasan Province, Iran. Currently, Odontobuthus includes two species in Iran, Odontobuthus doriae Thorell, 1876, which is restricted to high elevations of the central Iranian Plateau and Odontobuthus bidentatus Lourenço & Pezier, 2002 from the Zagros Mountains. The results of morphological comparisons, univariate and multivariate statistical analyses and phylogenetic analysis of COI sequence data clearly confirm a deep split between populations from the eastern Iranian Plateau and O. bidentatus Lourenço & Pezier, 2002 and O. doriae Thorell, 1876. Therefore, according to comparative morphological and molecular analyses, a new species, Odontobuthus tigari sp. nov. (♀♂) was described from eastern Iran. This addition represents the third species of this genus from Iran.

Myiasis is an infestation of tissue with the larval stage of dipterous flies. This condition mostly affects the skin but may also occur in certain body cavities. It can occur in either animals or humans and is caused by parasitic dipterous fly larvae feeding on the host's necrotic or living tissue. This disease rarely effects birds especially owls. In this study, infestation of an owl with cutaneous myiasis is reported. In October 2008, a wounded owl was referred by the environmental department of Chaharmahal-Bakhtiary province to the clinic of veterinary science at Shahrekord University in west central Iran. At the initial examination, clinical signs were extensive with a wound under the right wing. The wound was infested with 40 white conical maggots, 3-9 mm in length, which led to a diagnosis of myiasis in the owl. The maggots were carefully collected from the wound using sterile forceps and were kept in 70% ethanol and transferred to the laboratory of parasitology where the diagnosis was undertaken by the observation of posterior and anterior spiracle and cephalopharyngeal apparatus. According to key diagnostic features for maggots in birds, the larvae were identified as Lucilia sericata and Lucilia cuprina (Diptera: Calliphoridae). The wound was treated using usual acaricides, but due to the severity of the infestation and because of the delay in referring the animal to the clinic, it died 3 days post-treatment. This is the first report in Iran of an infestation of the Eurasian eagle owl (Bubo bubo) with L. sericata and L. cuprina (Diptera: Calliphoridae).
