RESUMO

Antimicrobial peptides (AMPs) are short biopolymers produced by living organisms as an immune system defense against infections. They have been considered as potential alternatives to conventional antibiotics. Experiments suggest that combining several types of different AMPs might enhance their antimicrobial activity more effectively than using single-component AMPs. However, a clear understanding of the underlying microscopic mechanisms is still lacking. We present a theoretical investigation of antibacterial cooperativity mechanisms involving several types of AMPs. It is argued that synergy results from intermolecular interactions when the presence of one type of AMP stimulates the association of another type of AMP to bacteria. It is found that increasing the number of different AMPs in the mixtures increases the number of such interactions, making them more efficient in eliminating infections. Our theoretical framework provides valuable insights into the mechanisms of antimicrobial action.

Antimicrobial peptides (AMPs), or defence peptides, are compounds naturally produced during immune responses of living organisms against bacterial infections that are currently actively considered as promising alternatives to antibiotics. Recent experimental studies uncovered that in many situations, combinations of different AMPs are much more successful in eliminating the bacterial pathogens than single peptide species. However, the microscopic origin of such synergistic activities remains not fully understood. We present and investigate a possible mechanism of synergy between AMPs. It is based on the idea that due to inter-molecular interactions, the presence of an AMP of one type stimulates the association of an AMP of another type, and this accelerates the overall association to the membrane, eventually killing the bacteria. This approach allows us to fully quantify the synergistic activities of AMPs, and it is successfully applied for several experimental systems. It is found that strong cooperativity can be achieved for relatively weak inter-molecular interactions, suggesting that the application of combinations of AMPs can be further rationally optimized to make it a powerful antibacterial treatment.

Antimicrobial peptides (AMPs) produced by multi-cellular organisms as their immune system's defence against microbes are actively considered as natural alternatives to conventional antibiotics. Although substantial progress has been achieved in studying the AMPs, the microscopic mechanisms of their functioning remain not well understood. Here, we develop a new theoretical framework to investigate how the AMPs are able to efficiently neutralize bacteria. In our minimal theoretical model, the most relevant processes, AMPs entering into and the following inhibition of the single bacterial cell, are described stochastically. Using complementary master equations approaches, all relevant features of bacteria clearance dynamics by AMPs, such as the probability of inhibition and the mean times before the clearance, are explicitly evaluated. It is found that both processes, entering and inhibition, are equally important for the efficient functioning of AMPs. Our theoretical method naturally explains a wide spectrum of efficiencies of existing AMPs and their heterogeneity at the single-cell level. Theoretical calculations are also consistent with existing single-cell measurements. Thus, the presented theoretical approach clarifies some microscopic aspects of the action of AMPs on bacteria.

In this communication, we report results of an experiment in which crude oil adsorbed on Teflon fabric is exposed to conditions expected in natural ocean-surface collection vehicle containers over a period of 3 months. Samples were recovered at designated time points and analyzed to determine degree of preservation of molecular signatures. Ratios of saturate hydrocarbons were preferentially preserved compared to those of aromatic compounds. Unsubstituted aromatic compounds such as naphthalene and dibenzothiophene were removed at higher rates relative to methyl-substituted homologues owing to differences in biodegradability and solubility. Ratios of important marker compounds persist over several months with sufficient fidelity for accurate determination of source-rock facies and thermal maturity. Collection and storage of oil samples on small pieces of adsorbent Teflon mesh broadens the opportunity space for collection of organic geochemistry data. Deployment of automated samplers equipped with such Teflon meshes aboard autonomous surface vehicles and human-operated vessels promises cheaper, better coverage of geochemical data in seep, slick, and spill surveys conducted for research, exploration, monitoring, and emergency response efforts.

This study aimed to assess the knowledge of antiretroviral (ARV) treatment and the associated factors in HIV-infected patients in Vietnam. We conducted a cross-sectional descriptive study of 350 human immunodeficiency virus (HIV)/acquired immunodeficiency syndrome (AIDS) patients being treated with ARV at outpatient clinics at Soc Trang, Vietnam, from June 2019 to December 2019. Using an interview questionnaire, patients who answered at least eight out of nine questions correctly, including some required questions, were considered to have a general knowledge of ARV treatment. Using multivariate logistic regression to identify factors associated with knowledge of ARV treatment, we found that 62% of HIV-infected patients had a general knowledge of ARV treatment, with a mean score of 8.2 (SD 1.4) out of 9 correct. A higher education level (p < 0.001); working away from home (p = 0.013); getting HIV transmitted by injecting drugs or from mother-to-child contact (p = 0.023); the presence of tension, anxiety, or stress (p = 0.005); self-reminding to take medication (p = 0.024); and a high self-evaluated adherence (p < 0.001) were found to be significantly associated with an adequate knowledge of ARV treatment. In conclusion, education programs for patients, as well as the quality of medical services and support, should be strengthened.

This study was conducted to determine the prevalence and determinants of medication adherence among patients with HIV/AIDS in southern Vietnam. METHODS: A cross-sectional study was conducted in a hospital in southern Vietnam from June to December 2019 on patients who began antiretroviral therapy (ART) for at least 6 months. Using a designed questionnaire, patients were considered adherent if they took correct medicines with right doses, on time and properly with food and beverage and had follow-up visits as scheduled. Multivariable logistic regression was used to identify determinants of adherence. KEY FINDINGS: A total of 350 patients (from 861 medical records) were eligible for the study. The majority of patients were male (62.9%), and the dominant age group (≥35 years old) accounted for 53.7% of patients. Sexual intercourse was the primary route of transmission of HIV (95.1%). The proportions of participants who took the correct medicine and at a proper dose were 98.3% and 86.3%, respectively. In total, 94.9% of participants took medicine appropriately in combination with food and beverage, and 75.7% of participants were strictly adherent to ART. The factors marital status (odds ratio (OR) = 2.54; 95%CI = 1.51-4.28), being away from home (OR = 1.7; 95%CI = 1.03-2.78), substance abuse (OR = 2.7; 95%CI = 1.44-5.05), general knowledge about ART (OR = 2.75; 95%CI = 1.67-4.53), stopping medication after improvement (OR = 4.16; 95%CI = 2.29-7.56) and self-assessment of therapy adherence (OR = 9.83; 95%CI = 5.44-17.77) were significantly associated with patients' adherence. CONCLUSIONS: Three-quarters of patients were adherent to ART. Researchers should consider these determinants of adherence in developing interventions in further studies.

In January 2020, we identified two severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)-infected patients in a familial cluster with one person coming from Wuhan, China. The complete genome sequences of two SARS-CoV-2 strains isolated from these patients were identical and 99.98% similar to strains isolated in Wuhan. This is genetically suggestive of human-to-human transmission of SARS-CoV-2 and indicates Wuhan as the most plausible origin of the early outbreak in Vietnam. The younger patient had a mild upper respiratory illness and a brief viral shedding, whereas the elderly with multi-morbidity had pneumonia, prolonged viral shedding, and residual lung damage. The evidence of nonsynonymous substitutions in the ORF1ab region of the viral sequence warrants further studies.
