RESUMO

A type of electrochemical biosensors based on magnetic Fe3O4/α-Fe2O3 heterogeneous nanosheets was constructed to detect Tau proteins for early diagnosis and intervention therapy of Alzheimer's disease (AD). Firstly, Fe3O4/α-Fe2O3 heterogeneous nanosheets were fabricated as the substrate to realize magnetic self-assembly and magnetic separation to improve current response, and Fe3O4/α-Fe2O3@Au-Apt/ssDNA/MCH biosensors were successfully constructed through the reduction process of chloroauric acid, the immobilizations of aptamer (Apt) and ssDNA, and the intercept process of 6-Mercapto-1-hexanol (MCH); the construction process of the electrochemical biosensor was monitored using Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS), and the factors affecting the current response of this sensor (concentration of Fe3O4/α-Fe2O3@Au and Apt/ssDNA, incubation temperature and time of Tau) were explored and optimized using differential pulse voltammetry (DPV). Analyzing the performance of this sensor under optimal conditions, the linear range was finally obtained to be 0.1 pg/mL-10 ng/mL, the limit of detection (LOD) was 0.08 pg/mL, and the limit of quantification (LOQ) was 0.28 pg/mL. The selectivity, reproducibility and stability of the biosensors were further investigated, and in a really sample analysis using human serum, the recoveries were obtained in the range of 93.93 %-107.39 %, with RSD ranging from 1.05 % to 1.94 %.

AIM: To explore the mediating effect of work engagement and the moderated mediating effect of emotional workload on the relationship between job demands and job performance among nurses. BACKGROUND: Nurses work in a high-demand situation that could affect their job performance. However, previous studies have reported an inconsistent relationship between job demands and job performance. The underlying mechanism of how job demands influence job performance remains unclear. METHODS: An online cross-sectional survey was conducted with a convenience sample of 893 nurses from 14 cities in Sichuan Province between November and December 2021. Data were collected using the Job Demands Scale, Job Performance Scale, Utrecht Work Engagement Scale, and emotional workload subscale of the Questionnaire on the Experience and Evaluation of Work. Bootstrap and simple slope methods were used to test a moderated mediation model using Hayes' PROCESS macro. The STROBE reporting guidelines were utilized. RESULTS: Job demands had a positive effect on job performance, and this effect was mediated by work engagement. Emotional workload moderated the indirect relationship between job demands and job performance. Specifically, the positive effect of job demands on job performance via work engagement was attenuated in nurses with a high emotional workload. CONCLUSION: This study sheds light on the complex relationship between job demands and job performance. Work engagement and emotional workload deserve more attention to improve nurses' performance. IMPLICATIONS FOR NURSING AND NURSING POLICY: Policymakers and nurse managers should make efforts to develop and implement strategies to foster nurses' work engagement, reduce their emotional workload, and further help nurses efficiently deal with job demands.

OBJECTIVES: Despite early mobilisation as a key component of enhanced recovery after surgery pathways for pancreatic surgery, the implementation of early mobilisation remains unsatisfactory. What factors influence the implementation of early mobilisation from the lens of all stakeholders is unclear. The aim of this study was to identify the influencing factors of early mobilisation in pancreatic surgery from the perspective of patients, family members and health professionals. DESIGN: A qualitative descriptive design using individual interviews and focus groups. SETTING AND PARTICIPANTS: Twenty-two patients undergoing pancreatic surgery, 10 family members and 10 healthcare professionals from a large university teaching hospital in China. METHODS: We collected data on participants' views on factors influencing early mobilisation after pancreatic surgery. Two researchers independently reviewed the transcripts and emergent coding. The data were analysed using qualitative content analysis. RESULTS: Three main categories that influenced the implementation of early mobilisation in pancreatic surgery were identified: (1) attitude towards early postoperative mobilisation (eg, perceived advantages or disadvantages of early mobilisation), (2) subjective norm (eg, impact from health professionals, family members and fellow patients) and (3) perceived behavioural control (eg, knowledge, abilities, resources and environment). CONCLUSION: Factors influencing early mobilisation are diverse and multidimensional. The successful implementation of early mobilisation requires the dedication of both patients and healthcare professionals.

PURPOSE: The purpose of this study was to develop the career growth scale for nurses (CGSN) and evaluate its psychometric properties. METHODS: This study was conducted in four phases: (1) creating a pool of potential items through a qualitative design; (2) developing a preliminary scale using a modified two-round Delphi process; (3) refining the preliminary scale to finalize the scale using a cross-sectional survey; and (4) evaluating the psychometric properties of the final scale using another cross-sectional survey. A sample of 858 registered nurses from 12 general hospitals was recruited for this study. RESULTS: The final scale consisted of 17 items categorized into three factors: career goal progress, professional ability and attribute improvement, and career promotion and prestige increase. The three factors accounted for 75.4% of the observed variance in career growth. The overall Cronbach's α was .96, and the intraclass correlation coefficient was .92. The content validity index was .97. Confirmatory factor analysis showed acceptable model fitness. CONCLUSIONS: These results showed that the CGSN has good psychometric properties and can be used to evaluate specific career growth among nurses. This new instrument can further help nurse managers and clinical nurses themselves assess career growth and identify unsatisfactory aspects of growth, thereby designing tailored training programs and evaluating the effectiveness of such interventions.

The aim of the present study was to explore the diagnostic value of a deep convolutional neural network (DCNN) model for the diagnosis of pulmonary nodules in adolescent and young adult patients with osteosarcoma. For the present study, 675 chest CT images were retrospectively collected from 109 patients with clinically confirmed osteosarcoma who underwent chest CT examination at Hangzhou Third People's Hospital (Hangzhou, China) from March 2011 to February 2022. CT images were then evaluated using the DCNN and manual models. Subsequently, pulmonary nodules of osteosarcoma were divided into calcified nodules, solid nodules, partially solid nodules and ground glass nodules using the DCNN model. Those patients with osteosarcoma who were diagnosed and treated were followed up to observe dynamic changes in the pulmonary nodules. A total of 3,087 nodules were detected, while 278 nodules were missed compared with those determined using the reference standard given by the consensus of three Experienced radiologists., which was analyzed by two diagnostic radiologists. In the manual model group, 2,442 nodules were detected, while 657 nodules were missed. The DCNN model showed significantly higher sensitivity and specificity compared with the manual model (sensitivity, 0.923 vs. 0.908; specificity, 0.552 vs. 0.351; P<0.05). In addition, the DCNN model yielded an area under the curve (AUC) value of 0.795 [95% confidence interval (CI), 0.743-0.846], outperforming that of the manual model (AUC, 0.687; 95% CI, 0.629-0.732; P<0.05). The film reading time of the DCNN model was also significantly shorter compared with that of the manual model [mean ± standard deviation (SD); 173.25±24.10 vs. 328.32±22.72 sec; P<0.05)]. The AUC of calcified nodules, solid nodules, partially solid nodules and ground glass nodules was calculated to be 0.766, 0.771, 0.761 and 0.796, respectively, using the DCNN model. Using this model, the majority of the pulmonary nodules were detected in patients with osteosarcoma at the initial diagnosis (69/109, 62.3%), and the majority of these were found with multiple pulmonary nodules instead of a single nodule (71/109, 65.1% vs. 38/109, 34.9%). These data suggest that, compared with the manual model, the DCNN model proved to be beneficial for the detection of pulmonary nodules in adolescent and young adult patients with osteosarcoma, which may reduce the time of artificial radiograph reading. In conclusion, the proposed DCNN model, developed using data from 675 chest CT images retrospectively collected from 109 patients with clinically confirmed osteosarcoma, may be used as an effective tool to evaluate pulmonary nodules in patients with osteosarcoma.

Aims: To investigate adherence to oral anticoagulants among patients after mechanical heart valve (BHV) replacement and further examine the mediating role of medication belief in the relationship between knowledge and medication adherence. Background: The number of patients who undergo BHV replacement has increased in recent years. Short-term anticoagulant therapy is recommended for patients after BHV replacement. However, little is known about adherence to oral anticoagulant therapy and the underlying mechanisms among patients with BHV replacement. Methods: A cross-sectional study was conducted between September 2022 and November 2022. A convenience sample of 323 patients who underwent BHV replacement was recruited from a tertiary public hospital in Southwest China. Data were collected by using the 8-item Morisky Medication Adherence Scale, Beliefs about Medicines Questionnaire-specific, and the Knowledge of Anticoagulation Questionnaire. The mediation model was tested by Hayes's PROCESS macro. The STROBE checklist was used. Results: Approximately 17.3% of participants had low adherence, 47.1% had medium adherence, and only 35.6% reported high adherence to oral anticoagulants. Knowledge and necessity beliefs were positively related to medication adherence, while concern beliefs were negatively correlated with medication adherence. Medication belief mediated the relationship between knowledge and adherence to oral anticoagulants. Conclusion: Patients with BHV replacement demonstrated relatively low adherence to oral anticoagulant therapy. Efforts to enhance medication adherence should consider improving patients' knowledge and medication beliefs.

Achieving high spin polarization transport and a pure spin current is particularly desired in spintronics. We use a sawtooth graphene nanoribbon (STGNR) and its derived five-member ring structure (5-STGNR) to design new spin caloritronic devices, since they have been successfully prepared experimentally and have an interface with no lattice distortion. By using first-principle calculations combined with the non-equilibrium Green's function approach, we have studied the spin caloritronic transport properties of several STGNR-based devices, including the structures with symmetrical and asymmetrical edges, and found some excellent spin caloritronic properties, such as spin polarization, magnetoresistance and the spin Seebeck effect. By introducing a temperature difference, giant magnetoresistance and spin Seebeck effects are achieved in a heterojunction with a symmetrical edge, whereas spin polarization is more effective in a heterojunction with an asymmetrical edge. Meanwhile, the metal-semiconductor-metal junction, which is composed of STGNRs with a symmetrical edge, exhibits approximately 100% spin polarization and produces a perfect thermally induced pure spin current at room temperature. Our results indicate that the devices consisting of a sawtooth graphene nanoribbon and its derived five-member ring structure are promising novel spin caloritronic devices.

The CYP2C19*2 gene carriers and non-carriers are closely related to the dosage of clopidogrel. To correctly guide the use of clopidogrel and promote individualized therapy, an ultra-sensitive electrochemical biosensor was developed for the detection of CYP2C19*2 gene. The heterogeneous α-Fe2O3/Fe3O4 nanosheets were prepared via the hydrothermal-calcination process, and the preparation parameters were optimized. The average diameter and thickness of the nanosheets were approximately 150 nm and 53 nm, respectively; and the saturation magnetization was 80.2 emu/g. The α-Fe2O3/Fe3O4@Au nanosheets were prepared by sodium borohydride reduction method, and self-assembled to the electrode surface with magnetic field. Ultra-sensitive detection of CYP2C19*2 gene was realized through the recognition ability of strong single base mismatching of peptide nucleic acid and signal amplification effect of magnetic α-Fe2O3/Fe3O4@Au nanosheets. Under optimal detection conditions, the current had a good linear correlation with the negative logarithm of CYP2C19*2 gene concentration in the range 1 pM-1 nM, and the detection limit was 0.64 pM (S/N = 3). Meanwhile, the electrochemical signals of target DNA and incomplete complementary DNA were detected. The constructed biosensor exhibited good selectivity, reproducibility, and stability, providing a promising strategy for the detection of other gene mutations by electrochemical biosensors.

Considering the high cost and tedious process of gene sequencing, there is an urgent need to develop portable and efficient sensors for the TP53 gene. Here, we developed a novel electrochemical sensor that detected the TP53 gene using magnetic peptide nucleic acid (PNA)-modified Fe3O4/α-Fe2O3@Au nanocomposites. Cyclic voltammetry and electrochemical impedance spectroscopy confirmed the successful stepwise construction of the sensor, especially the high-affinity binding of PNA to DNA strands, which induced different electron transfer rates and resulted in current changes. Variations in the differential pulse voltammetry current observed during hybridization at different surface PNA probe densities, hybridization times, and hybridization temperatures were explored. The biosensing strategy obtained a limit of detection of 0.26 pM, a limit of quantification of 0.85 pM, and a wide linear range (1 pM-1 µM), confirming that the Fe3O4/α-Fe2O3@Au nanocomposites and the strategy based on magnetic separation and magnetically induced self-assembly improved the binding efficiency of nucleic acid molecules. The biosensor was a label-free and enzyme-free device with excellent reproducibility and stability that could identify single-base mismatched DNA without additional DNA amplification procedures, and the serum spiked experiments revealed the feasibility of the detection approach.

Background: Previous researchers have demonstrated that the work practice environment influences nurses' engagement; however, few studies have explored the mechanisms that explain the links between them. Objectives: To examine whether basic psychological needs and organizational commitment mediate the relationship between the work practice environment and work engagement. Methods: A cross-sectional survey was conducted with a sample of 893 nurses from 14 cities in Sichuan Province of China between November 2021 and December 2021. Data were collected online using the Chinese version of the Practice Environment Scale of the Nursing Work Index, Basic Needs Satisfaction in General Scale, Organizational Commitment Scale, and Utrecht Work Engagement Scale. The Pearson correlation analysis and multiple mediation model were used to analyze the data. Results: The Pearson correlation analysis showed that work practice environment, basic psychological needs, and organizational commitment were positively associated with work engagement. The positive relationship between work practice environment and work engagement was mediated by basic psychological needs and organizational commitment [B = 0.505, SE = 0.032, 95% CI (0.442, 0.566)]. Conclusions: The study substantially contributes to the existing knowledge by revealing the mechanisms of fostering work engagement among nurses and explaining why the work practice environment influences work engagement.

Magnetic Fe3O4 nanoparticles were prepared via a simple hydrothermal method and utilized to load paclitaxel. The average particle size of Fe3O4 nanoparticles was found to be 20.2 ± 3.0 nm, and the calculated saturation magnetization reached 129.38 emu/g, verifying superparamagnetism of nanomaterials. The specific surface area and pore volume were 84.756 m2/g and 0.265 cm3/g, respectively. Subsequently, Fe3O4@mSiO2 nanoparticles were successfully fabricated using the Fe3O4 nanoparticles as precursors with an average size of 27.81 nm. The relevant saturation magnetization, zeta potential, and specific surface area of Fe3O4@mSiO2-NH2-FA were respectively 76.3 emu/g, -14.1 mV, and 324.410 m2/g. The pore volume and average adsorption pore size were 0.369 cm3/g and 4.548 nm, respectively. Compared to free paclitaxel, the solubility and stability of nanoparticles loaded with paclitaxel were improved. The drug loading efficiency and drug load of the nanoformulation were 44.26 and 11.38%, respectively. The Fe3O4@mSiO2-NH2-FA nanocomposites were easy to construct with excellent active targeting performance, pH sensitivity, and sustained-release effect. The nanoformulation also showed good biocompatibility, where the cell viability remained at 73.8% when the concentration reached 1200 µg/mL. The nanoformulation induced cell death through apoptosis, as confirmed by AO/EB staining and flow cytometry. Western blotting results suggested that the nanoformulation could induce iron death by inhibiting Glutathione Peroxidase 4 (GPX4) activity or decreasing Ferritin Heavy Chain 1 (FTH1) expression. Subsequently, the expression of HIF-1α was upregulated owing to the accumulation of reactive oxygen species (ROS), thus affecting the expression of apoptosis-related proteins regulated by p53, inducing cell apoptosis.

AIM: To examine the mediating effect of basic psychological needs on the relationship between perceived organizational support and work engagement among nurses. BACKGROUND: The satisfaction of basic psychological needs is crucial for breeding and sustaining individuals' intrinsic motivation. Little is known about the underlying motivational mechanisms that explain the relationship among perceived organizational support, basic psychological needs, and work engagement in a nursing context. METHODS: This was a cross-sectional online survey. A sample of 858 nurses from 12 hospitals was surveyed on their perceived organizational support, basic psychological needs, and work engagement. Structural equation models and bootstrapping methods were used to examine the hypotheses. STROBE reporting guidelines were utilized. RESULTS: Perceived organizational support was positively associated with basic psychological needs and work engagement. Basic psychological needs were positively associated with work engagement. Basic psychological needs mediated the relationship between perceived organizational and work engagement. CONCLUSION: Perceived organizational support may enhance work engagement by fulfilling the basic psychological needs of nurses. IMPLICATIONS FOR NURSING AND NURSING POLICY: Basic psychological needs deserve more attention in nursing organizations. Managers should seek optimal strategies to fulfill nurses' needs for autonomy, competence, and relatedness to stimulate their intrinsic motivation to enhance work engagement.

A label-free electrochemical aptasensor based on magnetic α-Fe2O3/Fe3O4 heterogeneous hollow nanorods was developed for the detection of cancer antigen 125 (CA125). Magnetic α-Fe2O3/Fe3O4 heterogeneous hollow nanorods were successfully prepared by the hydrolysis-calcination method, functionalized with polyethyleneimine (PEI), and modified with HAuCl4 to form magnetic α-Fe2O3/Fe3O4-Au nanocomposites with a layer of 5 nm gold nanoparticles (Au NPs) on the surface. The magnetic α-Fe2O3/Fe3O4-Au nanocomposites were used to fix the DNA-aptamer probe to amplify the current signal. The intensity of the current signal was proportional to the concentration of CA125, indicating that the sensor was a turn-on sensor. The linear range of the electrochemical aptasensor was 5-125 U/mL (R2 = 0.9975), and the limit of detection (LOD) was 2.99 U/mL. The electrochemical aptasensor exhibited favorable specificity, reproducibility, and stability. The analytical performance of the aptasensor in real serum samples was also investigated.

In order to develop the adsorption application of penta-graphene (PG) to N2O gas molecule, we calculated the sensing properties of PG and Ni-doped PG to N2O molecule via first-principles calculations. Based on the calculated adsorption energy, charge transfer, band gap, density of states and partial density of states, we observed that this gas molecule was weakly physically adsorbed on the surface of intrinsic PG, while the adsorption behaviors on the surface of Ni-doped PG were greatly influenced by the doping sites and adsorption orientations. With the Ni atom doped at the sp2 hybridized carbon site, strong chemical adsorption between the gas molecule and the substrate was induced. The adsorption structure of the N2O molecule with its N atom close to the substrate exhibited better stability. Moreover, an external perpendicular electric field could enhance the adsorption performance of the N2O molecule and adjust the charge transfer between the molecule and substrate. Our results broaden the adsorption applications of PG and indicate that Ni-doped PG is a potential candidate for N2O gas sensors.

AIMS: To explore nurses' experiences and perceptions of career growth and its influencing factors, as career growth is more closely associated with individual attitudes and behaviours in the new career era. DESIGN: A qualitative descriptive design was used. METHODS: Thirty-one nurses from 10 secondary and 8 tertiary hospitals in Sichuan Province of China were purposively selected to participate in this study. The data were collected using individual semi-structured face-to-face interviews. Two researchers independently reviewed the transcripts and emergent coding. The data were analysed using qualitative content analysis. FINDINGS: The nurses' perceptions of career growth fully described the nurses' experience and occurred in five dimensions: career promotion, career goal progress, professional ability and quality improvement, professional identity development and increase in personal prestige. The factors influencing career growth were identified at the organizational, individual and social levels. Career growth in nursing was complex, changed over time and showed several specific characteristics compared with other careers. The nurse-specific symbol of career growth was professional identity development, which reflected career progression characteristics. CONCLUSIONS: Career growth is a multi-dimensional concept with varying influencing factors. The meaning of career growth for nurses is distinct from that for employees in other careers. IMPACT: Nursing managers should guide nurses to comprehensively assess their career growth from multiple perspectives, and professional identity development deserves more attention. Both organizations and individuals should take responsibility for career management to pursue career growth.

In this work, the magnetic α-Fe2O3/Fe3O4 heterogeneous nanotubes were successfully prepared by solvent hydrothermal-controlled calcination method. The effects of additive concentration, hydrothermal temperature and time on morphology of products were investigated. The α-Fe2O3/Fe3O4 nanotubes with a saturation magnetization of 50 emu/g were prepared calcinated at 600 °C for 4 h using 0.8 g of glucose. Their average length, the outer and inner diameters were around 240 nm, 178 nm and 145 nm, respectively. The α-Fe2O3/Fe3O4 heterogeneous nanotubes coated with water-soluble liposome were applied for targeted delivery of curcumin. The release of curcumin inside the hollow structure of the nanocomposites could be triggered and effectively sustained represented a process of slow release. The encapsulation efficiency of curcumin in the α-Fe2O3/Fe3O4-CUR@LIP nanocomposites reached 82.1 ± 0.9%. MTT assays demonstrated that blank carriers had excellent biocompatibility and application of magnetic field significantly elevated the cytotoxicity of α-Fe2O3/Fe3O4-CUR@LIP nanocomposites on MCF-7 cell. Electrochemical experiment and Prussian blue staining indicated that the α-Fe2O3/Fe3O4@LIP nanocomposites could aggregate in cells to promote the internalization of curcumin. Magnetic α-Fe2O3/Fe3O4-CUR@LIP nanocomposites and curcumin enhanced the expression of reactive oxygen species in MCF-7 cells and induced apoptosis by fluorescence detection. Flow cytometry and western blot verified that the α-Fe2O3/Fe3O4@LIP nanocomposites under magnetic field enhanced cells late-apoptosis by adjusting the expression of apoptosis-related proteins.

Phyllodes tumour (PT) of breast is a rare biphasic neoplasm. Recent next generation sequencing analyses had revealed novel genetic alterations in PT but lacked a further characterisation of their relationship to different PT features and outcome. Here, using targeted sequencing, we examined a panel of 90 recurrently altered or cancer related genes in 88 PT samples (including 49 benign, 25 borderline and 14 malignant PT). Twenty-three genes showed alterations in at least 8.0% of cases. Alterations were significantly higher with an increasing grade of PT (p=0.033), particularly for copy number alterations. The top ten alterations were TERT promoter (58.0%), MED12 (53.4%), RARA (22.8%), FLNA (19.3%), SETD2 (15.9%), SYNE1 (18.2%), PCLO (15.9%), KMT2D (14.3%), CDKN2A (15.9%) and DNAH11 (14.8%). Alterations in CDKN2A/B, EGFR, TP53, PIK3CA, PTEN and ARID1B (p≤0.039) were associated with a higher grade. Analysing alterations based on common pathways indicated a significant correlation of cell cycle pathway and epigenetic alterations with a higher PT grade (p=0.036 and 0.075 respectively). Interestingly, recurrences were not correlated with tumour grade, but related to the presence of RARA mutation (p=0.011) and the absence of alterations in epigenetic pathway (p=0.031). Analysis of synchronous pair of PT showed more differences in gene mutations with divergent MED12 mutation. By contrast, the recurrent samples showed similar genetic alterations as the primary tumours. In summary, we characterised genetic alterations in PTs of different grades and confirmed the recurrent alterations observed in earlier studies. In addition, current data implicated the roles of cell cycle, epigenetic and RARA changes in PT recurrence and tumourogenesis.

Conductive and porous nitrogen-rich materials have great potential as supercapacitor electrode materials. The exceptional efficiency of such compounds, however, is dependent on their larger surface area and the level of nitrogen doping. To address these issues, we synthesized a porous covalent triazine framework (An-CTFs) based on 9,10-dicyanoanthracene (An-CN) units through an ionothermal reaction in the presence of different molar ratios of molten zinc chloride (ZnCl2) at 400 and 500 °C, yielding An-CTF-10-400, An-CTF-20-400, An-CTF-10-500, and An-CTF-20-500 microporous materials. According to N2 adsorption-desorption analyses (BET), these An-CTFs produced exceptionally high specific surface areas ranging from 406-751 m2·g-1. Furthermore, An-CTF-10-500 had a capacitance of 589 F·g-1, remarkable cycle stability up to 5000 cycles, up to 95% capacity retention, and strong CO2 adsorption capacity up to 5.65 mmol·g-1 at 273 K. As a result, our An-CTFs are a good alternative for both electrochemical energy storage and CO2 uptake.

In order to develop low-power-consumption electronic devices, we studied a heterojunction composed of a 5,4-sawtooth graphene nanoribbon and 6,6,12-graphyne nanoribbon by using first-principles calculations combined with the non-equilibrium Green's function approach. Some spin caloritronic and spintronic devices can be realized based on the heterojunction, exhibiting excellent and multifunctional properties: a net spin current and perfect spin Seebeck effect are generated, a thermal induced and bias induced colossal magnetoresistance is obtained, and a spin filter with 100% spin polarization and a spin rectification with a high rectification ratio are acquired. Our results indicate that the proposed heterojunction is a promising candidate for multi-effect and low-power-consumption electronic devices.
