RESUMO

ABSTRACT: 99m Tc-PYP/DPD/HDMP cardiac scintigraphy has a pivotal role in the diagnosis of ATTR cardiac amyloidosis. The combined findings of a Perugini visual score of 2 or 3 in the scan and the absence of monoclonal proteins in blood and urine are highly specific for the diagnosis of ATTR cardiac amyloidosis without a tissue biopsy. We report a case of mitral annular and valve calcification accurately identified in the SPECT/CT, but which could be misinterpreted as ATTR cardiac amyloidosis if only acquiring planar and SPECT images.

RESUMO

Little is known about the ecology and distribution of mobulid rays along Brazil's extensive coastline. Here we report opportunistic sightings of manta rays (Mobula cf. birostris) in the Brazilian Amazon estuaries and the Great Amazon Reef System. These sightings consist of manta ray individuals stranded in tide pools, caught in artisanal fisheries, and footage obtained with a submersible. Future investigations on the spatial, temporal, and environmental drivers of manta rays' distribution on the northern Brazilian coast and the threats posed by fishing gear are warranted.

The fitness benefits of social life depend on the ability of animals to affiliate with others and form groups, on dominance hierarchies within groups that determine resource distribution, and on cognitive capacities for recognition, learning and information transfer. The evolution of these phenotypes is coupled with that of neuroendocrine mechanisms, but the causal link between the two remains underexplored. Growing evidence from our research group and others demonstrates that the tools available in zebrafish, Danio rerio, can markedly facilitate progress in this field. Here, we review this evidence and provide a synthesis of the state-of-the-art in this model system. We discuss the involvement of generalized motivation and cognitive components, neuroplasticity and functional connectivity across social decision-making brain areas, and how these are modulated chiefly by the oxytocin-vasopressin neuroendocrine system, but also by reward-pathway monoamine signaling and the effects of sex-hormones and stress physiology.

Cerebral palsy (CP) can be considered the most frequent childhood physical disorder. The severity and type of dysfunction depend on the brain injury. Movement and posture are the most affected areas. CP is a lifelong condition, and parenting a child with this disorder brings additional challenges (e.g., dealing with grief) and needs (e.g., information). Identifying and characterizing their challenges and needs are pivotal to enrich the knowledge in this field and help draw more suitable support for parents. Interviews with 11 parents of children with CP attending elementary school were conducted. The discourse was transcribed, and a thematic analysis was performed. Three themes emerged from the data: (i) challenges of parenting a child with CP (e.g., internal challenges), (ii) crucial needs for parents to cope with a child with CP (e.g., information), and (iii) the intersection between challenges and needs of parents of children with CP (e.g., unawareness). Regarding the challenges and needs characterization, lifespan was the most frequent period of child development, and the microsystem was the context of life most reported. The findings may inform the design of educational and remediation interventions to support families of children with CP attending elementary school.

Hormones regulate behavior either through activational effects that facilitate the acute expression of specific behaviors or through organizational effects that shape the development of the nervous system thereby altering adult behavior. Much research has implicated the neuropeptide oxytocin (OXT) in acute modulation of various aspects of social behaviors across vertebrate species, and OXT signaling is associated with the developmental social deficits observed in autism spectrum disorders (ASDs); however, little is known about the role of OXT in the neurodevelopment of the social brain. We show that perturbation of OXT neurons during early zebrafish development led to a loss of dopaminergic neurons, associated with visual processing and reward, and blunted the neuronal response to social stimuli in the adult brain. Ultimately, adult fish whose OXT neurons were ablated in early life, displayed altered functional connectivity within social decision-making brain nuclei both in naive state and in response to social stimulus and became less social. We propose that OXT neurons have an organizational role, namely, to shape forebrain neuroarchitecture during development and to acquire an affiliative response toward conspecifics.SIGNIFICANCE STATEMENT Social behavior is developed over the lifetime of an organism and the neuropeptide oxytocin (OXT) modulates social behaviors across vertebrate species, and is associated with neuro-developmental social deficits such as autism. However, whether OXT plays a role in the developmental maturation of neural systems that are necessary for social behavior remains poorly explored. We show that proper behavioral and neural response to social stimuli depends on a developmental process orchestrated by OXT neurons. Animals whose OXT system is ablated in early life show blunted neuronal and behavioral responses to social stimuli as well as wide ranging disruptions in the functional connectivity of the social brain. We provide a window into the mechanisms underlying OXT-dependent developmental processes that implement adult sociality.

Aliarcobacter butzleri is an emergent enteropathogen that can be found in a range of environments. This bacterium presents a vast repertoire of efflux pumps, such as the ones belonging to the resistance nodulation cell division family, which may be associated with bacterial resistance, as well as virulence. Thus, this work aimed to evaluate the contribution of three RND efflux systems, AreABC, AreDEF and AreGHI, in the resistance and virulence of A. butzleri. Mutant strains were constructed by inactivation of the gene that encodes the inner membrane protein of these systems. The bacterial resistance profile of parental and mutant strains to several antimicrobials was assessed, as was the intracellular accumulation of the ethidium bromide dye. Regarding bacterial virulence, the role of these three efflux pumps on growth, strain fitness, motility, biofilm formation ability, survival in adverse conditions (oxidative stress and bile salts) and human serum and in vitro adhesion and invasion to Caco-2 cells was evaluated. We observed that the mutants from the three efflux pumps were more susceptible to several classes of antimicrobials than the parental strain and presented an increase in the accumulation of ethidium bromide, indicating a potential role of the efflux pumps in the extrusion of antimicrobials. The mutant strains had no bacterial growth defects; nonetheless, they presented a reduction in relative fitness. For the three mutants, an increase in the susceptibility to oxidative stress was observed, while only the mutant for AreGHI efflux pump showed a relevant role in bile stress survival. All the mutant strains showed an impairment in biofilm formation ability, were more susceptible to human serum and were less adherent to intestinal epithelial cells. Overall, the results support the contribution of the efflux pumps AreABC, AreDEF and AreGHI of A. butzleri to antimicrobial resistance, as well as to bacterial virulence.

Several strategies have been adopted to optimize biodiversity conservation. The use of fauna and flora species as flagships can help increase public commitment and raise funds for their conservation. However, species choices must be well balanced, considering not only intrinsic aspects (e.g., risk of extinction and ecological role), but also social, cultural, and economic aspects of the region where the flagship will be adopted. Brazil is one of the countries with the greatest diversity on the planet. Nevertheless, there are several challenges associated with natural resources conservation. Flagships have been adopted informally for decades throughout the country but there are no efforts to evaluate these strategies results for the preservation of the selected species. The aim of this study was to carry out an extensive survey on regional flagship species, their conservation status, level of endemism, and domestic legislation currently in force for their protection or human use. A total of 62 flagships were identified, with at least one species of flora and one of fauna for each of the 27 Brazilian Federative Units. The animals most commonly used as regional symbols are birds, followed by mammals. Flora species used as regional symbols are quite diverse. However, they are all somehow used by humans. As for legal protection, there are government measures for endangered species. Nevertheless, most species used as regional symbols are not at risk of extinction and, for this reason, do not receive any legal protection. Moreover, while some flagships are endemic, others are alien species, indicating that this criterion is not considered when choosing regional symbols. In the overall analysis it was noted that species choice could be optimized to highlight the national biodiversity. Despite efforts to establish legal measures for their preservation, few effective results have been observed. Therefore, improved effective measures for their conservation should be adopted to guarantee the effectiveness of flagship species for biodiversity conservation.

Abstract: Elasmobranchs exhibit the biggest population declines among vertebrates, being considered one of the groups with the highest risk of extinction. The Brazilian Amazon Coast (BAC) is considered a priority area for elasmobranch conservation, as many species are endemic to the region, and most of them are threatened with extinction. The present study made a scientometric analysis using the IRAMUTEQ method (Interface de R pour les Analyses Multidimensionnelles de Textes et de Questionnaires) to evaluate the trends of research with elasmobranchs in BAC in the last four decades. Ten research trends were identified, highlighting "Biodiversity", "Reproduction", "Trophic Ecology" and "Conservation". However, most of the publications found are aimed at understanding the diversity and abundance of species in the region, with the number of studies focused on reproduction, feeding and other aspects of the biology and ecology of elasmobranchs in the BAC being less expressive. Although the research area "Conservation" stood out in the search, the amount of relevant information so that management and conservation measures can be implemented is still low. Lastly, despite following the global trend of studies with elasmobranchs between 2000 and 2010, investigations into the BAC need to advance, especially considering the poor level of basic information on most species (Biology and population dynamics) and the incomplete knowledge of specific composition of elasmobranch species in the entire area, aspects relevant to conservation and more applied studies that rely on more recent methodologies and technologies.


Resumo: Elasmobrânquios apresentam os maiores declínios populacionais entre os vertebrados, sendo considerado um dos grupos com maior risco de extinção. O Litoral Amazônico Brasileiro (BAC) é considerado uma área prioritária para conservação de elasmobrânquios, pois várias espécies são endêmicas da região, e a maioria delas estão ameaçadas de extinção. O presente estudo fez uma análise cientométrica aplicando o método IRAMUTEQ (Interface de R pour les Analyses Multidimensionnelles de Textes et de Questionnaires) para avaliar as tendências de pesquisas com elasmobrânquios no BAC nas últimas quatro décadas. Dez tendências de pesquisa foram identificadas, destacando-se "Biodiversidade", "Reprodução", "Ecologia trófica" e "Conservação". Entretanto, a maioria das publicações encontradas é voltada para o conhecimento da diversidade e abundância das espécies na região, sendo o número de estudos voltados para a reprodução, alimentação e outros aspectos da biologia e ecologia de elasmobrânquios do BAC menos expressivo. Apesar da área de investigação "Conservação" ter se destacado na busca, a quantidade de informações relevantes para que medidas de manejo e conservação possam ser implementadas ainda é baixa. Por fim, apesar de seguir a tendência mundial de estudos com elasmobrânquios entre 2000 a 2010, as investigações sobre o BAC precisam avançar, especialmente considerando o baixo nível de informações básicas sobre a maioria das espécies (biologia e dinâmica populacional) e o conhecimento incompleto da composição específica das espécies de elasmobrânquios em toda a área, aspectos relevantes para a conservação e estudos mais aplicados e que contem com metodologias e tecnologias mais recentes.

Abstract: Recent statements from the Brazilian federal government indicate that impacting economic activities, particularly commercial shrimp farming, are being encouraged in mangrove areas in the near future. Alterations of the National Action Plan and legal instruments that partially protected mangrove ecosystems have created an even weaker legal framework than previously existed. Such changes are leading Brazil far from the global call to conserve mangroves and from the Aichi targets and United Nations Sustainable Development Goals. Unfortunately, the loss of mangrove ecosystems and their ecosystem services will negatively impact living standards for Brazilians in coastal areas.


Resumo: As recentes determinações do governo Brasileiro indicam que atividades econômicas impactantes para os manguezais, particularmente a carcinocultura, serão incentivadas num futuro próximo. Alterações no Plano de Ação Nacional e em outros instrumentos legais, que parcialmente protegiam os manguezais, fragilizaram ainda mais os meios legais para proteção dessas áreas. Essas mudanças fazem o Brasil seguir na contra-mão da demanda global pela conservação dos manguezais e dos objetivos de desenvolvimento sustentável traçados pela Organização das Nações Unidas, bem como das metas de Aichi para a biodiversidade. Consequentemente, a iminente perda dos ecossistemas de manguezais impactará negativamente o padrão de vida dos Brasileiros que vivem em áreas costeiras.

Oxytocin-like peptides have been implicated in the regulation of a wide range of social behaviors across taxa. On the other hand, the social environment, which is composed of conspecifics that may vary in their genotypes, also influences social behavior, creating the possibility for indirect genetic effects. Here, we used a zebrafish oxytocin receptor knockout line to investigate how the genotypic composition of the social environment (Gs) interacts with the oxytocin genotype of the focal individual (Gi) in the regulation of its social behavior. For this purpose, we have raised wild-type or knock-out zebrafish in either wild-type or knock-out shoals and tested different components of social behavior in adults. GixGs effects were detected in some behaviors, highlighting the need to control for GixGs effects when interpreting results of experiments using genetically modified animals, since the genotypic composition of the social environment can either rescue or promote phenotypes associated with specific genes.

Social living animals need to recognize the presence of conspecifics in the environment in order to engage in adaptive social interactions. Social cues can be detected through different sensory modalities, including vision. Two main visual features can convey information about the presence of conspecifics: body form and biological motion (BM). Given the role that oxytocin plays in social behavior regulation across vertebrates, particularly in the salience and reward values of social stimuli, we hypothesized that it may also be involved in the modulation of perceptual mechanisms for conspecific detection. Here, using videoplaybacks, we assessed the role of conspecific form and BM in zebrafish social affiliation, and how oxytocin regulates the perception of these cues. We demonstrated that while each visual cue is important for social attraction, BM promotes a higher fish engagement than the static conspecific form alone. Moreover, using a mutant line for one of the two oxytocin receptors, we show that oxytocin signaling is involved in the regulation of BM detection but not conspecific form recognition. In summary, our results indicate that, apart from oxytocin role in the regulation of social behaviors through its effect on higher-order cognitive mechanisms, it may regulate social behavior by modulating very basic perceptual mechanisms underlying the detection of socially-relevant cues.

Sociality is a complex phenomenon that involves the individual´s motivation to approach their conspecifics, along with social cognitive functions that enable individuals to interact and survive. The nonapeptide oxytocin (OXT) is known to regulate sociality in many species. However, the role of OXT in specific aspects of sociality is still not well understood. In the present study, we investigated the contribution of the OXT receptor (OXTR) signalling in two different aspects of zebrafish social behaviour: social preference, by measuring their motivation to approach a shoal of conspecifics, and social recognition, by measuring their ability to discriminate between a novel and familiar fish, using a mutant zebrafish lacking a functional OXTR. Although oxtr mutant zebrafish displayed normal attraction to a shoal of conspecifics, they exhibited reduced social recognition. We further investigated whether this effect would be social-domain specific by replacing conspecific fish by objects. Although no differences were observed in object approach, oxtr mutant fish also exhibited impaired object recognition. Our findings suggest that OXTR signalling regulates a more general memory recognition of familiar vs novel entities, not only in social but also in a non-social domain, in zebrafish.

Existing literature has analyzed homework characteristics associated with academic results. Researchers and educators defend the need to provide quality homework, but there is still much to be learned about the characteristics of quality homework (e.g., purposes, type). Acknowledging that teachers play an important role in designing and assigning homework, this study explored teachers' perspectives regarding: (i) the characteristics of quality homework and (ii) the characteristics of the homework tasks assigned. In the current study, mathematics teachers from elementary and middle schools (N = 78) participated in focus group discussions. To enhance the trustworthiness of the findings, homework tasks assigned by 25% of the participants were analyzed for triangulation of data purposes. Data were analyzed using thematic analysis for elementary and middle school separately. Teachers discussed the various characteristics of quality homework (e.g., short assignments, adjusted to the availability of students) and shared the characteristics of the homework tasks typically assigned, highlighting a few differences (e.g., degree of individualization of homework, purposes) between these two topics. Globally, data on the homework tasks assigned were consistent with teachers' reports about the characteristics of the homework tasks they usually assigned. Findings provide valuable insights for research and practice aimed to promote the quality of homework and consequently students' learning and progress.

This study explored mathematics teachers' conceptions of the homework feedback focusing on four key aspects: definition, purpose, types, and perceived impact. Forty-seven teachers from elementary and middle schools participated in six focus groups. Data were analyzed using content analysis. To enhance the trustworthiness of findings, classroom observations were used for triangulation of data. Participants conceptualized homework feedback in three directions (i.e., teachers' feedback provided to students, students' feedback provided to teachers, and homework self-feedback), being teachers' monitoring of students' learning the purpose reported by most teachers. Participants also reported the types of homework feedback more frequently used in class (e.g., checking homework completion, checking homework on the board), and their perceived impact on students. Findings provide valuable information to deepen the understanding of the homework feedback process, which may help develop new avenues for future research.

Proper response to stress and social stimuli depends on orchestrated development of hypothalamic neuronal circuits. Here we address the effects of the developmental transcription factor orthopedia (Otp) on hypothalamic development and function. We show that developmental mutations in the zebrafish paralogous gene otpa but not otpb affect both stress response and social preference. These behavioral phenotypes were associated with developmental alterations in oxytocinergic (OXT) neurons. Thus, otpa and otpb differentially regulate neuropeptide switching in a newly identified subset of OXT neurons that co-express the corticotropin-releasing hormone (CRH). Single-cell analysis revealed that these neurons project mostly to the hindbrain and spinal cord. Ablation of this neuronal subset specifically reduced adult social preference without affecting stress behavior, thereby uncoupling the contribution of a specific OXT cluster to social behavior from the general otpa-/- deficits. Our findings reveal a new role for Otp in controlling developmental neuropeptide balance in a discrete OXT circuit whose disrupted development affects social behavior.

BACKGROUND: The goal of this research was to study the weight of student variables related to homework (intrinsic homework motivation, perceived homework instrumentality, homework attitude, time spent on homework, and homework time management) and context (teacher feedback on homework and parental homework support) in the prediction of approaches to homework. METHOD: 535 students of the last three courses of primary education participated in the study. Data were analyzed with hierarchical regression models and path analysis. RESULTS: The results obtained suggest that students' homework engagement (high or low) is related to students' level of intrinsic motivation and positive attitude towards homework. Furthermore, it was also observed that students who manage their homework time well (and not necessarily those who spend more time) are more likely to show the deepest approach to homework. CONCLUSIONS: Parental support and teacher feedback on homework affect student homework engagement through their effect on the levels of intrinsic homework motivation (directly), and on homework attitude, homework time management, and perceived homework instrumentality (indirectly). Data also indicated a strong and significant relationship between parental and teacher involvement


ANTECEDENTES: el objetivo de esta investigación fue estudiar el peso de variables del alumno relacionadas con los deberes escolares (motivación intrínseca, percepción de utilidad, actitud, tiempo dedicado y gestión de ese tiempo) y del contexto (feedback del profesor y apoyo parental) en la predicción de los enfoques de trabajo en los deberes. MÉTODO: participaron 535 estudiantes de los tres últimos cursos de Primaria. Los datos fueron analizados en base a modelos de regresión jerárquica y path analysis. RESULTADOS: los resultados sugieren que la mayor o menor implicación del alumno en la realización de sus deberes escolares está relacionado con el grado de motivación intrínseca y una actitud positiva hacia ellos. También se observa que los estudiantes que gestionan bien su tiempo dedicado a los deberes (y no necesariamente los que dedican más tiempo) son quienes más profundamente trabajan en ellos. CONCLUSIONES: el apoyo parental y el feedback del profesor inciden sobre la implicación de los estudiantes a través de su efecto sobre los niveles de motivación intrínseca (directamente), y de la actitud, gestión del tiempo y percepción de utilidad (indirectamente). También sugieren una relación fuerte y significativa entre la implicación parental y de los profesores en los deberes escolares

Carotid body (CB) stimulation by hypercapnia causes a reflex increase in ventilation and, along with the central chemoreceptors, this prevents a potentially lethal systemic acidosis. Control over the CB chemoafferent output during normocapnia and hypercapnia most likely involves multiple neurotransmitters and neuromodulators including ATP, acetylcholine, dopamine, serotonin and adenosine, but the precise role of each is yet to be fully established. In the present study, recordings of chemoafferent discharge frequency were made from the isolated in vitro CB in order to determine the contribution of adenosine, derived specifically from extracellular catabolism of ATP, in mediating basal chemoafferent activity and responses to hypercapnia. Pharmacological inhibition of ecto-5'-nucleotidase (CD73), a key enzyme required for extracellular generation of adenosine from ATP, using α,ß-methylene ADP, virtually abolished the basal normocapnic single fibre discharge frequency (superfusate PO(2) ~ 300 mmHg, PCO(2) ~ 40 mmHg) and diminished the chemoafferent response to hypercapnia (PCO(2) ~ 80 mmHg). These effects were mimicked by the blockade of adenosine receptors with 8-(p-sulfophenyl) theophylline. The excitatory impact of adenosinergic signalling on CB hypercapnic sensitivity is most likely to be conferred through changes in cAMP. Here, inhibition of transmembrane, but not soluble adenylate cyclases, reduced normocapnic single fibre activity and inhibited the elevation evoked by hypercapnia by approximately 50 %. These data therefore identify a functional role for CD73 derived adenosine and transmembrane adenylate cyclases, in modulating the basal chemoafferent discharge frequency and in priming the CB to hypercapnic stimulation.

BACKGROUND: Homework is a universal practice used in schools, and is commonly related to academic achievement. According to literature, parental homework involvement has positive and negative aspects, depending on parents’ behaviors. METHOD: Assuming a phenomenographic perspective, this study examined 4th graders’ parents’ conceptions of their involvement in homework. With the purpose of mapping the parents’ various conceptions of homework involvement, 32 semi-structured interviews were conducted and analyzed. RESULTS: The results show that parents’ conceptions of homework involvement have a positive meaning, and focus primarily on the role played in the promotion of academic learning by (a) fostering their children’s autonomy, (b) exerting control over their learning, and (c) providing them with emotional encouragement (when children struggle with difficulties). CONCLUSIONS: Given that parents perceive their involvement in their children’s homework as important, it is necessary to promote parent-teacher collaboration and parent-training workshops to improve the quality of parental homework involvement.

BACKGROUND: Sedative-analgesics are often given to newborn infants and are known to affect many components of the autonomic nervous system. While morphine is most frequently used, α-2 adrenergic receptor agonists are being increasingly used in this population. Alpha-2 adrenergic receptors agonists also have anti-shivering properties which may make it a desirable drug to give to infants undergoing therapeutic hypothermia. The aim of this study was to systematically compare two different classes of sedative-analgesics, morphine, a µ-opioid receptor agonist, and clonidine an α-2 adrenergic receptor agonist on breathing, metabolism and core body temperature (CBT) in neonatal rodents. METHODS: Breathing parameters, oxygen consumption (VO2) and carbon dioxide production (VCO2), were measured prior to, 10 and 90 min after intraperitoneal (IP) administration of morphine (2, 10 or 20 mg/kg), clonidine (40, 200 or 400 µg/kg), or saline in Sprague-Dawley rat pups at postnatal day 7 (p7) while continuously monitoring CBT. RESULTS: Morphine reduced the respiratory rate, VO2 and VCO2 greater than clonidine at all dosages used (p<0.05, morphine vs. clonidine, for all metabolic and respiratory parameters). Furthermore, morphine induced prolonged respiratory pauses, which were not observed in animals treated with clonidine or saline. Morphine caused hypothermia which was dose dependent, while clonidine stabilized CBT in comparison to saline treated animals (p<0.0001). CONCLUSION: In the newborn rat, morphine causes profound respiratory depression and hypothermia while clonidine causes minimal respiratory depression and stabilizes CBT. All together, we suggest that clonidine promotes autonomic stability and may be a desirable agent to use in infants being treated with therapeutic hypothermia.
