Associated gas flaring has several consequences on the environment. This study was aimed at assessing the impact of gas flaring on soil enzymes and plant antioxidant activities from gas flare-bearing communities in Nigeria. Soil and plant samples were obtained from farmlands in Ukwa West and Izombe gas flaring sites, as well as unpolluted site from Olokoro (used as control). The level of activities of soil urease, dehydrogenase, phosphatases, plant antioxidant enzymes and lactate dehydrogenase (LDH) of selected plants (Gnetum africanum [GA], Piper guineense [PG], Gongronema latifolium [GL], Pterocarpus mildbraedii [PM]) were evaluated using standard methods. The results showed that the activities of urease were significantly higher (P < 0.05) in soil from Ukwa site than Izombe and the control soil. Dehydrogenase (DHA) and phosphatases recorded higher activities (P < 0.05) for Izombe soil than in Ukwa compared with the control. For plants, superoxide dismutase (SOD), glutathione S-transferase (GST) and glutathione peroxidase (GPx) recorded a significant (P < 0.05) higher activities in all the plants assayed from Ukwa site than Izombe and the control site. The activities of GPx from GA and PG plants at Izombe site were not significant (P > 0.05) when compared with the control, except for PM and GL which recorded a significant decrease (P < 0.05) in GPX and SOD activities, respectively. The activities of catalase enzyme also decreased significantly (P < 0.05) in all plants grown at Ukwa, while an increase was seen for GA and PM grown at Izombe compared with control. The overall variability in enzymes activities is an indication that soil ecosystem and plants are altered significantly by the stress load from the gas flaring pollutants which could serve as bio-indicators for assessing ecological risks and bioremediation.

Sickle cell disease is a group of diseases inherited through the gene and it affects the haemoglobin in the red blood cell. This study investigated the methanol seed extract of Buchholzia coriacea for possible in vitro anti-sickling effects and also determined the effect of Mucuna pruriens seed extract on the solubility and oxygen-binding rate of sickle cell haemoglobin. Sickle cell blood was collected from sickle cell disease patients with subsequent addition of 2% sodium metabisulphite to cause more sickling. Varying concentrations of the seed extracts (50%, 25%, 12.5% and 6.25%) were added to the pre-treated blood for these in vitro assays. The results showed that the extract of Buchholzia coriacea significantly (P < 0.05) inhibited sickling at all concentrations with the highest percentage inhibition of 73.3 ± 5.8, reversed sickled erythrocytes at all concentrations with the highest percentage reversal of 83.3 ± 5.8 and significantly (P < 0.05) inhibited polymerisation at all concentrations used in comparison to the parallel control. The extract of Mucuna pruriens seed significantly (P < 0.05) increased the solubility of sickle haemoglobin at 50%, 25%, 12.5% and 6.25% concentrations, increased Fe2+/Fe3+ ratio from 1.7 (control) to 12.2 (50% concentration) and reduced osmotic fragility (at 12.5% and 6.25% concentrations) when compared with parallel control. The results indicate the feasibility of the seed extracts as promising agents in the management of sickle cell disease.

OBJECTIVE: Cyanide is a highly toxic compound, and the consumption of products containing cyanide is a significant public health concern. Conversely, ß-carotene possesses essential nutritional attributes for human health, therefore the characterisation and quantification of both compounds in food products is fundamental. Herein, cyanide and ß-carotene levels in two flours produced from the roots of two varieties of cassava (Manihot esculenta crantz), namely UMUCASS-38(TMS 01/1371) and NR-8082, and their associated food products were detected and quantified. RESULTS: The cyanide content of NR-8082 and UMUCASS-38 flours was determined at 18.01 ± 0.01 ppm and 17.02 ± 0.02 ppm (mean ± SD), respectively. These flours contained significantly higher (p < 0.05) than the residual cyanide levels determined in the cookies and cake produced therefrom with levels of 10.00 ± 0.00 ppm and 7.10 ± 0.14 ppm (mean ± SD), respectively. The levels of ß-carotene determined in both the cake and cookie samples varied significantly (p < 0.05). The highest levels of ß-carotene at 6.53 ± 0.02 µg/g (mean ± SD) were determined in raw roots of UMUCASS-38. While NR-8082 levels of ß-carotene were less than UMUCASS-38 at 1.12 ± 0.02 µg/g (mean ± SD). Processing the roots into flour reduced the ß-carotene content to 4.78 ± 0.01 µg/g and 0.76 ± 0.02 µg/g (mean ± SD) in UMUCASS-38 and NR-8082 flours, respectively. Cookies and cake produced from flour derived from the UMUCASS-38 variety had (mean ± SD) 2.15 ± 0.01 µg/g and 2.84 ± 0.04 µg/g of ß-carotene, respectively.

BACKGROUND: Obesity is a growing public health problem arising from energy imbalance. The effect of 10% dietary incorporation of Vernonia amygdalina (VA) leaves into high-fat diets on some biological markers of adiposity and dyslipidaemia was investigated. METHODS: Experimental diets consisted of the following – CD (control diet); HFD (high-fat diet); and HFD- VA (HFD containing 10% Vernonia amygdalina leaves) supplementation. Fifteen male Wistar rats were randomly divided into three groups of five animals each. After twelve weeks of feeding, serum lipid profile, blood glucose concentrations, body weight, adiposity index, feed intake, fecal loss and relative organ weight were investigated. RESULTS: Vernonia amygdalina (VA) inhibited HFD-induced weight gain and adiposity in rats. HFD-induced obese rats showed a significant increase in the levels of serum TG and TC compared to rats on a normal diet. However, the levels of serum TG, TC, LDL-C in HFDVA rats reduced significantly relative to the levels in HFD rats. Our results indicate that HFDVA reversed fatty infiltration leading to decreased body weight and fat tissue mass in the rats. CONCLUSIONS: These results suggested that incorporation of Vernonia amygdalina into high-fat diets may have therapeutic potentials for obesity and related metabolic disorders. Further studies to explore its possibility as an alternative pharmacologic agent to treat obesity are warranted.

CONTEXT: Aqueous and methanol extracts of Vernonia amygdalina Del. (Asteraceae) (AEVA and MEVA, respectively) leaves are reported to possess anti-obesity properties, exerted through unknown mechanisms. OBJECTIVE: This study investigated the effects of AEVA and MEVA on relevant hormones and enzymes in high-fat diet (HFD)-induced obese rats. MATERIALS AND METHODS: Forty-two Wistar rats were placed into seven groups. The test groups received 100 mg/kg.bw AEVA (AEVA100), 500 mg/kg.bw AEVA (AEVA500), 50 mg/kg.bw MEVA (MEVA50) and 200 mg/kg.bw MEVA (MEVA200), respectively. The positive control (PC) group received 20 mg/kg.bw Orlistat, while the negative control (NeC) and normal control (NoC) groups received distilled water. The extracts were given orally daily for 12 weeks. Thereafter, the concentrations/activities of relevant hormones/enzymes in their sera were determined. RESULTS: Insulin concentrations (ng/ml) in the test groups ranged from 1.08 ± 0.01 (AEVA100) to 1.09 ± 0.01 (AEVA500). They were all similar (p > .05) to the NoC and PC controls. Leptin concentrations (pg/ml) in the test rats ranged from 0.02 ± 0.01 (AEVA500) to 0.03 ± 0.00 (MEVA50), and were all similar to the NoC group. The ghrelin concentrations of only the AEVA500 and MEVA200 groups were similar to those of the PC group (0.10 ± 0.01 pg/ml). AEVA100 and MEVA200 resulted in adiponectin concentrations (ng/ml) of the rats (0.27 ± 0.04 and 0.28 ± 0.04 respectively) that were similar to the PC group. The activities of lipoprotein lipase and the concentrations of intestinal amylase in the test rats were similar to values obtained for the control groups. CONCLUSION: Appetite regulation may be the mechanism through which the weight-loss properties of AEVA and MEVA are expressed.

The metabolism of yellow cassava (variety TMS 01/1368) was investigated in male albino rats fed a diet containing yellow cassava for 7 to 28 days. There were significant increases (P < 0.05) in total and free cyanide and thiocyanate in the sera and urine samples of the experimental rats compared with the control, significant increases (P < 0.05) in serum glucose, alanine aminotransaminase, aspartate aminotransaminase, and alkaline phosphatase levels of the experimental rats compared with the control, significant decreases (P < 0.05) in serum albumin of the experimental rats compared with the control, but no significant differences (P > 0.05) in the serum total proteins of the experimental rats compared with the control. The experimental rats treated for 7, 14, 21, or 28 days exhibited body weight decreases of 5.11%, 11.10%, 19.16%, and 24.18%, respectively, whereas the control group showed 9.17% gain in body weight. Total and free cyanide concentrations were detected in the liver, kidney, and heart of most of the rats in both the experimental and control groups, except for free cyanide in the control group that was not detected. Metabolism of the yellow cassava variety in experimental rats was capable of exposing the animals to cyanide, underscoring the need for its proper processing before consumption by humans.

AIM: This study aims to investigate the effect of unripe plantain (Musa paradisiaca) on markers of hepatic dysfunction in streptozotocin induced diabetic rats. METHODS: Blood glucose; relative liver weight (RLW); relative kidney weight (RKW); relative heart weight (RHW); relative pancreatic weight (RPW); serum and hepatic serum aspartate transaminase (AST), alanine transaminase (ALT), and alkaline phosphatase (ALP); serum amylase, lipase, total, and conjugated bilirubin; and chemical analysis of the test feed were determined using standard techniques. RESULTS: The diabetic rats had significant alteration (P < 0.05) of blood glucose; RLW; RKW; RPW; serum and hepatic AST, ALT, and ALP; serum total and conjugated bilirubin; and serum lipase activities compared with nondiabetic while these parameters were significantly improved (P < 0.05) in the rats fed unripe plantain. There were no significant differences (P > 0.05) in the RHW of the rats in the three groups, as well as significant decreases (P < 0.05) in the amylase levels of the diabetic rats compared with the nondiabetic, but there was nonsignificant increase (P > 0.05) in the amylase levels of the rats fed unripe plantain compared with the nondiabetic rats. The test and standard rat feeds contained considerable amount of proteins, carbohydrates, fats, phenols, and crude fiber. CONCLUSION: Amelioration of acute pancreatitis by unripe plantain could play a key role in its management of diabetes and related complications.

CONTEXT: Although unripe plantain (Musa paradisiaca) and ginger (Zingiber officinale) are used as single plants to manage diabetes mellitus in Nigeria, the possibility of combining them in a typical diabetic diet and the glycemic response elicited as a result of such combination has not been investigated. OBJECTIVE: To determine the effect of unripe plantain and ginger on serum total proteins, albumin, creatinine and urea levels of streptozotocin induced diabetic rats. METHODS: Twenty four male albino rats were used and were divided into 4 groups of 6 rats each. Group 1 (non-diabetic) received standard rat feeds; Group 2 (diabetic) received standard rat feeds; Group 3 received unripe plantain pellets and Group 4 received unripe plantain+ginger pellets. RESULTS: There were significant increases (P=0.045) of both serum urea and creatinine, but significant decreases (P=0.045) of both serum total protein and albumin levels, in Group 2 rats compared with Group 1. There were significant decreases (P=0.033) of both serum urea and creatinine levels of Group 3 and 4 rats compared with Group 2. In addition, there were significant increases of both serum total protein and albumin levels (P=0.033) in Group 3 rats compared with Group 2, but the comparison of serum total protein and albumin levels between Group 4 and Group 2 did not reach the significant level (P=0.056 and P=0.065 for serum total protein and albumin levels, respectively. CONCLUSION: Combination of unripe plantain and ginger at the ratio used in the management of renal dysfunction in diabetics was not very effective compared with unripe plantain alone.
