Avaliou-se o valor nutricional do capim-elefante verde colhido aos 56, 84 e 112 dias de idade, por meio do consumo e da digestibilidade em ovinos, da degradabilidade in situ e da cinética da fermentação ruminal. O capim-elefante colhido aos 56 dias proporcionou maiores valores de consumo da MS (1204,81g/dia), da EB (4910,41kcal/dia), da ED (3436,21kcal/dia) e da PB (130,90g/dia). Para o capim-elefante colhido aos 56 dias, ainda foram observados os maiores valores de digestibilidade da MS (70,64%), da EB (70,11%), da PB (68,96%), da FDN (71,4%) e do conteúdo ED (2,86Mcal/kg MS). Com o avanço da idade de corte, houve redução da degradabilidade potencial da MS (77,99% x 68,33%), da PB (84,80% x 56,52%) e da FDN (72,18% x 64,33%), para as idades de corte de 56 e 112 dias, respectivamente. O capim cortado aos 56 dias apresentou maior valor de produção cumulativa de gases (210,50mL/g de MS) e de degradabilidade da matéria seca (63,9%) após 96 horas de fermentação. O capim-elefante verde deve ser colhido aos 56 dias de rebrota em razão das características nutricionais apresentadas.(AU)


The nutritional value of fresh elephant grass harvested at 56, 84 and 112 days of age was evaluated by intake and digestibility in sheep, in situ degradability, and ruminal fermentation's kinetics. The elephant grass harvested at 56 days provided higher DM intake (1204.81g/day), GE (4910.41kcal/day), DE (3436.21kcal/day), and CP (130.90g/day) values. Furthermore, the highest DM (70.64%), GE (70.11%), DE (2.86%) and CP (68.96%) digestibility values were observed at 56 days. With the increase in the harvest age there was a reduction in the DM (77.99 x 68.33%), CP (84.80 x 56.52%) and NDF (72.18 x 64.33%) potential degradability in the treatments 56 and 112 days, respectively. Higher values of cumulative gas production (210,50mL/g DM) and DM degradability for elephant grass were observed at 56 days, after 96 hours of fermentation. The fresh elephant grass should be harvested at 56 days of regrowth due to the nutritional characteristics found in this study.(AU)

RESUMO

We generalize the linear discrete dimensional scaling approach for the repulsive Hubbard model to obtain a nonlinear scaling relation that yields accurate approximations to the ground-state energy in both two and three dimensions, as judged by comparison to auxiliary-field quantum Monte Carlo (QMC) data. Predictions are made for the per-site ground-state energies in two and three dimensions for n (filling factor) and U (Coulomb interaction) values for which QMC data are currently unavailable.

Alcoholic patients are more susceptible to Strongyloides stercoralis infection. The chronic use of alcohol raises the levels of endogenous corticosteroids, which regulates the development of larvae and stimulates the differentiation of rhabditiform into infective filariform larvae, thus inducing internal autoinfection. Therefore, early diagnosis is important to prevent severe strongyloidiasis. The aim of this study was to evaluate the efficacy of parasitological methods, according to the parasite load and the number of stool samples, for diagnosis of S. stercoralis infection, as well the peripheral blood eosinophil count in alcoholic patients. A total of 330 patients were included in this study. The diagnosis was established using three parasitological methods: agar plate culture, Baermann-Moraes method and spontaneous sedimentation. Peripheral eosinophilia was considered when the level was >600 eosinophils/mm3. The agar plate culture (APC) had the highest sensitivity (97.3%). However, the analysis of multiple samples increased the sensitivity of all parasitological methods. The sensitivities of the methods were influenced by the parasite load. When the larval number was above 10, the sensitivity of APC was 100%, while in spontaneous sedimentation the sensitivity reached 100% when the larval number was above 50. In the present study, 15.4% of alcoholic patients infected with S. stercoralis (12/78) had increased peripheral blood eosinophil count (above 600 eosinophils/mm3). For an efficient parasitological diagnosis of S. stercoralis infection in alcoholic patients, repeated examination by two parasitological methods must be recommended, including agar plate culture due to its higher sensitivity. Moreover, S. stercoralis infection was associated with eosinophilia, mostly in patients excreting up to 10 larvae/g faeces.

The genus Paracoccidioides is composed of thermal dimorphic fungi, causative agents of paracoccidioidomycosis, one of the most frequent systemic mycoses in Latin America. Mitochondria have sophisticated machinery for ATP production, which involves metabolic pathways such as citric acid and glyoxylate cycles, electron transport chain and oxidative phosphorylation. In addition, this organelle performs a variety of functions in the cell, working as an exceptional metabolic signalling centre that contributes to cellular stress responses, as autophagy and apoptosis in eukaryotic organisms. The aim of this work was to perform a descriptive proteomic analysis of mitochondria in Paracoccidioides lutzii yeast cells. After mitochondria fractionation, samples enriched in mitochondrial proteins were digested with trypsin and analysed using a NanoUPLC-MSE system (Waters Corporation, Manchester, UK). Ours results revealed that the established protocol for purification of mitochondria was very effective for P. lutzii, and 298 proteins were identified as primarily mitochondrial, in our analysis. To our knowledge, this is the first compilation of mitochondrial proteins from P. lutzii, to date. Copyright © 2016 John Wiley & Sons, Ltd.

A leishmaniose visceral (LV) é uma doença infecciosa crônica frequentemente fatal causada pela Leishmania infantum chagasi nas Américas. A enfermidade pode acometer vários órgãos, determinando diferentes manifestações clínicas. Contudo o envolvimento do coração raramente tem sido reportado em cães infectados por Leishmania sp. Dessa forma, descreve-se um caso de miocardite crônica com repercussões clínicas e patológicas em um cão naturalmente infectado por Leishmania infantum chagasi. A positividade para Leishmaniose Visceral foi determinada pela presença de anticorpos antiLeishmania sp. nos testes sorológicos (RIFI, ELISA e DPP) e confirmada por visualização de formas amastigotas de Leishmania sp. em punção aspirativa do linfonodo poplíteo. O exame cardiovascular revelou alterações radiográficas, eletrocardiográficas, na pressão arterial e nos biomarcardores cardíacos. Após eutanásia, amostras de tecido cardíaco foram avaliadas histologicamente e submetidas à imunomarcação, onde foi observado infiltrado mononuclear (plasmo-histiolinfocitário), com presença de estruturas arredondadas de coloracão marrom-amareladas (imunomarcadas), indicando formas amastigotas de Leishmania infantum chagasi no miocárdio. Os aspectos etiopatogênicos da leishmaniose visceral sobre o miocárdio neste caso podem estar relacionados tanto à presença do parasita quanto à resposta "reacional inespecífica" do tecido à agressão do parasita no organismo. Todavia ainda não se sabem se as cepas de Leishmania infantum chagasi da região semiárida paraibana apresentam algum tropismo por tecido cardíaco ou se induzem a reação imunológica cruzada, com implicações clínicas.


Visceral leishmaniasis (VL) is a chronic, often fatal infectious disease caused by Leishmania infantum chagasi in the Americas. The disease can affect many organs and may express different clinical forms. However, the involvement of the heart has rarely been reported in dogs infected by Leishmania sp. Thus, we describe a case of chronic myocarditis with clinical and pathological effects in a dog naturally infected by Leishmania infantum chagasi. Positivity for Visceral Leishmaniasis was determined by the presence of anti-Leishmania sp. in serological tests (IFAT, ELISA and DPP) and confirmed by visualization of amastigote forms of the parasite in the popliteal lymph node aspiration samples. The cardiovascular clinical examination showed changes in the radiographic, ECG, blood pressure and heart biomarkers. After euthanasia, heart tissue samples were histologically examined and underwent our immunohistochemistry assessment, where mononuclear infiltrate was observed (plasma cells, lymphocytes and macrophages) with the presence of rounded brownish-yellow (immunomarked) cells, indicating amastigotes of Leishmania infantum chagasi within the myocardium. The etiopathogenic aspects of visceral leishmaniasis in the myocardium in this case may be related either to the presence of the parasite or the "nonspecific reactive" response of the tissue attributable to the aggression of the parasite in the body. However, it is not known if the strains of Leishmania infantum chagasi found in the semi-arid of Paraíba have some tropism for cardiac tissue or if they induce immunological cross-reaction with clinical implications.

The prevalence of Toxoplasma gondii in free-ranging chickens is a good indicator of the prevalence of T. gondii oocysts in the soil because chickens feed from the ground. The prevalence of T. gondii in 152 free-range chickens (Gallus domesticus) from 22 municipalities in 7 northeastern states (Pernambuco, Rio Grande do Norte, Maranhão, Bahia, Ceará, Sergipe, and Alagoas) of Brazil was determined. Antibodies to T. gondii were assayed by the modified agglutination test (MAT); 81 (53.3 %) chickens had titers of 1:5 in 26, 1:10 in 9, 1:20 in 4, 1:40 in 1, 1:80 in 6, 1:160 in 6, 1:320 in 13, 1:640 in 6, 1:1,280 in 3, 1:2,560 in 6, and 1:5,120 or higher in 1. Hearts and brains of 81 seropositive chickens were bioassayed individually in mice. Toxoplasma gondii was isolated from 23 chickens with MAT titers of 1:5 or higher; the isolates were designated TgCKBr165-187. Five isolates killed all infected mice. Results indicate widespread contamination of rural environment in Brazil with T. gondii oocysts.

Goats are economically important in many countries, and little is known of caprine toxoplasmosis in Brazil. Antibodies to Toxoplasma gondii were assayed in the sera of 143 goats from 3 Brazilian states, using modified agglutination test (MAT titer > or = 1:25); 46 (32.2%) tested positive. Samples of brain, heart, diaphragm, and masseter of seropositive animals were pooled, digested in pepsin, and bioassayed in mice. Viable T. gondii specimens were isolated from tissue homogenates of 12 goats; the isolates were designated TgGtBr1-12. Ten of the 12 isolates killed 100% of infected mice, indicating that goats can harbor mouse-virulent T. gondii and, hence, can serve as a source of infection for humans.

Sheep are important in the epidemiology of Toxoplasma gondii infection but little is known of ovine toxoplasmosis in Brazil. Antibodies to T. gondii were assayed in sera of 495 sheep from 36 counties of São Paulo State, Brazil, using the modified agglutination test (MAT titer > or =1:25) and found in 120 (24.2%). Samples of brain, heart, and diaphragm of 82 seropositive sheep were pooled, digested in pepsin, and bioassayed in mice. Toxoplasma gondii was isolated from tissue homogenates of 16 sheep and the isolates were designated TgShBr1-16. Six of the 16 T. gondii isolates killed 100% of infected mice. Results indicate that asymptomatic sheep can harbor mouse-virulent T. gondii, and hence they can serve as a source of infection for humans.

RESUMO

By shifting the reference system for the local-density approximation (LDA) from the electron gas to other model systems, one obtains a new class of density functionals, which by design account for the correlations present in the chosen reference system. This strategy is illustrated by constructing an explicit LDA for the one-dimensional Hubbard model. While the traditional ab initio LDA is based on a Fermi liquid (the three-dimensional interacting electron gas), this one is based on a Luttinger liquid. First applications to inhomogeneous Hubbard models, including one containing a localized impurity, are reported.

RESUMO

Apresentamos um metodo colorimetrico direto, simples, sensivel, preciso e exato para a determinaçao do fosfato inorganico em amostras de soro e urina. Os reagentes sao estaveis, facilmente preparados e economicos. A reaçao se passa entre o fosfato e a soluçao molibdico-sulfurica para formar o complexo de fosfomolibdato, que e reduzido a azul de molibdenio pelo sulfato de p-metilaminofenol (ELON) na presença de dietanolamina, um potente solubilizante do precipitado formado. Os resultados de dosagens obtidos com o novo metodo apresentam excelente correlaçao com os do metodo de desproteinizaçao de GOMORY (6-7) e com 2 tecnicas diretas industrializadas
