Environmental exposures and gene-exposure interactions are the major causes of some diseases. Early-life exposome studies are needed to elucidate the role of environmental exposures and their complex interactions with biological mechanisms involved in childhood health. This study aimed to determine the contribution of early-life exposome to DNA damage and the modifying effect of genetic polymorphisms involved in air pollutants metabolism, antioxidant defense, and DNA repair. We conducted a cohort study in 416 Colombian children under five years. Blood samples at baseline were collected to measure DNA damage by the Comet assay and to determine GSTT1, GSTM1, CYP1A1, H2AX, OGG1, and SOD2 genetic polymorphisms. The exposome was estimated using geographic information systems, remote sensing, LUR models, and questionnaires. The association exposome-DNA damage was estimated using the Elastic Net linear regression with log link. Our results suggest that exposure to PM2.5 one year before the blood draw (BBD) (0.83, 95 %CI: 0.76; 0.91), soft drinks consumption (0.94, 0.89; 0.98), and GSTM1 null genotype (0.05, 0.01; 0.36) diminished the DNA damage, whereas exposure to PM2.5 one-week BBD (1.18, 1.06; 1.32), NO2 lag-5 days BBD (1.27, 1.18; 1.36), in-house cockroaches (1.10, 1.00; 1.21) at the recruitment, crowding at home (1.34, 1.08; 1.67) at the recruitment, cereal consumption (1.11, 1.04; 1.19) and H2AX (AG/GG vs. AA) (1.44, 1.11; 1.88) increased the DNA damage. The interactions between H2AX (AG/GG vs. AA) genotypes with crowding and PM2.5 one week BBD, GSTM1 (null vs. present) with humidity at the first year of life, and OGG1 (SC/CC vs. SS) with walkability at the first year of life were significant. The early-life exposome contributes to elucidating the effect of environmental exposures on DNA damage in Colombian children under five years old. The exposome-DNA damage effect appears to be modulated by genetic variants in DNA repair and antioxidant defense enzymes.

Objetivos: Determinar la predisposición a recibir esta vacuna contra SARS-CoV-2. Materiales y métodos: Se aplicó un diseño observacional de corte transversal en la población adulta del Paraguay entre mayo y octubre 2022. se aplicó el cuestionario de Kotta et al previamente validado, el cual fue difundido por redes sociales. Resultados: Se incluyeron 303 encuestados, con edad media 34 ± 12 años y predominio del sexo femenino (64,0%). En la muestra, 51,8% padeció COVID-19 y 97,3% ya recibió al menos una dosis de la vacuna. Se detectó que 58,4% aceptada la vacuna, 17,8% vacilaba en recibirla y 23,7% la rechazaba. La aceptación fue más frecuente en los varones (p 0,05). Conclusión: En el momento epidemiológico de disponibilidad universal de la vacuna y habiendo aún sujetos afectados por COVID-19, el rechazo a la misma fue 23,7%.
Objectives: To determine the predisposition to receive this vaccine against SARS-CoV-2. Materials and methods: An observational cross-sectional design was applied in the adult population of Paraguay between May and October 2022. The previously validated questionnaire of Kotta et al was applied, which was disseminated through social networks. Results: 303 respondents were included, with a mean age of 34 ± 12 years and predominance of the female sex (64.0%). In the sample, 51.8% suffered from COVID-19 and 97.3% have already received at least one dose of the vaccine. It was detected that 58.4% accepted the vaccine, 17.8% hesitated to receive it and 23.7% rejected it. Acceptance was more frequent in males (p 0.05). Conclusion: At the epidemiological moment of universal availability of the vaccine and with subjects still affected by COVID-19, rejection of it was 23.7%.

Equus members exhibit very divergent karyotype, genetic plasticity, and significant differences in their reproductive physiology. Despite the fact that somatic cell nuclear transfer and intracytoplasmic sperm injection (ICSI) has gained relevance in the last few years in horses, few reports have been published exploring ovum pick up (OPU) and in vitro maturation (IVM) of cumulus-oocyte complexes (COCs) in donkeys. Yet, some donkey species and breeds are considered endangered, and these assisted-reproductive technologies could help to preserve the genetic of valuable individuals. In this study, we tested the hypothesis that supplementation with jenny preovulatory follicular fluid (PFF) during IVM could improve oocyte developmental competence in the donkey. For this, in vitro nuclear maturation rates, cumulus cell expansion, and embryo development after ICSI of donkey COCs matured in culture media supplemented with fetal bovine serum (FBS) or donkey PFF, with a known metabolomic profile, were assessed. Time-lapse imagining was performed after ICSI of horse and donkey oocytes. Eight OPU sessions were done in five jennies with an average recovery rate of 69.2% (n = 45 COCs). Although lower cumulus cells expansion was observed in oocytes of PFF group (P = 0.0010), no significant differences were described in nuclear maturation rates and preimplantation embryo development between groups. Donkey ICSI embryos showed similar morphokinetics to horse ICSI embryos. Our study shows that supplementing IVM media with FBS or donkey PFF supports nuclear maturation and early preimplantation embryo development after ICSI in donkeys. To our knowledge, the present study is the first report of ICSI, time-lapse imaging and in vitro blastocyst production in donkey.

Tumor organoids are defined as self-organized three-dimensional assemblies of heterogeneous cell types derived from patient samples that mimic the key histopathological, genetic, and phenotypic characteristics of the original tumor. This technology is proposed as an ideal candidate for the evaluation of possible therapies against cancer, presenting advantages over other models which are currently used. However, there are no reports in the literature that relate the techniques and material development of tumor organoids or that emphasize in the physicochemical and biological properties of materials that intent to biomimicry the tumor extracellular matrix. There is also little information regarding the tools to identify the correspondence of native tumors and tumoral organoids (tumoroids). Moreover, this paper relates the advantages of organoids compared to other models for drug evaluation. A growing interest in tumoral organoids has arisen from 2009 to the present, aimed at standardizing the process of obtaining organoids, which more accurately resemble patient-derived tumor tissue. Likewise, it was found that the characteristics to consider for the development of organoids, and therapeutic responses of them, are cell morphology, physiology, the interaction between cells, the composition of the cellular matrix, and the genetic, phenotypic, and epigenetic characteristics. Currently, organoids have been used for the evaluation of drugs for brain, lung, and colon tumors, among others. In the future, tumor organoids will become closer to being considered a better model for studying cancer in clinical practice, as they can accurately mimic the characteristics of tumors, in turn ensuring that the therapeutic response aligns with the clinical response of patients.

In the agricultural sector, companies involved in the production of plastic greenhouses are currently searching for a suitable covering adapted for every climate in the world. For this purpose, this research work has determined the chemical, radiometric and mechanical properties of 53 polymeric films samples from Europe and South America. The chemical tests carried out with these samples were elemental analysis (C, H and N) and FT-IR spectrometry. The radiometric properties here studied were the transmission, absorption and reflection coefficients along the spectrum between 300 and 1100 nm. For the mechanical properties, tensile strength, tear strength and dart impact strength, tests were carried out. Finally, all these data were collected, and a multivariate statistical analysis was carried out using the SPSS statistical to group the samples into statistical groups adapted to specific climatic regions. The elemental analysis and FT-IR spectrometry allowed group the samples into nine groups. The samples were grouped according to their chemical (elemental analysis), radiometric and mechanical properties by multivariate analysis. The dendrogram separated five very different groups in terms of number of samples. These groups have specific chemical, radiometric and mechanical characteristics that separate them from the rest. These groups make it possible to narrow down the applications and correlate with the radiometric properties to see in which geographical area of the world they are most effective in increasing yields and achieving higher quality production.

Peru has been severely affected by the COVID-19 pandemic. By January 2022, Peru had surpassed 200â¯000 COVID-19 deaths, constituting the highest death rate per capita worldwide. Peru has had several limitations during the pandemic: insufficient testing access, limited contact tracing, a strained medical infrastructure, and many economic hurdles. These limitations hindered the gathering of accurate information about infected individuals with spatial resolution in real time, a critical aspect of effectively controlling the pandemic. Wastewater monitoring for SARS-CoV-2 RNA offered a promising alternative for providing needed population-wide information to complement health care indicators. In this study, we demonstrate the feasibility and value of implementing a decentralized SARS-CoV-2 RNA wastewater monitoring system to assess the spatiotemporal distribution of COVID-19 in three major cities in Peru: Lima, Callao, and Arequipa. Our data on viral loads showed the same trends as health indicators such as incidence and mortality. Furthermore, we were able to identify hot spots of contagion within the surveyed urban areas to guide the efforts of health authorities. Viral decay in the sewage network of the cities studied was found to be negligible (<2%). Overall, our results support wastewater monitoring for SARS-CoV-2 as a valuable and cost-effective tool for monitoring the COVID-19 pandemic in the Peruvian context.

RESUMEN: El objetivo del estudio fue comparar el déficit propioceptivo a través del Joint position sense (JPS) y Force steadiness en pacientes con reconstrucción del ligamento cruzado anterior (LCA) injerto hueso-tendón patelar-hueso (HTH) 6 a 12 meses postcirugía. Participaron 15 pacientes (13 hombres y 2 mujeres, 25,5 ± 1,3 años) con reconstrucción de LCA con autoinjerto HTH y 20 personas sin lesión del LCA (19 hombres y 1 mujer, 24,1 ± 0,8 años). Para evaluar la sensación de posición de la articulación de la rodilla se midió la Joint position sense (JPS) en tres rangos: 0°-30°, 31°-60° y 61°-90° y la sensación de fuerza del cuádriceps fue evaluada con la prueba Force steadiness (FS) al 15 % de la contracción voluntaria máxima (CVM), ambas pruebas realizadas 6 a 12 meses post cirugía. Los resultados mostraron que no hubo diferencias estadísticamente significativas en la sensación de la posición articular (JPS 0°-30°) (p=0.564) y 31°-60° (p=0.681), mientras que en el rango 61°-90° (p=0.003) existieron diferencias estadísticamente significativas. En las mediciones de sensación de fuerza del cuádriceps (FS al 15 % CVM) entre los pacientes operados de LCA técnica HTH y el grupo control no hubo diferencias estadísticas (p= 0.987) La sensación de la fuerza del cuádriceps medida con la prueba FS al 15 % CVM no presentaría déficit entre los 6 a 12 meses en pacientes post operados de LCA al ser comparados con sujetos sin lesión ni cirugía de este ligamento. Se concluye que la sensación de la posición articular medida con la prueba JPS en en tres rangos articulares de pacientes con reconstrucción de LCA injerto HTH 6 a 12 meses post cirugía sólo mostró alteraciones en el rango de 61°- 90° al ser comparado con el grupo control, lo cual indica que la sensación de la posición articular presenta un déficit en este rango específico.
SUMMARY: The aim of the study was to compare the proprioceptive deficit through the Joint position sense (JPS) and Force steadiness in patients with anterior cruciate ligament (ACL) bone-patellar tendon-bone graft (PTH) reconstruction 6 to 12 months post-surgery. Fifteen patients (13 men and 2 women, 25.5 ± 1.3 years) with ACL reconstruction with HTH autograft and 20 persons without ACL injury (19 men and 1 woman, 24.1 ± 0.8 years) participated. To assess knee joint position sensation, Joint position sense (JPS) was measured in three ranges: 0°-30°, 31°- 60° and 61°-90° and quadriceps strength sensation was assessed with the Force steadiness (FS) test at 15 % of maximal voluntary contraction (MVC), both tests performed 6 to 12 months post surgery. The results showed that there were no statistically significant differences in joint position sensation (JPS 0°-30°) (p=0.564) and 31°-60° (p=0.681), while in the range 61°-90° (p=0.003) there were statistically significant differences. In the quadriceps strength sensation measurements (FS at 15 % CVM) between the patients operated on ACL HTH technique and the control group there were no statistical differences (p= 0.987). The quadriceps strength sensation measured with the FS test at 15 % CVM would not present a deficit between 6 to 12 months in post- operated ACL patients when compared to subjects without injury or surgery of this ligament. It is concluded that the joint position sensation measured with the JPS test in three joint ranges of patients with ACL reconstruction HTH graft 6 to 12 months post surgery only showed alterations in the range of 61°- 90° when compared to the control group, indicating that the joint position sensation presents a deficit in this specific range.

High-throughput sequencing studies have shown the important role microbial communities play in the male reproductive tract, indicating differences in the semen microbial composition between fertile and infertile males. Most of these studies were made on human beings but little is known regarding domestic animals. Seminal bacteria studies made in stallions mostly focus on pathogenic bacteria and on their impact on reproductive technology. However, little is known about stallion commensal seminal microflora. That ultimately hinders our capacity to associate specific bacteria to conditions or seminal quality. Therefore, the aim of this study was to characterize the seminal microbial composition of 12 healthy, fertile stallion using next-generation sequencing. Hypervariable region V3 was chosen for bacterial identification. A total of nine phyla was detected. The most abundant ones were Bacteroidetes (46.50%), Firmicutes (29.92%) and Actinobacteria (13.58%). At family level, we found 69 bacterial families, but only nine are common in all samples. Porphyromonadaceae (33.18%), Peptoniphilaceae (14.09%), Corynebacteriaceae (11.32%) and Prevotellaceae (9.05%) were the most representative ones, while the Firmicutes phylum displayed the highest number of families (23, a third of the total). Samples showed high inter-subject variability. Findings previously described in other species notably differ from our findings. Families found in human such as Lactobacillaceae, Staphylococcaceae and Streptococcaceae only represented a 0.00%, 0.17% and 0.22% abundance in our samples, respectively. In conclusion, Porphyromonadaceae, Prevotellaceae, Peptoniphilaceae and Corynebacteriaceae families are highly represented in the seminal microbiome of healthy, fertile stallions. A high variation among individuals is also observed.

Despite the efforts focused on manufacturing biological engineering scaffolds for tissue engineering and regenerative medicine, a biomaterial that meets the necessary characteristics for these applications has not been developed to date. Bacterial nanocellulose (BNC) is an outstanding biomaterial for tissue engineering and regenerative medicine; however, BNC's applications have been focused on two-dimensional (2D) medical devices, such as wound dressings. Given the need for three-dimensional (3D) porous biomaterials, this work evaluates two methods to generate (3D) BNC scaffolds. The structural characteristics and physicochemical, mechanical, and cell behaviour properties were evaluated. Likewise, the effects of the pore size and surface area in the mechanical performance of BNC biomaterials and their cell response in a fibroblast cell line are discussed for the first time. In this study, a new method is proposed for the development of 3D BNC scaffolds using paraffin wax. This new method is less time-consuming, more robust in removing the paraffin and less aggressive toward the BNC microstructure. Moreover, the biomaterial had regular porosity with good mechanical behaviour; the cells can adhere and increase in number without overcrowding. Regarding the pore size and surface area, highly interconnected porosities (measuring approximately 60 µm) and high surface area are advantageous for the biomaterial's mechanical properties and cell behaviour. © 2018 Wiley Periodicals, Inc. A new analysis was used to evaluate the available number of sperm from each subpopulation after each centrifugation protocol. Frozen/thawed semen samples were divided into the following after-thawing treatments: uncentrifuged control (UDC), sperm washing (SW) and two colloid centrifugation procedures (Equipure, SLC-E, and Androcoll, SLC-A). Percentage of total and progressive motility (TM and PM), as well as sperm motility kinematics, distribution of motile sperm subpopulations, and recovery rates, were statistically compared among treatments. The SLC treatments showed higher (P < 0.001) TM and PM than UDC and SW. Following each SLC procedure, different percentages of the subpopulation with the most vigorous and progressive sperm (sP4) were obtained. SLC-A recovered a larger number of sperm belonging to sP4 than SLC-E, but not significantly higher than SW. From a practical point of view, sperm washing, the standard centrifugation procedure for equine semen processing, recovered the same amount of fast and progressive sperm as colloid centrifugation, apparently the best treatment according to traditional analysis. In conclusion, samples processed by SLC have higher motility percentages than SW and UDC but, after combining the available number of sperm, SLC and SW techniques are equally efficient in recovering sperm from the most vigorous, fast and progressive motile subpopulation (sP4).

The presence of cyanobacterial blooms and cyanotoxins in water presents a global problem due to the deterioration of ecosystems and the possibility of poisoning in human and animals. Microcystin LR is the most widely distributed cyanotoxin and liver cells are its main target. In the present study, HepG2 cells were used to determine DNA damage of three crude extracts of cyanobacterial blooms containing MC-LR, through comet assay. The results show that all extracts at a concentration of 500⯵gâ¯mL-1 caused low damage in hepatocytes exposed for 24â¯h, but produced total mortality even at low concentrations at 48â¯h. Moreover, balloons corresponding to cell apoptosis were found. Through HPLC/MS, MC-LR was detected in all samples of cyanobacterial blooms at concentrations of (5,65⯵gâ¯ml-1) in sample 1, (1,24⯵gâ¯ml-1) in sample 2 and (57,29⯵gâ¯ml-1) in sample 3. In addition, in all samples high molecular weights peaks were detected, that may correspond to other microcystins. Besides, the cytotoxic effect of a cyanobacterial bloom and some of its chromatographic fractions from the crude extracts were evaluated in U-937, J774, Hela and Vero cell lines, using the enzymatic micromethod (MTT). The highest toxicity was detected in U-937â¯cells (LC50â¯=â¯29.7⯵gâ¯mL-1) and Vero cells (LC50â¯=â¯39.7⯵g mL-1). Based on these results, it is important to remark that genotoxic and cytotoxicity assays are valuable methods to predict potential biological risks in waters contaminated with blooms of cyanobacteria, since chemical analysis can only describe the presence of cyanotoxins, but not their biological effects.

La competencia léxica como parte de la competencia comunicativa contempla una serie de elementos entre los cuales el dominio de la valencia léxica constituye uno de los factores que permite distinguir los hablantes nativos de un idioma de aquellas personas que lo estudian como idioma extranjero o segunda lengua. Este artículo expone algunas consideraciones metodológicas para la implementación de un folleto sobre valencia léxica en Inglés con Fines Médicos para el perfeccionamiento del tecnolecto de los estudiantes de 5to año de la carrera de Medicina, elaborado a partir de las necesidades cognoscitivas expresadas por parte de alumnos y profesores. El folleto facilita la adquisición y práctica de términos que enriquecen el tecnolecto; de esta forma se contribuye a un aprendizaje contextualizado, rápido y más eficaz por parte de los estudiantes. Además del carácter práctico de la propuesta, su fundamentación teórica puede servir de referencia para la elaboración de otros folletos(AU)
Lexical competence, as part of communicative competence, takes into account a series of elements among which mastering lexical valence is one of the factors allowing to distinguish native language speakers from people who study a foreign or a second language. The article presents methodological considerations for the implementation of a booklet on lexical valence in English for Medical Purposes which, developed from students and professors' cognitive needs, aims to improve fifth year medical students' technolect. It makes terms enriching technolect easier to practice and to acquire; therefore contributing to a contextualized, fast and more efficient learning by students. Apart from the practical character of the proposal, its theoretical basis can be used as reference for the development of other booklets(AU)

INTRODUCTION:: Cutaneous leishmaniasis (CL) is a tropical disease that affects millions of individuals worldwide. The current drugs for CL may be effective but have serious side effects; hence, alternatives are urgently needed. Although plant-derived materials are used for the treatment of various diseases in 80% of the global population, the validation of these products is essential. Gelatin capsules containing dried Artemisia annua leaf powder were recently developed as a new herbal formulation (totum) for the oral treatment of malaria and other parasitic diseases. Here, we aimed to determine the usefulness of A. annua gel capsules in CL. METHODS:: The antileishmanial activity and cytotoxicity of A. annua L. capsules was determined via in vitro and in vivo studies. Moreover, a preliminary evaluation of its therapeutic potential as antileishmanial treatment in humans was conducted in 2 patients with uncomplicated CL. RESULTS:: Artemisia annua capsules showed moderate in vitro activity in amastigotes of Leishmania (Viannia) panamensis; no cytotoxicity in U-937 macrophages or genotoxicity in human lymphocytes was observed. Five of 6 (83.3%) hamsters treated with A. annua capsules (500mg/kg/day) for 30 days were cured, and the 2 examined patients were cured 45 days after initiation of treatment with 30g of A. annua capsules, without any adverse reactions. Both patients remained disease-free 26 and 24 months after treatment completion. CONCLUSION:: Capsules of A. annua L. represent an effective treatment for uncomplicated CL, although further randomized controlled trials are needed to validate its efficacy and safety.

INTRODUCTION: During malaria infection, both parasite and host are under the effects of oxidative stress due to the increased production of reactive oxygen species, which can induce DNA damage by its genotoxic effects. OBJECTIVE: To evaluate genotoxic effects in human lymphocytes in a cohort of patients with malaria from Medellin and Quibdó. METHODS: We performed an observational cross sectional study in 100 individuals with malaria and 100 healthy controls. Patients infected with Plasmodium consulting the Institute Colombiano of Medicina Tropical of Medellin and the Hospital Ismael Roldán Valencia of Quibdó were included. Genotoxic effects (genetic damage) was analysed by electrophoresis using alkaline single cell gel (Commet assay). RESULTS: The average of tail length of malaria samples (26.9±9.8) was significantly higher than of controls (14.8±3.2) (p<0.01). CONCLUSION: In our study population, malaria infection was associated with increased genotoxicity, while other variables such as smoking, antimalarial treatment, and occupation were not.
