Consumption of sucrose-sweetened drinks (SSDs) during pregnancy and breastfeeding can lead to various health and metabolism issues, but the potential impact on neurodevelopment and long-term effects remains unclear. This study aims to examine how maternal consumption of SSDs during gestation and lactation influences anxiety and depression-related behavior in adult offspring. Adult female CD-1 mice were randomly assigned to a control group (CG) or a sucrose group (SG) 2 weeks before gestation. The SG had 2 h of access to an SSD (15% w/w, 0.6 kcal/ml) for 2 weeks before mating, during pregnancy, and throughout lactation, totaling 8 weeks. Adult offspring were then evaluated for depressive-related behaviors and anxiety-related behaviors. Our findings reveal that perigestational consumption of SSDs does not lead to offspring presenting behaviors related to depression, but it does increase swimming behavior. However, maternal consumption of SSDs could impact the fighting response due to a diminished motivational component. In contrast, perigestational consumption of SSDs has apparent effects on anxiety-related behavior. Furthermore, female offspring appeared to be particularly vulnerable, exhibiting a higher anxiety index compared with controls. These findings indicate that females could be more vulnerable to the effects of maternal consumption of SSDs, being more susceptible to the presence of anxiety-related behaviors.

Maternal nutrition and physical activity during pregnancy and lactation can modify offspring development. Here, we investigated the effects of maternal aerobic exercise (AE) and Western diet (WD) on brain development, cognitive flexibility, and memory of progenies. Sixteen adult female mice were assigned to AE or sedentary groups (SED) and fed a balanced diet (BD) or WD. Offspring were categorized into four groups: WD + AE, WD + SED, BD + AE, and BD + SED. The AE group showed enhanced spontaneous alternation in the T-maze test, suggesting an improvement in working memory and tasks related to cognitive flexibility. The novel object recognition (NOR) test showed that the BD + AE pups improved their absolute discrimination and discrimination index at 24 h, which suggests a delay in memory consolidation without affecting evocation. WD + SED showed poorer discrimination and recognition memory. The pups of AE mothers had better efficiency in short-term memory, whereas WD offspring showed low performance in long-term memory. Interestingly, exercise improved tasks related to cognitive flexibility, regardless of the diet. These findings indicate that maternal diet and physical activity modify offspring development and suggest that maternal AE during pregnancy could be a beneficial intervention to counteract the adverse effects of WD by improving spatial memory and cognitive flexibility in offspring.

The early stage of development is a vulnerable period for progeny neurodevelopment, altering cytogenetic and correct cerebral functionality. The exposure High-Fat Diet (HFD) is a factor that impacts the future mental health of individuals. This review analyzes possible mechanisms involved in the development of mood disorders in adulthood because of maternal HFD intake during gestation and lactation, considering previously reported findings in the last five years, both in humans and animal models. Maternal HFD could induce alterations in mood regulation, reported as increased stress response, anxiety-like behavior, and depressive-like behavior. These changes were mostly related to HPA axis dysregulations and neuroinflammatory responses. In conclusion, there could be a relationship between HFD consumption during the early stages of life and the development of psychopathologies during adulthood. These findings provide guidelines for the understanding of possible mechanisms involved in mood disorders, however, there is still a need for more human clinical studies that provide evidence to improve the understanding of maternal nutrition and future mental health outcomes in the offspring.

Maternal care in the rat is an ancient behavioral response to specific multisensory inputs widely integrated in a complex forebrain, limbic and brain stem network to meet the basic needs of the young. Early undernutrition interferes with the morphofunctional organization of the brain, including maternal circuitry. The late-emerging effects of pre- and neonatal undernutrition on nest building and pup retrieval by lactating Wistar rats were correlated with dendritic arbor and perikaryon measurements (Golgi-Cox) in layer II pyramidal neurons of the anterior cingulate cortex, layer III pyramidal neurons of the medial prefrontal cortex and multipolar basolateral amygdala neurons examined on lactation days 4 and 12. In the underfed group, pregnant F0 dams received different percentages of a balanced diet. After birth, prenatally underfed (F1) pups continued the undernutrition by remaining with a nipple-ligated mother for 12 h. Weaning occurred at 25 days of age, and pups were subsequently provided an ad libitum diet. At 90 days of age, F1 dams were maternally tested. Early underfed dams showed significant reductions in nest building and prolonged retrieval latencies for grasping pups by inappropriate body areas. The behavioral alterations were concurrent with highly significant reductions in the somatic cross-sectional area and perimeter, spine density and dendritic crossings of cingulate cells and medial prefrontal cortical pyramids, as well as smaller effects on amygdala neurons. The anatomical findings suggest different postsynaptic organizations that may affect the neuronal excitability stages for the integration and encoding of cues triggering the altered maternal response components of early underfed dams.
