RESUMO

Riparian tetragnathid spiders are used as biosentinels of aquatic contamination because they are specialized feeders of aquatic emergent insects and are also prey items for terrestrial predators (e.g., birds). Analysis of both trophic position (e.g., stable nitrogen isotopes) and contaminant concentrations are needed to utilize tetragnathids as biosentinels, which can present challenges when collecting enough biomass to reach analytical detection limits, due to their relatively small size. The purpose of this study was to investigate the impacts of a controlled diet source on spider biomass, egg laying and stable isotope values (δ13C and δ15N). Diet significantly influenced the biomass and egg laying of tetragnathids, with some spiders losing up to 50% of their biomass in a single egg-laying event. δ13C had a faster turnover rate in the whole-body of spiders compared to legs, which is important, as spider legs are presently used as surrogates for whole-body δ13C values.

Environmental exposure data are a key component of chemical and ecological assessments, supporting and guiding environmental management decisions and regulations. Measures taken to protect the environment based on exposure data can have social and economic implications. Flawed information may lead to measures being taken in the wrong place or to important action not being taken. Although the advantages of harmonizing evaluation methods have been demonstrated for hazard information, no comparable approach is established for exposure data evaluation. The goal of Criteria for Reporting and Evaluating Exposure Datasets (CREED) is to improve the transparency and consistency with which exposure data are evaluated regarding usability in environmental assessments. Here, we describe the synthesis of the CREED process, and propose methods and tools to summarize and interpret the outcomes of the data usability evaluation in support of decision-making and communication. The CREED outcome includes a summary that reports any key gaps or shortcomings in the reliability (data quality) and relevance (fitness for purpose) of the data being considered. The approach has been implemented in a workbook template (provided as Supporting Information), for assessors to readily follow the workflow and create a report card for any given dataset. The report card communicates the outcome of the CREED evaluation and summarizes important dataset attributes, providing a concise reference pertaining to the dataset usability for a specified purpose and documenting data limitations that may restrict data use or increase environmental assessment uncertainty. The application of CREED is demonstrated through three case studies, which also were used during beta testing of the methodology. As experience with the CREED approach application develops, further improvements may be identified and incorporated into the framework. Such development is to be encouraged in the interest of better science and decision-making, and to make environmental monitoring and assessment more cost-effective. Tetragnathid spiders have been used as sentinels to study the biotransport of contaminants between aquatic and terrestrial environments because a significant proportion of their diet consists of adult aquatic insects. A key knowledge gap in assessing tetragnathid spiders as sentinels is understanding the consistency of the year-to-year relationship between contaminant concentrations in spiders and sediment, water, and macroinvertebrates. We collected five years of data over a seven-year investigation at a PCB contaminated-sediment site to investigate if concentrations in spiders were consistently correlated with concentrations in sediment, water, and aquatic macroinvertebrates. Despite significant year-to-year variability in spider PCB concentrations, they were not correlated with sediment concentrations (p = 0.186). However, spider PCB concentrations were significantly, positively correlated with PCB concentrations in water (p < 0.0001, annual r2 = 0.35-0.84) and macroinvertebrates (p < 0.0001; annual r2 = 0.59-0.71). Analysis of covariance (ANCOVA) showed that spider PCB concentrations varied consistently with water (ß = 0.63) and macroinvertebrate PCB concentrations (ß = 1.023) among years. Overall, this study filled a critical knowledge gap in the utilization of tetragnathid spiders as sentinels of aquatic pollution by showing that despite year-to-year changes in PCB concentrations across environmental compartments, consistent relationships existed between spiders and water and aquatic macroinvertebrates.

Merolimnic insects can accumulate and transport considerable amounts of aquatic contaminants to terrestrial systems. The rate of contaminant biotransport, termed insect-mediated contaminant flux (IMCF), depends on emergent insect biomass and contaminant accumulation, both functions of environmental concentration. We developed a mathematical model of IMCF and apply it to three ecotoxicological studies obtained through the US Environmental Protection Agency's ECOTOX database to determine at which concentration maximum IMCF occurs. Model results demonstrate that the maximum IMCF depends on competing rates of biomass loss and contaminant accumulation and does not necessarily occur at the highest insect or environmental contaminant concentration. In addition, modeling results suggest that sublethal contaminant effects (e.g., decreased growth) on insect biomass can be an important and potentially underappreciated control on IMCF. Polychlorinated dibenzo-p-dioxin and polychlorinated dibenzofuran (PCDD/F) are persistent, toxic, and bioaccumulative. Currently, PCDD/F monitoring programs primarily use fish and birds with potentially large home ranges to monitor temporal trends over broad spatial scales; sentinel organisms that provide targeted sediment contaminant information across small geographic areas have yet to be developed. Riparian orb-weaving spiders, which typically have small home ranges and consume primarily adult aquatic insects, are potential PCDD/F sentinels. Recent studies have demonstrated that spider tissue concentrations indicate the source and magnitude of dioxin-like chlorinated compounds in contaminated sediments, including polychlorinated biphenyls (PCBs). Our aim in the present study was to assess the utility of riparian spiders as sentinels for PCDD/F-contaminated sediments. We measured PCDD/F (total [Σ] and homologs) in surface sediments and spiders collected from three sites within the St. Louis River basin (Minnesota and Wisconsin, USA). We then compared (1) patterns in ΣPCDD/F concentrations between sediment and spiders, (2) the distribution of homologs within sediments and spiders when pooled across sites, and (3) the relationship between sediment and spider concentrations of PCDD/F homologs across 13 stations sampled across the three sites. The ΣPCDD/F concentrations in sediment (mean ± standard error 286 591 ± 97 614 pg/g) were significantly higher than those in riparian spiders (2463 ± 977 pg/g, p < 0.001), but the relative abundance of homologs in sediment and spiders were not significantly different. Spider homolog concentrations were significantly and positively correlated with sediment concentrations across a gradient of sediment PCDD/F contamination (R2 = 0.47, p < 0.001). Our results indicate that, as has been shown for other legacy organic chemicals like PCBs, riparian spiders are suitable sentinels of PCDD/F in contaminated sediment. Aquatic ecosystems around the world are contaminated with a wide range of anthropogenic chemicals, including metals and organic pollutants, that originate from point and nonpoint sources. Many of these chemical contaminants have complex environmental cycles, are persistent and bioavailable, can be incorporated into aquatic food webs, and pose a threat to the health of wildlife and humans. Identifying appropriate sentinels that reflect bioavailability is critical to assessing and managing aquatic ecosystems impacted by contaminants. The objective of the present study is to review research on riparian spiders as sentinels of persistent and bioavailable chemical contaminants in aquatic ecosystems. Our review of the literature on riparian spiders as sentinels suggests that significant progress has been made during the last two decades of research. We identified 55 published studies conducted around the world in which riparian spiders (primarily of the families Tetragnathidae, Araneidae, Lycosidae, and Pisauridae) were used as sentinels of chemical contamination of lotic, lentic, and estuarine systems. For several contaminants, such as polychlorinated biphenyls (PCBs), Hg, and Se, it is now clear that riparian spiders are appropriate sentinels. However, many contaminants and factors that could impact chemical concentrations in riparian spiders have not been well characterized. Further study of riparian spiders and their potential role as sentinels is critical because it would allow for development of national-scale programs that utilize riparian spiders as sentinels to monitor chemical contaminants in aquatic ecosystems. A riparian spider sentinel program in the United States would be complementary to existing national sentinel programs, including those for fish and immature dragonflies. The ecological threshold of toxicological concern (ecoTTC) is analogous to traditional human health-based TTCs but with derivation and application to ecological species. An ecoTTC is computed from the probability distribution of predicted no effect concentrations (PNECs) derived from either chronic or extrapolated acute toxicity data for toxicologically or chemically similar groups of chemicals. There has been increasing interest in using ecoTTCs in screening level environmental risk assessments and a computational platform has been developed for derivation with aquatic species toxicity data (https://envirotoxdatabase.org/). Current research and development areas include assessing mode of action-based chemical groupings, conservatism in estimated PNECs and ecoTTCs compared to existing regulatory values, and the influence of taxa (e.g., algae, invertebrates, and fish) composition in the distribution of PNEC values. The ecoTTC continues to develop as a valuable alternative strategy within the toolbox of traditional and new approach methods for ecological chemical assessment. This brief review article describes the ecoTTC concept and potential applications in ecological risk assessment, provides an overview of the ecoTTC workflow and how the values can be derived, and highlights recent developments and ongoing research. Future applications of ecoTTC concept in different disciplines are discussed along with opportunities for its use.

There is growing concern over the health of North American birds, with evidence suggesting substantial population declines. Spiders are prominent dietary items for many bird species and mediate the transfer of contaminants to arachnivorous birds that consume them. Few studies have investigated the potential risk the spider exposure pathway poses to these birds because most studies have focused on piscivores. In the present study, we developed new chronic and acute As, Cd, Cu, Pb, Ni, Se, Zn, and MeHg spider-based avian wildlife values (SBAWVs) for multiple adult and nestling birds (primarily passerines) and then used the newly generated SBAWVs to characterize the risk to birds across 2 study areas: 1) 5 reaches in the southern Appalachian Mountains, an area with substantial mercury deposition but minimal anthropogenic impact, and 2) 4 reaches adjacent to the Emory River, an area impacted by the largest fly coal-ash spill in US history. We identified MeHg and Cu, Pb, Se, and Zn as contaminants of potential concern (COPC) at the Appalachian Mountain and Emory River study areas, respectively, based on dietary exposure of aquatic contaminants via riparian spiders. The identification of COPC at both study areas due to dietary spider exposure is notable not only because the spider exposure pathway has largely been uninvestigated at these sites but also because the aquatic systems in both areas have been studied extensively. Significant differences in MeHg concentrations were detected among spider taxa and suggest that the selection of spider taxa can impact risk characterization. These results indicate that the spider exposure pathway is important to consider when assessing potential risk, particularly for passerine birds. Millipedes are among the most important processors of leaf litter in temperate forests. Through consumption of leaf litter, millipedes may be exposed to mercury that accumulates in leaf tissues prior to senescence. To investigate mercury uptake in millipedes, Narceus spp. were collected from a remote site in the southern Appalachian Mountains, an area known to receive high mercury deposition. Additionally, aquatic primary consumers (larval caddisflies and stoneflies), brook trout (Salvelinus fontinalis) and rainbow trout (Oncorhynchus mykiss) were collected from the same site for comparisons of mercury concentrations and percent methylmercury. Bioaccumulation factors for millipedes were 18.5 and 20.2 for total and methylmercury, respectively. At this site, the mean THg concentration in millipedes was ~ 10 × greater than both brook trout and rainbow trout and ~ 200 × greater than that of aquatic primary consumers. Millipede THg concentrations ranged from 222 to 1620 ng/g ww in an area where EPA fish consumption criteria (300 ng/g MeHg in fish tissue, ww) were not exceeded. The mean percent methylmercury in millipedes was 1.4%, suggesting these animals were accumulating large quantities of inorganic mercury.

Tetragnathid spiders (Tetragnatha spp.) found in riparian habitats have recently been used as bioindicators of sediment contamination and insect-mediated contaminant flux. We investigated whether sexual dimorphism (size and behavior) influenced the female:male ratio in composite samples, stable isotope ratios (carbon [δ13C], nitrogen [δ15N]), and Hg concentrations in the southern United States. Additionally, we explored whether biomass for contaminant analysis could be preserved by using the legs of tetragnathids as a surrogate for whole-body δ13C and δ15N signatures. We found that female tetragnathids were significantly larger than male spiders and represented a larger proportion of spiders collected at all sites. However, despite the difference in size between sexes, no differences in growth dynamics, isotopic signatures (δ13C and δ15N), or mercury concentrations were observed. It was determined that the leg of a tetragnathid can accurately represent the stable isotope signature of an entire spider.

The susceptibility of Tennessee's Appalachian Mountains to anthropogenic stressors has remained largely uninvestigated likely due to a lack of known point source contamination. However, a growing body of scientific evidence suggests that depositional inputs can lead to concerning levels of contamination, even in remote areas. To investigate potential concerns, water quality parameters, contaminants in water (nitrogen, TSS, and metals), and contaminants in eastern brook trout (mercury, polychlorinated biphenyls [PCBs], organochlorine [OC] pesticides, dioxins, furans, and phthalates) were measured in four Appalachian Mountain streams from 2015 to 2017. Concentrations were compared to literature and/or model-derived (e.g., biotic ligand model) threshold values to determine whether levels exceeded those acceptable for stream health. Dioxins and furans were detectable in fish tissue at all sites with an average 2,3,7,8-tetrachlorodinbenzodioxin toxicity equivalence (TEQ) of 0.0015 ng/kg. Concentrations of PCBs, phthalates, and organochlorine pesticides were never above analytical quantitation limits, although several OC pesticides (e.g., alpha-chlordane) were detectable in fish. Aluminum concentrations in water were found at levels shown previously to cause mortality in brook trout during acidic rain events. The average whole-body methylmercury concentrations in fish among sites were 0.037 ± 0.003 µg/kg and were on average 75 ± 2% of total mercury.

Flexible, rapid, and predictive approaches that do not require the use of large numbers of vertebrate test animals are needed because the chemical universe remains largely untested for potential hazards. Development of robust new approach methodologies and nontesting approaches requires the use of existing information via curated, integrated data sets. The ecological threshold of toxicological concern (ecoTTC) represents one such new approach methodology that can predict a conservative de minimis toxicity value for chemicals with little or no information available. For the creation of an ecoTTC tool, a large, diverse environmental data set was developed from multiple sources, with harmonization, characterization, and information quality assessment steps to ensure that the information could be effectively organized and mined. The resulting EnviroTox database contains 91 217 aquatic toxicity records representing 1563 species and 4016 unique Chemical Abstracts Service numbers and is a robust, curated database containing high-quality aquatic toxicity studies that are traceable to the original information source. Chemical-specific information is also linked to each record and includes physico-chemical information, chemical descriptors, and mode of action classifications. Toxicity data are associated with the physico-chemical data, mode of action classifications, and curated taxonomic information for the organisms tested. The EnviroTox platform also includes 3 analysis tools: a predicted-no-effect concentration calculator, an ecoTTC distribution tool, and a chemical toxicity distribution tool. Although the EnviroTox database and tools were originally developed to support ecoTTC analysis and development, they have broader applicability to the field of ecological risk assessment. We investigated polychlorinated biphenyl (PCB) contamination at the Ashtabula River (northeast OH, USA) area of concern following remedial dredging using araneid and tetragnathid spiders. The PCB concentrations remain elevated in the area of concern compared with reference conditions. Patterns of contamination were strikingly similar between taxa, but were higher in tetragnathids at the most contaminated sites. Spider PCB homolog distributions identified 2 PCB sources to the area of concern. Based on these findings, we recommend situations where these taxa can be used alone, in concert, or combined into a composite "spider" sample to assess environmental contamination. Fecal indicator bacteria, such as Escherichia coli, are frequently monitored in recreational waterbodies as indicators of potential fecal pathogen presence and exposure. The present watershed analysis investigated the influence of season on E. coli concentration (MPN/100 mL) and loading (MPN/day) measurements for inland waters at baseflow conditions. The master dataset collected during 2007-2012 for three watersheds included 896 E. coli (Colilert) samples with simultaneous flow taken for a subset (39 %) of samples. The outcomes for grouped watersheds were reflected in most cases for individual watersheds. Concentration- and loading-based results were highest in summer and spring, and lowest in the winter and fall, respectively. The comparison of these two measurement techniques (concentration and loading) highlight the impact of flow data during baseflow conditions for inland waters and reveal that caution should be used when inferring one method's results from another.

Biomagnification of methylmercury in aquatic systems can cause elevated tissue mercury (Hg) and physiological stress in top predators. Mercury is known to affect stress hormone levels in mammals, birds and fish. In this study, the effects of maternally-transferred methylmercury on the stress physiology of Northern Water Snake (Nerodia sipedon) neonates were tested. Gravid females were dosed via force-fed capsules during late gestation with 0, 0.01, or 10 µg methylmercury per gram of body mass. Plasma corticosterone levels and leukocyte differentials were analyzed in baseline and confinement-stressed neonates from all dose levels. Neither Hg nor confinement stress had a significant effect on leukocyte differentials nor was Hg related to corticosterone levels. However, stress group neonates showed lower heterophil/lymphocyte ratios and this study was the first to show that neonate N. sipedon can upregulate CORT in response to stress. These results indicate that N. sipedon may be somewhat tolerant to Hg contamination.

Hypothesis-based no-effect-concentration (NOEC) and regression-based x% effect concentration (ECx) values are common statistical approaches used to summarize ecotoxicological effects. Controversy over the NOEC model has prompted a movement toward discontinuation of the NOEC in favor of ECx, but the best x% effect surrogate for NOEC has not yet been determined. Historically, 10% and 20% effect concentrations (EC10 and EC20) have been treated as NOEC analogs. Given these measurements' importance to ecotoxicology, further understanding of the relationships between NOEC and EC10 or EC20 is crucial. In the present study, a metadataset of daphnid chronic toxicity tests was compiled to analyze the strength and significance of NOEC:EC10 and NOEC:EC20 relationships. The impact of endpoint (e.g., mortality, reproduction) and test condition parameters (e.g., pH, temperature) on NOEC:EC10 and NOEC:EC20 was evaluated. Mortality endpoints were most sensitive 51% of the time, with growth and reproductive endpoints constituting the remainder, underscoring the value of using multiple endpoints to evaluate toxic effects rather than relying on reproduction as the a priori most sensitive endpoint. When test condition parameters were less restricted (e.g., pH, hardness), the NOEC:EC20 association was more robust, suggesting that variability introduced by test implementation increased variability in ECx calculation. The analysis revealed that, overall, EC10 was a more suitable analog than EC20 for NOEC. Recommendations include refinement and reporting of the test parameters pH and hardness to minimize variability in ECx calculation.

On December 22, 2008, a dike containing coal fly ash at the Tennessee Valley Authority Kingston Fossil Plant (TN, USA) failed, and within months, dredging operations began to remove ash-contaminated sediments. The purpose of this study was to investigate differences in the bioaccumulation of metals in three mussel species during and after dredging operations. Mussels were caged for approximately 1 year during dredging and after, and then mussel condition index values and As, Cd, Cr, Pb, Ni, Se, Hg, U, Fe, Mg, Al, Sb, Ba, Be, Co, Cu, Mn, Mo, Ag, Sr, Tl, V, and Zn concentrations in soft tissue were determined via inductively coupled plasma-mass spectrometery. Overall, the differences observed in metal bioaccumulation and mussel health suggest that mussels in the immediate downstream area of the dredging site may have been impacted, as evidenced by a significant decrease in mussel condition index values, but that this impact did not result in increased tissue concentrations of metals.

RESUMO

Tools exist to evaluate large ecotoxicity databases for risk assessment purposes, but these tools are less useful for alternative analytical purposes. In the present study, the authors developed the Stepwise Information-Filtering Tool (SIFT), a strategic method to select relevant, reliable data from a large ecotoxicity database; demonstrated utility in a case study of chronic toxicity data for statistical endpoint comparison purposes; and evaluated SIFT by comparison with 2 existing data evaluation methods.

RESUMO

Monitoring expression of the developmentally regulated genes shh, sox2, and tnnt1 by reverse transcriptase polymerase chain reaction (RT-PCR) allows determination of the rate of embryogenesis in zebrafish (Danio rerio) embryos without direct visual observation. The utility of combining this approach and morphological methods during toxicity studies was demonstrated with embryos developing at either 28.5 °C or 24.5 °C and with embryos exposed to sublethal doses of silver nanoparticles.
